5.Generation of reference fingerprint using principal component analysis

AIM: A novel approach to generate reference fingerprint, named principal component analysis method(PCA), was proposed in this paper. METHODS: Chromatographic finerprint of Tongbiding Injection and multi-wavelength chromatographic fingerprint of Fructus Gardenia were served as examples. Their corresponding reference fingerprints were yielded via PCA, and by comparison with average method and median method were also performed. RESULTS: Reference fingerprint, yielded by PCA based on least-squares principle, was essentially the linear combination of original chromatographic fingerprints. It had a better capability of maintaining and summarizing the data information of original chromatographic fingerprints and optimized properties, and was very similar to one generated by average method or median method. Abnormal fingerprints, if were possibly present in population of fingerprints, would be conveniently found via PCA method, and treated by using appropriate methods to eliminate the effects on reference fingerprint yielded. CONCLUSION: PCA method can be served as an effective approach to produce the reference fingerprint.
6.Dynamic changes of cardiovascular regulating factors in rats after aerobic exhaustive exercise.

OBJECTIVETo study the changes of cardiovascular regulating factors in rats during recovery of aerobic exhaustive exercise.
METHODSSixty male Wistar rats were randomly divided into control group, 1 h-exercise group, 3 h-exercise group, exhausted group, 2 h-recovery group and 12 h-recovery group. The rats were killed at corresponding times for each group after an 8-week-long treadmill training, and the levels of NO, ET, ANP and TXB2 in plasma were measured in each group.
RESULTSNO/ET ratio of 1 h-exercise group was significantly higher than that in control group (P < 0.01), while it was significantly decreased in 3 h-exercise group and exhausted group (P < 0.05). ANP contents in rat plasma were significantly higher in 3 h-exercise group, exhausted group and 2 h-recovery group than that in control group (P < 0.05 or P < 0.01). The concentration of TXB2 in plasma was significantly increased in 3 h-exercise group, exhausted group and 2 h-recovery group (P < 0.05).
CONCLUSIONChanges in cardiovascular regulating factors after exhaustive exercise may lead to deficiency of coronary circulation blood/oxygen supply, which may cause exercise-induced fatigue.

7. Relationship between polymorphisms of related genes in the folic acid metabolic pathway and the survival of gastric cancer patients treated with capecitabine combined with paclitaxel

Objective: To investigate the association of polymorphisms of related genes in folic acid metabolic pathway and the tumor candidate genes with the survival of gastric cancer patients treated with capecitabine combined with paclitaxel. Methods: Ninety-three patients with pathological diagnosis of gastric cancer were treated with capecitabine plus paclitaxel-based chemotherapy. The gene polymorphisms of tumor necrosis factor ( TNF) A308G (rs1800629), methylenetetrahydrofolate reductase ( MTHFR) C677T (rs1801131) and A1298C (rs1801133), methionine synthase ( MTR) A2756G (rs1805087) and methionine synthase reductase ( MTRR) A66G (rs1801394) were detected by TaqMan-MGB probe typing method. The median survival time (MST) of patients with different genotypes was compared, and the effects of various factors on the prognosis were evaluated. Results: The median follow-up period was 29.6 months. The MST of 93 patients was 34.93 months. The MST of patients with MTHFR 1298CA and 1298CC were 47.50 and 22.91 months, respectively. The log-rank test revealed that the polymorphism of MTHFR 1298C/A had marginally significant correlation with the survival of patients (χ2=3.447, P=0.062), and other gene polymorphisms were not associated with the survivals. The gender, drinking and operation were the major prognostic factors for gastric cancer patients receiving chemotherapy. Conclusion: Detection of MTHFR 1298CA polymorphism may predict the survival of gastric cancer patients receiving chemotherapy of capecitabine combined with paclitaxel.
8.Moxibustion Improved Transcutaneous Oxygen Tension and Exercise Capacity in Lower Limbs of Peripheral Arterial Disease.

OBJECTIVETo observe the effects of moxibustion and treadmill exercise on transcutaneous oxygen tension and exercise capacity in lower limbs of peripheral arterial disease (PAD).
METHODSTotally 58 mild-to-moderate PAD patients were assigned to the control group (18 cases), the moxibustion group (20 cases), and the treadmill exercise group (20 cases) by random digit table. Patients in the control group received conventional drug therapy for 12 weeks. Patients in the moxibustion group and the treadmill exercise group additionally received moxibustion [at Zusanli (ST36), Sanyinjiao (SP6), Yongquan (KI1)] and treadmill exercise respectively, once per day, 5 times per week for 12 weeks in total. Ankle-Brachial Index (ABI) , transcutaneous oxygen tension (TcPO₂), 6-min walking test (6MWT), and walking impairment questionnaire (WIQ) were assessed before and after treatment.
RESULTSCompared with the control group and the same group before treatment, there was no statistical difference in ABI in the moxibustion group and the treadmill exercise group (P > 0.05). But TcPO₂, 6MWT, and WIQ were obviously elevated (P < 0.01). Besides, 6MWT and WIQ assessment of the treadmill exercise group were better than that of the moxibustion group (P < 0.01) after intervention.
CONCLUSIONMoxibustion and treadmill exercise could improve the exercise capacity and TcPO₂of lower limbers in PAD patients.

9.The Effects and Mechanism of Lidocaine on Evoked-Bursting Firing of Injured Dorsal Root Ganglion Neurons

Objective To study the effects and current mechanism of low concentration of lidocaine on evoked-bursting (EB) firing of dorsal root ganglion (DRG) neurons in rat model of chronic compression (CCD) of DRG . Methods Twenty-four SD rats were divided into normal control group (n=12) and CCD model group (n=12). CCD group was treated with chronic oppression on L4 and L5 DRG with L shape bar. Normal control group received no treatment. In vivo intracellu?lar recording was used to record the incidence of EB and the effect of lidocaine on subthreshold membrane potential oscilla?tion (SMPO). Patch clamp recording was used to record the effect of lidocaine on persistent sodium current (INaP). Results The incidence of EB increased in CCD group( 45.97%, 57/124), which was significantly different when compared with nor?mal group (χ2=26.810, P<0.01). The magnitude of SMPO, INaP and EB were inhibited in a reversible way by lidocaine (50μmol/L). Conclusion The low concentration of lidocaine might play an analgesic effect in peripheral nervous system by se?lectively inhibiting INap, which participates in SMPO formation.
