1.Arthroscopic synovectomy in chronic synovitis of the knee in patients with hemophilia A

[Objective] To introduce a method of arthroscopic synovectomy in chronic synovitis of the knee in patients with hemophilia A and to evaluate its clinical value.[Method]Arthroscopic synovectomy was performed in 25 patients with hemophilia A from Sept.2002 to Jan.2007.Sufficient factor replacement was used and systematic rehabilitation exercise was emphasized.HSS scores were used to evaluate the efficiency of the operation.Insall standard was used to determine the difference between groups according to Outerbridge grading of the cartilage.[Result]Twenty-three patients were followed up.The mean period of follow-up was 27 months.The average frequency of hemarthrosis reduced from 23.4 times per year preoperatively to 9.8 times per year postoperatively(P
2.Comparative study of femoral tunnel enlargement after hamstring anterior cruciate ligament reconstruction with different graft fixations

[Objective]To compare the changes in femoral bone tunnel after arthroscopically assisted hamstring ACL reconstruction with different graft fixations and investigate the relation with clinical effect.[Method]Eighty-eight cases in three groups were included and underwent hamstring ACL reconstruction with different femoral graft fixations for average 16.5 months follow-up evaluation.Twenty-six cases in group A received Endobutton fixation,twenty-two cases(group B) received interference screw,and forty cases(group C)received Rigidfix system.Femoral bone tunnel diameters at the articular line level,the widest point,1 cm away from tunnel aperture on sagittal MRI were compared at one week postoperatively.Lysholm score was used to evaluate the clinical effect.[Result]Tunnel enlargement after ACL surgery was noted in the three groups.At the femoral widest point and 1 cm away from tunnel aperture,the tunnel enlargement was larger in group B and smaller in group C.At the femoral articular line level,the bone tunnel enlargement in group C was smallest.There was no difference between those of groups A and B.The shape of femoral tunnel was lineal in groups A and C,and conical in group B.No significant difference was found by Lysholm score.[Conclusion]The type of graft fixation is a major factor in the development of tunnel enlargement after ACL surgery.No correlation between graft fixation,bone tunnel enlargement and clinical effect has been found.
3.A comparative study of tunnel enlargement after anterior cruciate ligament reconstruction with autologous and allogenic tendons

Objective To investigate the changes of bone tunnel and differences of clinical outcomes after anterior cruciate ligament (ACL) reconstruction with hamstring tendons from autografts and allografts. Methods The study involved in 61 patients with ACL injury undergone arthroscopic ACL reconstruction from June 2008 to November 2009.According to ACL graft differences,61 patients were assigned to two groups,ie,Group A ( reconstruction with tendon allografts,n =27 ) and Group B ( reconstruction with tendon autografts,n =34).MRI examination was performed one week and 6-12 months post-operatively to measure the sagittal bone tunnel diameters at the aperture location,at the location of 1cm away from the aperture and at the widest location of tunnel.Taking the bone tunnel diameter at each location one week post-operatively as the measurement criteria,the differences of bone tunnel diameters at each corresponding location were analyzed statistically.Lysholm score was used to evaluate the clinical effects in the two groups. Results Group A and B were followed up for average 8.4 months and 8.5 months,respectively.Pre-operative clinical data of the two groups had no significant difference.MRI follow-up showed that the tunnel diameters at femoral side and tibial side both had different degree of enlargement.Meanwhile,the enlargement degree at the tibial side was larger than that at the femoral side,with no statistical differences of the two groups regarding tunnel enlargement of the three measurement locations at femoral and tibial sides.Joint stability in all patients was good.The two groups showed no significant difference in Lysholm score after operation ( P > 0.05 ). Conclusions Graft difference is a factor for bone tunnel enlargement after ACL surgery.Autologous and allogenic tendons show no difference in their role in bone tunnel enlargement after ACL surgery or correlation with clinical effects.
4.A Survey on Present Situation of Intelligence among Boarding School-age Children of the Va Nationality

Objective To analyze the present situation of intelligence of boarding school-age children of the Va nationality.Methods Multi-stage random sampling method was used to select 650 boarding school-age children from 10 primary schools in ShuangJiang county, and their level of intelligence was estimated by Raven's STANDARD progressive Matrices (SPM) .Results The level of intelligence of boarding school-age children of the Va nationality was significantly lower than the norm ( <0.05), the prevalence of Mental Deficiency was 19.9%, and it didn't show significant differences in different age and gender ( >0.05) .Conclusions The present situation of intelligence of boarding school-age children of the Va nationality is serious and should be improved as quickly as possible.
5.Investigation on Dietary Nutrition Status of 115 Patients with Graves Disease in Yunnan Province

