1.Apathy and related factors in patients with Parkinson's disease

Objective To investigate the prevalence,clinical traits and correlates of apathy in patients with Parkinson' s disease ( PD ).Methods Seventy-seven cases of PD patients and 40 health controls were recuited in the present study.Apathy was assessed using Lille Apathy Rating Scale(LARS) and related factors,such as motor function,cognition,depression,age,sex,education level,disease duration,levodopa equivalent doses (LED) and use of antipsychotic drugs were also evaluated.Four kinds of scales,Unified Parkinson' s Disease Rating Scale (UPDRS),Hoehn-Yahr staging,Montreal Cognitive Assessment (MoCA),Hamilton Depression Rating Scale (HAMD) were employed to evaluate the motor function,cogniton and depression,respectively.Results The prevalence of apathy and depression in the 77 PD patients were 49.4% ( 38/77 ) and 46.8% (36/77),respectively.Of the 77 PD patients,11 cases had apathy without depression (14.3% ); 9 cases had depression without apathy( 11.7% ) and 27 cases had apathy combined with depression( 35.1% ).Among the PD patients with apathy,the cognitive type is the most common one,accounting for 86.8% (33/38).There were significant statistical differences in education level,UPDRS- Ⅱ/Ⅲ scores,Hoehn-Yahr staging,MoCA and HAMD scores between apathy ( n =38 ) and no-apathy(n =39) PD patients(t =2.309,-3.144,-4.000,-3.217,2.649,-3.909,all P<0.05).According to the unifactoral Logistic regression analysis of apathy correlates,UPDRS-Ⅱ/Ⅲ scores,HoehnYahr staging and HAMD scores were the risk factors for apathy while education level and MoCA were the protective factors for apathy.In the multiple Logistic regression analysis,HAMD scores,UPDRS-Ⅱ scores and education level were included.Conclusions The prevalence of apathy in PD patients was significantly higher than healthy controls.Apathy could be present without depression in PD patients,and was associated with education level,motor dysfunction degree,cognitive impairment and depression severity.
2.EFFECT OF THREE TYPES OF CHELATING AGENTS ON EXCRETION AND DISTRIBUTION OF MERCURY

After the mice had been injected im203HgCl2 185 kBq/mouse, they were injected ip with chelating agents. There were 15 chelating agents of 3 types,of which the effect of eliminate mercury of pyridoxine derivative, benzenesul-fonate cysteine and sodium dimercaptosuccinate ( DMS-Na) were better than the others. The urine excretion of 208Hg was increased and the ratio were 4.6,11.5 and 22 respectively in comparison with the control group.The minimum 203Hg content in organs of mice was in the DMS-Na group,the next was the benzenesulfonate cysteine group,but there no significant difference between these 2groups.
3.Evaluation of retinal function after retinal transplantation for light-induced retinal degeneration in miniature pigs

Objective To investigate the possibility that transplanting the full-thickness neonatal piggy retinas that are completely differentiated but immature improves the retinal function after light-induced retinal degeneration in pigs.Methods Retinas from normal Guangxi Bama pigs aged 1-6 days were used as donor tissues.Neuroretina-RPE cografts were obtained from newborn pigs by using excimer laser for microablation of choroidal tissue and transplanted into the subretinal cavity of adult Bama pigs after light-induced retinal degeneration through vitrectomy and retinotomy.On days 5-7 and in 1st to 5th month after retinal transplantation,the survival of the cografts in the recipients and whether the host retinas have rejection were observed by ophthalmoscope,colour fundus photography and fundus fluorescein angiography,and the amplitude and lantency of N1,P1 waves between different periods were measured by mfERG.Results The retinal transplantation was performed in 15 eyes of 8 Bama pigs after light-induced retinal degeneration.The subretinal transplantation of the cografts was performed successfully in 11 eyes,with the operation success rate of 84.6%(11/15).In host retina,the gray-black graft inside transplantation bed could be seen clearly in 1st to 2nd month after transplantation and the leaked fluorescence in transplants was checked with FFA.The vertical comparison between different periods showed the amplitude of N1,P1 waves of retinal transplants rose with the extension of the survival time,and the areas where active response was observed were ring 2 and ring 3;but the latency of N1,P1 waves was shortened significantly in each ring as compared with that before operation,especially in late survival period.Conclusion The function measurement and the observation of living body together confirm the transplantation of completely differentiated retina from newborn pigs improves the retinal function of pigs after light-induced retinal degeneration.
4.Activation of endogenous retinal stem cells in RCS rats during onset and development of retinal degeneration

Objective To investigate whether endogenous retinal stem cells can be activated in the retina of Royal College of Surgeons (RCS) rats during the onset and development of retinitis pigmentosa. Methods The RCS-p+ rats with inherited retinal dystrophy were divided into 3 groups: the initial stage group (15-day RCS rats), the mid-stage group (30-day RCS rats) and the advanced stage group (90-day RCS rats) according to the severity of degeneration (n=4 in each group). RCS-rdy+p+ rats without retinal degeneration served as controls, and divide into three groups (15-day control, 30-day control, 90-day control) matched with RCS-p+ rats. A transcription factor (Chx10) expressed by embryonic retinal progenitors was detected using immunofluorescence and Western blotting. Results All of the retinal layers in the three control groups and in the 15-day RCS rats did not express Chx10, while the positive expression was observed in the photoreceptor layers of the 30-day and 90-day RCS rats. 5.Traditional Chinese medicine basic theory of Yin and Yang and experimental study on clinical quantization comparisonin of divergent and convergence medicine in treatingacute urticaria

