1.Mechanism of paeonol combined with paeoniflorin against myocardial ischemia injury:based on proteomics.

The study aims to investigate the effect of the compatibility of paeonol and paeoniflorin(hereinafter referred to as the compatibility) on the expression of myocardial proteins in rats with myocardial ischemia injury and explore the underlying mechanism of the compatibility against myocardial ischemia injury. First, the acute myocardial infarction rat model was established by ligation of the anterior descending branch of the left coronary artery. The model rats were given(ig) paeonol and paeoniflorin. Then protein samples were collected from rat cardiac tissue and quantified by tandem mass tags(TMT) to explore the differential proteins after drug intervention. The experimental results showed that differential proteins mainly involved phagocytosis engulfment, extracellular space, and antigen binding, as well as Kyoto encyclopedia of genes and genomes(KEGG) pathways of complement and coagulation cascades, syste-mic lupus erythematosus, and ribosome. In this study, the target proteins and related signaling pathways identified by differential proteomics may be the biological basis of the compatibility against myocardial ischemia injury in rats.

2.Effect of cytotoxicity on normal cells and anti-tumor activity on tumor cells of cerium nitrate in vitro

Objective To investigate the cytotoxicity on normal cell lines of cerium nitrate, the inhibitory effect on tumor cell lines and the effect on cell cycle in vitro. Methods The cytotoxicity of cerium nitrate on FL, L929 and inhibition rate on Hela, SGC-7901, B16, Lewis, K562, and H_(22) in vitro were detected with the method of MTT. The effect of cerium nitrate on cell cycle of SGC-7901 was studied by one-fluorescence dying method on flow cytometer. Results Cerium nitrate did not show the toxic activities on FL and L929 cell lines when the dose of cerium nitrate was it could noticeably inhibit the growth and proliferation of tumor cells nitrate inhibited the growth and proliferation of tumer cells including HeLa, SGC-7901, B16 and K562; cerium nitrate had a noticeably inhibitory effect on the growth and proliferation of H_(22) cell lines in vitro on Conclusion Cerium nitrate has the property of low toxicity and high selectivity on normal cells, but it can inhibit growth and proliferation of many kinds of tumor cell lines by adjusting cell cycle.
3.Effect of maple syrup on fatigue resistance in mice

Objective To investigate the effect of maple syrup on fatigue resistance in mice. Methods Totally forty Kunming mice were divided randomly into normal control group, low-dose maple syrup group (1.25g/kg), medium-dose maple syrup group (2.50g/kg), and high-dose maple syrup group (5.00g/kg). Mice were continuously given water and maple syrup through mouth for 30 days. Various groups swam for 10min. The contents of blood lactic acid were measured at swimming 0min, 10min and resting 30min. The contents of hemoglobin, blood lactic acid, serum urea nitrogen, muscle glycogen, and liver glycogen of various group mice were detected after resting 30min. 4.Prediction of drug bioavailability by genetic algorithm and artificial neural network.

AIMTo set up an artificial neural network system and optimize by genetic algorithm (GA) to predict drug bioavailability.
METHODSGenetic algorithm was used to optimize weights of the artificial neural network. The optimal solution of the artificial neural network model at a specific condition was obtained using the good search ability of genetic algorithm in order to predict drug bioavailability. Volume, refractivity, lgP(c), hydration, polarizability, E(HOMO) and E(LUMO) are inputs of the drug bioavailability prediction neural network, and its output is average drug bioavailability.
RESULTSThe prediction precision of average drug bioavailability of the GA- neural network model is 95.9%.
CONCLUSIONThis model can be used in the forecasting of drug bioavailability.

5.Supplementation of Fermented Barley Extracts with Lactobacillus Plantarum dy-1 Inhibits Obesity via a UCP1-dependent Mechanism.

OBJECTIVE:
We aimed to explore how fermented barley extracts with Lactobacillus plantarum dy-1 (LFBE) affected the browning in adipocytes and obese rats.
METHODS:
In vitro, 3T3-L1 cells were induced by LFBE, raw barley extraction (RBE) and polyphenol compounds (PC) from LFBE to evaluate the adipocyte differentiation. In vivo, obese SD rats induced by high fat diet (HFD) were randomly divided into three groups treated with oral gavage: (a) normal control diet with distilled water, (b) HFD with distilled water, (c) HFD with 800 mg LFBE/kg body weight (bw).
RESULTS:
In vitro, LFBE and the PC in the extraction significantly inhibited adipogenesis and potentiated browning of 3T3-L1 preadipocytes, rather than RBE. In vivo, we observed remarkable decreases in the body weight, serum lipid levels, white adipose tissue (WAT) weights and cell sizes of brown adipose tissues (BAT) in the LFBE group after 10 weeks. LFBE group could gain more mass of interscapular BAT (IBAT) and promote the dehydrogenase activity in the mitochondria. And LFBE may potentiate process of the IBAT thermogenesis and epididymis adipose tissue (EAT) browning via activating the uncoupling protein 1 (UCP1)-dependent mechanism to suppress the obesity.
CONCLUSION
These results demonstrated that LFBE decreased obesity partly by increasing the BAT mass and the energy expenditure by activating BAT thermogenesis and WAT browning in a UCP1-dependent mechanism.

