ObjectiveTo observe prophylactic effect and reliability of warfarin n preventing recurrant ischemic stroke.Methods168 patients identified as ischemic stroke admitted in Chaozhou Central Hospital from March 2001 to May 2004 were randomly divided into 2 groups,warfarin group and ticlopidine group (set as control).82 cases were in warfarin group,receiving warfarin 0.75~2.25mg/d orally.Prothrombin and international normalized radio (INR) were monitored,and INR was 1.5~2.0.86 cases were in ticlopidine group;they received 250mg/d ticlopidine orally.All patients were followed up every monty for 3 years,and recurrent ischemic stroke and drug side-effect were documented.ResultsThere were 8 cases (9.76%) of recurrence in warfarin group and 26 cases (30.02%) of recurrence in ticlopidine group.There was statistically significant difference between the two groups (P

4.Research progress on mechanism and premedication regimen of taxanes hypersensitivity reactions

Hypersensitivity reactions ( HSRs) are the serious problems of taxanes in the clinical appli- cation. These reactions range in severity from mild flushing and itching to bronchospasm, dyspnea and signifi-cant alterations in blood pressure with loss of consciousness or not, and death in some rare cases. Nevertheless, HSRs are often reported only sporadically in clinical trials or as case reports. The vague or inconsistent termi-nology used to describe these reactions may reflect our poor understanding of their pathophysiology. This article will reviews the advancement of taxanes HSRs.
5.Total hip arthroplasty via a direct anterior minimally invasive approach aided by lower extremity tractor for hip diseases

Objective To evaluate the short-term clinical results of total hip arthroplasty(THA) through a direct anterior minimally invasive approach aided by lower extremity tractor for hip diseases.Methods Between August 2013 and August 2015,115 patients (138 hips) were admitted for hip diseases.They were 71 men and 44 women,aged from 28 to 77 years (average,59.4 years).Their body mass indexes ranged from 19.7 kg/m2 to 28.9 kg/m2,averaging 24.7 kg/m2.Their hip diseases included avascular necrosis of the femoral head in 56 cases (with an average disease course of 15.7 months),hip osteoarthritis in 35 cases (with an average disease course of 12.6 months),hip dysplasia in 14 cases (with an average disease course of 2.8 years) and inflammatory arthritis in 10 cases (with an average disease course of 6.8 years).All the patients were treated by primary THA through a direct anterior minimally invasive approach and assistance of lower extremity tractor.Results All the 115 patients (138 hips) were followed up for an average of 31.6 months (from 12 to 40 months).Their average length of incision was 9.4 cm,average operative time 115.9 min,average blood loss 210.2 mL,and average hospital stay 7.5 days.Their postoperative imaging results were as follows:the angles of acetabular abduction averaged 41.6° (from 37° to 56°) and the angles of acetabular anteversion 18.1° (from 11° to 23°).Their average Harris hip score one year postoperatively was 91.4 points (from 80 to 96 points).Split fracture of the proximal femur happened in one patient (one hip) (0.7%) intraoperatively;injury to lateral femoral cutaneous never was observed in 6 patients (7 hips) (5.1%).Complications like deep infection,hip dislocation,deep vein thrombosis or prosthetic loosening were not observed during follow-ups.Conclusions THA with assistance of lower extremity tractor through a direct anterior minimally invasive approach can lead to fine short-term clinical results,including quick recovery of hip function and decreased intraoperative and postoperative complications.
6.Construction of eukaryotic expression plasmid carrying interleukin-18 receptor alpha,beta and establishment of cell line stably expressing interleukin-18 receptor beta

AIM:To construct human interleukin-18 receptor ?(IL-18R?) and interleukin-18 receptor ?(IL-18R?) eukaryotic expression vectors named pcDNA3.1-zeocin/IL-18R? and pcDNA3.0/IL-18R? respectively,and then stably transfect pcDNA3.0/IL-18R? into 293 cells in order to establish the cell line stably expressing IL-18R? and cell model which responds IL-18 signal transduction in vitro. METHODS:The experiment was performed at the laboratory of Department of Immunology,Peking University Health Science Center from June 2006 to May 2007. DNA encoding IL-18R? or IL-18R? was amplified and cloned into pcDNA3.1-zeocin or pcDNA3.0 vector,respectively. The recombinant of pcDNA3.0/IL-18R? was transfectd into 293 cells. After screening culture by G418,stable transfected cell line was established and the expression of IL-18R? was identified by Western-blot assay. The stable cell line with the combinant eukaryotic expression vector pcDNA3.1-zeocin /IL-18R? to establish the IL-18 signaling pathway,which was detected by NF-?B-dependent Luciferase. RESULTS:The eukaryotic expression vectors of IL-18R? and IL-18R? were constructed successfully. The gene of IL-18R? stably expressed in 293 cells and cell model of IL-18 signaling transduction in vitro could be established in these stable 293 cell lines. CONCLUSION:The establishment of NF-?B-activated cell model in vitro which responds to IL-18 can provide solid foundation for further experimental studies on IL-18 signaling transduction pathway.
7.Icariin promotes the osteogenic differentiation of mesenchymal stem cells:a good candidate for bone defect repair

