1.Effects of Staurosporine on the Proliferation and Apoptosis of Human Cholangiocarcinoma QBC-939 Cells

Objective To study the effects of protein kinase C (PKC) inhibitor staurosporine (STS) on the proliferation and apoptosis of human cholangiocarcinoma QBC-939 cells and to explore its possible mechanism. Methods CCK-8 was used to detect the effects of PKC inhibitor STS on the proliferation of human cholangiocarcinoma QBC-939 cells. The effects of STS on the ultrastructural characteristics of QBC-939 cells were observed by routine transmission electron microscopy (TEM). The apoptosis rate and the cell cycle distribution of QBC-939 cells were detected by flow cytometry. The expression of cyclin B1,Cdk1 and p-Cdk1 in QBC-939 cells was detected by Western blotting. Results STS could significantly inhibit the proliferation of QBC-939 cells in a dose-dependent manner (P <0. 05) and the half inhibitory concentration ( IC50 ) of QBC-939 cells at 24th and 48th h was 334 nmol·L-1 and 118 nmol · L-1 , respectively. TEM observed that STS could induce typical apoptotic bodies and super-microstructural changes of QBC-939 cells. By Annexin V-FITC/ PI double labeling flow cytometry,we found that the apoptotic rate of QBC-939 cells after treatment with STS for 0,12,24 and 48 h was (10. 16±4. 52)% ,(22. 35±2. 19)% ,(34. 27±2. 30)% and (59. 70±5. 97)% ,respectively. By flow cytometry,compared with the control group,STS could significantly increase apoptosis rate of QBC-939 cells,decrease the percentage of cells in G0 / G1 phase and increase the percentage of cells in G2 / M phase (P<0. 05). Western blotting proved that the expression levels of cyclin B1 and Cdk1 proteins in the STS-treated QBC-939 cells were significantly decreased (P<0. 05),while the expression level of p-Cdk1 protein in the STS-treated QBC-939 cells was significantly increased ( P < 0. 05 ). Conclusion STS can significantly inhibit cell proliferation and induce apoptosis of human cholangiocarcinoma QBC-939 cells and the mechanism may be related to cell cycle arrest at G2 / M phase.

6.Introduction to connection between chemotherapy tolerance, metastasis and micro-environment in breast cancer

This review article briefly described the interaction and inner link between chemotherapy tolerance and metastasis of breast cancer.The common pathways of occurrence of drug resistance and metastasis were analyzed with formation and outcome of breast cancer epithelial-mesenchymal transition (EMT) and with enrichment and function of breast cancer stem cells.The chemotherapy which play positive role in clinical practice was pointed out that may cause negative effects by changing tumor microenvironment.Those evidences show that the theory on tumor microenvironment will help to formulate the research strategies for preventing and reversing resistance of breast cancer to drug and metastasis.
7.Investigation and Evaluation of Selenium Nutrition in Huaibei Inhabitants

Objective To evaluate the selenium nutrition level of Huaibei inhabitants and to supply valid data to Chinese Microelement Nutrition Database. Methods 513 healthy people living in Huaibei for more than 10 years were selected randomly from September 2003 to June 2004. The selenium content in hair was measured by catalytic polarographic method to delimit selenium richness region. According to the selenium content in hair,the intake of selenium from food was estimated. The investigation results were evaluated by the standard of Chinese Dietary Reference Intakes. Results The selenium content in hair was (0.390 3?0.104 5) ?g/g in Huaibei inhabitants,only 51.9% of the people reached the proper level. The average intake of selenium of Huaibei inhabitants was (31.92?9.93) ?g/d,which showed that the selenium nutrition level of Huaibei inhabitants was lower than the standard of Chinese Dietary Reference Intakes except children between 0 to 6 years old. The percent which satisfied RNI and EAR standard occupied 19.9% (102/513) and 32.9% (169/513) respectively,which were also much lower than the 50% of physiological necessary level. Conclusion The selenium intake from food in Huaibei inhabitants was scarce obviously. To increase the selenium intake properly will be a wise selection.
8.EFFECT OF NALOXONE ON .KEMORRHAGIC SHOCK RELATED TO SYMPATHO-ADRENOMEDULLARY SYSTEM

Morphine antagonist naloxone (3mg/kgiv ) induced a rise in blood pressure,pulse pressure & respiratory rate in the rat under hemorrhagic shock. It also improved the survival rate. ?Receptor antagonist pheno-xybenzamine ( 1 mg/kg iv ) or adrenalectomy abolished the beneficial effects of naloxone. Reserpine ( 1 mg/kg ip for 5 d) which depleted catecholamines of periphal sympathetic nervous system could not abolish naloxone effects. Resrpine plus adrenalectomy abolished naloxone actions again. It is suggested that the effect of naloxone on the blood pressure is mainly due to release of catecholamine from adrenalmedu-llary. The results of isolated adrenal perfusion indicated that naloxone could not induce a rise in catecholamine concentration of perfused liquid. It is suggested that the action of naloxone on catecholamine release may not be a result of direct action upontbe chromaffin cells.
9.EBP50:a Novel Cancer Suppressor Protein

EBP50(ERM-binding phosphoprotein-50),a multifunctional adapter protein with 358 amino acids and two PDZ domains, regulates cell growth and migration. Lines of evidences indicate that it is a potential cancer suppressor protein. Loss of heterozygosity (LOH) and intragenic mutation of the ebp50 gene have been found in both primary breast tumors and breast cancer cell lines. EBP50 suppresses the breast cancer cell proliferation via its interaction with many tumor suppressor protein including PTEN, SYK, MERLIN, etc. Here the molecular structure of EBP50, signal pathway regulated by EBP50, and the relationship between breast cancer development and EBP50 are discussed.
10.Application of Endopore implant in inadequate alveolar jaw bone

From March 2004 to June 2007, the effect of the 32 Endopore implants which were placed in 25 patients with the inadequate alveolar jaw bone treated in the Implant Center of Department of Oral and Maxillofacial Surgery, the Affiliated Stomatological Hospital of Anhui Medical University were retrospectively analyzed. All patients were treated with Endopore implants of 5 mm ? 7 mm without any bone grafts simultaneously. Second-stage restoration was conducted 3-6 months later. Regular follow-up was between 1 and 4 years. All of the 32 implants gained osseointegration and were restored. During the follow-up, no implant loosed or lost. No inflammation was observed in the soft tissue around the implants. All the patients were satisfied with the final prostheses. With the unique root surface of porous microscopic titanium particle and the shorter design, Endopore implants can be used in the posterior area of inadequate alveolar jaw bone. They could improve the indication of the dental implant.
