Objective To research the spatiotemporal pattern of N-cadherin expression during development of rat cardiomyocytes and age-related changes of N-cadherin expression. Methods Using immunohistochemical method,myocardial N-cadherins distribution was investigated in fetal rat and postnatal development(1 postnatal day to old rat),and quantitied by HIPAS-1000 computerized image analytical instrument. Results The expression of N-cadherin was located in myocardium of atrial and ventricule and septum interventriculare and papillary muscles.The N-cadherin immunolabeling was found in myocyte membranes and within cytoplasm in fetal rat heart.From neonatal to infant rat,the pattern of N-cadherin immunolabeling changed progressively,from being dispersed over the entire cell surface as in the fetal to the transverse terminals of the myocytes,toward the distribution within the intercalated disk.From young to old rat heart,the typical N-cadherin was located in transversely orientated intercalated disk.The percentage of N-cadherin immune postive area in rat ventricular myocardium showed a progressively changement with age.Conclusion The present paper demonstrated that the N-cadherin expression exists progressively with ages and the changing pattern of N-cadherin is closely related with development of the intercalated disk in rat myocardium.The mechanical coupling provided by adherens junctions is essential for the stable cell-cell contact.

And then, cultured bottle was maintained in water bath at 37℃ and performed with microwave radiation in dark chamber, whose reflectivity was about zero. It was regarded as radiation group, and the average power density of radiation was 30 mW/cm2; in addition, the duration of radiation was 30 minutes.Cells did not deal with microwave radiation were regarded as control group. Phosphorylation level of NF-E2-related factor-2 was measured at 2, 4, 8 and 24 hours after radiation with immune coprecipitation-autoradiography technique and dealt with semi-quantitative analysis with gel scanning system. Cells in the control group were analyzed directly. ②Active analysis and expressional measurement of protein kinase C: Cells in the radiation group and the control group were dealt with the same cultured method, condition, radiation styles, dosage and environment as mentioned above. At 2, 4, 8 and 24 hours after radiation, cells were split to extract plasma and membrane protein. Furthermore, activity of protein kinase C was measured with r-32P-ATP labeled liquid scintillation spectrometer; gray value of protein strap was dealt with semi-quantitative analysis with gel scanning system; staining degree of plasma was observed after immunocytochemical staining of protein kinase C. In addition, cells in the control group were measured and observed directly.MAIN OUTCOME MEASURES: ① Phosphorylation level of NF-E2-related factor-2 in radiation group and control group; ②Results of active analysis and expressional measurement of protein kinase C in radiation group and control group.RESULTS: ① Phosphorylation level of NF-E2-related factor-2 in radiation group and control group: Gray value of NF-E2-related factor-2 was higher in radiation group than that in control group at 2, 4 and 8 hours after radiation.Phosphorylation level of NF-E2-related factor-2 reached the peak at four hours after radiation. In addition, results of semi-quantitative scanning analysis showed that, at 2, 4 and 8 hours after radiation, phosphorylation level of NF-E2-related factor-2 was increased 33%, 261% and 141% in radiation group as compared with that in control group,respectively (t = 2.974, 4.209, 4.047, P ＜ 0.05), and then, fallen down to normal value 24 hours later. ② Results of active analysis and expressional measurement of protein kinase C in radiation group and control group: At 2, 4 and 8 hours after microwave radiation, expression of protein kinase C in radiation group was higher than that in control group,especially at the 4 hour. In addition, at 24 hours after radiation, expression of protein kinase C recovered the normal value. Results of immunocytochemical staining showed that staining of plasma was deeper in radiation group than that in control group at 4 hours after radiation. Moreover, results of r-32P-ATP labeled liquid scintillation spectrometer also suggested that, at 2, 4 and 8 hours after radiation, activity of protein kinase C was increased 36%, 93% and 47% in radiation group as compared with that in control group, respectively (t =2.801, 3.654, 3.035, P ＜ 0.05). And then, activity of protein kinase C was decreased after 24 hours, otherwise, activity of protein kinase C reached the peak at 4 hours after radiation.CONCLUSION: Microwave radiation can strengthen the phosphorylation level of NF-E2-related factor-2 in vascular endothelial cells during a special period; meanwhile, it can also cause the increase of expression of protein kinase C.Time effect of activity of protein kinase C is coincidence with phosphorylation level of NF-E2-related factor-2.

