Objective To determine the contents of hypaconitine and mesaconitine in Shubining Granules. Methods The HPLC method was established for the determin ation of hypaconitine and mesaconitine in Shubining Granules. The HPLC was perfo rmed on Akasil C18(4.6 mm?250 mm,5 ?m) column,mobile phase consisted of meth anol-0.1 %triethylamine (70 ∶30),column temperature 30 ℃,and the flow rare was 1.0 mL/min.The detection wavelength was 230 nm.Results A good linearity w as obtained in the range of 0.126～2.520 ?g of Hypaconitine and 0.152～3.04 ?g of Mesaconitine.The average recovery of Hypaconitine was 101.12 %.The average recovery of Mesaconitine was 99.68 %. Conclusion This method is simple and qui ck,and can be used as quality control of Shubining Granules.

Objective To develop a quantitative method to determine vitexin in Cajanus cajan (L.) Millsp. Methods The chromatographic conditions were as follows: column C18(250 mm? 4.6 mm, 5 ? m) , mobile phase being MeOH ∶ 1 % HAC (25 ∶ 75) for 20 mins and then being MeOH ∶ 1 % HAC (30 ∶ 70) after 20 mins, flow rate at 1.0 mL/min, and wavelength at 339 nm. Results The good linearity of this method was in the range of 0.104 4～ 0.522 ? g (r=0.999 4), and the average recovery of vitexin was 100.11 % ( RSD =0.63 % ).Conclusion The method is simple and sensitive with good stability, it can be suitable for the quality control of vitexin in Cajanus cajan (L.) Millsp..

Objective Brain natriuretic peptide(BNP) is released from the cardiac ventricles in response to increased wall tension. The prognostic significance of blood brain natriuretic peptide in Chinese patients with cardiovascular disease has not been established. The purpose of this study was to investigate the value of brain natriuretic peptide for predictin g cardiac death within 1 month in Chinese patients with cardiovascular disease. Methods One hundred and seven inpatients with cardiovascular disease, whose blood brain natriuretic peptide concentration were measured within 1 - 3 days of admission, using triage BNP test, were divided into 2 groups: the survival and the non-survival, according to the results of 1was positively correlated with heart rate, left ventricular end-diastolic dimension, history of heart failure and old myocardial infarction (r=0.28, P=0. 000 4; r=0.49, P＜0. 000 5; r=0.39, P＜0. 000 5; r=Area under the curve of the receiver-operating-characteristic(ROC) of brain natriuretic peptide to predict cardiac death at 1 month in patients with cardiovascular disease was 0.89%, 95% confidence interval 0.79-0. 98, P＜0. 000 5; stepwise logistic regression analysis indicated that brain natriuretic peptide (≥755pg/mL) was the only independent predictor of cardiac death at 1 month in patients with cardiovascular disease (OR= 17.6, 95 % confidence interval, 8.7- 66.5, P＜0. 000 5 ). Objective To explore the effects of walking exercise on glycometabolism,dynamic blood pressure and the quality of life of patients with both hypertension and type 2 diabetes on the basis of conventional drug treatment.Methods Sixty-two patients with both hypertension and diabetes who could support taking walking exercise of more than 5,000 steps/d were randomly divided into a walking exercise group (32 cases) and a control group (30 cases).Both groups were given conventional drug treatment (including valsartan,acarbose and metformin).Those in the walking exercise group took more than 10,000 steps/d of aerobic exercise while the patients in the control group were just given normal community care.This continued for a period of 3 months.Fasting plasma glucose (FPG),glycated hemoglobin-A1C (HbA1c),fasting insulin (FINS),the homeostasis model of assessment for insulin resistence index (HOMA-IR),the homeostasis model of assessment for insulin sensitivity (HOMA-IS),dynamic blood pressure parameters and quality of life were observed.Results In the walking exercise group,the FPG,HbA1c,FINS,HOMA-IR,HOMA-IS,dynamic blood pressure and quality of life indicators were all significantly different after 3 months of daily walking exercise compared with either baseline or the control group.Conclusion Accompanied by conventional drug therapy,10,000 steps/d of walking exercise can improve the glucose metabolism,dynamic blood pressure and quality of life of patients suffering from mild hypertension and type 2 diabetes.

OBJECTIVE:To systematically evaluate the clinical efficacy and safety of Compound Huangbo liquid combined with chemical medicine in the treatment of ulcerative colitis (UC),and to provide evidence-based reference.METHODS:Re trieved from CBM,Wanfang Database,VIP,CJFD,PubMed,EMBase and Cochrane Library,randomized controlled trials(RCTs) about Compound Huangbo liquid combined with chemical medicine (trial group) vs.chemical medicine alone (control group) in the treatment of UC were collected.Meta-analysis was performed by using Rev Man 5.3 statistical sofetware after literature scanning,data extraction and quality evaluation according to Cochrane reviewer's handbook 5.1.0.RESULTS:A total of 8 RCTs were included,involving 737 patients.The results of Meta-analysis showed that,response rate of trial group was significantly higher than that of control group,with statistical significance [OR=4.69,95 ％ CI(3.00,7.34),P＜0.001];there was no statistical significance in the incidence of ADR between 2 groups [OR=0.94,95 ％ CI (0.48,1.84),P=0.86].CONCLUSIONS:Compound Huangbo liquid combined with chemical medicine is better than chemical medicine alone for UC,and both have similar safety.

