1.Effects of GSRb3 on GABA in brain tissue of rats with injury of hypoxia

Objective To observe the influence of ginsenoside Rb3(GSRb3) on ?-aminobutyric acid (GABA) in brain tissue of rats with injury of hypoxia.Methods 20 rats were randomly divided into A, B, C, D and E group. The models of cerebral anoxia in group A and C were made by hypopiesia and hypoxia, and the models of hypoxia-reoxgenation were made in group B and D. 24 h and 1 h before establishment of models, ginsenoside Rb3 was injected peritoneally in rats of group C and D, respectively. The number and form of GABA-like immunoreactive neurons in hippocampus CA1 area of rats after brain hypoxia-reoxgenation were investigated with immunohistochemistry.Results (1)Compared with group E, GABA-like immunoreactive neuron density in CA1 area of group A and B decreased, presented with light staining and absence of prominency. GABA-like immunoreactive neuron density in CA1 area in group C and D was significantly higher than that in group A and B, but the shape was similar to group E. (2) The number of GABA-like immunoreactive neuron in CA1 area was 7.7?2.83 (group A), 10.1?2.08 (group B), 30.9?2.02 (group C), 33.1?4.2 (group D) and 16.9?1.05 (group E), respectively. Objective To study the effects of the monomers of ginsenoside Rb1 (GSRb1) on the concentrations of intracellular calcium in ischemic neurons of rats.Methods Hippocampus neurons from embryo of rats were cultured in vitro, then placed into normal extracellular fluid (normal control group), simulated ischemic extracellular fluid (ischemia group), simulated ischemic extracellular fluid without calcium (ischemia without calcium group) and simulated ischemic extracellular fluid with different concentration of GSRb1. The fluorescence intensity of intracellular calcium in each group was measured by laser scanning confocal microscope technique, and the corresponding percentage was calculated by comparison with normal control.Results Compared with normal control group, fluorescence intensity of intracellular calcium increased by ( 73.5?10.31)% in ischemia group, ( 4.5?2.58)% in ischemia without calcium group,( 20.2?3.41)%, ( 13.6?2.98)%,( 10.5?3.62)% and ( 12.7?4.51)%, respectively, in simulated ischemic extracellular fluid with different concentration of GSRb1 groups (20, 40, 60 and 80 ?mol/L). 4.Arthroscopic reconstruction of posterior cruciate ligament using hamstring tendon transfer and e-val uation of its clinical evaluation

Objective The present was to report the methods of arthroscopic reconstruction of poste-rior cruciate ligament by a two-strand semitendinosus tendon autograft and evaluate its clinical results.Method From June1998through June2002,28cases had undergone arthroscopic restoration of a torn pos-terior cruciate ligament,including22female and6male patients with an average age of21years(range,18to31years).The time-interval between the sustained injury and the surgery was25days to3months.The causes of injury of posterior cruciate ligament were sport activity in6cases,traffic accident in18cases and fall down in4cases.All of the patients suffered from knee pain,knee laxity and atrophy of quadriceps femoris.The posterior drawer test was positive in26cases preoperatively,in which6cases were associated with positive anterior drawer test.The Lachmans test was positive in all of28cases preoperatively.The piv-ot shift test was positive in12cases preoperatively.The concomitant injuries consisted of anterior cruciate ligament injury in6cases,meniscal tear in8cases and medial collateral ligament injury in5cases.Results Follow-up period extended from27to48months (mean,33months).According to the international knee documentation committee(IKDC)activity grades,grade D were26cases,and grade C were2cases preoper-atively,while postoperatively,grade A were9cases,grade B16cases,and grade C3cases at the last fol-low-up.The clinical assessment judging by Lysholm knee functional scoring system,the average scores be-fore operation was65.5?3.6;however the scores increased to an average of92.7?3.6postoperatively.Con-clusion Arthroscopic reconstruction of injured posterior cruciate ligament using hamstring tendon transfer passes through the tunnel more smoothly than the bone-patellar tendon-bone because of no bone blocks,furthermore,the tunnel disappears after the screw is absorbed to avoid the impact of the ligament the inside tunnel and prevent the tunnel extending gradually to cause the ligament laxity and knee instability.The at-tachment of the tendon on the outlet of tibial tunnel and the rigid anchoring of ligament on the outlet of femoral tunnel could provide enough strength for constructed posterior cruciate ligament.So the patients could begin postoperative rehabilitation as soon as possible and recover functional activity earlier.

TOF-MS has been more and more widely used in clinical and scientific research due to its advantages such as fast and easily operated.Diabetic nephropathy is the most common complications of diabetes.But clinical existing detection meth-ods to some extent have some hysteresis.The applications and developments of TOF-MS coupled with other proteomics technologies provided new ideas and methods for early diagnosis and prognosis of diabetic nephropathy.In this paper,the lat-est research results in nearest years about the application of TOF-MS to diabetic nephropathy will be reviewed.

Objective:To explore the effect of siRNA targeting myeloid cell leukemia-1(Mcl-1)on the biological behavior of salivary adenoid cystic carcinoma cells.Methods:The chemically synthesized Mcl-1-siRNA was transfected into salivary adenoid cystic carci-noma SACC-2 cells.The expression levels of Mcl-1-mRNA and Mcl-1protein were examined by Real-time PCR and western blotting respectively.MTT assay,transwell chamber and flow cytometry were used to determine the effect of Mcl-1-siRNA on SACC-2 cell pro-liferation,migration and apoptosis.Results:Compared with the control group,liposome group and NC-siRNA group,SACC-2 cell proliferation rate of Mcl-1-siRNA group was obviously slowed down.48 h after transfection,the migration of SACC-2 cells in Mcl-1-siRNA group(39 ±9.0)were lower than that in control group(69 ±6.0).The apoptosis rate of Mcl-1-siRNA group(8.6%)was sig-nificantly higher than that in control group(1.9%).Conclusion:Silence Mcl-1 can inhibit cell proliferation and migration and pro-mote apoptosis of salivary adenoid cystic carcinoma cells.

