2.Mobilization of peripheral blood stem cells in mice with short-duration and high-dose recombinant human granulocyte colony-stimulating factor

BACKGROUND: The prompt and efficient peripheral blood stem cell (PBSC) mobilization is necessary for repairing and regenerating injured tissues. Recombinant human granulocyte colony-stimulating factor (rhG-CSF) or its combination with a large dose of chemotherapy is usually utilized for PBSC mobilization, but the regimen requires a long time and a complex procedure. OBJECTIVE: To modify the common PBSC mobilization, and investigate mobilization effect of PBSC in KM mice with short-duration and high-dose rhG-CSF. DESIGN, TIME AND SETTING: A randomized controlled animal trial was carried out in the laboratory of Hematology, the Second Affiliated Hospital to Dalian Medical University from September 2006 to March 2007. MATERIALS: Twenty-four KM male mice of 8 months old were divided into 3 groups according to the administration: conventional regimen group, short-duration and high-dose group, and saline control group. METHODS: Mice in the conventional regimen group were injected subcutaneously with rhG-CSF (0.2 ?g one time, twice a day for 6 days). Mice in the short-duration and high-dose group were injected with equal volume of normal saline at first 4 days, and then with rhG-CSF (2.5 ?g one time, twice a day for 2 days). Mice in the saline control group were injected subcutaneously with equal volume of normal saline for 6 days. MAIN OUTCOME MEASURES: Peripheral blood white blood cell (WBC) and colony-forming unit-granulocyte macrophage in KM mice were counted. RESULTS: WBC in the peripheral blood of KM mice increased rapidly in conventional regimen group and short-duration and high-dose group. No obvious change of WBC was observed in the control group. The number of colony-forming unit-granulocyte macrophage in conventional regimen group and short-duration and high-dose group was significantly elevated over 50 folds than control group (P 0.05). CONCLUSION: There is a good efficacy and feasibility in PBSC mobilization of KM mice with short-duration and large-dose rhG-CSF, and it is a successful amelioration of PBSC mobilization of mice peripheral blood.
3.Psychological conditions of patients in different stages of in vitro fertilization-embryo transfer cycles

BACKGROUND:With the improvement of in vitro fertilization-embryo transfer technology, patients with in vitro fertilization-embryo transfer are facing more and more psychological stress. To completely understand their mental features and to explore effective mental intervention are important problems faced workers of reproductive medicine.
OBJECTIVE:To investigate the psychological condition of in vitro fertilization-embryo transfer patients.
METHODS:A total of 92 in vitro fertilization-embryo transfer patients, who entered the cycle from October to
November 2012, were enrol ed in this study. The psychological conditions of these patients were evaluated using Symptom Checklist-90.
RESULTS AND CONCLUSION:Al the factor scores of the Symptom Checklist-90 in patients with in vitro fertilization-embryo transfer achieved significantly higher levels than national adult norm on their first day of in vitro fertilization-embryo transfer cycles (P<0.05), with the exception of Psychiatric state factor. Meanwhile, in addition to Psychiatric state, differences of al the factors of Symptom Checklist-90 in patients with in vitro fertilization-embryo transfer were statistical y significant in different stages of in vitro fertilization-embryo transfer cycles at the first day of entering cycle, the day of obtaining ovum, and the seventh day of embryo transfer, (P<0.05). Interpersonal relationship, Depression and Anxiety scores were increased at the first day of entering cycle, the day of obtaining ovum, and the seventh day of embryo transfer (P<0.05). These results showed that patients with in vitro fertilization-embryo transfer tend to have poor psychological status, which is a problem in patients during the different stages of in vitro fertilization-embryo transfer cycles and becomes aggravated during egg retrieval and waiting for the outcome. Interventional measures should be taken in different stages to improve patients’ mental health.
4.Bone marrow mesenchymal stem cells for epidermal and skin appendage regeneration

