1.Immunological tolerance of rat pancreatic islet transplantation induced by transfusion of donor apoptotic splenocytes

Objective To investigate the effect of transfusion of allogeneic apoptotic splenocytes on immunological tolerance to pancreatic islets transplantation in rat. Method Recipient SD rats were rendered diabetic mellitus by a single intraperitoneal injection of streptozotocin (STZ). The apoptosis of donor Wistar rat splenocytes was induced by X-irradiation with electron linear accelerator. Pancreatic islets were isolated by intraductal collagenase P, and were purified by Ficoll-400 discontinuous density gradient after culturing for 1-2 days from donor Wistar rat. The diabetic SD rats were randomly divided into 4 groups, and respectively received infusion of Hanks solution, donor normal splenocytes, donor apoptotic splenocytes, and donor necrotic splenocytes via dorsal veins of penis, and then they received pancreatic islets from the same donor rat underneath their renal subcapsular space 7 days later. Mixed lymphocyte reaction (MLR) was used as immunological parameter of transplantation tolerance besides the observation of blood glucose level and allograft survival time. Islet graft failure was defined as the presence of a plasma glucose concentration of at least 11.0mmol/L on 2 successive days. Results The pre-infusion of donor apoptotic splenocytes significantly prolonged the survival time of allograft as shown by the median survival time (MST) of graft of 31days, and the longest survival time of 42 day, and it apparently decreased the rejection response in recipient SD rat in one-way mixed lymphocyte culture. Conclusion Transfusion of donor apoptotic splenocytes could induce immunological tolerance of allogenic islets transplantation in rat.
3.Effect of different time administration of parecoxib sodium on postoperative analgesic efficacy in patients undergoing thoracic surgery

Objective To investigate the effect of different time administration of parecoxib sodium on the postoperative analgesic efficacy in patients undergoing thoracic surgery. Methods This was a prospective,randomized, double-blind, placebo-controlled study. Sixty ASA Ⅰ orⅡ patients aged 17-83 yr undergoing pulmonary lobectomy were randomly allocated to one of 3 groups (n=20 each):A, B and C groups. Group A received normal saline 2 ml at 30 min before skin incision and the end of operation. Group B received iv parecoxib sodium 40 mg at 30 min before skin incision and normal saline 2 ml at 30 min before the end of operation. Group C received normal saline 2 ml at 30 min before skin incision and iv parecoxib sodium 40 mg at 30 min before the end of operation. All the patients received patient-controlled intravenous analgesia with morphine and VAS score was maintained≤3. The patients were followed up after operation.The morphine consumption, patients' global evaluation of the postoperative analgesia (0-100, 0=worst pain, 100=no pain), nausea and vomiting, body temperature , volume o chest drainage, hepatic, renal and blood coagulation function were recorded. Results Compared with group A, the morphine consumption was significantly reduced, the patient' s satisfaction score increased and body temperature decreased in B and C groups(P<0.05 or 0.01). There was no significant difference in the morphine consumption, patient's satisfaction score and body temperature between B and C groups(P>0.05). No significant difference was found in the parameters of hepatic, renal and blood coagulation function, volume of chest drainage and incidence of nausea and vomiting among the three groups(P>0.05).Conclusion When postoperative analgesia is assisted with iv parecoxib sodium 40 mg given at 30 min before skin incision or at 30 min beforethe end of operation,the efficacy is similar,and both can improve the postoperative analgesic efficacy of morphine and reduce fever after operation in patients undergoing thoracic surgery.
4.Optimization of the Extraction Technology of Ardicrenin from Ardisia crenata by Orthogonal Experiment

OBJECTIVE: To optimize the extraction technology of ardicrenin from Ardisia crenata.METHODS: The content of ardicrenin was determined by RP-HPLC. The extraction technology was optimized by orthogonal experiment with the extraction rate of ardicrenin as evaluation index and with the volume fraction of ethanol,the extraction temperature,extraction time and extraction times as factors.RESULTS: The optimum extraction technology was determined as follows: 80%ethanol was used as solvent;the extraction temperature was 80 ℃;the extraction time was 20 min and the extraction was conducted for 3 times.The extraction rate of ardicrenin was 6.04%.CONCLUSION: The optimized extraction technology is feasible and reproducible,and it provides theoretical basis for mass extraction of the ardicrenin from A.crenata.
5.Simultaneous determination of five flavonoids in Duchesnea indica at five picking time by HPLC

AIM To establish an HPLC method for the simuhaneous content determination of five flavonoids in Duchesnea indica (Andr.) Focke at five picking time (April,May,June,July and August).METHODS The analysis of D.indica methanol extract was performed on a 25 ℃ thermostatic Agilent ZORBAX SB-C18 column (250 mm×4.6 mm,5 μm),the mobile phase comprising of acetointrile-0.1% methanoic acid flowing at 1.0 mL/min in a gradient elution manner,and the detection wavelength was set at 350 nm.RESULTS Rutin,hyperoside,isoquercitrin,celereoin and kaempferol showed good linear relationships within their own ranges (R2 ≥0.998 6),whose average recoveries were 97.1%-101.5% with the RSDs of 1.37%-2.37%.The contents of five constituents in samples at different picking time exhibited obvious differences,among which lutin and hyperoside contents were the highest in June,isoquercitrin content was the highest in July,and celereoin and kaempferol contents were the highest in August.CONCLUSION The suitable picking time of D.indica is June and July.
6.Change of immunological response and cell proliferation in breast augmented by polyacrylamide hydrogel injection

Objective:To study the change of immunological response and cell proliferation in breast tissues augmented by polyacrylamide hydrogel injection(PHI).Methods:The expression of CD68,CD25 and PCNA in 20 breast tissues with indurations,12 without indurations after breast augmentation by PHI,and 10 normal breast tissues was examined by immunohistochemistry P-V6000; analysis was also done by H-E staining.Results:Hyperplasia of fibrous tissue and infiltration of inflammatory cells and macrophages were found in the breast and adjacent tissues 3-8 years after PHI.Positive cells of CD68,CD25 and PCNA hardly existed in the normal tissues,but the breast tissues around the polyacrylamide hydrogel had many positive cells of CD68 and PCNA,especially in cases with indurations;there were significant differences between the 3 groups(P
7.Application of magnetic resonance imaging in diagnosis and treatment of pelvic neoplasms

Objective To study the value of magnetic resonance imaging(MRI) in the diagnosis and treatment of pelvic neoplasms. Methods The patients who were suspected of malignant pelvic neoplasms were performed MRI.The surgical approach was determined according to the MRI results.The MRI results were compared with operation findings and pathological reports. Results The MRI results were consistent with operation findings and pathological reports in 22 cases highly suspected of pelvic neoplasms and 15 cases with abnormal endometrial echo.Therapeutic regimen was designed for 5 cases with uterine cervix cancer according to MRI stages,and favourable surgical curative effects were obtained. Conclusion MRI provides a reliable guideline for surgical approach and may improve the outcomes.
