1.Mesenchymal stem cells as cellular vehicles for tumor gene therapy

The conventional approach of systemic viral gene delivery is often associated with limited therapeutic efficacy and toxicity because of a lack of tumor specificity. Mesenchymal stem cells have the tumortracking abilities which provide possibility that the stable and high level of anti- cancer gene expression could be acquired within local tumor and a novel approach for better tumor-targeted gene delivery.
2.Several Problems in Quality Control of PACS

Objective To discover the main points of PACS quality control through basic study and advance some solution ways which include hardware and software improving. Methods Combined computer and network tech with hardware and software,like tech of network security,to increase the validity of PACS quality control. Results The archives and communications could be more accurate and safe by use of hardware and software and ROC theory. Conclusion The importance of quality control of PACS in the practical work is progressing and latest tech of hardware and software should be used to increase the quality control of PACS.
3.Clinical analysis of testicular torsion with the primary symptom of abdominal pain in adolescents and boys

Objective To evaluate the characteristics of the testicular torsion patients with the first symptom of acute abdominal pain.Methods From October 1998 to May 2014, 8 adolescent or boys with testicular torsion presenting acute abdominal pain instead of scrotal pain as their primary symptom were retrospectively reviewed.The average age was 12 (3-16) years.An acute abdominal pain presented firstly in all cases, which accompanied by nausea and vomiting in 4 cases, groin pain in 2 cases, fever in 1 case. In genital examination, the scrotum skin was red and swollen, while the testis was in transverse position and tenderness.The testicular and homolateral abdominal pain got worse when we pulled the involved testis. Doppler ultrasound examination revealed that the blood supply of the involved testis decreased or disappeared.Results In the 8 cases, 5 cases were not examined their testicles or scrotum during the first evaluation, which were found the testicular necrosis during the following exploratory surgery, and orchidectomy was performed. In the other 3 cases, external genital were examined during the first evaluation, and the scrotal and testicular abnormalities were detected.Doppler ultrasonography showed abnormality in testicular blood supply.Testicular torsion was proved during the emergent operation, and the 3 testes were salvaged.The mean duration time from symptom onset to operation was 4 h in the salvaged group, while the average time was 28 h in the orchiectomy group.The mean follow-up time was 18 ( 10-36) months.In the 3 cases whose testis was saved, 1 affected testis was atrophied, and the blood supply of the other two was good.No severe complications occurred in the other 5 cases.Conclusions Testicular torsion should be considered in adolescents and boys with acute lower abdominal pain.The routine external genital examination should be performed when a boy or adolescent complains of abdominal pain.
4.Suppression of experimental choroidal neovascularization utilizing anti-FLK1 monoclonal antibody

· AIM: To investigate the possibility of FLK-1 as a therapeutic agent for choroidal neovascularization (CNV) in mouse by FLK1 monoclonal antibody.· METHODS: CNV was induced in C57BL/6 mouse eyes by krypton laser photocoagulation. Preoperatively and on the 3rd, 6th and 9th day after photocoagulation, the animals were intraperitoneally injected with 500μg FLK1 monoclonal antibody. Choroidal fluorescein angiography and histopathological examination were used to assess the development of experimental CNV quantitatively 10 days after laser treatment.· RESULTS: Laser-induced CNV developed in all lesions on the 10th day after the operation. The development of CNV was significantly inhibited in experimental group, indicated by choroidal fluorescein angiography and histopathological examination (P<0.001).· CONCLUSION: Our findings suggest that FLK-1 may be a promising agent for anti-angiogenesis therapy for CNV.
5.Expression of FLK-1 in laser-induced choroidal neovascularization in mouse

AIM: To investigate the expression of vascular endothelial growth factor receptor 2 (VEGFR-2, also known as FLK-1) in laser-induced choroidal neovascularization (CNV) in mouse.METHODS: CNV was induced in C57BL/6 mouse eyes by krypton laser photocoagulation. Choroidal fiuorescein angiography and histopathological examination were used to assess the development of experimental CNV. Cryostat sections from lesions on day 10 after laser treatment and normal eyes were prepared for Immunohistochemistry for FLK-1.RESULTS: Laser-induced CNV developed in all lesions on day 10. The expression of FLK-1 was detected in endothelial cells, retinal pigmented epithelium (RPE)-like cells and fibroblast-like cells in neovascular lesions. In normal adult mouse retinas, FLK-1 expression was mainly observed in RPE cells, inner nuclear and ganglion cell layers.CONCLUSION: Our findings demonstrated that expression of FLK-1 may play a role in the formation of laser-induced CNV in mice, which suggest that FLK-L may be a promising potential target for antiangiogenesis therapy for CNV.
6.Effects of RNAi of MBPˉ1 gene on proliferation of gastric cancer SGCˉ7901 cell line

