1.Determination of the Dissolution of Perospirone Hydrochloride Tablets

OBJECTIVE:To establish a method for the determination of the dissolution of perospirone hydrochloride tablets(PHT).METHODS:The dissolution of PHT was determined in accordance with Chinese Pharmacopeia(2005 Edition) using water as solvent with the rotation speed at 50 r?min~(-1).The sample was taken at 30 minutes with its absorbability detected at a wavelength of 316 nm determinated by UV.Then the dissolution was calculated.RESULTS:The linear range of perospirone hydrochloride was 2~20?g?mL~(-1)(r=0.999 9)with an average recovery at 99.76%(RSD = 0.27%).The dissolutions of 3 batches of samples were all above 80%.CONCLUSION:The method was proved to be simple and reliable, and it can be used to determine the dissolution of PHT.
2.Application of clinical risk index for babies in low birth weight preterm infants

Clinical risk index for babies( CRIB) is applied in low birth weight preterm neonates to assess the initial severity of illness,predict mortality risk rates,evaluate their own performance,and audit the performance between different medical institutions.CRIB plays an important role in the progressive develop-ment of neonatology.The scoring rules and advantagse of CRIB scorni g system were introud ced in this arti-cle.Teh scoring system is able to correctly predict mortaliyt probabilities and long-term neurodevelopmental outcomes for low birth weight preterm infants.
3.Simultaneous Determination of 9 Common Inorganic Anions in Huanglian Shangqing Tables by Accelerat-ed Solvent Extraction-ion Chromatography

OBJECTIVE:To establish the method for simultaneous determination of 9 common inorganic anions in Huanglian shangqing tablet. METHODS:The accelerated solvent extraction-ion chromatography was adopted. Inorganic anions were deter-mined by Ion Pac AS11-HC anion exchange column,protected by Ion Pac AG11-HC column and eluted by hydroxide potassium so-lution(gradient elution)at the flow rate of 1.2 mL/min. The column temperature was set at 30 ℃,elution time was 18 min,and sample volume was 25 mL. RESULTS:The linear ranges of fluorinion,formate ion,nitrite ion,bromide ion,nitrate ion,sulfate ion,oxalate ion and phosphate ion were 0.1-5 mg/L(r=0.9990-0.9999). The limits of quantitation were 0.020,0.078,0.030, 0.058,0.052,0.068,0.084,0.064,0.074 mg/L,and the limits of detection were 0.005,0.024,0.008,0.017,0.015,0.022, 0.026,0.020,0.021 mg/L,respectively. RSDs of precision,stability and repeatability were all lower than 4.0%;recoveries were 80.00%-125.08%(RSD ranged 0.97%-2.47%). CONCLUSIONS:The method is simple,precise,stable and repeatable,and can be used for 9 common inorganic anions in Huanglian shangqing tablet.
4.Treatment of Lithotripsy for Acute Renal Failure Resulting from Upper Urinary Calculi

Objective To study and evaluate the therapeutic safety and effect of frequency-doubled-double-pulse laser(FREDDY)lithotripsy for treating upper urinary calculi combined with acute renal failure.Methods The clinical data of 32 cases treated by frequency-doubled-double-pulse laser(FREDDY)lithotripsy were retrospectively analyzed.Results After the operation the serum BUN and Cr levels in the patients got close or returned to normal and the free rate of the stones was up to 90.6%(29/32).Conclusions The ureteroscopic frequency-doubled-double-pulse laser(FREDDY)lithotripsy has the advantage of safety,high efficiency and less trauma for treating the upper urinary tract obstruction combined with acute renal failure.It can also deal with the bilateral ureteral stones at one time.It can be the first choice when the condition is proper.

Strengthening the ethical education on oral and maxillofacial surgeon and right education on patients are in favor of development of tissue engineering in the oral and maxillofacial surgery.

7.Construction and identification of eukaryotic expression vector of bcl-2 siRNA

AIM: To construct eukaryotic expression vector of small interfering RNA(siRNA) specific to bcl-2 and investigate the effect of recombinant plasmid on suppressing bladder cancer cell growth.METHODS: siRNA of bcl-2 gene was designed according to the principle of RNAi-based medicine, and was converted into cDNA coding expression of small hairpin RNAs(shRNA) of siRNA. The cDNA was synthesized and inserted into plasmid pGenesil-1. The recombinant eukaryotic expression vectors of pGenesil-1545 and pGenesil-1555 were controlled by the U6 promoter of RNA polymerase Ⅲ, identified by the restriction map and the sequence analysis, and transfected into T24 cells. After T24 cells were transfected for 72 h, expression of bcl-2 mRNA was assayed by RT-PCR; and MTT was used to observe the proliferation of T24 cells.RESULTS: The recombinant plasmids of pGenesil-1545 and pGenesil-1555 were identified by the restriction map and the sequence analysis. The sequences completely coincided with the designs. The expression of the bcl-2 mRNA in T24 cells transfected with recombinant plasmid decreased nearly 80%, and the growth of T24 cells was suppressed significantly.CONCLUSION: The siRNA eukaryotic expression vector against bcl-2 gene is successfully constructed. It effectively downregulates the expression of bcl-2 in T24 cells and suppresses the cell growth.
8.The effect of modified electroconvulsive therapy and lithium on oxidative stress status in bipolar disorder patients with depression episode

