2.Expression of Legionella pneumophila virulence genes under biofilm condition

Objective To investigate the physiological state of L. pneumophila in biofilm. Methods Genes previously identified as good markers for the transmissive and replicative phases of the L. pneumophila life cycle during growth in Acanthamoeba castellanii were examined for their expression fold change in the sessile cells as compared to planktonic cells using real-time RT PCR. Results Mature L. pneumophila biofilms were formed at 37t in 75 cm2 cell culture treated flasks for 18 days. The ratio of gene (mip, flaA and fliA) expression in post-exponential cells compared to exponential cells is 0. 53, 4. 45 and 3. 67. The exponential phase cultures display replicative traits and post-exponential bacteria express transmissive factors. The ratio of gene (mip, flaA and fliA) expression in sessile cells compared to exponential cells is 4.42, 5.24 and 16.21, while the sessile cells compared to exponential cells is 8.39, 1. 18 and 4. 43, respectively. Conclusion The violence gene expression of L pneumophila in biofilm is unique.
3.The prevalence of vitamin D deficiency in intensive care unit and its influence on prognosis

Objective To investigate the prevalence of vitamin D deficiency in medical intensive care unit (ICU) and its relationship with severity of disease and prognosis.Methods A prospective study was performed to evaluate vitamin D status in 216 patients admitted to the medical intensive care unit.The incidence of hypovitaminosis D was observed.Acute Physiology and Chronic Health Evaluation Ⅱ (APACHE Ⅱ) score,days kept in ICU and on ventilator,main laboratory findings,and mortality rate were compared among patients with different serum 25-hydroxyvitamin D [25 (OH) D] levels.Potential risk factors for mortality were analyzed by multivariate logistic regression analysis.Results One hundred and fifty-three patients (70.8%) developed hypovitaminosis D.25 (OH) D deficiency was identified in 95 (44.0%),25 (OH) D insufficiency in 58 (26.8%),and 25 (OH) D sufficiency in 63 (29.2%) patients.APACHE Ⅱ score,positive blood culture,the incidence of multiple organ dysfunction syndrome(MODS),and mortality rate were higher in deficiency group compared with the other two groups [APACHE Ⅱ score:deficiency group 25 (20,28) score,insufficiency group 22 (17,26) score,sufficiency group 19 (18,20) score,P<0.01 ; positive blood culture:deficiency group 18.9%,insufficiency group 13.8%,sufficiency group 3.2%,P=0.015 ; MODS:deficiency group 48.4%,insufficiency group 43.1%,sufficiency group 25.4%,P=0.025; mortality rate:deficiency group 40%,insufficiency group 24.1%,sufficiency group 15.9%,P =0.003].25 (OH)D levels were negatively correlated with APACHE Ⅱ score and mortality rate (r =-0.325,P<0.01 ; r=-0.276,P<0.01,respectively).Analysis by multiple logistic regression demonstrated that 25 (OH) D deficiency (OR =3.005,95 % CI 1.321-5.875,P =0.008) was independent risk factor for mortality.Conclusions This study demonstrates that vitamin D deficiency is highly prevalent among patients admitted into ICU.Vitamin D deficiency is associated with disease severity,and seems to be an independent risk factor for mortality.
4.Control and testing methods for blood bump in heart assistant and replacement devices.

Using extracorporeal circulation for cardiopulmonary bypass is the first step for the successful cardiac surgery, and the blood pump is the key component in extracorporeal circulation devices, the control and measurement of mechanical-fluid parameters such as flow rate and pressure are very important to guarantee the successful cardiac surgery. This paper reviews several control methods of the blood pump based on the flow rate/pressure, heart rate, ventricular work and blood assist index, and also discusses the direct and indirect measurements of the flow rate and pressure. This review concludes with the main possible trends for the further development of the blood pump control and testing methods.

5.Magnetic field numerical calculation and analysis for magnetic coupling of centrifugal blood pump for extracorporeal circulation.

