2.Effect of astragalus complanatus on liver free radical metabolism in rats of exercise training

Objective To study the effect of astragalus complanatus on the metabolism of free radicals in liver and exercise performance in models of exhaustive training rats.Methods After 7 d adaptive training,we randomly divided rats into three groups: quiet control group(A),sport training group(B) and astragalus complanatus training group(C).We measured rat liver indices such as the activities of total superoxide dismutase(T-SOD),glutathione peroxidase(GSH-Px),catalase(CAT) and content of malonaldehyde(MDA).Results The activity of astragalus complanatus groups(C) T-SOD(78.17?0.17),GSH-Px(35.42?1.68) and CAT(17.61?0.05) in liver tissue increased and the content of MDA(6.70?0.53) decreased remarkably compared with that in sport training group(B).Conclusion As an efficient free radical scavenger,astragalus complanatus can strongly enhance anti-oxidative status and improve exercise performance in trained rats.
3.Effect of quercetin on hemoglobin and blood biochemical indexes in rats following exhausted exercise

Totally 40 male SD rats were randomly divided into sedentary group (G1, n =8), exercise group (G2, n =16) and exercise with drug group (G3, n =16). Animal models were established by treadmill training. At the last time for training, rats in the G2 and G3 were separately divided into control group (n =8) and exhausting exercise group (n =8). Rats in the G3 were orally injected with quercetin 50 mg/kg by intragastric administration everyday at 3 hours before training, and others in the G1 and G2 were fed with the same amount of saline by gavage. Hemoglobin (Hb) content, activities of serum lactate dehydrogenase (LDH), alanine aminotransferase (ALT) and aspartate aminotransferase (AST) were determined in exhausted rats. The results showed that quercetin could remarkably restrain the decrease of Hb content and the increase of activities of ALT, AST and LDH after exhausted exercise.
4.EFFECT OF HIPPOPHAE RHAMNOIDES OIL ON METABOLISM OF FREE RADICALS AND ULTRASTRUCTURE OF MYOCARDIUM IN MICE

Objective: The effect of oil of Hippophae rhamnoides (OHR) supplements on sGOT,metabolism of free radical and ultrastructure of myocardium was observed in mice.Methods: 70 male ICR mice were randomly divided into six groups: sedentary control group (SCG), sedentary+OHR group (SHG), immediately after exhaustive exercise in control group (IEC) and in OHR group (IEO), 24 hours recovery after exhaustive exercise in control group (REC) and in OHR group (REO). After 6 w swimming program with gradually increasing intensity, these mice carried out an exhaustive swimming before sacrifice. Serum GOT, SOD and MDA in myocardium were measured and the changes of ultrastructure of myocardium were observed by transmission electron microscope (TEM). Results: 1) Serum GOT was significantly different among these groups except between SCG and SHG, IEC and REC. 2) SOD was significantly different among the above groups except between SCG and SHG, IEO and REO. 3) The concentration of MDA was significantly different among the above groups except between IEC and IEO, REC and REO. 4) The integrity of mitochondrion, rough surfaced endoplasmic reticulum and myofilament in myocardium was better in OHR groups than in control groups and the pathological tendency of these ultrastructures was lighter in OHR groups compared with control groups. Conclusion: OHR supplementation can reduce the damage of myocardium caused by exercise and enhance antioxidative status of myocardium; OHR supplementation can protect the ultrastructure of myocardium.
5. Effects of aerosol inhalation of recombinant human keratinocyte growth factor 2 on the lung tissue of rabbits with severe smoke inhalation injury

Objective:
To investigate the effect of recombinant human keratinocyte growth factor 2 (rhKGF-2) on lung tissue of rabbits with severe smoke inhalation injury.
Methods:
A total of 120 New Zealand rabbits were divided into 5 groups by random number table after being inflicted with severe smoke inhalation injury, with 24 rats in each group. Rabbits in the simple injury group inhaled air, while rabbits in the injury+phosphate buffer solution (PBS) group inhaled 5 mL PBS once daily for 7 d. Rabbits in injury+1 mg/kg rhKGF-2 group, injury+2 mg/kg rhKGF-2 group, and injury+5 mg/kg rhKGF-2 group received aerosol inhalation of 1 mg/kg, 2 mg/kg, and 5 mg/kg rhKGF-2 (all dissolved in 5 mL PBS) once daily for 7 d, respectively. On treatment day 1, 3, 5, and 7, blood samples were taken from the ear central artery of 6 rabbits in each group. After the blood was taken, the rabbits were sacrificed, and the tracheal carina tissue and lung were collected. Blood pH value, arterial oxygen partial pressure (PaO2), arterial blood carbon dioxide pressure (PaCO2), and bicarbonate ion were detected by handheld blood analyzer. The expressions of pulmonary surfactant-associated protein A (SP-A) and vascular endothelial growth factor (VEGF) in lung tissue were detected by Western blotting. Pathomorphology of lung tissue and trachea was observed by hematoxylin-eosin staining. Data were processed with analysis of variance of two-way factorial design and Tukey test.
Results:
(1) Compared with those in simple injury group, the blood pH values of rabbits in the latter groups on treatment day 1-7 had no obvious change (
