1.Clinical analysis of recombinant human granulocyte macrophage colony-stimulating factor gel in the treatment of burn wounds

Objective To analyze the clinical efficacy of recombinant human granulocyte macrophage colony stimulating factor gel in the treatment of burn wounds.Methods 80 cases of burn patients in our hospital from February 2016 to February 2017 received treatment, were randomly divided into control group and observation group, 40 cases in each group, the control group given routine treatment, and the observation group was treated with recombinant human granulocyte macrophage colony stimulating factor gel on the basis of the treatment of the control group.The clinical effects of the two groups were compared and analyzed, including the healing time, the total effective rate of wound healing, the healing rate of the wound, and the related adverse reactions.Results The patients in, the healing time, total wound healing efficiency,and wound healing rate of the control group were significantly better than those of the control group(P<0.05), and no adverse reactions occurred in the two groups.Conclusion Burns were treated with recombinant human granulocyte macrophage colony stimulating factor gel treatment can effectively improve the treatment of patients with total efficiency, shorten the healing time, and can improve the healing rate, the treatment has a very important meaning and value, worthy of promotion and application in clinical widely.
2.Effects of breviscapine on the renal structure, function and PKC-mRNA and its protein expression in brain-dead BA-Ma mini pigs

Objective To investigate the effects of breviscapine on the renal structure, function and PKC-?mRNA and its protein expression in brain-dead BA-Ma mini pigs. Methods Fifteen BA-Ma mini pigs were randomly divided into 3 groups: brain-dead group (group A), breviscapine pretreatment group (group B), and control group (group C), 5 pigs in each group. The brain-dead models were established by increasing intracranial pressure in a modified, slow and intermittent way. At 3, 6, 12, 18 and 24 h after the initial brain death, serum BUN, Cr, TNF-?, IL-1?, and IL-6 were determined. At 3, 6, 12, and 24 h, the changes of renal tissues were observed by HE staining, and the expression of PKC-?mRNA and protein was detected by RT-PCR and immnohistochemistry respectively. The ultrastructural changes of hepatic cells were observed under electron microscopy. 3.Superimposition: a key word in neonatal acute respiratory distress syndrome

The Montreux definition established in 2017 made it clear that the neonates can not be excluded from the diagnosis of acute respiratory distress syndrome (ARDS), which supported the urge of emphasizing the specificity and importance of neonatal ARDS (nARDS) in the past ten years in China. Neonatal idiopathic respiratory distress syndrome is caused primarily by insufficient pulmonary surfactant system, which pathologically and clinically presented as typical respiratory distress syndrome. While the causes of nARDS often coexisted with underlying conditions, and its pathological and clinical features are a superposition of both respiratory distress syndrome and underlying conditions. Therefore, superimposition is the key to understanding the etiology, pathology, diagnosis, and treatment of nARDS, which is crucial for optimizing the clinical practice of nARDS.
4.Cloning of MSI-78 Gene in Escherichia coli DH5? and Identification of Positive Recombinant

OBJECTIVE To clone the MSI-78 gene for the purpose of providing evidence for further studies in prokaryotic expression and activities of antimicrobial peptides. METHODS According to the amino acid sequences of MSI-78,the MSI-78 gene was designed favorable for the Escherichia coli codons. After EcoRⅠand PstⅠ disgestion,cohesive ends were added to both ends respectively and the MSI-78 gene was synthesized by chemical methods. Then,the MSI-78 gene was ligated with pUC-18,transformed into the E. coli DH5?. Through filtration of ? complementary screening,the positive recombinant was finally identified by enzyme digestion of ECORⅠand ECORⅠ/PstⅠ and by PCR. RESULTS The MSI-78 gene was ligated with pUC-18 and transformed into the E. coli DH5?. As a result,MSI-78 gene was cloned in E. coli DH5? successfully. CONCLUSIONS The cloning of the MSI-78 gene provides evidence for further studies of its prokaryotic expression and activities of antimicrobial peptides.
5.High-density seeding affects transforming growth factor-beta1-induced epithelial-mesenchymal transition of HaCaT cells