BACKGROUND:Recent studies have shown that icari n is a good bone-inducing factor that can promote the osteogenic differentiation of mesenchymal stem cel s, providing a new hope for the treatment of bone defects. OBJECTIVE:To review research achievements in pharmacological effects of icari n effects on bone tissue metabolism as wel as its effect to promote osteogenic differentiation of mesenchymal stem cel s. METHODS:The first author retrieved CNKI and PubMed databases for relevant Chinese and English literatures using keywords of“icariin, stem cell, osteogenesis”, respectively. Articles regarding icariin, stem cells, osteogenesis were included, and repetitive studies were excluded. Totally 754 articles were retrieved initially. In accordance with inclusion and exclusion criteria, 41 articles were included in result analysis. RESULTS AND CONCLUSION:As the main ingredient of Herba epimedi , icari n functions as a good osteogenetic growth factor to promote the osteogenic differentiation of bone marrow mesenchymal stem cel s. In recent years, icari n has been shown to promote adipose-, umbilical cord-, and periodontal ligament tissue-derived mesenchymal stem cel s to differentiate into osteoblasts. But such studies are less reported. Until now, mesenchymal stem cel s stil exhibit unsatisfactory osteogenic ability in in vivo experiments. Given this, osteogenetic growth factors contribute to the osteogenic differentiation of mesenchymal stem cel s. Therefore, the use of icari n is expected to provide a good strategy for bone defect repair.
8.Screening of proteins interacting with neuropeptide CART

Objective The objective was to look for cocaineand amphetamine-regulated transcript peptide(CART) receptors or proteins interacted with CART.Methods A cDNA library from rat brain was constructed.Then the BacterioMatch two-hybrid system was used to screening for proteins interacted with CART by using CART41-102 as bait.Results The cDNA library and bait pBT-CART41-102 were constructed successfully.The library harvested 3.37?106 individuals,and the reconstruction rate was 98.5%.5.01?106 cotransformed clones were screened,93 clones were conformtively identified to interacting with CART and were DNA sequenced.By bioinformation analysis,6 known proteins interacted to CART were identified.Other unknown interaction proteins with short exactly similiar motif to 22 membraneproteins or receptors were also identified.Conclusion CART might interact to proteins for energy balance,cell proliferation and enzymes related to protein processing and degrading.
9.Anti-DNase B used in the epidemiologic study of rheumatic fever in the schoolchildren

Objective To explore the epidemic conditions of rheumatic fever (RF) with a simple method at some regions.Methods Antistreptococcal DNase B test microtiter method was adopted to detect human sera antibody in natural populations who are responsible to RF.All 216 schoolchildren aged 10~12 years old were chose by the random cluster sampling.The schoolchildren′s serum antibody against DNase B in each season from september 1988 to August 1989 was detected,and compared with antistreptolysin “O” (ASO);furthermore,5 different regional schoolchildren′s serum antibody to DNase B was tested, and 100 schoolchildren aged 10~13 years old were chose in every region.Meanwhile,the RF incidence of 5~18 year schoolchildren was investigated for 4 years,and approximately 364 915 person times were inquired.All the results of the anti DNase B and ASO were compared with the schoolchildren′s RF incidence.Results ① The levels of the anti DNase B were higher during fall,winter and spring seasons,and lower in summer,which were coincided with the RF incidence,and linear relation was very good ( r =0 913, P
10.Clinical analysis of 57 cases of thyroid disease with concomitant focal lymphocytic thyroiditis

Objective To evaluate the methods of diagnosis and treatment of thyroid disease with concomitant focal lymphocytic thyroiditis(FLT), and explore the reasons for its confused with Hashimoto′s disease(HD).Methods During the recent 25 years, 207 patients underwent surgical trearment for pathologically diagnosed HD.Among this group, 143 cases of HD with other concomitant thyroid disease were retrospectively analysed.Results Of the 143 cases, 57 cases were found to have thyroid disease with concomitant FLT, and this was 27.5%(57/207) of the total HD group, or 39.9%(57/143) of the group with thyroid disease and concomitant HD.Intraoperative pathologic section revealed that focal lymphocytic infiltration was positive in 87.7%(50/57) of cases. The postoperative hypothyroidism occurrence rate was 19.3%(11/57), of which, 7 cases(7/57, 12.3%) were subclinical hypothyroidism.Conclusions The character of pathologic changes of thyroid disease with FLT and with HD was different. Intraoperative pathologic section can be helpful in the diagnosis of this condition and can have important significance as a guide to the scope of (surgical) resection of the thyroid gland.
