Objective To study the resistance status of helicobacter pylori (HP) to commonly used antibacterial drugs as well as clinical effects of quadruple therapy in the patients with chronic obstructive pulmonary disease (COPD) infected by Hp.Methods A total of 180 cases of patients with Hp infection in the inpatient department and outpatient department of our hospital from August 2012 to December 2014 were collected, including 90 cases of COPD (COPD group) and 90 cases of non-COPD (control group). The gastric antrum mucosal tissues in all cases were taken for conducting Hp culture and identification. The resistance situation to clarithromycin, metronidazole, amoxicillin, levofloxacin and furazolidone in each group was observed. In addition, the two groups were equally divided into subgroup A,B and C according to different medication schemes:which were given 3 kinds of quadruple medication scheme containing amoxicillin and clarithromycin (group A),amoxicillin and levofloxacin (group B) and amoxicillin and furazolidone (group C).Then the clinical effects were compared. Results The resistance rate of infected Hp strains to clarithromycin in the control group was 21.11%;which to metronidazole was 55.56%;which to amoxicillin was 3.33%;which to levofloxacin was 5.56%,while which in the COPD group were in turn 26.67%,57.78%,7.78% and 16.67% respectively. No strain was found to be resistant to furazolidone in the two groups. There was statistically significant difference in the resistance to levofloxacin and double antibiotic resistance between the two groups (P<0.05).The eradication rates for using 3 kinds of quadruple therapy in 3 subgroups of control group were 90.0% in the group A,96.7% in the group B and 100.0% in the group C,while which in the COPD group were 86.7%,80.0% and 96.7% respectively;the Hp eradication rate for adopting the group B scheme had statistical difference between the two groups (P<0.05).Conclusion The resistance rate of COPD patient affected by Hp to levofloxacin has an increasing trend,so its use should be cautious. The quadruple scheme containing amoxicillin+furazolidone can serve as the preferred scheme for the initial Hp eradication therapy in COPD patients affected by Hp.

In this article, an HPLC method for the contents determination of amino acids in Qihong Maitong injection was reported. In detailed, OPA-Fmoc pre-column derivatization was adopted, and related HPLC methods to determine the contents of amino acids was established. Linear relationship was well constructed for 17 amino acids through the method mentioned above. Briefly speaking, the optimized method was accurate and reproducible, and suitable for the determination of amino acids in Qihong Maitong injection and corresponding quality control.

BACKGROUND: The growth behaviors of cells on the biomaterials scaffold may be affected by the topography, pore size, wettability, porosity and other factors.OBJECTIVE: This research is aimed to observe the growth and proliferation of Schwann cells in silk fibroin scaffolds with different pore sizes. METHODS: Two kinds of silk fibroin scaffolds with different pore sizes were prepared, including a large pore size scaffold (pore size 50-60 μm) and a small pore size scaffold (pore size 10-20 μm). Schwann cells (R3 [33-10ras3]) served as seed cells and incubated in 37 ℃, 5% CO2 incubation box. When cells filled up the culture bottle bottom and formed a dense monolayer, they were digested and the cell concentration adjusted, then Schwann cells were seeded onto the surface of the porous silk fibroin scaffolds with different pore sizes. After seven days of co-culture, the growth and proliferation of Schwann cells were observed under scanning electron microscope.RESULTS AND CONCLUSION: The growth of Schwann cells on the surface of silk fibroin scaffolds with different pore sizes was varied. On the surface of small pore size scaffold (10-20 μm), the cell density was low, while the phenotype of cells was bipolar, cells arranged in parallel or linked as the cell chains. On the surface of large pore size scaffold (50-60 μm), more cells could be seen, but most of the cells were in the shape of single sphere, cells clustered on the surface of the porous scaffold or aggregated as a bunch of grape at the bottom of pores. Only few cells were bipolar and lied on the ridge between the pores. The result showed that the pore size of porous silk fibroin scaffolds is an influential factor for the growth and adhesion of Schwann cells. Schwann cells are conducive to grow on the scaffolds with pore size larger than cell body diameter.

Objective To investigate the expression and prognostic significance of CD151, c-Met and integrin alpha 3, alpha6 in pancreatic ductal adenocarcinoma (PDAC). Methods The expression of CD151, c-Met and integrin alpha3, alpha6 in 71 patients with PDAC and 10 samples of normal pancreas tissues were detected by immunohistochemistry, and the relationship between the expression of CD151, c-Met and integrin alpha 3, alpha 6 and the clinicopathological features, prognosis of these patients was analyzed. Results The positive expression rates of CD151, c-Met and integrin alpha 3, alpha 6 in PDAC were 81.69% (58/71) , 69.01% (49/71), 69.01% (49/71) and 84.51% (60/71) , and there was no expression in normal pancreas tissues. The expressions of CD151, c-Met were significantly associated with TNM stage and lymph node metastasis (P < 0.05). The expression of CD151 was positively correlated with the expressions of c-Met and integrin alpha3, alpha6 (r =0.583, P =0.000, r = 0.457;P =0.000, r = 0.671 ;P =0.000). Univariate analysis suggested the expression of CD151, c-Met and integrin alpha3, alpha6 was associated with survival (P<0.05). Multivariate analysis suggested the expression of CD151, c-Met was the independent prognostic factor for post-operative survival. Conclusions CD151, c-Met and integrin alpha3, alpha6 play a role in the development, metastasis and prognosis of PDAC, and they might be new markers to predict biological behavior and the prognosis of PDAC patients.

Antigen Rv2628 of Mycobacterium tuberculosis is associated with latent tuberculosis infection. In this study, Rv2628 was prokaryotic expressed and purified, its immunological characteristics was evaluated with macrophage cell line RAW264.7 and BALB/c mice. The results show that Rv2628 was mainly expressed in form of inclusion body confirmed by SDS-PAGE, and could react with rabbit anti-H37Rv polyclonal antibody detected by Western blotting assay, indicating that the protein had an effective immunoreactivity. The interactions between Rv2628 and macrophage cell line RAW264.7 confirmed that it could effectively induce cells to produce pro-inflammatory cytokines, the relative expression level of IL-6 mRNA was higher than the control group in 1-12 h. BALB/c mice were subcutaneously immunized with Rv2628 protein, the production of IFN-gamma and IL-4 in the spleen cells was determined by Sandwich ELISA, in the Rv2628 immunized group, the level of IFN-gamma was significantly higher than that of IL-4 (P < 0.000 1). It indicated the protein induced Th1-tendency immune responses. At the same time, Rv2628(11-30) peptide used as coating antigen, the murine serum antibody titer detected by indirect-ELISA was 1:1 600, which demonstrated that Rv2628 could also induce humoral immune responses. In summary, Rv2628 could induce specific pro-inflammatory cytokines, affectively induce strongly Th1-tendency immune response and humoral response, it could be a potential target for developing subunit vaccine against TB. In addition, it laid foundation for probing the cross-talk between M. tb and host.