BACKGROUND:Burned rat serum has been reported as an inducer that can induce differentiation of mesenchymal stem cells into epidermal cells.
OBJECTIVE:To induce in vitro differentiation of bone marrow mesenchymal stem cells into epidermal cells that were transplanted alone or combined with inducers for the repair of skin wound defect and epidermal reconstruction.
METHODS:Under aseptic environment, rat bone marrow was harvested to culture adherent cells using low-glucose Dulbecco’s modified Eagle’s medium. Culture cells at passage 4 were confirmed as mesenchymal stem cells by flow cytometry. Bone marrow mesenchymal stem cells were induced by Dulbecco’s modified Eagle’s medium/Ham’s nutrient mixture F-12 containing 20%burned rat serum to differentiate into epidermal cells that were identified by immunohistochemistry. Wistar rat models of ful-thickness skin wounds were prepared and divided into three groups. The 5-bromo-2-deoxyuridine-labeled autologous bone marrow mesenchymal stem cells and induced bone marrow mesenchymal stem cells were coated singly onto a rat model of burn wounds, and rat models of burn wounds with no treatment served as controls. Wound contraction and regeneration of epidermal cells and skin appendages were observed.
RESULTS AND CONCLUSION:After isolation and culture of cells for 24 hours, a few of adherent cells grew as fibroblast-like cells with fusiform shape. At 16 days, cells completely covered the bottom of bottle, exhibiting a fish or reticular arrangement. After detection by flow cytometry, cells were cultured in Dulbecco’s modified Eagle’s medium/Ham’s nutrient mixture F-12 containing 20%burned rat serum, and fusiform-shaped cells gradual y became round or oval cells. Flow cytometry analysis and immunocytochemistry results showed that cells expressed keratin, which were confirmed as epidermal cells. The results show that both the bone marrow mesenchymal stem cells transplantation alone or with necessary inductor is better for skin repair than natural healing, exhibiting a faster regeneration of skin and skin appendages. Bone marrow mesenchymal stem cells are deduced preliminarily to be involved in epidermis and hair fol icle regeneration, thereby improving skin healing.
5.Prevalence of preoperative malnutrition and application of nutrition support among gastrointestinal cancer patients in Beijing

ObjectiveTo investigate the prevalence of preoperative malnutrition and nutrition support among gastrointestinal cancer patients.MethodsA retrospective study was conducted based on the nutritional status and nutrition support of 336 gastrointestinal cancer patients who received elective radical surgery in Peking Union Medical College Hospital from April to December 2009.ResultsMalnutrition was found in 34.6% gastric cancer patients,23.8 % colorectal cancer patients,and 23 % esophageal and cardiac cancer patients.The patients with worse nutritional status were given longer duration of nutrition support and higher energy intake.Conclusion Malnutrition occurs in gastrointestinal cancer patients,yet sometimes treated with inappropriate nutrition support.
6.Study on the construction of evaluation indicator system of health education effect for our country′s hospitalized patients with diabetes

Objective To construct a scientific and feasible evaluation indicator system of health education effect of our country′s hospitalized patients with diabetes. Methods On the basis of literature review and semi-structured interview combined with group discussion, alternative index system were determined,and then the Delphi method were used for two round consultation with 30 diabetes experts to finally determine indicator evaluation system. Results Positive coefficient of the two rounds consultation were 86.7% (26/30) and 100.0% (26/26) respectively, experts′ authority coefficient was 0.889,and the Kendall W coefficients of concordance were 0.447,0.369 and 0.302 respectively (P<0.01). The evaluation indicator system of health education effect was consisted of 6 fisrt-level indicators, 24 second-level indicators and 55 third-level indicators. Conclusions The evaluation indicator system of health education effect is reliable and scientific, which can provide objective and quantitative standards for health education effect of hospitalized patients with diabetes in our country.
7.Expression of Bcl-2 and P53 Gene Proteins on Leukemia Cells and Its Correlation with Chemotherapeutic Efficacy

