1.Effect of myricetin on migation and invasion ofhepatoma SMMC-7721 cells

Aim To investigate the effects of myricetin on the migration and invasion of hepatoma SMMC-7721 cells.Methods SMMC-7721 cells were treated with different concentrations of myricetin.Migration and invasion of SMMC-7721 cells were examined by Wound healing and Transwell, and the expression of E-cadherin and N-cadherin was detected by RT-qPCR and Western blot.Results Myricetin may inhibit the viability of SMMC-7721 cells via different concentration(10~40 μmol·L-1);furthermore, myricetin might also inhibit the migration and invasion of hepatoma SMMC-7721 cells.Meanwhile with the increasing of myricetin concentration, both filopodia and lamellipodia formation was reducd, and 7721 cells displayed in more integrity.And data from RT-qPCR and Western blot demonstrated that myricetin may up-regulate the expression of E-cadherin, simultaneously, down-regulate N-cadherin in SMMC-7721 cells.Conclusion Myricetin may influence the cell migration and invasion through up-regulating the expression of E-cadherin, simultaneously, down-regulating N-cadherin and activate cytoskeleton remolding of SMMC-7721.
2.Percutaneous vertebroplasty versus percutaneous kyphoplasty for the treatment of old vertebral osteoporotic compression fracture

BACKGROUND:Pereutaneous vertebroplasty and percutaneous kyphoplasty as the minimal y invasive treatment techniques have been widely used in the treatment of vertebral osteoporotic compression fracture, but there are controversies, and each method has its advantages and disadvantages.
OBJECTIVE:To compare the clinical efficacy between percutaneous vertebroplasty and percutaneous kyphoplasty in the treatment of vertebral osteoporotic compression fracture.
METHODS:100 patients with vertebral osteoporotic compression fracture were col ected, among them, 40 patients received percutaneous vertebroplasty, and 60 patients received percutaneous kyphoplasty. The visual analog scale score was used to assess the pain degree of the patients before treatment;the visual analog scale score was also used at 1 week after treatment. The X-ray film was taken to measure the ratio between anterior and rear edges of vertebrae.
RESULTS AND CONCLUSION:The visual analog scale scores in the two groups were significantly decreased after treatment, and there was significant difference when compared with that before treatment (P<0.05);there was significant difference in the restore of vertebral height in two groups when compared with that before treatment, but there was no significant difference in the orthopedic effect between two groups (P<0.05). The incidence of complications of bone cement leakage after treatment was 35%in the pereutaneous vertebroplasty group and 12.5%in the percutaneous kyphoplasty group, and there was significant difference in the incidence of complications between two groups (P<0.05). Pereutaneous vertebroplasty and percutaneous kyphoplasty can significantly relieve the pain of the patients with vertebral osteoporotic compression fracture, but there is no significant difference in the orthopedic effect between pereutaneous vertebroplasty and percutaneous kyphoplasty.
3.Effect of Compound Rhubarb Mixture on contraction of isolated small intestinal smooth muscle of rabbit

OBJECTIVE: To investigate the effect of Compound Rhubarb Mixture on contraction of isolated small intestinal smooth muscle of rabbit. METHODS: Serum containing Compound Rhubarb Mixture was prepared. The contraction curves of isolated small intestinal smooth muscle before and after administration of the serum were recorded by Medlab biological information collecting system. RESULTS: The serum containing Compound Rhubarb Mixture amplified the contraction amplitude of isolated small intestinal smooth muscle of rabbit. The rate of change of contraction amplitude was elevated significantly after administration, while the frequency of contraction did not change obviously. CONCLUSION: The serum containing Compound Rhubarb Mixture can enhance the contractive function of isolated small intestinal smooth muscle of rabbit.
4.Effect and mechanism of emodin on intestinal movement

AIM To examine the effect and mechanism of emodin on intestinal movement of constipation model. METHODS The movement of charcoal powder and the absorption of D-xylose into serum were observed. The electrical p otential difference(PD) related to Na + and glucose transport was measured across the wall of rev erted intestinal sacs. RESULTS Emodin 0 1, 0 2 g?kg -1 incr eased markedly the rate of charcoal powder moving and inhibited the absorption of D-xylose into serum. Emodin 0 2, 0 4, 0 8 and 1 6 g?L -1 decreased PD when ther e was glucose. However, emodin had little effect when glucose was free. CONCLUSION Emodin promotes intestinal movement and this effect is related to Na +,K +-ATPase.
5.Expressions of octamer-binding transcription factor 4 and phosphorylated-protein kinase B in colorectal cancer and their clinical significances

