1.Update on sublingual immunotherapy in patients with asthma due to house-dust mites

Sublingual immunotherapy(SLIT) has received increasing attention as a treatment for asthma due to house-dust mites. The precise mechanisms of SLIT remain unclear, but SLIT has been shown to modify the balance of Th1/Th2, increase IgG4 and decrease IgE. The allergen is captured within the oral mucosa by Langerhans-like dentritic cells, suggesting that such cells are prone to induce tolerance. Increasing data support the safety, efficacy and good compliance with SLIT. However, the precise mechanisms, optimal dosage and duration of SLIT require further investigation.
2.Advances in relationship between plexinC1 and lung diseases

PlexinC1 is a large transmembrane protein widely distributed,which belongs to receptor system. PlexinC1 is expressed by monocyte,T-cells,B-cells,dendritic cells,neutrophils,and platelets. Recent researches showed that plexinC1 was intensely associated with lung injury,asthma and pulmonary fibrosis. PlexinC1 binded to its corresponding ligand and activated cellular signal pathway,which then participating in pathophysiologic processes of diseases mentioned above. Reasonably,PlexinC1 influences the development, prognosis of lung diseases and would provide a new target in control and treatment of lung diseases. This pa-per reviewed simply the formation,distribution,function of plexinC1 and the role of it playing in the associat-ed lung diseases.

4.Observation on the effect of Liqidingchuan pill combined with Dexamethasone on the treatment of the patients with acute asthma

Objective To observe the clinical efficacy of LiQiDingChuan pill combined with Dexamethasone on the treatment of patients with acute asthma.Methods 93 patients with acute asthma from July 2015 to September 2016 were randomly divided into the observation group(47 cases)and the control group(46 cases).The control group were received dexamethasone, and the observation group were given LiQiDingChuan pill combined with Dexamethasone.The main symptoms disappeared time, forced expiratory volume in 1 second, peak expiratory flow rate value, hs-CRP, TNF-α and clinical efficacy were recorded and analyzed.Results ①After treatment, wheezing cough, chest tightness and wheezing disappeared time were(4.71±0.59)d,(2.85±0.49)d,(4.93±0.70)d in the observation group, which were shorter than(7.76±1.02)d,(5.16±0.74)d,(8.67±1.12)d in the control group, and the differences were statistically significant(P<0.05).② After treatment, FEV1 and PEF levels were(65.69±3.74)V/L,(218.92±16.09)L/s, which were better than(57.45±3.62)V/L,(176.34±15.14)L/s in the control group, and the differences were statistically significant(P<0.05).③ After treatment, hs-CRP and TNF-α levels were(6.76±0.78)mg/L,(7.99±1.87)pg/mL, which were better than(6.76±0.78)mg/L,(7.99±1.87)pg/Ml in the control group, and the differences were statistically significant(P<0.05).④The total effective rate in the observation group was 91.49%, which was significantly higher than 71.74% in the control group, and the differences were statistically significant(P<0.05).Conclusion Liqidingchuan pills combined with dexamethasone treatment has a good therapeutic effect on acute phase of asthma, which can reduce asthma related symptoms, reduce the inflammation level in the patients, optimize the forced expiratory volume in 1 second and peak expiratory flow rate.The combination is better than the single drug.
5.Preparation and Pharmaceutical Characterization of Lansoprazole Liposomes

OBJECTIVE:To prepare and characterize lansoprazole(LAP) cationic liposomes. METHODS:Liposomes were prepared by ethanol injection technique.An orthogonal test was utilized to optimize the formulation and preparation of LAP liposomes.The encapsulation efficiency was determined by ultrafiltration.The morphological examination of LAP liposomes was performed using transmission electron microscopy.The particle size and Zeta potential of the liposomes were measured.The release rate of LAP from liposomes was tested. RESULTS:The liposomes with spherical or ellipsoidal shape and better stability featured the encapsulation efficiency of(80?1.23)%,the mean partical size of (184?21)nm,and Zeta potential of (36.1?5)mV.The release kinetics in vitro obeyed first-order equation.The stability of LAP was better. CONCULSION:The selected formulation and preparation technic of lansoprazole liposomes were rational and stable and liposomes featured a sustained release in vitro.
6.Study on the Identification and Potency Determination of L-asparaginases from 2 Kinds of Strain

