1.Multifocal electroretinogram of rod and cone cells in patients with retinitis pigmentosa

Objective To observe the mutifocal electroretinogram (mfERG) characteristics of rod and cone cells in patients with retinitis pigmentosa (RP) and to evaluate the function of photosensory cell.Methods The mfERG recording technique for rod cell in eight normal subjects (eight eyes) were established and the influence of different brightness lightstimulus in P1 wave amplitude were analyzed.The cone and rod cells mfERG of 38 eyes in 19 patients were recorded and then calculated positive ratio from local signal-noise ratio.The average visual acuity and P1 wave amplitude density of cone mtERG in different types were compared and statistically analyzed.Meanwhile,the changes in P1 wave amplitude of cone and rod mfERG in four quadrants also compared and analyzed.Results Rod cell mfERG in normal subjects can be recorded stably by using blue flashes with low light intensity as 0.04 cd/m~2.In patients with RP,the cone and rod cells mfERG can he detectd 65.790./00 and 10.51% respectively.P1 wave amplitude density in type I of cone cell mfERG was significantly higher than that in type Ⅱ (t=5.21,P=0.000).There were no differences in average visual acuity (t=1.15,P=0.612).P1 wave amplitude density in type Ⅰ was negatively related to IogMAR visual acuity (r=-0.48,P=0.04 ).The comparison of rod and cone cells mfERG in local wave characteristics showed that P_1 wave amplitude densities had spatial relationship in each area.Conclusions The results suggested highly variable central responses in cone cell in RP patients,higher positive recorded ratio in cone cell than rod cell and spatial correspondence between the function of reserved cone and rod cells.
2.Transplantation of rabbit embryonic full-thickness retina into the subretinal space of rats

Objective To study the transplantation of embryonic full-thickness retina, including neural retina and retinal pigment epithelial (RPE), into the subretinal space of rats and to investigate the development of the grafts. Methods A total of 34 eyes from 3-week-old pigmented rabbit embryos were treated with the dispase to obtain pieces of the full-thickness retina, and then grafts were injected into the subretinal space of the right eyes of 20 Wistar rats and 14 RCS (Royal college of surgeons) rats. Host rats were sacrificed at 3, 7, 14, and 28 d after transplantation. Sections were stained with hematoxylin eosin and immunohistochemistry. Results The rabbit embryonic retina was capable of developing and differentiating in the subretinal space of the host rats. Conclusion The rabbit embryonic retina is capable of developing and differentiating in the subretinal space of host rats. Transplantation of xenografts of embryonic full-thickness retina is practicable, but the methods of transplantation need to be improved.
3.Study on family adaptability and cohesion in cerebral palsy children

Objective To explore the family adaptability and cohesion in cerebral palsy children.Methods A total of 225 children were selected in two hospitals of Anhui province from May to November in 2014.Their parents or family primary caregivers were interviewed using Family Adaptability and Cohesion Scale,Second Edition-Chinese version (FACESII-CV) and self-designed general information questionnaire.Results The actual family types of 225 cerebral palsy children families were:102 cases of intermediate type (45.3%),76 cases of extreme type (33.8%),47 cases of balance type (20.9%),while the ideal family types were:92 cases of intermediate type (40.9%),62 cases of extreme type (27.6%),71 cases of balance type (31.5%),it was significant between the actual and the ideal family types (x2=6.817,P<0.05).The actual and ideal scores of family adaptability and cohesion scale were (64.09 ± 12.23) points,(43.28 ± 9.91) points and (73.91 ± 9.31) points,(52.73 ± 7.16) points,respectively,the difference was significant (t=-9.049,-10.976,P<0.01).The scores in the family which the mother had a higher culture degree (F=7.075,8.499,P<0.01),the cerebral palsy children did not live with grandparents (t=5.925,4.138,P<0.05),the one-child family (t=5.370,4.652,P<0.05) were higher.Conclusions The relationship between family members are estranged in most cerebral palsy children families,the ability of family coping with stuff is low and the family function is poor,in addition,the family adaptability and cohesion are mainly affected by the mother's culture degree,whether live with grandparents,the number of children.All above these are need to be intervented early to improve family function.
4.Study of choroidal circulation in macular area in high myopia

Objective To observe the characteristics of the choroidal circulation in the macular area in patients with high myopia. Methods Fundus fluorescein angiography (FFA) and indocyanine green angiography (ICGA) were performed in 25 cases (44 eyes) with high myopia, 16 cases (20 eyes) of low to moderate myopia, and 19 cases (24 eyes) of normal eyes using TOPCON50IA fundus fluorescein photographic system. The filling time of fluorescence in the choroid at the earlier stage and the filling time of fluorescence in the choroidal vessels in each group were compared. The relationship of the abnormal fluorescence configuration and the blood perfusion in the choroidal capillary vessels with the corrected visual acuity was analyzed. 5.Effects of genistein on the growth of human ovarian carcinoma transplanted subcutaneously in nude mice

Objective To investigate the effects of genistein used alone and in combination with cisplatin on the treatment of human ovarian carcinoma transplanted subcutaneously in nude mice. Methods Nude mouse model of human ovarian carcinoma transplanted subcutaneously was established. A total of 30 mice were divided into 5 groups: control group (0 04% saline), genistein group (subcutaneous injection of genistein at 0 2 mg/kg and 0 4 mg/kg), cisplatin group (intravenous injection of cisplatin at 4 mg/kg), and genistein plus cisplatin group. The growth of the subcutaneously transplanted tumor and changes of mouse body weight in each group after treatment using different medication regimens were observed at 7, 14, 21, and 28 d. Histopathological examination was also conducted. Results Genistein (0 4 mg/kg) had significantly inhibitory effect on the transplanted xenograft growth in vivo . Tumor volume, tumor weight, and T/C ratio (mean volume of treated group/mean volume of control group) decreased. Significantly enlarged necrotic areas were found in genistein treated group. The weight loss of nude mice after treatment with genistein was not significantly different from in the control group. Conclusion Genistein has the inhibitory effect on the growth of human ovarian carcinoma transplanted subcutaneously in nude mice. Genistein (0 4 mg/kg) in combination with cisplatin (4 mg/kg) has a synergistic effect on xenograft of human ovarian cancer cell line SKOV 3 in nude mice. The results provide the evidence for the potential usefulness of genistein for the prevention and treatment of human ovarian carcinoma.