Objective To investigate the expressions of octamer-binding transcription factor 4 (Oct4) and phosphorylated-protein kinase B (p-Akt) proteins in colorectal cancer and their clinical significances, in order to explore the roles of Oct4 and p-Akt in the staging and grading of colorectal cancer. Methods Immunohistochemical technique was used to examine the expression of Oct4 and p-Akt proteins in 78 cases of colorectal cancer in Wuxi Second Hospital of Traditional Chinese Medicine and Wuxi First People's Hospital from January 2011 to December 2016. Relationship between expressions of Oct4 and p-Akt proteins and clinicopathological parameters were analyzed. Results The positive rate of Oct4 in tumors was 74.36 % (58/78), which was obviously higher than that in normal tissues [35.90 % (20/78), χ2= 23.32, P < 0.01]; and the positive rate of p-Akt in tumors was 67.95 % (53/78), which was obviously higher than that in normal tissues[28.21 %(25/78),χ2=24.68,P <0.01].The double positive and negative expression rate of these two proteins accounted for 80.8 %(63/78), with a linear positive correlation (r= 0.455, P < 0.000 1). In 78 cases of colorectal cancer, the expression of Oct4 protein was correlated with histological grade, lymph node metastasis, and Duke staging (all P < 0.05), and the expression of p-Akt protein was correlated with histological grade and lymph node metastasis (both P < 0.05). The multivariate logistic regression analysis showed that the expression of Oct4 protein was related to histological grade and Duke staging(both P<0.05),and the expression of p-Akt protein was only related to lymph node metastasis (P<0.05). Conclusion The combined detection of Oct4 protein and p-Akt protein has reliable and important clinical significance for judging the histological grade,lymph node metastasis and Duke staging of colorectal cancer.
6.Effect of Gubitongxiao granules in a mouse model of steroid-induced necrosis of the femoral head

BACKGROUND:Glucocorticoids can inhibit the expression of hub genes in the parathyroid hormone type Ⅰ receptor(PTH1R)/protein kinase A(PKA)signaling axis and interfere with the osteogenic and angiogenic differentiation of bone marrow mesenchymal stem cells,leading to the disruption of blood supply in bone and bone tissue structures.Previous studies of the research team showed that Gubitongxiao granules can induce blood vessel formation and inhibit osteoblast apoptosis,which has a certain effect on the prevention and treatment of steroid-induced femoral head necrosis. OBJECTIVE:To observe the therapeutic effect of Gubitongxiao granules in a mouse model of steroid-induced femoral head necrosis,and to explore its mechanism from the PTH1R/PKA signaling axis. METHODS:An animal model of steroid-induced necrosis of the femoral head was established by intraperitoneal injection of lipopolysaccharide and gluteal muscle injection of prednisolone acetate.After identification by nuclear magnetic resonance method,60 mice that were successfully modeled were divided into model group,Gubitongxiao granule group and Tongluo Shenggu capsule group,with 20 mice in each group.Another 12 normal mice were used as control group.The corresponding groups were intragastrically given the corresponding drugs for 12 weeks,and then the samples were taken under anesthesia.Histomorphology of femoral head samples was observed by hematoxylin-eosin staining.Enzyme-linked immunosorbent assay was used to detect the serum levels of bone alkaline phosphatase,type Ⅰ amino-terminal extension peptide,parathyroid hormone,osteocalcin and alkaline phosphatase.Western blot and RT-qPCR were used to detect PTH1R,PKA,myocyte enhancer factor 2,sclerostin and guanylate-binding protein activity-stimulating peptide at protein and gene expression levels,respectively. RESULTS AND CONCLUSION:Gubitongxiao granules may reduce the serum PTH level in mice,inhibit the activation of the PTH1R/PKA signal axis,further up-regulate the protein expressions of sclerostin and myocyte enhancer factor 2,and increase the levels of bone alkaline phosphatase,type Ⅰ amino-terminal extension peptide,osteocalcin and alkaline phosphatase in mice,thus improving femoral head necrosis,which is comparable to the intervention effect of Tongluo Shenggu capsules.It is speculated that Gubitongxiao granules may prevent and treating hormonal femoral head necrosis by regulating the PTH1R/PKA signaling axis.
7.Preliminary study on polymerized human cord hemoglobin increasing the chemosensitivity of subcutaneous xenografts of breast cancer in nude mice

