Objective: To investigate the protective effects and underlying mechanisms of sodium pyruvate (SP) on RBC storage lesions using an oxidative damage model. Methods: Six units of leukocyte-depleted suspended RBCs (discarded for non-infectious reasons within three days post-collection) were randomly assigned to four groups: negative control (NS), positive control (PS), experimental group 1 (SP1), and experimental group 2 (SP2). Oxidative stress was induced in the PS group by the addition of hydrogen peroxide (H

), while SP1 and SP2 received SP supplementation at different concentrations (25 mM and 50 mM, respectively) in the presence of H

. After 1 hour of incubation, RBC morphology was assessed microscopically, and biochemical indicators including glutathione (GSH), malondialdehyde (MDA), methemoglobin (MetHb), adenosine triphosphate (ATP), and Na
/K
-ATPase activity were measured. Results: RBCs in the PS group exhibited pronounced morphological damage, including cell shrinkage and echinocyte formation, whereas both SP-treated groups showed significantly reduced structural injury. SP treatment led to elevated GSH levels and decreased concentrations of MDA and MetHb, suggesting attenuation of oxidative stress. Additionally, SP enhanced intracellular ATP levels and Na
/K
-ATPase activity, thereby contributing to membrane stability. Notably, the SP2 group (50 mM) demonstrated superior protective effects compared to SP1 (25 mM). Conclusion: Sodium pyruvate effectively attenuates oxidative storage lesions in RBCs, primarily through its antioxidant properties, energy metabolism supporting ability, and celluar membrane stabilizing function. These findings suggest SP as a promising additive for enhancing the quality and safety of stored RBCs.
3.Investigation of Goiter Prevalence in Shaanxi Province

Objective To know the prevalent trend of goiter in Shaanxi province and to provide the basic data for making the control strategy. Methods In 2002, with the population proportionate sampling, the investigation was conducted according to China National Criteria for Diagnosis and Grade Division of Endemic Goiter GB16004-1995. Results The prevalence rate of goiter was 6.52%, the number of patients with visible goiter was 317 000 in Shaanxi province. Conclusion It was not proper to evaluate the population iodine nutrition by the goiter rate of children aged 8-10 years, it may aggrandize the serious degree of IDD. Up to now, the number of patients with visible goiter is decreased by 659 000 and no new Cretinism patient has been found in Shaanxi.
4.Determination of Tween-80 CMC in Chinese Herbal Medicine Injections by Ultraviolet Spectrophotometry

Ultraviolet spectrophotomety was used to determine Tween-80 critical micell concentration (CMC) in different Chinese herbal medicine injections. Studies on the concentration of iodine solution and wavelength suitable for the determinntion were carried out. This method is quick and accurate and can be used for the quality control of such preparation.

Objective To observe the effects of embedding implantation of collagen and Tetramethylpyrazine in acupoints and electro-acupuncture on the expression of HSP70 in hippocampus of rat with focal cerebral ischemia reperfusion. Methods SD rats were randomly divided into normal group, sham operation group, model group, electro-acupuncture group, embedding medicine in acupoints group. Model of local ischemia was made by middle cerebral artery occlusion, preparation of implantation collagen and electro acupuncture and Tetramethylpyrazine was embedded in the acupoints of Dazhui and Neiguan two-sided. The expression of HSP-70 protein in hippocampus of different time point (24, 72, 120 h) was assayed by immunohistochemistry. Results There were only few positive cells of HSP-70 in normal group and sham operation group. The expression of HSP-70 in each therapy group were all heightened, and achieved the peak 24 h later and then declined in brain tissue after focal cerebral ischemiareperfusion, the differences were relatively prominent compared with the same phase model group (P

Objective To observe the influence of bone marrow mesenchymal stem cells (BMSCs) transplantation on the expression of alpha-amino hydroxymethyl-oxazole propionic acid (AMPA) receptors GluR1 and GluR2 in rats with spinal cord injury (SCI) so as to investigate the potential anti- chronic stress mechanism of BMSCs transplantation in treatment of rats with spinal cord injury.Methods A total of 48 adult male SD rats were equally divided into three groups:control group,treatment group and model group.The rats in the model and treatment group underwent lower thoracic SCI with the modified Allen' s method,and the rats in control group received only laminectomy.At day 7 after thoracic SCI,100 μl of Hank's buffered saline solution containing 1.0 × 106 BMSCs was injected into the subarachnoid space from L4-L5 intervertebral space in the treatment group and control group,and the same amount of Hank' s buffered saline solution was injected in the model group.The motor function of the rat posterior limbs was assessed by Basso-Beattie-Bresnahan (BBB) scale before and after operation.Half of the rats were anesthetized at days 14 and 28 postoperatively to harvest brains which were frozen and cut in a cryostat to detect the expressions of GluR1 and GluR2 proteins by immunohistochemistry.Results After BMSCs transplantation,the motor function of the posterior limbs in the treatment group was improved progressively.At day 14 after transplantation,the number of GluR1-positive cells of the model and treatment group was higher than that of the control group in the hippocampus CA1 region (P <0.05,P <0.01 respectively) ; GluR2-positive cells had the similar tendency,without significant difference(P > 0.05 ).At the 28th day after transplantation,GluR1 positive cells of the model group were higher than those of the control group in CA1,CA3,DG regions and those of the treatment group in CA1,CA3 regions (P <0.05,P <0.01,respectively) ; GluR1 positive cells of the model and treatment group were higher than their counterpart at day 14 after grafting procedure,with significant difference (P <0.05,P <0.01,respectively).GluR2 positive cells of the treatment group were higher than those of the control group in the basolateral amygdale (BLA) (P <0.05 ) and had similar tendency with GluR1 expression in other regions ( P > 0.05 ).Conclusion BMSCs transplantation implies a potential antichronic stress mechanism of SCI rats,since it can improve the motor function of posterior limbs in rats with lower thoracic SCI and regulate the expressions of AMPA receptor GluR1 and GluR2.
7.Study on the Quality Standard of Fructus arctii Concentrated Granules