OBJECTIVE: To systematically evaluate therapeutic efficacy and safety of Compound kushen colon-release capsule versus related chemical drugs in treatment of ulcerative colitis (UC).METHODS: Retrieved from PubMed, Embase, Cochrane library, China Journal Full-text Database (CJFD), VIP, CBM and Wanfang database, RCTs about therapeutic efficacy (response rate, TCM syndrome, therapeutic efficacy of mucosal lesion) and safety (the incidence of ADR) of Compound kushen enteric-release capsule versus related chemical drugs in the treatment of UC were collected. Meta-analysis was conducted by using Rev Man 5. 3 statistical software after data extraction and quality evaluation with Cochrane 5. 1. 0. RESULTS: A total of 9 RCTs were included, involving 649 patients. The results of Meta-analysis showed that response rate [OR=2. 16, 95％CI(1. 28, 3. 63), P=0. 004] and therapeutic efficacy of mucosal lesion [OR=2. 42, 95％ CI(1. 07, 5. 47), P=0. 03] of Compound kushen colon-release capsule were significantly higher than that of chemical drugs in the treatment of UC, with statistical significance. There was no statistical significance in therapeutic efficacy of TCM syndrome [OR=1. 70, 95％CI(0. 87, 3. 32), P=0. 12] or the incidence of ADR between 2 groups. CONCLUSIONS: Therapeutic efficacy of Compound kushen colon-release capsule is better than that of related chemical drugs in the treatment of UC with similar safety.

Objective: To investigate the effect of astragalus polysaccharides (APS) on proliferation, invasion, apoptosis and drugresistance of cisplatin-resistant colorectal cancer (CRC) HT-29/DDP cells through regulating miR-20a/TGFBR2 axis, and to explore the possible mechanism. Methods: Human CRC HT-29 cells and HT-29/DDP cells were used as non-drug resistant and resistant cell models, respectively; HT-29/DDP cells were randomly divided into four groups, including untreated (HT-29/DDP) group, APS treatment group, miR-20a mimics + APS group, and si-TGFBR2 + APS group. qPCR and Western blotting were applied to detect the expressions of miR-20a and TGFBR2 in HT-29/DDP cells treated with different concentrations ofAPS (0, 0.5, 1.0, 1.5 and 2.0 mg/ml). Subsequently, dual luciferase reporter gene assay was used to verify whether TGFBR2 was a target gene of miR-20a. In addition, CCK-8, Transwell andAnnexin V-FITC/PI double staining were applied to examine the effect ofAPS on proliferation, invasion and apoptosis of HT29/DDP cells. Furthermore, subcutaneous HT-29/DDP cell xenograft model was established on nude mice, and the effect ofAPS on the growth of transplanted tumor was observed. Results: APS significantly inhibited the proliferation of HT-29/DDP cells in a dose-dependent manner (P<0.01). Meanwhile, the expression of miR-20a was down-regulated in HT-29/DDP cells treated with APS, while the expression of TGFBR2 was significantly up-regulated (all P<0.01). Additionally, dual luciferase reporter gene assay result showed that TGFBR2 was a direct target of miR-20a in HT-29/DDP cells and its expression was suppressed. Furthermore, APS could enhance the drug sensitivity of HT-29/DDP cells through downregulating the inhibitory effect of miR-20a on TGFBR2 expression, thereby suppressed proliferation and invasion, and induced apoptosis of HT-29/DDP cells in vitro and in vivo. It was also found that this effect was related with the suppression of PCNA and Bcl-2 proteins and promotion of Bax and Caspase-3 proteins. Conclusion: APS reverses the resistance of HT-29/DDPcells to cisplatin by down-regulating the inhibitory effect of miR-20a on TGFBR2 expression.

Objective A HPLC-quantitative analysis of multi-components by single marker(QAMS)was established to determine 5 ingredients including uracil,uridine,hypoxanthine,inosine and guanosine in Periplaneta americana.Methods Separation took place on a Agilent ZORBAX SB-Aq column(250 mm×4.6 mm,5 μm)by gradient elution of methanol-0.01 mol·L-1 potassium dihydrogen phosphate at 20℃with a flow rate of 1.0 mL·min-1.The detection wavelength was 260 nm and the injection amount was 10 μL.The relative correction factors(fa/b)was calculated for the other four components with uridine as an internal standard.The content of 5 ingredients in 10 batches of Periplaneta americana was determined by QAMS.Results were compared with those of external standard method(ESM).Results Five nucleosides showed good linear relationships in their own ranges(r＞0.999 5),and the average recoveries ranged from 97.0%to 100.8%.The relative correction factors of uracil,hypoxanthine,inosine and guanosine were 0.908 0,1.005 3,1.969 5 and 1.303 4,respectively.Conclusion The established method is accurate and stable.It can provide theoretical reference for the quality control of Periplaneta americana.
