1.Determination of telomerase activity in carcinoma of bile duct and its clinical significance

Objective To explore the relationship between te lomerase activity and bile duct carcinoma and its significance in clinical diag nosis. Methods Telomerase activities were examined in 23 cases of bile duct carcinoma, 5 cases of carcinoma adjacent tissues and 5 cases of nor mal bile duct tissues respectively with telomerase PCR-ELISA method. Results Telomerase activity was detected in 18 of 23 cases (78.3%) of b ile duct carcinoma, and was not found in 5 cases of carcinoma adjacent tissues a nd 5 cases of normal bile duct tissues. The detection rate of telomerase activit y had no correlation with patients' age, sex, tumor site and size but significan tly correlated to metastasis of tumor (P<0.05). Conclusion The level of telomerase activity was significantly higher in bile duct carcino ma and may be served as one of the clinical marker for malignant neoplasm becaus e of its spsecificity.

Objective To study the effect of somatostatin analogue (SMS201-995,SMS) on apoptosis of human cholangiocarcinoma SK-ChA-1 cells and apoptosis-regulated gene bcl-2 and bax. Methods The changes of apoptosis and bcl-2/bax gene expression were detected by DNA gel electrophoresis, flowcytometric analysis and immunohistochemistry, in situ hybridization methods in cholangiocarcinoma SK-ChA-1 cells after treated with SMS(100?ng/ml) for 6, 12 and 24 hours.Results The characteristic ladder was not detected by agarose gel electrophoresis after treated with SMS(100?ng/ml) for 6 hours. When the cells were treated for 12 and 24 hours, the ladder of DNA electrophoresis was observed. While the cells were treated with SMS for 6, 12 and 24 hours, the percentage of annexin V-positive cells were (22?5)% ,(39?7)% and(58?10)%. Besides,SMS inhibited bcl-2 protein expression and increased bax protein and mRNA expression. Conclusion SMS can induce SK-ChA-1 cell apoptosis mainly with regulation of bcl-2 and bax gene expression.

Xanthone and its derivatives occupy a large part of the family of natural polyphenolic compounds with various biological and pharmacological activities.In recent years (from 2006 to 2011),it was reported that 127 xanthones were discovered from plants and fungi using various modem separation methods including silica gel/polyamide column chromatography,HPLC,high-speed counter-current chromatography,high-performance centrifugal partition chromatography,etc.Since total synthesis and structure modification for xanthone and its derivatives have been given attention worldwide,we introduced the synthetic methods of xanthone skeletons as well.Unfortunately,to date,there are still weaknesses in current methods of separation and synthesis,which need to be improved.This review,to a certain extent,provides necessary foundation for the further research and development of medicines containing xanthone and its derivatives.
6.Effects of somatostatin analogues on the cell cycle and the related gene expression in human cholangiocarcinoma cell line SK-ChA-1

Objective To investigate the effects of somatostatin analogue (SMS201 995, SMS) on the cell cycle of human cholangiocarcinoma SK ChA 1 cells and cell cycle regulated gene cyclinD1 and p16. Methods SK ChA 1 cells were synchronized by serum starvation. Cells at different phases were harvested at 48 h after SMS treatment of the synchronized SK ChA 1 cells. The distribution of cell cycles was analyzed by flow cytometry. The expression levels of cyclinD1 and p16 gene were detected by SABC immunohistochemistry and in situ hybidization. Objective:To analyze the clinical characteristics and diagnosis and treatment of children with Kasabach-Merritt phenomenon (KMP).Methods:A retrospective analysis was conducted on the clinical data and follow-up data of 8 patients diagnosed KMP in Beijing Children′s Hospital, Capital Medical University from January 2016 to January 2019.The clinical data included laboratory examination, diagnosis, treatment and prognosis.Results:Among the 8 children with KMP, 6 cases were male and 2 cases were female.The median onset age was 4 (0-17) months, 2 cases of neonatal onset.The median onset to the diagnosis time was 59 (34-140) days; 6 cases with bone destruction; 6 cases had misdiagnosis and mistreatment history, they were misdiagnosed as idiopathic thrombocytopenic purpura, Evans syndrome, abnormal bone and joint development; 4 cases were Kaposiform hemangioendothelioma; 8 cases were used alone or combined with the application of hormones, Sirolimus, and Vincristine, 7 patients underwent interventional therapy.All patients survived with a median follow-up period of 487 (112-1 033) days.Median time of platelet count returned to normal was 24.5 (7-60) days, and median time of fibrinogen returned to normal was 20 (7-30) days.Median time of D-dimer dropped to a normal was 105 (40-240) days.Conclusions:Children with concurrent platelet count and coagulation abnormalities should be considered with KMP.Doctors need to identify the potential visceral vascular lesions.Early diagnosis and treatment are important, which can improve the clinical prognosis of patients.