【Objective】 To investigate the effect of polymerized human cord hemoglobin (PolyCHb) on the chemosensitivity of human breast cancer MCF-7 cell subcutaneous xenografts in nude mice and its mechanism. 【Methods】 The MCF-7 cells in exponential growth phase were collected and made into suspension cells at a density of 5×107 cells/mL.Subsequently, the cells were inoculated subcutaneously in the right limb of 18 BALB/c-nu nude mice with 0.2 mL cells per mouse to establish subcutaneous xenograft.When the tumor volume reached about 100 mm3, they were randomly divided into chemotherapy group: doxorubicin 5 mg·kg-1, once/week; chemotherapy + PolyCHb group: in addition to doxorubicin (chemotherapy group), PolyCHb 600 mg·kg-1, 3 times/week; the control group: normal saline 90 mg·kg-1, once/week; all were injected through tail vein continuously for 4 weeks.From the day of injection (d 0), the tumor volume of each group of nude mice was measured every 3 days, and the tumor growth curves were drawn accordingly.After 38 days, the tumor growth observation was completed.The tumor was removed and weighed to calculate the tumor inhibition rate.HE staining, immunohistochemistry and TUNEL method were used to observe the pathological changes of tumor tissue, detect the expression of HIF-1α, and detect tumor cell apoptosis respectively.The content of reactive oxygen species (ROS) of each group was determined by fluorescence staining. 【Results】 The tumor volume (mm3) of chemotherapy + PolyCHb group, chemotherapy group and the control group at day 38 were 196.35±103.45 vs 316.29±62.88 vs 519.42±177.33 (P<0.05), and the tumor inhibition rate (%) of chemotherapy + PolyCHb treatment group and chemotherapy group was 62.20 vs 39.11, respectively.HE staining and TUNEL detection showed that cell necrosis and apoptosis in the growth area of tumor tissue increased in chemotherapy + PolyCHb group.Immunohistochemistry and fluorescence staining showed that HIF-1α expression in chemotherapy + PolyCHb group decreased and reactive oxygen species (ROS) content increased. 【Conclusion】 PolyCHb increases the chemosensitivity of subcutaneous xenograft in nude mice with breast cancer, and its mechanism may be related to the increase of ROS in tumor tissue and the promotion of tumor cell apoptosis.

9.Preparation technology of platelet-rich-plasma lysate combined with hyaluronic acid microneedles and wound healing effect

【Objective】 To investigate the application prospect of platelet-rich plasma lysate (PL) microneedles in the treatment of wound healing by optimizing the preparation process and evaluating the activity of platelet growth factor. 【Methods】 Firstly, hyaluronic acid (HA) was used as needle matrix material, and the concentration of HA was preliminarily determined by investigating the fluidity of HA solution, shape of needle, pressure change performance and puncture performance.Combined with the results of HA dissolution in PL, mechanical properties of PL microneedles and skin puncture, the preparation process of platelet-rich plasma lysis microneedles (PLMN) was optimized to investigate the growth factor activity of PLMN and evaluate the effect of PLMN on wound healing in rats. 【Results】 When the concentration of HA was 30 mg/mL, the solution had good fluidity.The HA microneedles could pierce three layers of sealing film, with the puncture rate at 70%, and the solution had good mechanical properties.When HA was dissolved in PL at a concentration of 30 mg/mL at 4℃, the solution had no crystallization and was with good fluidity.The prepared PLMN showed good needle shape and no bending under HD microscope and scanning electron microscope.In vitro transdermal simulation results showed that PLMN could penetrate 4 layers of sealing membrane, and the puncture rate of the first three layers was more than 85%.In vivo lysis experiment showed that PLMNs could be completely dissolved within 10 min after being inserted into the body.Forty minutes later, the micropores left after the insertion of the microneedle were almost closed, without redness, swelling, erythema and other adverse reactions.In vitro and in vivo transdermal experiments showed that clear blue holes were visible after PLMNs penetrated the skin stain, indicating that the microneedles could pierce the skin barrier.Growth factor detection results showed that the content of platelet-derived growth factor (PDGF-BB) and transforming growth factor (TGFβ1) in platelet lysate were (17.67±1.23) ng/mL and (105.95±2.16) ng/mL, respectively.After PLMNs were prepared and stored at room temperature for 1 week, the content of PDGF-BB(ng/mL) and TGFβ1(ng/mL) were 15.48±2.12 and 56.26±1.53, respectively.Compared with PL(vs 17.67±1.23 and 105.95±2.16), the content of PDGF-BB and TGFβ1 decreased, but still maintained the activity of growth factors.The results of wound healing experiment in rats showed that the wound healing rate (%) of PLMN group on the third day was significantly different from that of the control group(52.51±6.15 vs 33.31±3.60, P<0.05). 【Conclusion】 The PLMN prepared in this experiment has good mechanical properties and can successfully pierce the skin cuticle.It can also maintain the activity of growth factors, and has a good effect on wound healing in rats, which provides a new idea for minimally invasive treatment in the field of tissue repair.
