1.Clinical Study on Self-made Wuhuang Tang Combined with Western Medicine for Type Ⅲ Prostatitis

OBJECTIVE:To study the action mechanism of the self-made Wuhuang tang(Chinese medicine)in the treatment of type Ⅲ prostatitis(chronic nonbacterial prostatitis,CNBP)so as to promote the action mechanism of traditional Chinese medicine to the molecular level.METHODS:90 patients with CNBP were divided into two groups(n=45 each):one group received western medicine alone,another group received integrated Chinese and western medicine(ICWM)(Wuhuangtang plus western medicine).The indicators such as the expressed prostatic secretions(EPS)routine,IL-1?,and TNF-? before and after the treatment were analyzed statistically.RESULTS:The levels of IL-1? and TNF-? in ICWM-treated group decreased significantly and the clinical symptoms of these patients improved significantly as compared with those treated with western medicine alone.CONCLUSION:The self-made Wuhuang tang can markedly alleviate the symptoms of CNBP patients and decrease levels of IL-1? and TNF-? in patients' EPS,thus it is an effective clinical empirical prescription for CNBP.
2.EXPERIMENTAL OBSERVATION FOR METABOLIC CHANGES IN GLUCOSE AND LACTIC ACID IN BIPEDICLED SEPARATION DELAYED FLAP

Bi-polar,bi-pedicled separation delayed flaps were designed on the backs of New Zealand rabbits with the application of a medical silicon membrane as the separation material. Values of glucose and lactic acid, PtcO 2 were measured separately at proximal,middle and distal areas of the flaps after the distal pedice was cut at 0,1,4,7,14,21 days,respectively, after the formation of flaps. After flap delaying,PtcO 2 in separated flap was always lower than that of the mormal control,accompanied with glucose decrease and lactic acid increase, although there were significant differences in different areas of the flap. The survived area of the flap increased after 7 days. Furthermore,all the length of the flap survived after 21 days,although the level of glucose in the flap was still lower and lactic acid was still higher.The results suggested that by separation and delaying the flap may adapt gradually to hypoxia via lowered metabolism.For flap survival,the threshold value of glucose in flap is 0 01mmol/L , lactic acid is 0 47mmol/L,and PtcO 2 is 3mmHg .
3.RELATION BETWEEN CHANGE OF CAPILLARY PRESSURE AND SURVIVAL IN BI-PEDICLED SEPARATION DELAY FLAP

Relation between change of capillary pressure, survival length in bi pedicled separation delay flap was observed. Bi polar, bi pedicled separation delay flaps were designed on the backs of New Zealand rabbits. Medical silicon membrane was used as separative material. Values of capillary pressure were measured at proximal, middle and distal points. Survival length increased with delayed time, capillary pressure at middle was lower in the bi pedicled separation delay flap. It is concluded that capillary pressure is increased greatly by separation delay, and there is a linear relation between capillary pressure and survival length of flaps.
4.Relationship Between Complications and Prognosis of Chronic Severe Hepatitis

Objective To explore the relationship between complications and prognosis of chronic severe hepatitis(CSH).Methods The clinical data of 520 patients with CSH treated in our hospital were retrospectively analyzed.Results The most common complications of CSH were ascites,electrolyte disturbance and spontaneous peritonitis,and their total occurring rate was over 50%.Among all the complications,cerebral edema and cerebral hernia had the worst prognosis,failure in treatment and mortality were 100%.Secondary were followed by hepato-renal syndrome,upper digestive tract hemorrhage and hepatic encephalopathy that resulting to effectless rate and mortality of 90% .The effectiveless rate and mortality rose along with increase in number of complications .Meanwhile,the severity of hepatic encephalopathy was closely related to mortality in the patients.Conclusions Effective treatment of complications of CSH and reduction of occurring rate of the complications are very important for decreasing mortality of patients with CSH.
5.Rhodiola polysaccharide effect on spermatogonial stem cell proliferation in vitro