6.CD137 signaling promotes angiogenesis through regulating macrophage M1/M2 polarization.

Objective: To investigate whether CD137 signaling can promote angiogenesis via regulating macrophage M1/M2 polarization. Methods: (1) The primary peritoneal macrophages in mice induced by 3% thiglycollate broth were divided into three groups: control group, CD137 signaling activated group and CD137 signaling inhibited group. Various specific markers of M1 and M2 macrophages were detected to observe the phenotype change of macrophages, and the macrophages protein expression of CD137, CD86 and CD206 was detected by flow cytometry (FCM). The protein and mRNA expression of induced nitric oxide synthase (iNOS), arginase Ⅰ(Arg-1) was determined by Western blot and RT-PCR, respectively. The secretion levels of IL-12 and IL-10 in culture supernatant of macrophages were detected by ELISA. (2) Macrophages were co-cultured with the endothelial cells (bEnd.3), and macrophages were implanted in the upper chamber, endothelial cells were implanted in stromal glue of the lower chamber. The experiment was divided into three groups: the control group, CD137 signaling activated group and peroxisome proliferator-activated receptor-γ (PPAR-γ) inhibited group, and tube formation ability of endothelial cells in each group was determined. Results: (1) The purity of primary peritoneal macrophages in mice was (97.93±1.31)%. The expression of CD137 on the surface of macrophages was (97.40±2.70)%. (2) Compared with control group, the mRNA and protein expression levels of Arg-1 were significantly increased and the mRNA and protein expression of iNOS were significantly decreased in CD137 signaling activated group (all P<0.05). Compared with CD137 signaling activated group, the mRNA and protein expression of Arg-1 were significantly lower and the mRNA and protein expression levels of iNOS were significantly higher in CD137 signaling inhibited group (all P<0.05). FCM results showed that the average fluorescence intensity of CD206 was higher, while the average fluorescence intensity of CD86 was lower in CD137 signaling activated group than in control group (P<0.05, P<0.01, respectively); the expression of CD206 was significantly lower, while the expression of CD86 was higher, in the CD137 signaling inhibited group than in CD137 signaling activated group (P<0.05, P<0.01, respectively). ELISA results showed that the secretion of IL-10 was higher, and the secretion level of IL-12 was significantly lower in CD137 signaling activated group than in control group (both P<0.01); the secretion of IL-10 was significantly lower and the secretion of IL-12 was significantly higher in CD137 signaling inhibited group than in CD137 signaling activated group (both P<0.05). (3) Values of the formation of tube length and branch number were both longer in CD137 signaling activated group than control group (P<0.05). The formation of the tube length and branch number were less in PPAR-γ inhibited group than in CD137 signaling activated group (P<0.05). Conclusion: CD137 signaling can promote angiogenesis by regulating macrophage M1/M2 polarization.

7.Layer-specific strain assessment on left ventricular function before and after PCI in patients with ST segment elevation myocardial infarction.