OBJECTIVE: To discuss the identification method of L-asparaginases prepared from E.coliASI.357 and Erwinia carotovora and the optimal enzymatic reaction conditions of potency determination.METHODS: HPLC method and isoelectric focusing electrophoresis were applied for the identification of L-asparaginase from 2 kinds of strain.The effects of category of buffer solution and pH value on enzymatic reaction of potency determination of L-asparaginase were investigated.RESULTS: HPLC chromatogram of L-asparaginases from E.coli ASI.357 was different from that from Erwinia carotovora.The retention time of the peaks were 11.0 min and 11.8 min.The isoelectric point (PI) of L-asparaginase produced from E.coliASI.357 was within 4.65~5.1 and that produced from Erwinia carotovora was within 7.1~8.20.The optimal enzymatic reaction conditions of potency determination of L-asparaginase produced from E.coliASI.35 were Tris-HCl (pH=9.0) as buffer and that produced from Erwinia carotovora was 0.2 mol?L-1 phosphate (pH=8.0) as buffer.CONCLUSION: The isoelectric point (PI) of L-asparaginases produced from 2 kinds of strain is different from each other as well as their optimal enzymatic reaction conditions of potency determination.The L-asparaginases from 2 kinds of strains should be controlled as 2 different categories.
9. Acute toxicity of total polymethoxyflavones from Bauhinia championii in mice and its effects on gastric ulcer

Objective To study the acute toxicity of total polymethoxyflavones from Bauhinia championii (PMFBC) in mice and its effect on experimental gastric ulcer. Methods The acute toxicity of PMFBC was investigated by Bliss method. Three kinds of gastric ulcer model including indomethacin, ethanol, and water immersion-restraint stress were used to observe the effect of PMFBC in mice. Results The LD50 value of ig administration PMFBC on mice was 1 252.319 mg/kg, which showed acute toxicity; PMFBC had protective effect on three gastric ulcer models, which could significantly reduce the ulcer area of the mice, and had a high inhibition rate. Conclusion PMFBC has obvious inhibition effect on the fomation of gastric ulcer in mice, which can be safty used in a certain dose range.
10.Effect of inositol requiring enzyme 1-mediated endoplasmic reticulum stress in liver cell apoptosis of experimental fulminant hepatic failure and its significance

Objective To study the role of inositol requiring enzyme 1 (IRE1)-mediated endoplasmic reticulum stress on hepatocyte apoptosis of experimental fulminant hepatic failure (FHF).Methods Thirty male depuratory Wistar rats were manufactured to be FHF model by peritoneal injection of D-galactosamine (D-GalN) and lipopolysaccharide (LPS),and 30 rats were injected peritoneally with 0.9% sodium chloride solution as controls.The apoptosis of liver cells was detected by flow cytometry.The protein and mRNA expressions of Caspase-12 and IRE1 in liver tissues were detected by immunohistochemistry and reverse transcriptation-polymerase chain reaction (RT-PCR).The independent samples were compared by Kruskal-Wallis H test.The comparison between two groups at the same time point was done by Mann-Whitney U test.The correlation analysis was done by rank correlation.Results The apoptotic rates of liver cells at 2,4,8 and 12 hours were increased over time in model group (x2 =25.475,P=0.01),which were higher than control group (U=0,P<0.01).The expressions of Caspase-12 and IRE1 proteins in liver tissues were upregulated in model group,while the expressions were not detected in control group.The expressions of Caspase-12 and IRE1 mRNA in model group were also increased over time and peaked at 8 h,then gradually decreased; the differences among different time points were statistically significant (x2 =23.983,x2 =24.820; both P<0.01),and all higher than control group (U=0,P<0.01).IRE1 was positively correlated with both Caspase-12 and hepatocellular apoptotic rate (r=0.733 and 0.715,respectively;both P<0.01).Caspase-12 was positively correlated with hepatocellular apoptotic rate (r=0.586,P<0.01).Conclusions IRE1 mediated endoplasmic reticulum stress on hepatocyte apoptosis is closely related to the development of FHF.The earlier intervention on endoplasmic reticulum stress pathway,the more protective effect in liver failure.