Objective To investigate the effects of c-myc promoter binding protein(MBP-1)gene expression silencing on the pro-liferation in vitro in human gastric cancer cell line SGC-7901.Methods The cells divided into three groups:blank control group (cells without transfecting gastric cancer cell),negative control group(cells transfecting missense sequence)and experimental group (cells transfecting MBP-1 shRNA).Two MBP-1 shRNA sequences and one negative control shRNA sequence were designed,syn-thesized and cloned into pSIREN-retroQ plasma.Then the recombinant plasmids were constructed and transfected into human gas-tric cancer SGC-7901 cells by Lipofectamine 2000.After selection of cells with puromycin for two weeks,stable cell line SGC-7901 expressing gene of MBP-1 specific shRNA was obtained.The expressions of MBP-1 mRNA and protein in SGC-7901 were deter-mined by the real time PCR and Western blot,respectively.The effects of altered expression of MBP-1 on the cell proliferation were measured by MTT cell proliferation assay.Results PCR and sequencing indicated that the recombinant plasmids pSIREN-retroQ was constructed.Then the recombinant plasmids were transfected into human gastric cancer SGC-7901 cells by Lipofectamine 2000. After selection of cells with puromycin for two weeks,stable cell line SGC-7901 expressing gene of MBP-1 specific shRNA was ob-tained.The relative expression level MBP-1 mRNA in the MBP-1 siRNA transfection group was significantly decreased compared with the blank control group(P <0.05).Compared with the blank group,the expression levels of MBP-1 protein in the experimental group also significantly decreased.The proliferation abilities of SGC-7901 cells at 48,72,96,120 h after MBP-1 siRNA transfection were significantly increased compared with the blank control group (P < 0.05 ).Conclusion Down-regulating the expression of MBP-1 can obviously promote the proliferation of human gastric cancer cell line SGC-7901.MBP-1 gene may become the new target of gene therapy for gastric cancer.
7.miR-141 suppresses cell proliferation by target down-regulating EphA2 in human oral squamous cell carcinoma

Objective To assess the effect of miR-141 on proliferation of human oral squamous cell carcinoma and target relationship between miR-141 and EphA2 .Methods pcDNATM6.2-GW-pre-miR-141 was constructed and identified by qRT-PCR.EphA2-WT and EphA2-MT sequences were respectively cloned into pmirGLO plasmid . The potential proliferation function of miR-141 on CAL27 cells was analyzed by MTT .The target relationship be-tween miR-141 and EphA2 was identified by Dual-Luciferase Assay System , qRT-PCR and Western blot .Results We constructed successfully the recombinant plasmids , including pcDNATM6.2-GW-pre-miR-141, pmirGLO-E-phA2-WT and pmirGLO-EphA2-MT, and the transfection efficiency of pre-miR-141 was increased in CAL27 cells compared to control group(P<0.001).miR-141 could suppress the proliferation of CAL27 cells(P<0.05). Furthermore, a significant reduction of luciferase activities of CAL27 cells co-transfected with pre-miR-141 and EphA2-WT(P<0.001).The mRNA(P<0.001) and protein expression levels of EphA2 were decreased in CAL27 cells transfected with pre-miR-141 .Conclusions Overexpression of miR-141 may suppress cell prolifera-tion by targeting at EphA2 in CAL27 cells.
8.Application of IRT Unfolding Model on "Autonomy in Learning Rating Scale for College Students"

Objective: Attempt to use generalized graded unfolding model to analyze the item of "Autonomy in Learning Rating Scale for College Students " adequacy of the scale designing and evidence probability of the usage of unfolding model in costructing tests. Methods: Based on randomly selected 600 graduates’ response of the scale, item analysis was carried on in 497 potent subjects. Results: Subjects whose trait continuum were between 2.5-4.0 and -3.5~-3.0 were short, and all the item locations were negative; all the item discriminations were above 0.5; the item information functions of 2, 6, 7, 19, 21, 25, 49, 50, 51 were below 0.4098; the fit plots of the item and test satisfied the requirements. Conclu- sion: Fit index of the whole scale is good; some items need to be revised or deleted; GGUM could be used in constructing attitude scales.
9.Diagnostic value of GenoType? MTBDRsl assay for the resistance to fluoroquinolones anti-tuberculosis drugs:a Meta-analysis

Objective To evaluate the diagnostic accuracy of GenoType? M TBDRsl for the resistance to fluoroquinolones anti‐tuberculosis drugs .Methods Systematic and comprehensive literature was searched in PubMed ,Embase ,Web of Science ,CBM , CNKI ,VIP and Wanfang database .The relative studies of GenoType? MTBDRsl to fluoroquinolones anti‐tuberculosis drugs were included .After quality assessment ,Meta‐Disc1 .4 software was used to analyze the data .Results A total of 16 trials ,involving 1 766 participants ,were included .The results of Meta‐analysis showed that the weighted sensitivity ,specificity ,positive likelihood ra‐tio ,negative likelihood ratio ,diagnostic odds ratio ,and the area under summary receiver operation curve were 0 .83 ,0 .96 ,17 .50 , 0 .20 ,108 .46 and 0 .934 9 ,respectively .Conclusion GenoType? M TBDRsl assay for the resistance to fluoroquinolones anti‐tuber‐culosis drugs might be with high sensitivity and specificity ,which could be recommended as efficacy diagnostic tool .