7.Therapeutic effects of concomitant exotropia surgery

Objective To observe the effects of strabismus surgery for treatment of concomitant exotropia and analyze the influential factors. Methods From 2000 to 2004, 95 patients hospitalized in our department underwent strabismus surgery and followed up for one week to 3 years. We retrospectively analyzed the deviation and superior sensory outcome at week 1 (95 cases) and week 6 (58 cases) after surgery. Results Seventy-four cases (82.11%) were completely corrected in one-week follow-up, and 46 cases (79.31%) in six-week follow-up. Nineteen cases (32.76%) recovered superior sensory. Conclusion Timely surgery of concomitant exotropia can improve not only the appearance but also the function of the eyes.
8.Preliminary application of the revised objective structure clinical examination stations

Objective To explore the feasibility and effectiveness of the revised OSCE stations.Methods 106 sstudents of grade 2008 and 2009 from Xinhua clinical medical school of Shanghai Jiaotong University School of Medicine were involved,32 of whom (experimental group) tool modified OSCE examination and 74 of whom (control group) tool traditional OSCE examination.The revised OSCE exam was the one which on the basis of original OSCE test station setup,improved the operating part of the test stand,adopted clinical cases as stem,asked the students to make possible diagnosis,and perform the corresponding operation.We evaluated the students based on the OSCE scores from the school exam and the overall performances in the national clinical skills competition.Comparison between the two groups was performed by using t test.Results All students who have received the new training methods agreed that it stood closer to the clinical setting.Revised-OSCE trained group of students test scores than traditional OSCE trained group of students,though it didn't have statistical significance.Students from the revised-OSCE group had the score of (80.50 ± 15.45),while the traditional group had the score of (78.30 ± 17.68).By taking the revised-OSCE trainings,all the students have improved their clinical skills and two years in a row to win the Shanghai Jiaotong University School of Medicine Clinical Skills Competition.Conclusions Case-based OSCE stations are well accepted.The new OSCE stations are much closer to clinical teaching and can make better objective assessment of students,so it plays a good role in improving medical students' overall performances.
9.Effect of mild hypothermia combined with hydrogen-rich saline on cerebral injury after cardiac arrest and resuscitation in rats

Objective To evaluate the effect of mild hypothermia combined with hydrogen-rich saline on cerebral injury after cardiac arrest and resuscitation in rats.Methods Healthy male Sprague-Dawley rats,aged 7-8 weeks,weighing 280-320 g,were divided into 5 groups (n=33 each) using a random number table method:sham operation group (group S),cardiac arrest and resuscitation group (group CAR),hydrogen-rich saline group (group H2),mild hypothermia group (group MH),and mild hypothermia plus hydrogen-rich saline group (group MH+H2).Cardiac arrest was induced with transoesophageal cardiac pacing followed by cardiopulmonary resuscitation to establish the cerebral injury model.Hydrogen-rich saline 5 ml/kg was intraperitoneally injected immediately after return of spontaneous circulation (ROSC) in H2 and MH+H2 groups,while the equal volume of normal saline was given instead in the other groups.The body temperature of rats was cooled down to 32-34℃ within 15 min starting from the time point immediately after ROSC and maintained for 4 h in MH and MH+H2 groups.Fifteen rats were selected at 24 h after ROSC to assess the neurological function score (NDS).Eighteen rats in each group were sacrificed at 24 h after ROSC,and brains were removed for microscopic examination of the pathological changes in hippocampal CA1 region after hematoxylin and eosin staining and for determination of pyramidal cell count and expression of glucose-regulated protein 78 (GRP78),C/EBP-homologous protein (CHOP),caspase-12,caspase-3,Bcl-2 and Bax in hippocampal CA1 region (by Western blot).Results Compared with group S,the NDS was significantly decreased,the pyramidal cell count was reduced,the expression of GRP78,CHOP,caspase-12,caspase-3 and Bax was up-regulated,and the expression of Bcl-2 was down-regulated in the other four groups (P<0.05).Compared with group CA-R,the NDS and pyramidal cell count were significantly increased,the expression of GRP78 and Bcl-2 was up-regulated,and the expression of CHOP,caspase-12,caspase-3 and Bax was down-regulated in H2,MH and MH+H2 groups (P<0.05).Compared with group H2 or group MH,the NDS and pyramidal cell count were significantly increased,the expression of caspase-3 and Bax was down-regulated,the expression of Bcl-2 was up-regulated (P<0.05),and no significant change was found in the expression of GRP78,CHOP and caspase-12 in group MH+H2 (P> 0.05).Conclusion Combination of mild hypothermia and hydrogen-rich saline offers enhanced efficacy in reducing cerebral injury after cardiac arrest and resuscitation over mild hypothermia or hydrogen-rich saline alone in rats.