Objective:To explore the effects of cysteine-rich 6 1 (Cyr6 1 )on biological behavior of human adenoid cystic carcinoma ACC-LM and ACC2 cells.Methods:The chemically synthesized Cyr6 1-siRNA was transfected into ACC-LM and ACC2 cells.Cell proliferation was measured by the MTT method,the invasive ability was evaluated by Transwell chamber assay,and cell apoptosis was analyzed using flow cytometry by double staining with Annexin V and propidium iodide.Results:Cyr61-siRNA significantly down-regu-lated Cyr61 protein expression in ACC-LMand ACC2 cells.Cyr61-siRNA markedly inhibited the proliferation and invasion of the cells, however,there was no significant difference in cell apoptosis between Cyr6 1-siRNA and control groups.Conclusion:Cyr6 1 promote the proliferation and invasion of adenoid cystic cancer cells.

Objective:To investigate the effect of c-fos on the proliferation and migration of oral squamous cell carcinoma and potential mechansism.Methods:The expression of c-fos,CyclinD1 and p16 in 60 oral squamous cell carcinoma samples and 60 oral mucosa tissue samples was examined by immunohistochemistry.HN6 and SCC9 cells were respectively transfected with siRNA-c-fos and siRNA-scramble,then were respectively divided into control group,siRNA-scramble group and siRNA-c-fos group.The mRNA and protein expressions of CyclinD1 and p16 were decteted,meanwhile cell proliferation and migration were tested.Results:Compared with the oral mucosa tissue samples,the expressions of CyclinD1 and c-fos were increased in the carcinoma samples,while the expression of p16 was reduced.Compared with control group,the expressions of CyclinD1 in siRNA-c-fos group were significantly reduced,while p16 enpression was increased,with the inhibition of cell proliferation and migration.Conclusion:c-fos may regulate pl6/CyclinD1 signaling pathways and promote the proliferation and migration of oral squamous cell carcinoma.
9.The effects of IGF-1 on the proliferation and apoptosis of cultured condylar chondrocytes

Objective:To investigate the effects of IGF-1 and IL-1β on the proliferation and apoptosis of cultured human condylar chondrocytes( CCs) of temporomandibular joint( TMJ) . Methods:Cultured CCs were derived from human TMJ condylar cartilage tis-sue, and identified by immunocytochemistry staining. The cultured cells were divided into 6 groups:control group, IL-1β(10 μg/L) group and IL-1 group (10 μg/L) + IGF-1 group (0, 1, 10, 50 and 100 μg/L, respectively). The cell proliferation ability was de-tected by MTT assay. The cell cycle and apoptosis were detected by flow cytometry. The expression of apoptosis-associated factors Bcl-2, Bax and p38 MAPK/NF-κB proteins were detected by Western blot. Results:Type II collagen was positively expressed in cultured CCs. IL-1β treatment decreased cell proliferation, increased cell apoptosis with concomitant increase of the percentage of early apopto-sis and late apoptotic cells, increased Caspase-3 expression, decreased Bcl-2/Bax ratio, and increased the expression of p38 MAPK/NF-κB proteins. Whereas, with 1-100 μg/L IGF-1 pretreatment, the proliferation ability and Bcl-2/Bax ratio of the cells were in-creased(P<0. 05), the apoptotic cells were decreased, the expression of Caspase-3 and p38 MAPK/NF-κB proteins was decreased ( P<0. 05) in a dose-dependent manner. Conclusion:IGF-1 may inhibit IL-1β-induced cell apoptosis and attenuate the activation of p38 MAPK/NF-κB of human condylar chondrocytes.
10.Clinical study of spiral stone basket assisted with FURL in treatment of unilateral upper ureteral stones for diameter>1.0 cm

Objective To investigate the clinical effects and safety of spiral stone basket assisted with FURL in the treatment of unilateral upper ureteral stones for diameter>1.0 cm. Methods 140 patients with unilateral upper ureteral stones for diameter>1.0 cm from January 2012 to December 2015 were randomly divided into control group (70 patients) with FURL used alone and observation group (70 patients) with spiral stone basket assisted application on the basis of control group;the perioperative clinical indicators, the lithotripsy success rate, the stone clearance rate, the stone removal rate and the postoperative complication incidence of both groups were compared. Results The operation time of observation group was signiifcantly longer than control group (P<0.05). There was no signiifcant difference in the blood loss volume in operation and hospitalization time between 2 groups (P>0.05). The lithotripsy success rate and the stone clearance rate of observation group was signiifcantly higher than control group (P<0.05). The stone removal rate of observation group was signiifcantly lower than control group (P<0.05). The postoperative complication incidence of observation group was no difference between 2 groups (P> 0.05). The total treatment expenses of observation group was signiifcantly fewer than control group (P<0.05). Conclusion Spiral stone basket assisted with FURL in the treatment of unilateral upper ureteral stones for diameter>1.0 cm can efifciently higher the stone removal effects, reduce the stone removal risk and not increase the postoperative complications incidence.
