1.Chinese medicinal compound Yinzhiku significantly inhibits hepatitis virus

Objective: To evaluate the anti-hepatitis effect of Yinzhiku Compound and obtain pharmacodynamic evidence for its clinical application.Methods: Using the model of HepG2.2.15 cells,we determined the inhibitory effects of the drug extract with water and that with alcohol on HbsAg and HbeAg from the HepG2.2.15 cells by serum pharmacological methods.Results: The inhibition of the two Abstracts on HbsAg reached the peak on the sixth day and gradually dropped afterwards,but still at a high level on the twelfth day.Their inhibitory effect on HbeAg,too,attained the highest level on the sixth day,but did not fall obviously afterwards and remained high even on the twelfth day.Conclusion: Yinzhiku Compound has a high inhibitory effect on both HbsAg and HbeAg,with no evident difference between the drug extract with water and that with alcohol.
2.Doxazosin induces HeLa cell apoptosis via transcription factor activator protein-2?

Objective To investigate the pro-apoptotic effects of an ?1-adrenergic inhibitor,doxazosin,in HeLa cells and the role of transcription factor activator protein-2? (AP-2?) in the process.Methods The HeLa cells were treated with doxazosin (0,10,40 or 60 ?mol/L) for 16 h,and cell apoptosis was detected by flow cytometric analysis.After doxazosin (60 ?mol/L) treated the HaLa cells,the cells transfected with AP-2? overexpressing constructs or an antisense oligonucleotide against AP-2? for 16 h,the apoptosis was analyzed with FCM.The expression of AP-2? and caspase-3 in above 3 groups of cells was detected by relative quantitative RT-PCR and Western blot analysis,respectively.Results Doxazosin increased the apoptotic rate and total cell death rate of the HeLa cells,and upregulated the expressions of AP-2? and caspase-3 in a dose-dependent manner.Overexpressing AP-2? improved the increased apoptotic rate and total cell death rate induced by doxazosin,and enhanced the increased expression of caspase-3.Whereas doxazosin-induced apoptosis and the total cell death in HeLa cells were decreased by antisense AP-2?,and antisense AP-2? in part abolished the increased effects of doxazosin on caspase-3 expression.Conclusion Doxazosin induces apoptosis in HeLa cells in a dose-dependent manner,and transcription factor AP-2? is functionally involved in this process.
3.Studies on Quality Standard of Niaoduqing Tablet

Objective: To study the quality standard of Niaoduqing Tablet, a preparation of Chinese herbal medicine. Methods: Thin layer chromatography (TLC) was used for the identification of Radix et Rhizoma Rhei, Radix Polygoni Multiflori, Radix Sophorae Flavescentis, Radix Salviae Miltiorrhizae and Rhizoma Chuanxiong in Niaoduqing Tablet. TLC scanning was used for the determination of astragaloside IV and HPLC for determination of paeoniflorin in Niaoduqing Tablet. Results: TLC identification were highly specific. A good linearity for astragaloside IV was ranged 1.006~9.064?g and that for paeoniflorin was ranged 0.176~0.880?g. The average recovery of astragaloside IV was 97.69 %( RSD= 2.13 %) and that of paeoniflorin was 97.47 %( RSD=0.87 %). Conclusion: This method can be used for the standard of quality control of Niaoduqing Tablet.
4.Research on mechanisms of deoxypodophyllotoxin-induced inhibition of cell proliferation and migration in human lung cancer NCI-H358 cells

Objective To investigate the effects and mechanism of deoxypodophyllotoxin on cell proliferation and mi?gration of human lung cancer NCI-H358 cells in vitro. Methods CCK-8 assay, flow cytometry assay, wound healing assay and DCFH-DA assay were used to detect the effects of deoxypodophyllotoxin on the proliferation, cells cycle, apoptosis, mi?gration and reactive oxygen species (ROS). The protein expressions of Cyclin B1, Cdc25c, CDK1, Caspase-3, p53, Bcl-2, MMP9, ERK1/2, p38MAPK and JNK were measured by Western blot assay, respectively. Results Deoxypodophyllotoxin inhibited cell proliferation and reduced migration in human lung cancer NCI-H358 cells. Flow cytometry analysis showed that treatment with deoxypodophyllotoxin resulted in cell cycle G2/M and S phase arrest, cell apoptosis and ROS production. The result of Western blot assay showed that protein expressions of Cyclin B1, Cdc25c, CDK1, Bcl-2 and MMP9 were down-regulated while Caspase-3 and p53 were up-regulated. Moreover, Deoxypodophyllotoxin treatment decreased the phosphory?lated levels of ERK1/2, p38MAPK and JNK obviously. Conclusion Deoxypodophyllotoxin could suppress the proliferation and migration of human lung cancer NCI-H358 cells in vitro, which is a potential anti-tumor drug.
5.Study of Isothermal Adsorption of Bovine γGlobulin on Nylon Affinity Membrane with LTryptophane Ligand by Batch Method