Objective According to the basic theory of Yin and Yang,acute urticaria should be treated with divergent medicine instead of convergence medicine.In order to prove the theory,we carry out the quantifiable and repeatability traditional Chinese medicine screening tests in acute urticaria patients,with double controls of experimental group (divergent Chinese medicine),control group (convergence Chinese medicine)end instrumental quality-control standard substance MEBE.Methods Germany MORA-Super bio-resonance instrument (medical mode) as adopted as objective index.32 cases of acute phase urticaria were selected and treated with divergent or convergence Chinese traditional medicine,to observe the results whether conform to traditional Chinese medicine basic theory of Yin and Yang (That is whether the acute phase urticaria should be treated with divergent Chinese medicine).Results Through the 32 cases repeat experiments,22 out of 23 kinds of Divergent Chinese medicine have been screened out in experimental group; while none has been screened out in 12 kinds of convergence Chinese medicine in control group.There was significant difference between two groups (P<0.01).The result was entirely consistent with TCM basic theory of opposite relationship between yin and yang.The top five divergent Chinese medicines were divaricate saposhnikovia root,cicada slough,catnip,shrub chastetree rruit,and lily magnolia.Conclusion Treated acute urticaria with divergent Chinese medicine conforms to the basic theory of traditional Chinese medicine.
6.Membrane properties of rat retinal ganglion cells during postnatal development

Objective To study the membrane properties of rat retinal ganglion cells during postnatal development, including the passive membrane properties and the action potentials evoked by depolarizing current injections and the relationship between the membrane properties and ages. Methods Whole cell patch clamp recordings of the retinal ganglion cells (RGCs) in retinal slices of postnatal rats (age ranging from postnatal days 7 to 30) were performed. Results (1) A total of 112 RGCs were recorded from postnatal rats. (2) The electrophysiological properties of RGCs changed significantly during development. The excitability of RGCs increased in an age-dependent manner. There was significant difference between before-eye-opening group(P7-13d) and after-eye-opening group(P14-30d) . (3) Three different discharge patterns of RGCs in response to sustained depolarizing current pulses were recorded: single-spike firing, transient firing and sustained firing. 7.Electrophysiological properties of retinal ganglion cells in Long Evans rats at different postnatal developmental stages

Objective To investigate the electrophysiological properties of retinal ganglion cells(RGCs) in Long Evans rats during different postnatal developmental stages so as to explore the intracellular mechanism of retinal neuron maturation. Methods Whole cell patch clamp recordings of RGCs in acute retinal slices of Long Evans rats (postnatal days 3 to 31) were performed. Depolarizing current pulses of different densities were inflicted on RGCs to evoke action potentials(AP) . Results A total of 94 RGCs, single spike cells, transient and sustained cells, were acquired in 33 rats. The proportion of the three types of TGCs was different. There was significant difference in electrophysiological properties. Conclusion During the period of postnatal development, gradually matured electrophysiological properties of RGCs and differences in amplitudes and frequencies of AP suggest that different types of RGCs may play different roles in coding and transmitting spatial and temporal information.
8.Comparison of electrophysiological and morphological properties of retinal ganglion cells in Long Evans and Wistar rats

Objective To compare the differences of the electrophysiological and morphological properties of retinal ganglion cells (RGCs) in Long Evans and Wistar rats. Methods Whole cell patch clamp recordings were made from RGCs in the acute retinal slices of Long Evans and Wistar rats aged postnatal 0-31 d. RGCs were stained with Lucifer Yellow diffused into cells during whole-cell recordings for the purpose of morphological study. The sections of rat retinas by HE staining were examined by light microscopy. Results The differences of electrophysiological properties in Long Evans and Wistar rats were not significant. There were pigment granules in retinal pigment epithelial cells of Long Evans but not in those of Wistar rats. Conclusion Long Evans rats can be the good animal model for visual and neural science research in place of Wistar rats.
9.Haplotype analysis of two families with X-linked retinitis pigmentosa

Objective To identify the disease locus in X-linked retinitis pigmentosa (RP) families using genetic linkage analysis. Methods Five microsatellite markers were selected from the RP2, RP3, RP6, RP23 and RP24 gene loci, respectively. Haplotype analysis for two X-linked RP families was performed to determine the critical region. Two-point linkage analysis was performed using MLINK program. Results In FYJ and ZCF X-linked RP families, the LOD score was 1.18 and 1.03 at DXS 993, 0.58 and -2.69 at DXS 1068, -2.33 and -2.45 at DXS 1214, -2.34 and -2.51 at DXS 8051, -2.23 and -2.62 at DXS 8043. Conclusion The phenotype of ZCF family is not caused by mutation of RP3, RP6, RP23, RP24 gene, and FYJ family may be linked to RP2 or RP3 gene.
10.Electron microscopic observation of Randall's plaque in patients with calcium oxalate stone

Objective To study the ultrastructure of the renal papillary Randall's plaque in calclum oxalate stone formers. Methods The 14 biopsy samples of the Randall's plaque in 12 patients with calcium oxalate stone undergoing PCNL for stone removal were obtained using endoscopic biopsy technique,followed by staining with hematoxylin-eosin or fixing with osmium tetroxide,and then the ultrastructure of the Randall's plaque was observed under light microscope and transmission electron microscope. Results In all 12 patients,72 renal papillae were examined.All kidReys were found to have papillary plaque and 7 of the patients had attached stones.Sixty-three papillae(87.5%)contained plaque.Calcium deposition was seen in the 12 renal papilla tissue by light microscopy.Transmission electron microscopy images of the 2 Randall's plaque samples showed several cluster of sharp and large crystals lied closer to the surface of Randall's plaque.The typical crystals were acicular with light profile. Conclusions Randall's plaque is an interstitial medullary and papillary deposit of calcium oxalate.The appearance of the deposition of calcium oxalate crystals lies upon Randall's plaque,which might be an explanation for the mechanism of calcium oxalate stone formation.