Objective: To evaluate the changes of left ventricular function in patients with ST segment elevation myocardial infarction (STEMI) before PCI and within 24 hours after PCI by layer-specific strain, and to explore the value of this new assessment method for quantitative monitoring on the myocardial function in STEMI patients. Methods: A total of 40 patients with acute anterior wall myocardial infarction, who underwent PCI in Affiliated Hospital of Jiangsu University during July 2017 to July 2018, were included in this prospective cohort study. According to the symptom to balloon time (STB), the patients were divided into STB ≤6 hours group (26 cases) and STB 6-12 hours group (14 cases). Echocardiography was performed before, immediately, 3 hours and 24 hours after PCI. Echocardiographic indexes including endocardial myocardial longitudinal strain (LS-endo), 18-segment full-thickness myocardial longitudinal strain (LS) of left ventricle and left ventricular global longitudinal strain (GLS) were measured. The mean LS-endo and LS values of myocardial segments in infarcted area (IALS-endo, IALS) and the mean LS-endo and LS values of myocardial segments in non-infarcted area (NIALS-endo, NIALS) were calculated. Results: There were 34 males and 6 females in this cohort and age was (62±10) years. In STB≤6 hours group, the IALS-endo value ((13.7±4.9)% vs. (10.0±2.7)%, P<0.05) and NIALS-endo value ((17.0±2.9)% vs. (14.6±2.9)%, P<0.05) were significantly higher at 24 hours after PCI than those before PCI. In the group of STB 6-12 hours, IALS-endo decreased immediately after PCI ((6.7±3.3)% vs. (11.9±6.5)%, P<0.05), and there was a rising trend at 3 hours after PCI (P>0.05). At 24 hours after PCI, the index was higher than that immediately after PCI ((13.6±8.4)% vs. (6.7±3.3)%, P<0.05). The NIALS-endo value was significantly higher at 24 hours after PCI than that before PCI ((17.1±2.1)% vs. (14.5±3.2)%, P<0.05). In the STB 6-12 hours group, the decrease rate of IALS-endo immediately after PCI was higher than that in the STB ≤6 hours group (93% (13/14) vs. 35% (9/26), P<0.001). In STB ≤6 hours group, the NIALS value at 24 hours after PCI was higher than that before PCI (P<0.05), and there was no significant difference in IALS, NIALS and GLS at other time points (P>0.05). Conclusions: Layered LS is superior to full-thickness LS and GLS in evaluating left ventricular function in STEMI patients. LS measured by echocardiography can continuously and quantitatively evaluate the changes of left ventricular myocardial function in STEMI patients before and after PCI.

8.IL-1β-Treated Bone Marrow Mesenchymal Stem Cells Enhances Osteogenetic Potential via NF-κB Pathway.

Obsjective:To investigate the effects of differentaction time of IL-1β on the osteogenic capacity of bone marrow mensenchymal cells(BMMSC) and the role of nuclear factor-κB (NF-kB) pathway.
METHODSBMMSC isolated from normal donors was treated with IL-1β for 1 or 7 days, respectively. Alkaline phosphatase (ALP) and alizarin red(AR) stainings were used to detect the osteogenic differentiation potential of BMMSC. The mRNA expression of EphB4, IGF-1 and OPG in BMMSC was measured by real-time PCR. The immunohistochemistry was employed to measure the expression of bone morphgenetic protein-2(BMP-2) and p-Smad1/5/8 in BMMSC. Furthermore, the Western blot was used for the detection of iκBα and phospho-iκBα (p-ikBα) in IL-1β-treated BMMSC. And the results of IL-1β-treated BMMSC were compared with control group.
RESULTSCompared with control group, the osteogenetic potential of IL-1β-treated BMMSC was enhanced, but the pro-osteogenic differentiation effect of IL-1β was remarkedly inhibited in the presence of NF-kB pathway inhibitor PDTC. The total ikBα level of IL-1β-treated BMMSC was lower (P<0.05), and phospho-iκBα (p-iκBα) level was higher (P<0.05). Besides, BMP-2 expression was higher (P<0.05) in the IL-1β-treated BMMSC, however, p-Smad1/5/8 protien level was not significantly different among IL-1β-treated for 1 d, 7 d and control groups (P<0.05). And the mRNA expression levels of IGF-1, EphB4 and OPG in BMMSC were up-regulated after IL-1β treatment (P<0.05). In addition, the osteoblastogenesis of BMMSC treated with IL-1β for 7 days was significantly different from those treated only for 1 day.
CONCLUSIONProlonging IL-1β treatment can enhance the osteogenetic differentiation of BMMSC more significantly. And this osteogenetic alteration of BMMSC occurs via its NF-κB pathway, but not via BMP-2/Smad pathway.
10. Doctor-patient trust and its influencing factors based on the view of patient family members

Objective To investigate the evaluation of the doctor-patient trust by family members based on the general interpersonal trust scale, and explore the influencing factors of the evaluation of doctor-patient trust by patient family members, and to provide references for improving the doctor-patient trust and the doctor-patient relationship. Methods A survey of 3010 patient family members in 10 hospitals from 5 provinces in China was conducted using a self-made questionnaire and a general interpersonal trust scale. Results Only 47.2% of the family members agreed that the doctor-patient trust degree was good, and the interpersonal trust score was (70.52±7.07) points. The age, monthly income level, life satisfaction of family members are the important factors affecting the evaluation of doctor-patient trust (P < 0.05). Conclusion The patient family members have low recognition of doctor-patient trust, and some targeted measures should be taken to improve the interpersonal trust level and life satisfaction of the family members, so as to further enhance the doctor-patient trust.
