2.Effect of high glucose on the expression of KLF6 in human lens epithelial cell

Background Epithelial-mesenchymaltransition (EMT)isamajorcontributortothe pathogenesis of posterior capsular opacification(PCO).Kruppel-like factor 6 (KLF6) is a zinc finger protein,which can be stimulated by high glucose in proximal tubule cells and involved in transforming growth factor beta (TGF-β)induced EMT of diabetic nephropathy.ObjectiveThis study was designed to investigate the effect of high glucose on the expression of KLF6 and its target genes( TGFB1,TGFBR1,COLIA1,HSP47) in human lens epithelial cells (LECs).MethodsHuman LECs(SRA01/04) were cultured and exposed to different concentration of glucose.The expressions of KLF6 mRNA and protein were analyzed by real time polymerase chain reaction( real time PCR) and western blot after treatment with high glucose.The expressions of KLF6 target genes were analyzed by real time PCR to evaluate the EMT of SRA01/04 cells.ResultsCompared with the control group(5.5 mmol/L),the relative mRNA levels of t-KLF6 and wt-KLF6 in SRA01/04 treated with high glucose(22.2,44.4,66.6 mmol/L) increased obviously (F =72.53,42.02,P<0.01 ).Then,the concentration of 22.2 mmol/L was used in the next experiments.The relative mRNA levels of t-KLF6 and wt-KLF6 increased to the peaks after treatment with high glucose for 12 h,and began to decrease after 24 h until lower levels after 48 h ( F =100.12,125.52,P < 0.01 ).Western blot showed that the expression of KLF6 protein was also upregulated by high glucose treatment.With the promotion of the expression of KLF6 gene,the relative mRNA levels of TGFB1,TGFBR1,COLlAl and HSP47 of treated cells also respectively increased after treatment for 12 h,and began to decrease after 24 h until nearly at the levels of the control groups after 48 h( F=6.73,162.35,64.39,12.05,P<0.05 ).ConclusionsIt was concluded that high glucose induced the expression of KLF6 in human LECs,and KLF6 transiently stimulated the expression of target genes TGFB1,TGFBRl,COLlAl and HSP47 which were mainly involved in the mechanism of EMT.
3.Determination of c-fos mRNA in U937 cell line with in situ hybridization

The levels of c-fos mRNA in U937 cell line were determined with in situ hybridization.Theresults showed that TPA induced the expression of c-fos gene with a relationship of dosage-ef-fect.It was also proved that GM-CSF,TNF,IL-9,IL-3 and IL-6 increased the expression of c-fos gene.
4.Clinical analysis of antibiotic associated diarrhea in children with severe bacterial pneumonia

Obiective To study the clinical characteristics and therapeutic measures of the antibiotic associated diarrhea (AAD) in children with severe bacterial pneumonia. Methods The information of antibiotic use history in children with severe bacterial pneumonia from January 2013 to June 2013 was retrospectively analyzed. Results The incidence rate of AAD was 70.59%in children with severe bacterial pneumonia. According to the frequency, the third-generation cephalosporins, penicillin and carbapenems were the major causes of AAD. The percentage of the age≤3 y, the duration of antibiotic usage>7 d and the number of antibiotics≥2 were significantly higher in AAD children than in non-AAD children (χ2=89.81~119.45, P<0.001). All the patients were improved after cessation or change of antibiotics. Conclusions The incidence of AAD in children with severe bacterial pneumonia is high, which is related to the age, the duration of antibiotic usage and the combination use of antibiotics.

Methods One hundred and fifty Han Chinese adults (ratio of male to female,1∶1)were randomly chosen and parameters for computer aided angular analysis of facial soft tissues included:lateral angles:∠Ns Prn Pgs;∠Ns Prn GsPgs;∠Ns Prn Sn; As Ns Bs;∠Ns Sn Pgs;∠Gs Ns Prn;∠Cm Sn Ls and ∠Li Bs Pgs. front angles: ∠Zyr Rc Mes; ∠Zyl Lc Mes; ∠Exr Exl Sn; ∠Exl Exr Sn; ∠Exr Sn Exl; ∠Enl Enr Sn; ∠Enr Enl Sn; ∠Enr Sn Enl; ∠Sn Chr Chl; ∠Sn Chl Chr and ∠Chr Sn Chl.Results Lateral angles ∠Ns Prn Sn and ∠Li Bs Pgs,in male were larger significantly than that those in female, but ∠Gs Ns Prn in male was smaller significantly than that in female. Front angles,∠Zyr Rc Mes and ∠Zyl Lc Mes,in male were larger significantly than those in female.Conclusion Angular analysis plays a unique role in the measurement of facial soft tissue profile. There is a difference of facial soft tissue profile between male and female Han Chinese.

9.The potential effects of Lactobacillus acidophilus on the prevention and treatment of neonatal mice infected with human rotavirus

Objective To investigate the potential effect of Lactobacillus acidophilus (L. acidophilus) on prevention and treatment of neonatal mice infected with human rotavirus (HRV). Methods Sixty 4-day-old kunming mice were randomly divided into control group. HRV infected group, L. acidophilus pretreated group (treated before HRV infection ) and L. acidophilus treated group(treated after HRV infection). The manifestation and pathological changes in small intestine of neonatal mice were observed. The HRV antigen in the feces and intestines were measured by ELISA and fluorescent-focus assay, respectively. Results The severity and duration of diarrhea as well as mortality in L. acidophilus pretreated group and treated group were lower than those in HRV infected group. The duration of HRV-antigens shedding following infection was considerably prolonged in HRV infected group compared to that in L. acidophilus pretreated group and treated group. Furthermore, decreased expression of HRV antigen and little pathological changes in intestinal mucosa were found in L. acidophilus pretreated group and treated group when compared with HRV infected group. Conclusion L. acidophilus may be used as an alternative approach for the prevention and treatment of neonatal mice infected with HRV.
10.A Review on Virtual Cystoscopy Techniques

Objective: Bladder cancer is the ninth cause of cancer deaths and has high recurrence rate after resection of the tumors. Cystoscopy is the current most accurate method for investigating the bladder abnormalities. However, it is expensive, uncomfortable and invasive. It is possible to induce bleeding, urinary-tract infection and even puncture of bladder. Advances in medical imaging and computer technologies make virtual cystoscopy a potential alternative. Methods: Computed tomography (CT) and magnetic resonance imaging (MR/) are the preferred imaging modalities for virtual cystoseopy to get clear structural images or (and) functional images of the bladder. The boundary of bladder is segmented manually or automatically, and then the bladder is reconstructed and displayed by surface rendering. Useful features are extracted from the images and expressed o nthe reconstructed bladder to provide more valuable diagnosis information for doctors. Results: Comparing with conventional cystuscopy, virtual cystuscopy is noninvasive, more convenient, flexible and can provide more useful diagnosis information as well. Conclusions: Virtual cystoscopy is a promising method of detection and reexamination of bladder cancer. So far, more researches are needed for the virtual cystoscopy before clinical application. It clinical and commercial value are under investigation.