BACKGROUND:To establish a rapid and effective method to obtain sufficient spermatogonial stem cels that can meet the clinical need is urgent to be solved in the spermatogonial stem cel transplantation.
OBJECTIVE:To study the effect of rhodiola polysaccharide on the proliferation of spermatogonial stem celsin vitro.
METHODS:Under sterile conditions, spermatogonial stem cels and Sertoli cels were isolated from the testis of mice, and spermatogonial stem cels were seeded onto the feed layer of Sertoli cels. Then, the co-cultured cels were assigned into experimental group 1 (simple cel culture medium), experimental group 2 (cel culture medium containing 150 mg/L rhodiola polysaccharide) and experimental group 3 (cel culture medium containing 150 mg/L rhodiola polysaccharide, 1 U/L leukemia inhibitory factor and 10 μg/L glial cel line-derived neurotrophic factor). After 7 days of co-culture, flow cytometry was used to detect cel proliferation in vitro, and cel viability and positive expression of GFRa-1, Thy-1 and C-kit were calculated.
RESULTS AND CONCLUSION:After 7 days of co-culture, the cels grew rapidly and presented with colony and thyrsiform growth, and the number of cel masses increased significantly, al of which were in line with the proliferative features of spermatogonial stem cels. The GFRa-1, Thy-1 and C-kit proteins were expressed in the cel membrane and cytoplasm, mainly in the cel membrane. The viability of spermatogonial stem cels and positive expression of GFRa-1 and Thy-1 were ranked as folows: experimental group 3 > experimental group 2 > experimental group 1, and there were significant differences between groups (P < 0.05). The positive expression of C-kit had no difference between experimental groups 1 and 2, but it was significantly higher in the experimental group 3 than the other two groups (P < 0.05). These findings indicate that rhodiola polysaccharide used alone or combined with leukemia inhibitory factor and glial cel line-derived neurotrophic factor can enhance the proliferative ability of spermatogonial stem celsin vitro.
6.Seminiferous capsule extract effect on the proliferation of spermatogonial stem cells

BACKGROUND:Chinese herbs for kidney nourishment can promote the proliferation and differentiation of spermatogonial stem cel s. OBJECTIVE:To verify the effect of seminiferous capsule extract on spermatogonial stem cel proliferation. METHODS:Spermatogonial stem cel s were isolated from the testis of male mice and synchronized by serum-free medium fol owed by an addition of 10, 50, 100 mg/L seminiferous capsule extracts. After 24 hours of culture, viability, proliferation and cel cycle of spermatogonial stem cel s were observed. RESULTS AND CONCLUSION:Compared with the control group, seminiferous capsule extracts promoted the cel number, viability and proportion at S stage. The number of BrdU-labeled spermatogonial stem cel s was increased significantly after intervention with seminiferous capsule extracts, especial y at the concentration of 50 mg/L. These findings indicate that seminiferous capsule extracts can promote the proliferation and viability of spermatogonial stem cel s.
7.Curcumin for propranolol-induced psoriasis-like lesions in guinea pigs: evaluation of therapeutic efficacy and effects on proliferating cell nuclear antigen expression

Objective To estimate the influence of curcumin on psoriasis-like lesions and proliferating cell nuclear antigen (PCNA) expression in guinea pigs,so as to investigate the therapeutic effect and mechanism of curcumin in psoriasis.Methods A model of psoriasis-like skin inflammation was established by applying propranolol 5% cream (4 times a day for 3 weeks) to the dorsal skin of ears of 30 guinea pigs,which were then equally classified into 5 groups:model group receiving no treatment,observation group receiving no treatment but observation for 2 weeks,model control group treated with intragastric 25% polyethylene glycol solution (1 rnl once a day) for 2 weeks,low-dose and high-dose curcumin groups treated with intragastric curcumin solution at 20 and 40 mg/kg per day respectively once a day for 2 weeks.Six guinea pigs receiving neither induction by propranolol nor treatment by curcumin or polyethylene glycol solution served as the normal control group.Skin specimens were harvested from the ears of guinea pigs in the normal control group after three weeks of breeding,in the model group immediately after the establishment of psoriasis-like model,in the observation group after 2 weeks of observation,and in the other 3 groups after 2 weeks of treatment.Subsequently,haematoxylin-eosin (HE) staining was conducted to observe histopathologic changes,and immunohistochemical assay to detect the expression of proliferating cell nuclear antigen (PCNA).Results Gross observation of the skin revealed that curcumin attenuated psoriasis-like skin manifestations in guinea pigs.There were significant differences in histopathologic scores (F=296.14,P< 0.01) and PCNA expression rate among the 6 groups (F =108.49,P < 0.01).Least significant difference (LSD) test showed that both histopathologic scores and PCNA expression rate were significantly higher in the model group than in the normal control group (6.42 ± 0.49 vs.0.92 ± 0.20,63.17% ± 5.47% vs.20.83% ± 2.99%,both P < 0.01).After 2 weeks of treatment,both low-dose and high-dose curcumin groups showed significantly lower histopathologic scores (4.25 ± 0.27 and 1.75 ± 0.42 vs.6.42 ± 0.49,both P< 0.01) and PCNA expression rate (43.50% ± 2.90% and 25.50% ± 3.74% vs.63.17% ± 5.47%,both P < 0.01) compared with the model group.Conclusion Curcumin can attenuate pathological manifestations of psoriasis-like lesions,and downregulate PCNA expression in guinea pigs.
8.Expression of RNF2 in breast carcinoma and its significance

