1.The Analysis of Results of Urine Protein Electrophorests for 108 Cases with Diabetic Nephropathy

Objective The separation of various proteins in urine was used to differentiate the types of urinary protein to determine the site of diabetic kidney damage and lesions. Methods We used sodium dodeeyl sulfonate - agarosegel electrophoresis(SDS - AGE) to detect the unconccntrated urine on 108 cases patients with diabetic nephropathy urine specimens analysis. Results The results of urinary protein electrophoretic of 108 patients displayed: 4 eases for renal tubular proteinuria, 10 eases for physiological proteinuria, 22 eases for glomerular proteinuria, 72 cases for mixed proteinuria. Conclusion Urine protein electrophoresis for diabetic nephropathy patients with positive can distinguish the various components, and to understand the degree of kidney damage and lesions. This method is low -cost, non - invasive and highly sensitive. The film can be stored. It is worthy to be popularized.
2.Operative cooperation and nursing of patients receiving holmium laser lithotripsy under percutaneous nephroscope

Objective To explore the operative cooperation and nursing points of holmium laser lithotripsy under percutaneous nephroscope. Methods A retrospective study was carried out to investi-gate the operative cooperation and nursing of 38 patients receiving holmium laser lithotripsy under percuta-neous nephroscope in our hospital from March 2006 to August 2007. Results Stones were completely cleared in 31 patients at first holmium laser lithotripsy,in 5 patients at second holmium laser lithotripsy,in 2 patients at third holmium laser lithotripsy. One patient complicated with water intoxication and septicemia. Operation time ranged from 50 to 190 minutes,blood loss volume reached 50 to 200 ml with a average of 100 ml.No blood transfusion was needed during the operations and no secondary hemorrhage was seen postoperation. Double J catheter was inserted and indwelled for 30 to 60 days with a average of 38 days. Postoperative follow-up was satisfying. Conclusions Holmium laser lithotripsy under pereutaneous nephroscope possesses the advantages of little trauma,hemorrhage and complication and quick recovery.It is the best choice for treatment of calculus in urinary system.

4.Effects of Lead Exposure on Study, Memory and Autonomic Activities of Mice

Objective To study the neurotoxicity of lead to mice and the mechanism. Methods Two months old(grown) and two weeks old(infancy) mice were employed, 1 ?l of 1 mol/L lead acetate was given by brain ventricle injection, 3 d, 5 d, 1 week, 2 weeks and 3 weeks after treatment with lead, the change of autonomic activities and the ability of study and memory of the mice were examined. Results The latency period of Lashley Ⅲ water maze prolonged significantly in grown group and the infancy group compared with the control groups at every time point of test. Conclusion Lead exposure can damage the function of study and memory of mice, and increase the frequency of autonomic activities of theirs.

6.Chinese medicinal compound Yinzhiku significantly inhibits hepatitis virus

Objective: To evaluate the anti-hepatitis effect of Yinzhiku Compound and obtain pharmacodynamic evidence for its clinical application.Methods: Using the model of HepG2.2.15 cells,we determined the inhibitory effects of the drug extract with water and that with alcohol on HbsAg and HbeAg from the HepG2.2.15 cells by serum pharmacological methods.Results: The inhibition of the two Abstracts on HbsAg reached the peak on the sixth day and gradually dropped afterwards,but still at a high level on the twelfth day.Their inhibitory effect on HbeAg,too,attained the highest level on the sixth day,but did not fall obviously afterwards and remained high even on the twelfth day.Conclusion: Yinzhiku Compound has a high inhibitory effect on both HbsAg and HbeAg,with no evident difference between the drug extract with water and that with alcohol.
7.Effect of synchronous chemotherapy combined with spleen polypeptide on immune function in patients with esophageal cancer and cardiac cancer after operation