Objective To examine the effect of modified electroconvulsive therapy (MECT) on the plasma oxida-tive stress level in bipolar depression. Methods Forty-two patients with bipolar depression were randomly divided into two groups. The intervention group (n=18) received antidepressants and 12 times MECT for 6 weeks and the control group (n=24) received antidepressants and Li2CO3 for 6 weeks. The Chinese versions of the 17 items Hamilton Depression Rating Scale (HAMD-17), Young Mania Rating Scale (YMRS), Clinical Global Impression Scale (CGI-S) and Treatment Emergent Symptom Scale (TESS) were used to assess participants at baseline and after 6 weeks of treatment. The plasma levels of superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GSH-Px) and malondialdehyde (MDA)were detected at baseline and after 6 weeks to assess the level of oxidative stress. Results Repeated measures analysis of variance showed that the plasma level of SOD was higher in MECT group than in Li2CO3 group (F=15.26, P<0.01), and the level of MDA was higher in Li2CO3 group (F=18.18, P<0.01). The interactive effect of group and time was significant in GSH-Px level (F=6.39, P=0.02). The level of GSH-Px was lower in MECT group than in Li2CO3 group after 6 weeks (P<0.05). The CAT level was higher in the response patients than in non-response patients after 6 weeks (P<0.05). Con-clusions Both MECT treatment and Li2CO3 treatment can alter oxidative stress levels in patients with bipolar depression. The mechanisms underlying its therapeutic regimen may correct the imbalance of the plasma CAT level.
9.Effects of Captopril in renal ischemia reperfusion injury

Objective To explore therapeutic mechanism of angiotensin-converting enzyme inhibitor (Captopril) in renal ischemia reperfusion (I/R) injury.Methods Wistar rats were randomly divided into sham operation group (Sham group),ischemia-reperfusion group (I/R group),captopril-treated group (CAP group),I/R model was made through ligating one side renal vessel.Renal function indexes including SCr and BUN were detected through biochemical analysis.Changes of renal tissue were observed by pathological section.ELISA detection was used to determine inflammatory cytokines such as IL-6,IL-8 and TNF-α.Activation of MAPK signaling pathway were analyzed by Western blot.Results After captopril treatment,SCr,BUN levels of the I/R group and the CAP group were higher than those of Sham group(P < 0.05),Scr,BUN concentrations of CAP group were lower than those of I/R group(P < 0.05).After captopril treatment,IL-6,IL-8,TNF-α concentrations of I/R group were higher than those of the Sham group(P <0.05);IL-6,IL-8,TNF-α concentrations of CAP group were lower than those of I/R group(P < 0.05).Renal tissue c-jun N-terminal kinase,extracellular signal-regulated protein kinases,P38 in I/R group and CAP group were phosphorylated,and there were no significant differences between the two groups.Conclusion Captopril couldimprove renal function to some degree,and reduce expression of inflammatory cytokines rather than MAPK inflammatory signaling pathway.
10.A comparsion study on the social functions promotion of different medicine treatment strategies on the patients with treatment-resistant depression

Objective To evaluate the effectiveness of different medicine treatment strategies on the social functions promotion on the patients with treatment-resistant depression (TRD). Methods 375 Patients with TRD were randomly grouped into 8 groups, and each group was received 8 weeks different treatment for paroxetine,venlafaxine, mirtazapine, paroxetine plus risperidone, paroxetine plus sodium valproate, paroxetine plus buspirone, paroxetine plus trazodone,or paroxetine plus thyroxine, respectively. The efficacy and social functions were evaluated with HAMD-17, SDSS and SF-36. Results There were significant difference in SDSS scores between 8th week and the baseline( P<0.01 ) , and for social functions factor scores of SF-36 there was significant difference between 4th ,8th week and the baseline in each groups( P<0.01 ). There were significant difference in social functions factor scores of SF-36 and subtracting scores between 4th and 8th week in all groups except group paroxetine and group venlafaxine(P < 0.05 or P < 0.01 ). There were significant difference in SDSS subtracting scores at 8th week among 8 groups( paroxetine plus risperidone group 7.05 ± 6.39, mirtazapine group 6.53 ± 4.75, paroxetine plusthyroxine group 5.14 ± 4.94, paroxetine group 5.13 ± 4.94 ,paroxetine plus trazodone group 5.00 ± 4.94, paroxetine plus sodium valproate group 4.60 ± 4.09, venlafaxine group 4.57 ± 4.18, paroxetine plus buspirone group 4.24 ± 4.95 ) ( Z = 2.076, P < 0.05 ), between group paroxetine plus risperidone and group venlafaxine , group paroxetine plus sodium valproate, group paroxetine plus buspirone,as group mirtazapine and group paroxetine plus buspirone(P< 0.05 ), respectively. The influencing factors on improving social functions are the severity, improvement of depressive symptoms and latest onset time. Conclusions These 8 treatment strategies all can promote social functions on the patients with TRD. But the intensity and chronological order of improvement werent the same among 8 groups. The influencing factors on improving social functions are the severity, improvement of depressive symptoms and latest onset time.