OBJECTIVE:To establish the quality standard of Fructus arctii concentrated granules. METHODS:TLC was con-ducted for the qualitative identification and HPLC was used for the content determination of arctiin in F. arctii concentrated gran-ules. The determination was performed on Dionex C18 column with mobile phase consisted of methanol-water(40∶60,V/V) at the flow rate of 1.0 ml/min. The detection wavelength was set at 280 nm,and the column temperature was 30 ℃. The samples size was 10 μl. RESULTS:TLC spectrum showed that the spots of F. arctii concentrated granules and arctiin reference medicinal material had the same color;the linear range of arctiin was 0.5-12.5 μg(r=0.999 8)with the average recovery of 98.41%(RSD=0.81%, n=9). The RSDs of precision,stability,repeatability tests were no more than 1.0%. CONCLUSIONS:The method is feasible and reproducible,and can effectively control the quality of F. arctii concentrated granules.
8.Clinical efficacy of Frey and Beger procedures for chronic pancreatitis

Objective To analyse clinical efficacy of Frey and Bger procedures for chronic pancreatitis. Methods Clinical data of seventy patients of chronic pancreatitis undergoing Frey (32 cases) and Beger Procedures (38 cases) in our hospital from January 1999 to June 2009 were retrospectively analyzed,the rate of pain relief and endocrine and exocrine function of the pancreas in longterm follow-up were compared with each other. Results Overall perioperative morbidity for Frey group and Beger group was 12. 5% and 7.9% respectively (P =0. 52); Total pain relief rate in long-term follow-up was 96. 8% and 94. 7% respectively (P =0. 57). Postoperative de novo diabetes mellitus was 12. 5% and 21% ( x2 = 0. 90, P = 0. 34), Rate of steatorrhea was 12. 5% and 18.6% respectively ( x2 = 0. 15, P =0. 70). Conclusions Frey and Beger procedures were equally effective in improving symptoms of chronic pancreatitis and there were no significant differences in perioperative morbidity, rate of pain relief and function of endocrine and exocrine of the pancreas.
9.Changes of plasma orexin-A level and behavior performance before and after operation in children with obstructive sleep apnea-hypopnea syndrome

Objective To explore the changes of plasma orexin-A level before and after operation in obstructive sleep apnea-hypopnea syndrome(OSAHS) children and its effect on their behavior performance.Methods 120 cases OSAHS children performed tonsillectomy and / or adenoidectomy and 30 cases normal children as control group.According to the AHI index,the OSAHS group was divided into mild group (5 times/h ≤ AHI < 20times/h,n=13),moderate group (20/h ≤ AHI <40/h,n=76),and severe group (AHI ≥ 40 times / h,n=31).And at the same time,according to the sensory integration ratings,OSAHS group was divided into normal group(n =30),mildly abnormal group (n =47),moderately abnormal group (n =28),severely abnormal group (n =15).Before operation and 6 months after operation,plasma orexin-A levels and children's sensory integration were measured.Results Plasma orexin-A level of the OSAHS group ((0.41 ± 0.06) μg/ml) was significantly higher compared with the control group((0.31±0.04) μg/ml) (P<0.01).In orexin-A level of different AHI groups before and after operation(mild group:(0.33±0.02) μg/ml vs (0.28± 0.03) μg/ml,moderate group:(0.39±0.04) μg/ml vs (0.29±0.03) μg/ml,severe group:(0.49±0.04) μg/ml vs (0.32± 0.02) μg/ml),there had significant differences (P<0.01).In OSAHS children,AHI index had positive correlation with preoperative plasma orexin-A level (r=0.803,P<0.01).There was a significant negative correlation between sensory integration scores and plasma orexinA level(r=-0.812,P<0.01).Conclusions Plasma orexin-A level of OSAHS children is closely related to the severity of OSAHS and the changes of their behavioral ability.And it may become a diagnostic plasma marker of OSAHS children.
10.Influence of Butylphthalide on Pharmacokinetics of Aspirin in Rats

OBJECTIVE:To study the influence of butylphthalide on the pharmacokinetics of aspirin in rats. METHODS:20 rats were randomly divided into control group(vegetable oil 0.4 ml/rat+aspirin 10 mg/kg)and trial group(butylphthalide 80 mg/kg+aspirin 10 mg/kg) intragastrically,once a day,for consecutive 10 days. Blood samples were collected before the last medication and 10,20,40,60,120,240,360,480,600 and 720 min after medication,0.2 ml each time. The blood concentration of drugs was determined by HPLC,and pharmacokinetics parameters were calculated by DAS 2.0 software. RESULTS:Main pharmacokinet-ic parameters of aspirin in control group vs. trial group were as follows as cmax of (28.68 ± 6.08) vs. (29.33 ± 4.25)μg/ml;t1/2 of (2.48±0.67)vs.(1.60±0.36)h;AUC0-720 min of(188.71±24.29)vs.(140.31±15.08)μg·h/ml;CL/F of(0.05±0.01)vs.(0.07± 0.01)L/(h·kg);there were significant differences in t1/2,AUC0-720 min and CL/F(P<0.05). CONCLUSIONS:Butylphthalide has no significant effect on the absorption and distribution of aspirin in rats,but can strengthen its metabolism and elimination.