Purpose To investigate the expression of RNF2 in breast disease tissues and cell lines,and to analyze the association between expression of RNF2 and clinicopathological characteristics in breast carcinoma.Methods Expression of RNF2 protein and mRNA levels was detected using immunohistochemistry of EnVision two-step and qRT-PCR in breast carcinoma and benign breast disease as well as in cell lines.Results RNF2 expression was sigmificantly higher in breast carcinoma tissue specimens compared with benign breast disease specimens (P <0.05).Besides,the expression of RNF2 protein was significantly associated to tumor size,lymph node status and TNM stage (P < 0.05 for both),but was not related to age,histological grade,the expression of ER,PR and HER-2 (P > 0.05 for both).Higher expression of RNF2 mRNA was detected in breast carcinoma cell lines compared with breast epithelial cell lines (P < 0.05).Conclusion RNF2 is overexpressed in breast carcinoma and can be a potential therapeutic target for breast carcinoma.
9.Expression of aquaporin 4 in contused brain tissue and its relationship with brain edema

Objective To observe the expression of aquaporin 4 (AQP-4) in contused brain tissue and its relationship with brain edema following brain trauma.Methods A retrospective case control analysis was made on 42 patients with severe brain trauma admitted from January 2015 to March 2016.There were 23 males and 19 females,aged from 23 to 62 years [(35.5 ± 5.6) years].Glasgow coma score (GCS) was 3-5 points in 7 patients,6-8 points in 23 and 9-10 points in 12.Brain tissue removed from the area 1 cm near the contusion during the cranial surgery were allocated to study group (n =42),while brain tissue removed far from the contusion after internal decompression were used as control (n =8).Ultrastructure of brain tissues was observed under electron microscope.Water content of brain tissue was measured by dry-wet weight method and expression of AQP-4 was measured by immunohistochemical method at postinjury hours < 6,6-12,12-24,24-72,72-96 and > 96.Results Morphology and structure of brain tissue in control group were normal.Whereas in study group,the intracellular and interstitial edema were obvious and morphological structure were damaged.Water content and AQP-4 expression in control group showed no obvious increase after operation(73.55 ±0.10,0.193 ±0.016).Water content in study group increased significantly compared to control group and reached the peak value (81.28 ± 0.56) at postinjury 24-72 hours (P < 0.01).AQP-4 expression in study group increased at postinjury 6 hours (0.242 ±0.023) and reached a peak at postinjury 24-72 hours (0.338 ± 0.013),with significant difference compared to control group (P < 0.05).Correlation analysis showed change of brain water content was positively correlated with expression level of AQP-4 (r =0.931,P < 0.01).Conclusion Expression of AQP-4 in the injured area of brain trauma is significantly increased along with the increase of water content,suggests that the upregulation of AQP-4 plays an important role in traumatic brain edema.
10.Expression and significance of BCL-6 and ZEB2 in breast cancer

Purpose To explore the biological significance of BCL-6 and ZEB2 in invasion,metastasis and prognosis of breast cancer.Methods The expressions of BCL-6,ZEB2 protein and mRNA were detected respectively in 228 cases of breast cancer and 80 cases of breast benign lesions by immunohistochemical SP two-step staining and situ hybridization.Result The expression levels of BCL-6,ZEB2 protein and mRNA in breast cancer tissues were significantly higher than in breast benign lesions (P < 0.05).The expressions of BCL-6 was positively correlated with tumor size,lymphatic metastasis,histological grade,TNM staging and HER-2 expression (P < 0.05).The expressions of ZEB2 was positively correlated with tumor size,lymphatic metastasis,TNM staging and HER-2 expression (P < 0.05).The overall survival and relapse-free survival of BCL-6 and ZEB2 positive expression were significantly less than the negative expression (P < 0.01).Conclusion The BCL-6 and ZEB2 are closely correlated with the evolution process of breast cancer,which may become important means for monitoring and warning the metastasis,invasion,and prognosis of breast cancer.
