1.Enlightenment of the holism in traditional Chinese medicine to research of onset and development of essential hypertension, diabetes and hyperlipemia

OBJECTIVE: According to the holism in traditional Chinese medicine (TCM) theory, this manuscript explores the relativity of pathogenesis among hypertension, diabetes and hyperlipemia.DATA SOURCES: We applied computer to search articles in English on pathogenesis ofhypertension, diabetes and hyperlipemia in Medline between January 1985 and December 2004 by words "hypertension, diabetes, hyperlipemia, pathogenesis" .Besides, we also used computer to search articles in Chinese on pathogenesis of hypertension, diabetes and hyperlipemia in Chinese magazine full text database between January 1990 and December 2004 by words "hypertension, diabetes, hyperlipemia, pathogenesis".STUDY SELECTION: Primarily, articles on pathogenesis of hypertension,diabetes and hyperlipemia were selected in TCM and modern medicine respectively. Accepted standard were: ① Random control clinic test with methods of single blind, double blind or no blind; ② Nonrandom control test;③ Fore-and-aft control test; ④ Chapter in monograph. Eliminated standard:repeated research.DATA EXTRACTION: Totally 28 articles on pathogenesis of hypertension,diabetes and hyperlipemia were found in TCM and modern medicine, however, 12 articles were selected according to accepted standard, and 16 articles were eliminated due to repeated content.DATA SYSTHESIS: Totally 12 selected articles were analyzed on pathogeneses of hypertension, diabetes and hyperlipemia in TCM and modern medicine, and hinted internal affiliation of these three diseases from view of molecular biology.CONCLUSION: These three diseases can be regarded as a whole entity for study. High blood pressure, high blood sugar and high lipemia could be treated as different forms of the entity or in different developing courses of the whole. In a certain course it might present as one of the forms specifically. It might manifest high blood pressure mainly in clinic, or high blood sugar, or high lipemia,also several forms at the same time. It will be beneficial to take the holism and visceral theory in TCM for reference in research of modern medicine.
2.Prenatal stress promotes formation of chronic stress-induced hippocampal amyloid β protein in offspring mice

Objective To explore whether prenatal stress promotes formation of chronic stress-induced hippocampal amyloid β (Aβ) protein in 6-month-old male offspring mice and its mechanism.Methods The APPswe/PSIdE9 double transgenic mice were divided into 4 groups according to the prenatal stress and offspring mice stress:prenatal control-offspring control group (CC group),prenatal control-chronic offspring stress group (CT group),chronic prenatal stress-offspring control group (TC group),and chronic prenatal stress-chronic offspring stress group (TT group) (n=18).The number of amyloid plaques in brains was checked using Congo red staining.ELISA was used to examine the hippocampus levels ofamyloid-β proteins (Aβ1-42 and Aβ1-40) in the offspring mice;β-site APP-cleaving enzyme 1 (BACE1) activity was detected using fluorospectrophotometry.Additionally,Western blotting were used to observe the expression levels of phosphorylated eukaryotic initiation factor 2α (p-eIF2α),phosphorylated protein kinase R [PKR]-like ER kinase (p-PERK),glucose-regulated protein 78 (Grp78) and β-site BACE1 in the hippocampus.Results As compared with that in the CC group,the number of amyloid plaques in brain in CT,TT and TC groups was increased.The expressions of p-eIF2α,p-PERK,Grp78,BACE1,Aβ1-40 and Aβ1-42 in the hippocampus of CT group were significantly increased as compared with those in the CC group (P<0.05).The expressions of p-eIF2α,p-PERK,Grp78,BACE1,Aβ1-40 and Aβ1-42 in the hippocampus of TT group were further significantly increased as compared with those in the CT group (P<0.05).There was no significant difference in BACE1 activity among the different groups (P>0.05).Conclusion The prenatal stress can promote the formation of hippocampal Aβ protein induced by chronic stress in 6-month-old male offspring mice,whose mechanism may be that prenatal stress aggravates hippocampal neurons endoplasmic reticulum stress,activates the PERK,then causes eIF2 alpha phosphorylation,and finally promotes BACE1 expression.
3.ABO genotyping by duplex amplification and oligonucleotide arrays assay.

OBJECTIVE:
ABO genotyping for forensic identification by oligonucleotide chip.
METHODS:
Oligonucleotide microarrays which could detect 3 different SNPs in exon 6 and exon 7 for ABO genotyping were used. Population studies on ABO was carried out in a sample of 115 unrelated Chinese Han individuals. The method was also applied to cases.
RESULTS:
The technique could identify 6 genotypes of ABO system. According to the results of population studies, no significant deviations from Hardy-Weinberg equilibrium could be found. The observed and expected heterozygosities were 0.591 and 0.616 respectively. The polymorphic information content was 0.544. The average exclusion probabilities in buos and trios was 0.188 and 0.344 respectively. The discrimination power is 0.777.
CONCLUSION
The data and case application demonstrated that ABO typing by oligonucleotide probe arrays was a useful technique for paternity testing and individual identification.

4.Application of Eurorad database in the teaching of radiology in standardized residency training: A case study of teaching film-reading

The teaching of radiology in standardized residency training needs a large number of case data to strengthen the subjective understanding and awareness of residents. The database built by residency training bases can meet the needs of teaching to a certain extent, but the conditions of training bases vary across regions, which makes it difficult to achieve homogeneity in the teaching of radiology. This article discusses the application of Eurorad database in the teaching of radiology in standardized residency training. This database is free of charge, reliable, and comprehensive and provides a large number of free reliable cases and images for teaching, covering both common and rare diseases. Moreover, it can also be used to cultivate the English ability and comprehensive quality of residents and help to establish a hierarchical training system for radiology and non-radiology residents, thereby promoting the improvement in the quality of standardized residency training. This article shows the potential value of Eurorad database in the teaching of radiology in standardized residency training, and comparative studies are needed in the future to further prove its effectiveness.