A new nylon affinity matrix with L-tryptoph ane as ligand was prepared for adsorption of bovine γ-globulin. Effects of tem perature, ionic strength and pH on affinity adsorption of γ-globulin on affinity membrane were studied by batch method. The results show that the interaction between ligand and γ-globulin includes mainly electrostatic and hydrophobic interaction. The affinity adsorption at optimum condition obeys Langmuir adsorption model with maximum adsorption capacity and minimum nonspecific adsorption. The space location between protein and ligand and the configuration of proteins wi ll change when deviating from this condition.
6.Effect of Isoliquiritigenin on C6 glioma cell proliferation and differentiation

Aim To investigate the effects of isoliquiri-tigenin(ISL)on C6 glioma cell proliferation and differ-entiation.Methods C6 glioma cells’viability and proliferation were respectively measured by SRB test. Colony formation of C6 glioma cells from different groups was assayed.After culturing the cells from each group,giemsa staining was used to observe cell mor-phology.RT-PCR was applied to detect mRNA expres-sion of GFAP.Western blot was applied to detect the expression of GFAP.Results ISL effectively inhibited the viability of C6 glioma cells when compared with the control group in a concentration-dependent manner (P<0.01).The morphological observation under light mi-croscope showed that:in the control group,most of the undifferentiated C6 cells showed long fusiform and po-lygonal shape.Compared to the control group,the C6 cells treated with ISL revealed alteration in morphology such as astrocytes with smaller smooth,round body and much finer longer,tapering processes.The cloning for-mation rate detection revealed that:the colonies in the control group semerged earlier and were larger than those experimental ones,the cloning formation rate was higher,while almost no effective cells colony emerged in ISL treated groups(P <0.01 ).Western blot and RT-PCR analysis showed that GFAP expression in the ex-perimental groups increased(P <0.01).Conclusion ISL may inhibit the proliferation of C6 glioma cells and induce their differentiation.
7.Standing at a table while training improves the upper limb function of hemiplegic stroke survivors

Objective To explore the effect of standing at a table while training on the upper limb function and muscle surface electromyography of hemiplegics.Methods Sixty hemiplegic persons were randomly divided into an experimental group and a control group,each of 30.The affected upper limbs of the experimental group were trained while standing at a table,while the control group was trained while sitting.Before the treatment,as well as after 2 and 4 weeks of treatment,both groups' motor functioning was evaluated using the Fugl-Mayer upper limb assessment (FMA),as well as muscle surface electromyography.Results Before the treatment there was no significant difference betweenthe two groups' average FMA scores.After 2 and 4 weeks of treatment it had increased significantly in the experimental group,but in the control group the increase was not significant until the fourth week.In terms of surface myography,significant differences were observed in the biceps femoris and gastrocnemius muscles of both groups after 2 weeks.Two weeks later there was further significant improvement in both groups except for the tibialis anterior muscles of the control group.The differences between the two groups were significant after two weeks in the electromyograms of the biceps femoris,gastrocnemius muscle and anterior tibialis.After four weeks the differences between the groups in all of the electromyograms were significant.Conclusion Compared with the traditional sitting position,standing at a table while training can effectively improve the muscle activity of the rectus abdominis and the spine so as to promote the recovery of movement in hemiplegic upper limbs.
8.Risk factors of recurrence after ultrasound-guided thermal ablation for treating uremia secondary hyperparathyroidism

Objective To observe the risk factors of recurrence after ultrasound-guided thermal ablation for treating uremia secondary hyperparathyroidism(SHPT).Methods Totally 59 patients with uremia SHPT who underwent ultrasound-guided thermal ablation were enrolled,including 23 cases with(relapse group)and 36 without SHPT recurrence(non relapsed group).Clinical data were compared between groups,univariate and multivariate logistic regression analysis were performed to screen independent risk factors of SHPT recurrence.Results There were significant differences of serum free thyroxine(FT4),urea,intact parathyroid hormone(iPTH)1 day after ablation,1 day decrease rate of iPTH,the maximum diameter of the largest nodule,ablation time,total ablation energy,energy to volume ratio and the proportion of 1 day decrease rate of iPTH≤90%between groups(all P<0.05).Higher urea,lower energy to volume ratio and 1 day decrease rate of iPTH≤90%were all independent risk factors of SHPT recurrence(all P<0.05).Conclusion Higher urea,lower energy to volume ratio and 1 day decrease rate of iPTH≤90%were independent risk factors of recurrence after ultrasound-guided thermal ablation for treating uremia SHPT.