Objective To probe into the feasibility of establishing a uniform reimbursement plan for hospitalization expenses under NRCMS in Nanjing city. Methods Hospitalization expenses in 2005 for peasants under NRCMS from the sour suburb districts and county in Nanjing were sampled. Based on this study, a reimbursement ratio was designed for such peasants as classified by "grades, sections and accumulated reimbursement". Results A uniform NVRMS reimbursement plan for hospitalization expenses may not be practical for all peasants in Nanjing. Yet such a plan of relative uniformity is worthy of experimenting as follows: no minimum for reimbursement;within the same expense range, reimbursement ratio may be measured and fixed accordingly among districts and county in the city, ranging 20%-70%;a uniform ceiling and reimbursement range are recommended. Conclusions A uniform city-wide plan requires by-step and planned implementations. With support from government finance, fund-raising gaps among these areas can be narrowed gradually, cutting back differences in their reimbursement plans in the end.

OBJECTIVE To study the possibility of applying Anerdian skin disinfectant for blood donor skin disinfection. METHODS Put Anerdian at 21℃,35℃ and 42℃ to exposure in air for 24h,48h and 72 h,detect the volatilization volume,effective iodine and appearance of disinfectant,and evaluate its effect on blood donor elbow by culture method. RESULTS At 21℃ exposure for 24 h the volatilization rate for Anerdian was 5%,effective iodine was 0.215%.At 35℃ and 42℃ exposure for 24 h its volatilization rate was more than 5%,and effective iodine was within the range of 0.18-0.22%.The average killing logarithm value of elbow skin natural bacteria in 30 donors was over 1. CONCLUSIONS Anerdian disinfectant is suitable for blood donor predonation skin treatment.

Objective To screen a gastric carcinoma subcell line with higher metastatic potential and to explore the relationship between the liver metastatic potential and liver microenvironment with the screened subcell line Methods A gastric carcinoma subcell line was screened via procedures of orthotopic transplantation of subcutaneous tumor of its parent cell line SGC 7901 in nude mice and primary tissue culture of its liver metastatic focus Expressions of bFGFR, TGF?R 1 and c Met(hepatocyte growth factor receptor) in both the subcell line and its parent cell line were determined by immunohistochemistry Results A gastric carcinoma cell subcell line(named SGC 7901LM 2) was established from its parent cell line SGC 7901 with higher metastatic potential, and the expression of c Met increased significantly in the screened subcell line Conclusion The screened gastric carcinoma subcell line with higher metastatic potential is helpful for the further studies of liver metastasis of gastric carcinoma The elevated expression of c Met may be helpful to the liver metastatic potential of SGC 7901LM 2

Objective To investigate the differences of connexin 43(Cx43)expression between adult and infant heart. Methods By using immunohistochemistry to observe the expression of Cx43. Results 1.The expression of Cx43 had a punclate distribution in cytoplasm and over the entire surface of the cardiocyte,and a few located at intercalated disk of atrial and ventricular myocardium in the infant heart.2.Cx43 positive granules distributed irregularly in cell side surface and cytoplasm as well as intercalated disk in adult atrium.Cx43 immunolabelling of adult ventricular myocardium was typically confined to the site of intercalated disk.3.The results of image analyzer showed that the amount of connexin43 expression was lower in the atrium than that of the ventricle in infant heart and atrium bigger than ventricle in adult heart.The expression of Cx43 was less in adult heart than that of infant heart.Conclusion The expression of Cx43 was mainly over the entire surface of the myocardium in infant heart.There were expression differences of Cx43 in human ventricular and atrial myocytes.The amount of Cx43 expression was higher in ventricle than that of atrium in infant heart and atrium bigger than ventricle in adult heart.It was less in adult heart than that of infant.It showed that the expression of Cx43 in human heart existed a developmental differences.

Objective:To analyze the clinical characteristics and diagnosis and treatment of children with Kasabach-Merritt phenomenon (KMP).Methods:A retrospective analysis was conducted on the clinical data and follow-up data of 8 patients diagnosed KMP in Beijing Children′s Hospital, Capital Medical University from January 2016 to January 2019.The clinical data included laboratory examination, diagnosis, treatment and prognosis.Results:Among the 8 children with KMP, 6 cases were male and 2 cases were female.The median onset age was 4 (0-17) months, 2 cases of neonatal onset.The median onset to the diagnosis time was 59 (34-140) days; 6 cases with bone destruction; 6 cases had misdiagnosis and mistreatment history, they were misdiagnosed as idiopathic thrombocytopenic purpura, Evans syndrome, abnormal bone and joint development; 4 cases were Kaposiform hemangioendothelioma; 8 cases were used alone or combined with the application of hormones, Sirolimus, and Vincristine, 7 patients underwent interventional therapy.All patients survived with a median follow-up period of 487 (112-1 033) days.Median time of platelet count returned to normal was 24.5 (7-60) days, and median time of fibrinogen returned to normal was 20 (7-30) days.Median time of D-dimer dropped to a normal was 105 (40-240) days.Conclusions:Children with concurrent platelet count and coagulation abnormalities should be considered with KMP.Doctors need to identify the potential visceral vascular lesions.Early diagnosis and treatment are important, which can improve the clinical prognosis of patients.