Objective To investigate the effect of synchronous chemotherapy combined with spleen polypeptide on immune function in patients with esophageal cancer and cardiac cancer after operation. Methods Sixty postoperative patients of esophageal and esophagogastric junction carcinoma were collected in the Fourth Hospital of Hebei Medical Universitythe from March 2013 to January 2014,and were randomly divided into two groups,including 30 cases were treated with simple chemotherapy as control group,and the other 30 cases were treated with chemotherapy combined with lineal polypeptides as experimental group. Results There were no significant differences in terms of CD3,CD4 and CD4/CD8 between the two groups before operation( P>0. 05) . After two cycles of chemotherapy,the CD3,CD4,CD4/CD8 of experimental group were all significantly higher than the control group((76. 08±8. 04)% vs. (38. 58±6. 40)%,(42. 91±7. 73)% vs. (42. 91±7. 73)%,1. 31 ±0. 21 vs. 1. 16±0. 22),and the differences were significant(P=0. 037,0. 022,0. 008). The survival quality of the experimental group was better than that of the control group ( P=0. 038 ) . The proportion of patients with WBC in the normal range of the experimental group was significantly higher than that in the control group (80. 0% vs. 53. 3%,P=0. 028). The adverse reactions of the digestive system in the experimental group were lower than those in the control group, the difference was statistically significant between the two groups ( acid reflux and heartburn:26. 7% vs. 63. 3%,P=0. 004;nausea and vomiting:33. 3% vs. 73. 3%,P=0. 002;loss of appetite:46. 7% vs. 80. 0%,P=0. 007) . Conclusion The treatment of the patients with esophageal cancer and cardiac cancer after chemotherapy with spleen peptides can improve immunity,and improve the quality of life of patients after chemotherapy and chemotherapy.
8.Expression of ?_3 -adrenoceptor and its function in human detrusor cells

Objective To study the expression of ? 3 adrenoceptor (? 3 AR) and its function in human detrusor cells. Methods Cellular functional studies,radioligand binding experiments and reverse transcription polymerase chain reaction were used to determine the expression and its influence on detrusor contractions. Results mRNA of 3 subtypes of ? ARs was detected. Non selective ? AR agonists and ? 3 AR decreased the contractions of detrusor cells induced by KCL, but both agonist and antagonists of ? 1 and ? 2 AR could not influence the contraction;BRL37344 concentration dependently displaced dihydroalprenolol binding to the membrane of detrusor cells. Conclusions The mRNA of subtypes of ? ARs is distributed in human detrusor cells,but ? 3 ARs is the main receptor mediating detrusor relaxation.
9.Location of DNase Ⅰ hypersensive sites exactly in the promoter region of CD133 in colonel cancer cell line SW480 cells by inverse-PCR

Objective To precious localize DNase Ⅰ hypersensive sites exactly in the promoter region of CD133 of cell line SW480 by inverse-PCR.Methods The colonel cancer cell SW480 nuclei were suspended in digested buffer,treated with DNase Ⅰ at the concentration of 10 U/ml for 10 min.The inversePCR was performed as follows.DNA treated by DNase Ⅰ was purified,fragmented with restricted enzyme EcoRI and Xmal Ⅰ.Then the ends were blunted,ligated by T4 ligase.PCR was performed,and production was sequenced.The restricted enzymes cut sites were near DNase Ⅰ cleavage sites.Results 9 DNase Ⅰ cut sites were identified in CD133 promoter region.The DNaseI hypersensitive sites all distributed in a region -300 bp--700 bp up to transcription start site.Conclusion The DNase Ⅰ cleavage sites could identified preciously by application of inverse-PCR.These sites locate in a region of-300 bp--700 bp up to transcription start site.
10.Development of Multiple ELISAs for the Detection of Antibodies against Classical Swine Fever Virus in Pig Sera

The major immunogenic proteins (Ems,E2 and NS3) of classical swine fever virus (CSFV) (Shimen strain) were expressed in E.coli and purified by affinity chromatography.The recombinant antigens were applied to develop multiple enzyme-linked immunosorbent assays (ELISAs) for the detection of specific antibodies in pig sera.Optimum cut-off values were determined by receiver operating characteristic (ROC) analysis after testing 201 sera of vaccinated pigs and 64 negative sera of unvaccinated piglets.The multiple ELISAs were validated with 265 pig sera yielding high sensitivity and specificity in comparison with the virus neutralization results.The results demonstrated that multiple ELISAs can be a valuable tool for the detection of CSFV infection and serological surveys in CSFV-free countries or for the evaluation of the antibody responses in pigs induced by a live attenuated C-strain vaccination.