Objective To learn dietary and nutritional status of the toxic diffuse goiter (GD) patients in Yunnan region,to investigate dietary nutrition intake levels of GD patients.Methods By using food frequency method and self-programmed food frequency table,GD patients' dietary nutrition was investigated.We calculated GD patients' intake of all kinds of food and a variety of nutrients per capita per day,and compared them with the suggested values in Chinese residents balanced diet pagoda and dietary nutrients reference intakes (RNIs);measured the patients' body weight,height and calculating body mass index in the field.Results (1) Dietary structure:GD patients' food intake kinds were various in Yunnan region,the intakes of cereals,tuber crops,meat,poultry,oils and fats and nuts were high;the intakes of vegetables,homonemeae,fruits,eggs,fish and shrimp,milk and milk products were low.(2) Energy and nutrient intake:the energy contribution from fats exceeded the recommended value,the energy contribution from carbohydrates was lower than the recommended value,the intakes of dietary fiber,vitamin A,thiamine,riboflavin,calcium and sodium were low,the intakes of vitamin E,phosphorus and iron were high.(3) Physical examination:GD patients' marasmus rate,overweight rate and obesity rate were 25.2%,7.0% and 1.7%,respectively.There was no statistically significant difference between males and females (P>0.05) Conclusion GD patients' food intake kinds are various in Yunnan region,but the dietary structure is unreasonable.
6.The effect of repetitive transcranial magnetic stimulation on the changes of depressive like behaviors in chronic unpredictable mild stress model rats and its possible mechanism

Objective To investigate the effect of repetitive transcranial magnetic stimulation (rTMS) on the depressive like behaviors and expression of brain derived neurotrophic factor (BDNF),IL-1β and NF-κB of hippocampal in chronic unpredictable mild stress (CUMS) rats.Methods Thirty-two adult male rats were randomly divided into four groups (n =8):Control group,Control + rTMS group,CUMS group and CUMS + rTMS group.The sucrose preference test,forced swim test and open field test were used to evaluate depressive like behaviors for each groups.In addition,the expression of BDNF,NF-κB and IL-1 β in hippocampal were detected by western blot and ELISA after behavioral test,respectively.Results 1.The effects of rTMS on depressive like behaviors of CUMS rats:in the sucrose preference test,the sucrose preference rate of CUMS rats (0.67 ± 0.06) was significantly lower than Control group (0.91 ± 0.04),which was higher in the CUMS + rTMS group (0.83 ±0.08).In the forced swim test,the immobility time of CUMS group ((26.88 ± 11.33) s) was longer than Control group ((15.22 ± 6.75) s) and CUMS + rTMS group ((18.41 ± 6.95) s).In the open field test,both the total distance travelled and number of central area entry times of CUMS group((849.165 ± 769.01) cm,(7.42 ± 5.68))were significantly shorter ((6224.81 ± 1403.2) cm) and smaller (22.86 ± 3.72) than Control group,and those of the CUMS + rTMS were longer ((4105.57 ± 1516.92)cm) and larger (21.25 ± 3.45).All the behavioral results were statistically significant (P< 0.05).And of all the aforementioned behavioral parameters,there were no significant differences between Control group and Control + rTMS group(P>0.05).2.The effects of rTMS on the hippocampal expressions of BDNF,NF-κB and IL-1β in CUMS rats:compared with Control group,the hippocampal expression of BDNF in CUMS rats was significantly decreased,while the expressions of NF-κB and IL-1β in the hippocampus were significantly increased (P< 0.05).Compared with CUMS group,the hippocampa expression of BD-NF in the CUMS + rTMS group was increased,and the expressions of NF-κB and IL-1β in the hippocampus was significantly decreased (P < 0.05).The expressions of BDNF,NF-κB and IL-1β had no differences between Control group and Control + rTMS group.Conclusion rTMS increased the expression of BDNF,reduced the production of NF-κB and IL-1β,and alleviated depressive like behaviors in CUMS rats.
7.Effects of sertraline on the cell viability and expression of tyrosine hydroxylase and phosphorylated ERK1/2 in NGF-induced PC12 cells

Objective To investigate the effect of sertraline on the viability and the expression of tyrosine hydroxylase (TH) and phosphorylated ERK1/2 in NGF-induced rat pheochromocytoma (PC12) cells.Methods NGF-induced PC12 cells were pretreated or directly treated with different concentrations of sertraline for 24 or 48 hours and the pretreated groups were then subjected to serum withdrawal condition. Then cell viability was determined by the cell counting Kit-8 (CCK-8). The expression of Tyrosine hydroxylase (TH) and pERK1/2in NGF-induced PC12 cells was determined by immunohistochemistry and western blot respectively. Results The viability of NGF-induced PC12 was improved after administration with sertra]ine. After 24h sertraline administration, the cells activity of PC 12 cells at 20μM ( 1.32 ± 0. 11 ) , 10μM ( 1. 17 ± 0.05 ) of direct effect, and 20μM ( 1.15 ±0.11 ) of protect effect increased dramatically as compared with control group. But high dose ( 50μM )sertraline express high toxic effect to PC12 cells. The expression of TH was increased by sertraline 20 μM at both 24h(ratio of TH/β-actin = 1.27 ±0.05) and 48h(ratio of TH/β-actin = 1. 23 ±0.08) compare with control group,and the expression of pERK1/2 also increased dramatically by sertraline 20 μM at both 24h (ratio of (pERK1/2)/β-actin = 1.41±0.05) and 48h( ratio of (pERK1/2)/β-actin = 1.40 ±0.06) compare with control group(P<0. 01, P < 0. 05). Immunohistochemistry showed similar results. Conclusion These data suggest that the neuroprotective effect of sertraline may play an important role in depression therapy, and this effect might be mediated by TH and pERK1/2 up-regulation.
8.The protective effects of gastrodin on the brain injury and the integrity of myelin in striatum after cerebral ischemia-reperfusion in mice