AIM To study the flavonoids from the leaves of Astragalus membranaceus (Fish.) Bge..METHODS The ethyl acetate and n-butyl alcohol fractions of 75% ethanol extract from A.membranaceus leaves were isolated and purified by silica,ODS and preparative HPLC column,then the structures of obtained compounds were identified by spectral data.RESULTS Thirteen compounds were isolated and identified as quercetin (1),quercetin-3-O-β-D-glucopyranoside (2),rhamnocitrin-3-O-β-D-glucopyranoside (3),rhamnocitrin-3-O-β-neohesperidoside (4),rhamnocitrin-3-O-3-D-glucopyranoside (1'''→2'')-β-D-apiofuranosyl (5),complanatuside (6),glycitein (7),4',7-dihydroxy-3-methoxy isoflavone (8),genistein (9),calycosin-7-O-β-D-glucopyranoside (10),genistin (11),glycitin (12),tiliroside (13).CONCLUSION Compounds 5,8,13 are isolated from genus Astragalus for the first time,and compound 2 is first isolated from this plant.
9.Changes in corneal morphology after seawater soaking following explosive injury in rabbits

Obiective: To observe the changes in corneal tissue after seawater soaking following explosive injury in rabbits and the effects of seawater soaking on the healing of cornea after explosive injury. Methods:Make 20 similar explosive injury models of rabbit's eyeball using 10 adult grey rabbits. For each rabbit, the right eye was in experiment and the left one was for comparison. After the injury, the experimental eyes were soaked in seawater for 30 minutes and the comparable eyes were soaked in 0.9% sodium chloride injection for the same time. All eyes were underwent slit lamp examinations, fluorescein dyeing, and ultrasonic cornea thickness metering on the corneas before the injury and 1,2,3,4,5,7,9,10,15 and 20 days after the injury respectively. At the same time, 2 rabbits were killed and the eyes were examined pathologically. The cornea thickness numbers were compared using self- comparison t test and grouping t test. Results: After seawater soaking following explosive injury, slit lamp microscope examination found that the corneas were edema and turbidity, the experimental eyes were stricter than the control eyes. The corneas of the experimental eye were vascularized 10 days after the injury. Fluorescein dyeing showed that more apparent pigmentations were found on the experimental eyes than that on the control ones.Comparative examination under the optical microscope showed that the corneal tissues of the experiment eyes changed distinctly, their inflammations were patency and healing was delayed and the vascularization degree became high after the injury. Cornea thickness of the experimental eyes became apparently thicker than that of the control eyes. The difference was statistically significant. Conclusion:Scars and vascularization of various degrees will appear in corneal stroma after the explosive injuries. Seawater soaking will have bad effects on the healing of the cornea injured in explosions.

Objective The purpose of this study was to provide practical anatomic data for the imaging diagnosis and surgical treatment of the disease of the subphrenic spaces. Methods Sectional anatomy of the subphrenic spaces on the coronal plane were investigated on 30 sets of serial coronal sections of superior abdomen of Chinese adult cadavers and MR imaging of 2 abdomen of cadavers. Results Subphrenic spaces were divided into perihepatic spaces and perisplenic spaces on the coronal plane.The space between the anterior margin of left gastropancreatic fold and the posterior layer of hepatogastric ligament is the only direct communication between the superior and inferior recesses of the lesser sac.Its communicating type is divided into three types on the coronal plane.The right layer of the gastrophrenic ligament is continuous with the posterior layer of the lesser omentum,the left layer of the gastrophrenic ligament is continuous with the right layer of the phrenicosplenic ligament and the posterior layer of the gastrosplenic ligament.Left gastropancreatic fold is continued to the left and right layers of gastrophrenic ligament upward.The bare area of stomach is located between the left and right layers of gastrophrenic ligament,its existing rate is 100%.The bare area of spleen is located among phrenicosplenic ligament,gastrosplenic ligament,splenorenal ligament and splenocoloic ligament.Its the most width exists at the splenorenal ligament.It is divided into the part of the splenic hilus and the splenorenal part.Its maximum is 2 64?1 16 cm,4 16?2 24 cm respectively. Conclusion The coronal plane is dominant for showing left gastropancreatic fold and the communication relationship between superior and inferior recesses of the lesser sac.The superior and inferior recesses of the lesser sac that may communicate with each other is 73 3 percent of all the cases studied
