Objective To observe the effects of interferon ? (IFN?) on thyroglobulin (TG) and thyroid peroxidase (TPO) gene expressions in FRTL5 cells induced by thyroid stimulating antibody (TSAb). Methods TSAb crude fraction was extracted by polyethylene glycol 4000. Recombinant rat interferon ? (0, 1, 10, 10 2, 10 3 U/ml) was added to the FRTL5 cells induced by TSAb, and then expressions of TG, TPO mRNA were measured by Northern blot, the cell growth was measured by 〔 3H〕 thymidine incorporation. Results (1)TSAb increased TG, TPO gene expressions, and 〔 3H〕 thymidine incorporation. (2)Interferon ?inhibitedTSAb inducedTG, TPOgeneexpressionsand〔 3H〕 thymidine incorporation. Conclusion Interferon ? inhibits the growth and function of thyrocytes induced by TSAb, which suggests that interferon ? might regulate thyroid function in Graves' disease.

OBJECTIVE To investigate the biological and molecular biological characteristics of Klebsiella pneumoniae in burn patients in order to give the first hand information for preventing and controlling of hospital acquired infections.METHODS The identification was done by Bio-Merieux ATB expression.Antimicrobial susceptibility test was performed with K-B method. The plasmid DNA was extracted by Alkaline Lysis, and separated by electrophoresis on the gel.The ESBLs detection was based on NCCLS.RESULTS The K.pneumoniae from the burn patients and the environment were sensitive to CIP,FOX and IPM,but showed resistance to the rest 12 antibiotics. The plasmid DNA profile analysis showed 3 types, and the relative molecular mass was approximately 4.7?106,3.6?106 and 2.0?106.The molecular biological characteristics showed these pathogens were ESBLs-producing K.pneumoniae, which was different from the control bacteria. At the same time, the pathogens caused the original infection were detected, and they were accordingly Staphylococcus aureus,Streptococcus pyogenes, and Pseudomonas aeruginosa.CONCLUSIONS The outbreak in burn patients is caused by ESBLs-producing K.pneumoniae, which has the same antibiotic resistance spectrum and plasmid DNA profile. This ESBLs-producing K.pneumoniae has the same origin. The pathogen might be transmitted by the case history clips and the door knobs. It was suggested that something must be done to enhance the antisepsis administration in order to prevent the hospital acquired infection.

[Abstract ] Objective The purpose of this study was to observe the effects of dl-3n-butylphthalide (NBP) sodium chloride injection post-processing on the expressions of X-inhibitor of apoptosis (XIAP) and Bcl-2/adenovirus E1B19kDa interacting protein 3 (BNIP3) in the hippocampus CA1 neurons of focal cerebral ischemia reperfusion (IR) rats, and to investigate the brain-protection mechanisms of NBP. Methods A total of65 adult male Sprague-Dawley rats were divided into five groups of equal number, sham op-eration, IR, and low-,medium -and high-dose NBP, according to the random number table. The IR models were established by modified ligation of the middle cerebral artery. The animals in the NBP groups received intra-abdominal injection of NBP at 2, 4, and 6 mg/kg, re-spectively.All the rats were sacrificed at 24 hours after modeling, neurological scores obtained by Zea Longa, the volume of infarction measured by TTC staining, the number of apoptotic cells counted by TUNEL, and the expressions of XIAP and BNIP3 detected by immunohistochemistry and real-time PCR. Results The neural function defect scores were markedly lower in low-, medium-and high-dose NBP groups than in IR model rats (P<0.05), with statis-tically significant differences among the three dose groups (P<0.05).The volume of infarction was remarkably higher in the low-dose than in the medium-and high-dose NBP groups (P<0.05).The number of apoptotic cells in the hippocampus CA1 neurons was de-creased in the NBP groups as compared with the IR models (P<0.05).The XIAP-and BNIP3-positive cells were significantly in-creased in the IR model rats as compared with the sham operation group ([22.31 ±0.94] and [60.13 ±2.59]/HP vs [3.07 ±1.43] and [5.78 ±0.44]/HP, P<0.05).In comparison with the IR models, the NBP-treated rats showed a progressively increased number of XIAP-positive cells in low-, medium-, and high-dose groups ([28.70 ±1.18], [32.79 ±0.88], and [37.01 ±1.24]/HP) (P<0.05) but a decreased number of BNIP3-positive cells in the three dose groups ([52.07 ±1.02], [40.30 ±2.00], and [31.04 ± 0.43]/HP) (P<0.05).Similarly, the expression of XIAP mRNA was up-regulated while that of BNIP3 mRNA down-regulated in the NBP treatment groups as compared with the IR model rats, both in a dose-dependent manner (P<0.05). Conclusion NBP post-processing has a neuroprotective effect on IR rats, which is associated with its impact on the expressions of XIAP and BNIP3.

This study is to establish a simple and practical co-culture method of cortical neurons and astrocytes of rats. The cortex of the new-born SD rats was digested by 0.125% pancreatic enzyme, and the differential adherence was applied to obtain the mixed cell suspension of neurons and astrocytes. A low concentration of cytarabine was used to inhibit the astrocytes in a moderate way to get neuronal and astrocyte co-culture. The morphological characteristics of the cells in different times were observed under the inverted microscope. The cells began to adhere the wall 2 h after the inoculation. Neurons and astrocytes grew in a good condition under the inverted microscope 9 days after the inoculation. The results of the immunofluorescence staining and Rosenfeld's staining indicated that the co-culture of neurons and astrocytes was successful and the ratio of neurons and astrocytes was close to 1:1. A new neurons and astrocytes co-culture method, which is simple and convenient, was successfully established. It will be an efficient method for the related researches about neuronal and astrocyte co-culture in vitro.

Objective To ratiocinate the formula of relationship between opening size of laminoplasty and the increment of sagittal canal diameter following double-door cervical laminoplasty and to predict the increment of sagittal canal diameter and the cross sectional area of canal according to the opening size of laminoplasty. Methods Twenty patients (12 males and 8 females) with multilevel cervical spondylotic myelopathy had undergone double-door cervical laminoplasty (C3-C7 in 9 patients and C3-C6 in 11 patients,89 segments) in our institution between September 2010 and January 2013.The formula describing the relationship between the opening size of laminoplasty and the increment of sagittal diameter was deduced. The parameters of pre-and post-operative computed tomography scans of 20 patients who had undergone laminoplasty surgery were measured by picture archiving and communication system (PACS) software, and the increment of sagittal canal diameter and the cross sectional area of canal were predicted when the opening size of laminoplasty were 6 mm,8 mm,10 mm,12 mm,14 mm and 16 mm according to the formula. Results Increment of sagittal canal diameter and canal area respectively showed significant difference in the same segment laminoplasty.Increment of sagittal canal diameter between various groups in the same segment (C3-C6) showed significant difference.Increment of sagittal canal diameter between the opening size of 14 mm and 16 mm in C7 laminoplasty showed no significant difference.Increment of sagittal canal diameter was increased steadily following C3-C7 double-door laminoplasty with opening sizes of 6 mm,8 mm,10 mm,12 mm,14mm and 16mm,but the increasing trend in sagittal canal diameter diminished gradually. Conclusion Increment of sagittal canal diameter and canal area following C3-C7 laminoplasty can be accurately predicted according to the opening size of laminoplasty by this formula. The formula can help operator to perform double-door cervical laminoplasty based on accurate individual laminoplasty opening size, which prevents inadequate or excessive opening.