To explore Bcl-2 and P53 gene proteins expression on human leukemia cells and their relationship with chemotherapeutic efficacy, Bcl-2 and P53 gene proteins expression was assayed by ABC immunohistochemical staining. Results showed that the expression rates of Bcl-2 and P53 gene proteins were 67% and 41% respectively in leukemia cells from 52 patients. While there was no difference of Bcl-2 protein level in ALL and ANLL, the P53 protein level was higher in ANLL than that in ALL (P < 0.05). When CML patients got into the blast crisis phase, the level of Bcl-2 and P53 proteins became very high. Compare with previously untreated AL, relapse/refractory AL patients had higher Bcl-2 and P53 protein level, lower marrow complete remission, and was easy to relapse. The expression of Bcl-2 and P53 protein could be used as new predictors of chemotherapeutic efficacy and prognosis in patients with leukemia. The high protein expression of Bcl-2 and P53 demonstrated that CML was conversion to blast crisis phase.
8.Investigation on professional maturity and career planning of nursing students

Objective To investigate professional maturity and career planning of nursing students and explore their correlation.Methods Totals of 138 nursing students were investigated by professional maturity scale and career planning scale.Results Both professional maturity and career planning of nursing students were average level,with the score of (3.08 ± 0.24),(3.47 ± 0.41),and professional maturity degree was positively correlated with career planning level (P < 0.05).Conclusions Professional maturity and career planning of nursing students have a certain degree of development,but there is still some gap to mature period,and the imbalance appears in various dimensions.Clinical nursing educators should strengthen the self-confidence of nursing students,and guide career planning according to the degree of professional
9.Reliability and validity of Chinese version of Pediatric Quality of Life Inventory TM 3.2 Diabetes Module for children and adolescents

Objective:To translate Pediatric Quality of Life Inventory TM 3.2 Diabetes Module (PedsQL TM 3.2-DM) for children and adolescents into Chinese and analyze the reliability and validity of it. Methods:Following the principle of Brislin translation, the source scale was translated by 2 supervisor nurses with master's degrees and the scale was back translated by 2 nursing experts with overseas study experience. The semantic adjustment of the scale was carried out by pre-surveys. By the convenient sampling method, a total of 175 children and adolescents with diabetes who were treated in Department of Endocrinology and Department of Pediatrics in ClassⅢ Grade A hospitals in Yantai and Qingdao from March to August 2019 were investigated and 175 effective scales were finally included to analyze the reliability and validity of the revised scale in children and adolescents with diabetes.Results:The Chinese version of PedsQL TM 3.2-DM had 33 items in total, including 4 dimensions such as disease-related symptoms, treatment compliance, disease-caused worry and communication. The Cronbach's α coefficient of Chinese version of PedsQL TM 3.2-DM was 0.927 and that of 4 dimensions were respectively 0.885, 0.910, 0.828 and 0.798. After assessment by panel of experts, the content validity index of Chinese version of PedsQL TM 3.2-DM was 0.97 and the content validity index of every item was from 0.80 to 1.00. A total of 4 common factors were extracted by exploratory factor analysis and the cumulative variance contribution rate was 59.539%. In addition, the factor loading of each item on its common factor was over 0.4. Conclusions:The Chinese version of PedsQL TM 3.2-DM has good reliability and validity, which can be used as a simple tool for self-assessment of quality of life in children aged from 8 to 19 years with diabetes.
10.Efficacy comparison of Outpatient Department education and inpatient self-management education in patients with type 2 diabetes

Objective To explore the effects of outpatient and inpatient self-management education in patients with type 2 diabetes and analyze the weak spot of diabetes mellitus health education. Methods We selected in a 1∶1 rate 110 newly diagnosed patients with diabetes mellitus in outpatient and endocrine ward from March 2013 to October 2013. Out-patients selectively received systematic outpatient education related to diabetes as one-to-one guidance by outpatient education nurses. Inpatients received diabetes self-management education as alternate one-to-one guidance and lectures undertaken by primary nurses and education nurses. Results One month after interventions, the compliance of diet, exercise, medications, and blood glucose monitoring in out-patients were all lower than that in inpatients (P <0. 01). Conclusions The effects of inpatient′s self-management education are better than outpatient education. Managers should analyze the weak spot of outpatient diabetes mellitus health education, and explore the effective methods of blood glucose monitoring education to improve patients′blood glucose monitoring compliance.
