1.Outcome of donor bone after vascularized allograft bone transplantation

AIM: To observe the outcome of donor bone after vascularized allograft bone transplantation under immunoregulation. METHODS: The experiment was conducted at the Microsurgery Institute, Medical College, Wuhan University from December 2002 to March 2004. Totally 21 male flap-eared Japanese rabbits were selected as donor and 42 female ones as recipients. Recipients were composed of 21 rabbits in the experimental group and 21 rabbits in the control group. ①Anastomotic vascularized massive shaft of femur allograft was performed in Japanese flap-eared rabbits of either sex. ②The experimental animals were assigned into experimental group (treated with ciclosporin A, 25 mg orally, once a day, for 3 weeks) and control group (no any immunosuppressive agent). ③X-ray and histology examinations were performed at months 1, 4 and 9 after operation. Bone grafts were analyzed with Sry semi-quantitative polymerase chain reaction. RESULTS: A total of 42 rabbits were involved in the result analysis. ①X-ray manifestation at different time points after rabbit transplantation in the two groups: X-ray showed typical fracture healing mode at different phases after operation in the experimental group, whereas the increase of bone graft density, and around by a mass of bony callus in the control group. ②Histology manifestation of rabbits in the two groups after operation: At different time phases it showed typical fracture healing mode in the experimental group after operation, and 9 months later the marrow cavity was filled with myeloid tissues, and the internal and external cortex was flat; Haversian canal had revered to the normal size; Bone cells appeared in the bone lacuna; Peripheral cortex was wrapped with fibrous tissues. In the control group, it showed inflammatory cell infiltration with rejection, bone marrow necrosis in marrow cavity, formation of fibra tissues, bone cell loss in bone lacuna, and the bone grafts were wrapped with woven bone that was from recipients. ③Outcome of Sry semi-quantitative PCR analysis: The Sry semi-quantitative PCR analysis showed that the relation between the ratio of male DNA and the postoperative period was positive; the ratio of experimental group at month 9 postoperative was near to 100%, while control group only reached 11.8%. CONCLUSION: Under the immunoregulation, bone graft can unite with the recipient bone in an earlier postoperative period and replace by recipient bone rapidly in vascularized allograft bone transplantation.
2.Content Determination of Residual Organic Solvents in Artesunate by Capillary GC

OBJECTIVE:To establish capillary GC method for the determination of residual organic solvents in artesunate,such as methanol and ethyl acetate.METHODS:Capillary GC was adopted and the temperature of flame ionization detector was 250℃.RESULTS:The linear range were 0.03~0.605 mg?mL-1 for methanol(r=0.999 8) and 0.05~1.002 mg?mL-1 for ethyl acetate(r=0.999 7).The average recovery rates were 98.7% for methanol(RSD=3.0%) and 99.1% for ethyl acetate(RSD=2.1%).Only the ethyl acetate was detected in samples.CONCLUSION:Established method is simple and accurate for the content determination of residual organic solvents in artesunate.
3.Short term efficacy of early postoperative intestinal microecological therapy for gastric cancer

Objective:To study the short-term effect of early application of intestinal microecological therapy after gastric cancer surgery.Methods:A retrospective analysis was made on 96 patients with early and middle stage gastric cancer who underwent surgical treatment in the department of gastrointestinal surgery of Shulan (Hangzhou) Hospital from June 1, 2020 to May 31, 2021. Among them, 48 patients in the observation group were given enteral nutrition support treatment in the early stage after operation and intestinal microecological preparation, while the control group of 48 patients in the early postoperative were given enteral nutrition support. The serum total protein, albumin, prealbumin and other nutrition related indexes and immune related indexes such as CD4 + , CD8 + , CD4 + /CD8 + were detected before and 7 days after operation. The clinical indexes such as the time of first anal exhaust and the incidence of infectious complications were recorded. Results:There was no significant difference between the two groups in preoperative serum total protein, albumin, prealbumin and immune related indexes such as CD4 + , CD8 + , CD4 + /CD8 + ( P>0.05). 7 days after operation, the above indexes in the observation group were significantly higher than those in the control group (all P<0.05). The anal exhaust recovery time of observation group was faster ( P<0.05); There was no significant difference in the incidence of postoperative infectious complications between the two groups ( P>0.05). Conclusions:Early application of intestinal microecological agents in patients with gastric cancer after operation can significantly improve the nutritional status and immune function, promote the recovery of intestinal function, and will not increase the incidence of complications.
4.Acupuncture combined with auricular point sticking therapy for post stroke depression:a randomized controlled trial.

OBJECTIVETo observe the clinical effects of acupuncture combined with auricular point sticking based on the western medication for post stroke depression (PSD).
METHODSSixty patients with PSD were randomly assigned into an acupuncture plus auricular application group (a combination group) and a medication group, 30 cases in each one. 20 mg paroxetine hydrochloride was prescribed orally in the medication group, once a day for continuous 8 weeks. Based on the above treatment, 30-minute acupuncture was used in the combination group for 8 weeks at Baihui (GV 20), Sishencong (EX-HN 1), Shenting (GV 24), Yintang (GV 29), Shenmen (HT 7), Neiguan (PC 6), Taichong (LR 3), Hegu (LI 4), Zusanli (ST 36), Sanyinjiao (SP 6) and Fenglong (ST 40), once the other day and three times a week. Auricular point sticking therapy for 8 weeks was applied at shenmen (TF), pizhixia (AT), xin (CO), and gan (CO), with pressing 3 times a day and once 3-5 days. The total score and each factor scores of Hamilton's depression scale (HAMD) were observed in the two groups before and after treatment, and Asberg's antidepressant side-effect rating scale (SERS) and clinical effect were evaluated.
RESULTSAfter treatment, the total HAMD scores of the two groups decreased compared with those before treatment (both<0.05), with better effect in the combination group (<0.05). The scores of the combination group after treatment were lower than those in the medication group, including the anxiety/somatization factor, sleep disturbance factor, hopelessness factor (all<0.05). The total effective rate of the combination group was 86.7% (26/30), which was better than 66.7% (20/30) of the medication group (<0.05). The SERS score of the combination group was lower than that of the medication group (<0.05).
CONCLUSIONSAcupuncture combined with auricular point sticking can improve the clinical symptoms and are effective and safe for PSD.