Objective To investigate the effects of gastrodin on neural function and the expression of myelin basic protein (MBP) and neurofilament high molecular weight (NFH) in the striatum during cerebral ischemiareperfusion in mice.Methods 36 Kunming mice were randomly divided into sham group,MCAO group and gastrodin (GAS) group.The middle cerebral artery occlusion(MCAO) was established by artery embolization.The mice in sham group were received fake surgery and saline,and the mice in MCAO and GAS group were exposed to MCAO,and received saline and GAS (100 mg/(kg · d)) injection,respectively,immediately after the operation for 7 days.On the 8th day of operation,the neurological severity scores of the mice were observed and the volume ratio of the cerebral infarction was estimated by triphenyl tetrazolium chloride (TTC) staining.Immunohistochemistry was used to detect the MBP and NF-H in the striatum.Results (1) The mice in MCAO group showed significant neurologic deficient in comparison with sham group,and the neurological severity scores of gastrodin group(3.13±0.64) were significantly higher than that(1.38±0.52) of MCAO group (P<0.05).(2) Results of TTC staining showed that the infarction volume was obviously larger in the injured cerebral tissue in MCAO group in comparison with sham group,and the volume ratio of the cerebral infarction significantly decreased after the intervention with GAS (P<0.05).(3) The integral optical density of MBP(272968.14±1215.23) and NF-H(12 142.73±47.16) in MCAO group decreased as compared to that((43 855.23±2434.16),(275 321.00±926.15)) in sham group and GAS group((321 531.2±2376.14),(106 135.73±598.15)) (P<0.05).Conclusion GAS can improve neural function of mice after middle cerebral artery occlusion,and it may play an important role in protecting myelin and nerve fibers of striatum.
9.Construction of EGFR gene G719S and T790M mutation vector and preliminary clinical application

Objective To construct mutant recombinant vector of epidermal growth factor receptor ( EGFR) gene G719S and T790M sites associated with cervical cancer, lay the foundation for the detection of EGFR gene mutation in cervical cancer. And using it to es-tablish a molecular switch platform to detect cervical cancer EGFR gene mutations. Methods Using the wild-type recombinant plasmid as template, the mutant fusion target fragment were amplified by overlap PCR, then connect this target fragment into the vector pMD19-T. The constructed mutant recombinant plasmid was finally transformed into competent cells E.coli DH5αfurther identified by PCR with bacterial solution and genome sequencing. Establishing the molecular switch for the detection of clinical cervical cancer samples. Re-sults The G719S and T790M mutations were successfully certified by genome sequencing, and the site-directed mutant vector was successfully constructed. In addition, a molecular switch detection platform was also successfully established for the detection of cervical cancer tissue DNA. Conclusion We successfully constructed an EGFR gene mutant recombinant vector by overlap PCR technique, which providing a new technical means for gene site-directed mutagenesis. And the molecular switch detection platform was successfully established based on it, which furnishing a new method for clinical detection of EGFR gene mutations.
10.Association between single nucleotide polymorphism at rs175048 locus of ATM gene and susceptibility to lung cancer in Han population in Hengyang, Hunan

Objective: To explore the association between the single nucleotide polymorphism (SNP) of rs175048 in ataxia telangiectasia mutated (ATM) gene and lung cancer susceptibility in Han population. Methods: A total of 225 cases of blood samples from lung cancer patients treated in Hospital of Traditional Chinese Medicine of Hengyang City and the Affiliated First Hospital of Nanhua University from October 2015 to August 2016 were collected as case group, and 128 cases of blood samples from healthy people were collected as the control. The polymorphisms of ATM rs175048 of above mentioned participants were detected by using the SNP sensitive On/Off Switch technique. The genotypes and allele frequencies were analyzed to compare the distribution difference between case group and control group as well as its association to the clinical features of lung cancer. Results: The genotype frequencies of AA, AT and TT of ATM rs175048 were 24.9%, 52.9%, 22.2% in case group and 42.2%, 42.2%, 15.6% in control group, respectively (all P< 0.01). Moreover, the frequencies of alleles A and T were 51.0%, 49.0% in case group, and 63.0%, 37.0% in control group (all P<0.01). Genotype TT might increase while genotype AT might decrease the risk of lung cancer. rs175048 SNP was significantly correlated with smoking, age, sex and family history (all P<0.05). Conclusion: rs175048 SNPis significantly associated with lung cancer, and TT genotype may increase the risk of lung cancer.