BACKGROUND: The cel density is one of the factors involved in the state of cel differentiation, and the effect of cel density on transforming growth factor-β1-induced epithelial-mesenchymal transition of HaCaT cells is stil unclear. OBJECTIVE: To observe the effect of cel density on transforming growth factor-β1-induced epithelial-mesenchymal transition of HaCaT cells. METHODS: HaCaT cells was seeded in 6-wel plates at low density of 103/cm2 and high density of 105/cm2 then treated by 2 μg/L transforming growth factor-β1 for 48 hours, thereafter observed the changes in cel morphology. The transcription levels of epithelial cadherin, tight junction protein-1, vimentin, neuronal-cadherin were detected by real-time PCR, and expression levels of epithelial cadherin and vimentin were detected by Western blot. RESULTS AND CONCLUSION: Cel gap of HaCaT cells grew larger after treated with transforming growth factor-β1 for 48 hours, and cel morphology was long spindle rather than polygonal in low-density group, while in high-density group without obvious morphological changes. The real-time PCR showed that the transcriptions of epithelial cel marker epithelial cadherin and tight junction protein-1 were suppressed when compared with the control group (P < 0.05), and the decreasing deree in the high-density group was higher than that in the low-density group (P <0.05), mesenchymal cel marker neuronal-cadherin and vimentin were upregulated in the high-density group and the low-density group when compared with the control group (P < 0.05), and there were no significant differences between high-density group and low-density group. Western blot results verified the changes of neuronal-cadherin and vimentin expression level. These results suggest that the high seeding density can inhibit transforming growth factor-β1-induced epithelial-mesenchymal transition of HaCaT cells.

7.Three-dimensional finite element analysis of angled abutments in anterior maxilla implant restoration

Objective To explore the effects of angled abutments on the anterior maxilla implant restoration. Methods We analyzed the biomechanical properties of implants of different sizes (Φ3.5 mm,4.0 mm and 4.5 mm in diameter;L11.5 mm and L13 mm in length)after connecting different angled abutments (0°,10°,20°,and 30°) using finite element method.Results The stresses and strains of loading parts of restorations increased and their distribution became more concentrated as the angle of abutment increased.Cortical bone of Φ3 .5 implants with smaller angle (10°or less)andΦ4.0 implants with abutments had the risk of overpassing the bone elastic threshold when the angle approached 30°.However,the cortical bone elastic deformation was within a safe range at all angles inΦ4.5 group.Conclusion We should consider the diameter of the implant when selecting angled abutments.The angled abutments are not suitable for small diameter implants.The bite force should be under control when needed. The larger angled abutments can be applied in the standard and major diameter implants and it is necessary to avoid occlusal overloading.

Objective:To study the influences of angled abutments connected with small diameter implants in anterior maxilla restora-tion.Methods:Simulation models of angled abutment with small diameter restoration in anterior maxilla were established,the biome-chanical properties of the models were studied by finite element method(FEM)analysis.Results:Following the increase of the abut-ment angle,the stress and strain of all parts increased linearly and more concentrated,while appropriate prosthetic crown could share some load.Cortical bone elastic deformation exceeded the threshold when the abutment angle was bigger than 8 °.Conclusion:With abutment angle increase,stresses and strains will increase and will be more concentrated,which is unfavorable for load conduction.Ap-propriate crown can reduce the adverse effects of angled abutment.Angled abutment should not be used for d 3.5 implant.
9.3D construction and printing of bone tissue engineering scaffolds based on CT data

Objective To study 3D construction and printing of bone tissue engineering scaffolds by bone defect modeling on Mimics software based on CT data. Methods CT data of jaw bone defect from patients were acquired and the images were segmented using threshold segmentation combined with region growing . The three dimensional model was reconstructed by Boolean operation. The individual 3D digital model was reconstructed with internal structure by combining with computer; preparing poly-lactic acid scaffold in virtue of 3D technology. Results Using the Mimics software, we successfully constructed a 3D digital reconstruction model of bone defect based on CT data. The constructed scaffold model with certain internal form and structure was matched with the bone defect of patients, and the constructed model was exported onto STL standard format, which may be in common use. Conclusion The 3D digital model of bone defect scaffolds may effectively be reconstructed based on the CT data using Mimics software and computer aided design.
10.Optimization of Water-extraction Technology of Compound Kuhuang Formulation by Orthogonal Test

OBJECTIVE:To optimize the water-extraction technology of Compound kuhuang formulation. METHODS:The wa-ter-extraction technology of Compound kuhuang formulation was optimized by orthogonal design,with the total contents of matrine and oxymatrine,the total content of prim-O-glucosylcimifugin and 5-O-methylvisamminol,the content of berberine hydrochloride and extract yield,and with the extraction time,amount of water(3 reflux extractions)and soaking time as the factors;and verifi-cation tests were conducted. RESULTS:The extraction time had significant effect on comprehensive evaluation value (P<0.05). The optimal water-extraction technology was as follows as extraction time of 120 min,water amount of 8,6,and 6 times as the amount of herbs,and soaking time of 20 min. In verification tests,the average total content of matrine and oxymatrine was 3.152 6 mg/g(RSD=1.03%,n=3),that of prim-O-glucosylcimifugin and 5-O-methylvisamminol was 4.977 2 mg/g(RSD=2.27%,n=3),the average content of berberine hydrochloride was 3.345 0 mg/g (RSD=1.19%,n=3) and the average extract yield was 49.23%(RSD=2.43%,n=3). CONCLUSIONS:The optimal water-extraction technology of Compound kuhuang formulation is stable and feasible.