This paper mainly studies the driving system of centrifugal blood pump for extracorporeal circulation, with the core being disc magnetic coupling. Structure parameters of disc magnetic coupling are related to the ability of transferring magnetic torque. Therefore, it is necessary to carry out disc magnetic coupling permanent magnet pole number (n), air gap length (L(g)), permanent magnet thickness (L(m)), permanent magnet body inside diameter (R(i)) and outside diameter (R(o)), etc. thoroughly. This paper adopts the three-dimensional static magnetic field edge element method of Ansys for numerical calculation, and analyses the relations of magnetic coupling each parameter to transmission magnetic torque. It provides a good theory basis and calculation method for further optimization of the disc magnetic coupling.

OBJECTIVE To compare the difference of Pseudomonas aeruginosa(PAE) biofilms formation in different conditions including incubated time,temperature,attaching materials,and flowing speed.METHODS PAE biofilms were established in a chemostat-coupled MRD and detected with the method of viable bacteria counting.The number of CFU/disc was measured in different culture time(8h,24h or 72h),different temperature(23℃ or 37℃),different adherent materials(glass or silicone),different flow velocities(30ml/h or 90ml/h) and different culture media(M63 medium,M-H croth or LB broth).RESULTS Keeping the other conditions invariable,the log10CFU/disc of viable bacteria in 8h,24h or 72h biofilms were 4.01?0.26,4.59?0.49 or 5.20?0.47,respectively(P

According to the clinical requirements of cardiopulmonary bypass surgery, this paper established a simulation system for cardiac surgery which consists of venous reservoir, variable balance chamber, blood suction bag, ventricle suction bag, resistance valves, pressure gauges and tubings. Using the proposed system, perfusionists can mimic the implementation of pre-established surgery strategy, predict various abnormal conditions in the operation, and accordingly take the urgent actions so as to improve the success rate of surgery and to ensure the safety of patients.

9.Different effect of Ceftazidime on the early and mature Pseudomonas aeruginosa biofilm bacteria

Objective To observe the effect of Ceftazidime on the early and mature Pseudomonas aeruginosa biofilm bacteria. Method The early and mature PA biofilm were formed through the chemostat combined Robbins device. Then 64 ?g/ml Ceftazidime was applied to the early and mature PA biofilm bacteria for 24 hours respectively. Scanning electronic microscope(SEM) was used to observe the effect of Ceftazidime on early and mature biofilm. The difference between the viable bacteria in early and mature biofilm was analyzed by statistical method. Result 24 hours after Ceftazidime was added, Pseudomonas aeruginosa in early biofilm was killed on the whole while there were still numbers of bacteria in mature biofilm. Statistical analysis demonstrates a great variance in viable counts. Conclusion Pseudomonas aeruginosa in mature biofilm is more tolerant to antibiotics than that in early biofilm.
10.Rapid identification of Nontuberculous Mycobacterium using matrix assisted laser desorption/ionization time of flight mass spectrometry

Objective The purpose of this study was to compare and develop the method for identification of Non-tuberculous Mycobacteria (NTM) using matrix assisted laser desorption/ionization time of flight mass spectrometry (MALDI-TOF MS),and evaluate the feasibility,accuracy and repeatability of MALDI-TOF MS to discriminate NTM.Methods Fifteen clinical strains were collected from January to March in 2012 and 68 environmental strains were retrospectively collected from 2011 to 2012.A protocol for sample pre-treatment and protein extraction was developed and utilized it to identify clinical and environmental isolates.The results from 16 s rRNA sequencing were served as control.Results Method A was more effective in protein extraction.Although all the three methods got the same species result,a total of 83 strains belonging to 10 distinct species grown in Middle brook 7H10 media were analyzed.All members of the NTM were identified accurately at the genus level and 80.7% (67/83) of strains could be identified at the species level.Six strains were identified at the complex level.81.9% (68/83) of NTM got high spectral scores.The identification of cultured colony could be completed in 1.5 hours.And it had good reproducibility.Conclusion MALDI-TOF MS can be used as a rapid and accurate method for identification of Mycobacteria in clinical microbiology laboratories,implying its good application prospects.
