1.The effects and mechanisms of BTBD10 on the proliferation of islet beta cell

ObjectiveTo explore the role of BTBD10 overexpression in the proliferation of insulinoma cell line INS-1and its mechanism. MethodsThe recombined expression plasmid of pcDNA4.0-BTBD10 was constructed by gene cloning technique and was transfected into INS-1 cell by lipofectamine 2000. The stable overexpression BTBD10 of INS-1cell was selected at 48th hour after transfection.INS-1 cell proliferation activity was measured by MTT method.The expression of BTBD10,protein kinase B(Akt),phospho-Akt(p-Akt),mammal target of rapamycin (mTOR) and phospho-mTOR (p-mTOR) were determined by Western blot.ResultsThe stable overexpression BTBD10 of INS-1 cell wassuccessfullyconstructed.OverproductionofBTBD10promotedbetacellproliferation.The phosphorylation of Akt and mTOR was increased and the ratio of p-Akt/Akt and p-mTOR/mTOR was enhanced in the INS-1 overexpressed by BTBD10.But the expression of total Akt and mTOR presented no obvious changes. Conclusion The overexpression BTBD10 of INS-1 cell could activate of Akt/mTOR signalling pathway via stimulating phospho-mTOR and Akt,and enhance overall cell protein translation,so as to promote proliferation of INS-1 cell.
2.Application of preformed orbital titanium mesh in reconstruction of orbital fracture.

OBJECTIVETo assess the effect of preformed orbital titanium mesh in the reconstruction of orbital fracture.
METHODTwenty-seven cases of blow-out orbital fracture were diagnosed in the period from January 2012 to July 2012, all with serious defect of the orbital wall and herniation of the orbital soft tissues. Preformed orbital titanium mesh was grafted to the traumatic orbital walls. Orbital axial and coronal CT and three-dimension CT scan examination were performed routinely in all cases preoperatively and postoperatively to evaluate the postoperative exophthalmos and orbital wall reconstruction.
RESULTSAll the patients were followed up for 3 months. Orbital wall reconstruction was successfully performed in all cases without serious complications. The postoperative enophthalmos was less than 3 mm in 23 of the 27 cases.
CONCLUSIONPreformed orbital titanium mesh allows accurate reconstruction of orbital wall fracture.

3.Effect of mitomycin C in reducing hypertrophic scar in rat traumatic osteomyelitis model

Objective To investigate the effect and mechanism of mitomycin C in reducing hypertrophic scar in rat traumatic osteomyelitis model.Methods A total of 120 Wistar rats were divided into control group (Group A,n =40),traumatic osteomyelitis group (Group B,n =40),traumatic osteomyelitis treated with Mitomycin C group (Group C,n =40),according to the random number table.The model of traumatic osteomyelitis was produced by Staphylococcus aureus.Muscle tissues around the focus were harvested at 15 d and 30 d postinjury.HE staining was used to observe the changes of muscle tissue structure.Immunohistochemistry was used to detect expression of transforming growth factor (TGF)-β1.Masson staining was used for collagen deposition evaluation.Western blot was used for detection of levels of TGF-β1 and collagen Ⅰ.Results HE staining revealed consistent alignment of fibers within the muscle in Group A.Fibrosis with the muscle was observed in both Group B and C,but the degree of muscle fiber disorder was decreased in Group C compared to Group B.Either 15 d or β0 d after injury,expressions intensity of TGF-β1,collagen fraction volume,and activation levels of TGF-β1 as well as collagen Ⅰ were higher in Group B and C than Group A,and all parameters were decreased in Group C compared to Group B (all P < 0.05).Conclusion Mitomycin C can reduce hypertrophic scar formation in traumatic osteomyelitis model,and the potential mechanism relates to downregulated TGF-β1 and collagen Ⅰ.
4.Research Progress on Relation Between Shift Work and Ovarian Cancer

The incidence rate of ovarian cancer ranks the third among female malignant tumors, the mortality rate ranks the first and the prognosis is poor. Ovarian cancer poses a serious threat to the health of women's lives. The incidence of ovarian cancer is related to heredity, endocrine disorders and adverse lifestyle factors. In recent years, some scholars have carried out the impact of lifestyle, especially shift work, on the occurrence and development of ovarian cancer. Shift work can lead to circadian disruption, which affects hormonal balance in the body and may be a risk factor for ovarian cancer. This review summarizes four cohort studies and two case-control studies on the relation between shift work and ovarian cancer. The conclusion of the studies is inconsistent, suggesting that further researches are needed.
5.A review of metformin in occurrence and prognosis of ovarian cancer

Ovarian cancer is one of the most serious gynecological tumors in the world, and its mortality ranks first in gynecological malignancies. The study found that metformin is not only used as a first-line oral hypoglycemic agent for the treatment of type 2 diabetes, but also has a certain inhibitory effect on the occurrence and development of ovarian cancer. In recent years, domestic and foreign scholars have carried out experimental and epidemiological studies on the relationship between metformin and ovarian cancer. This review included 7 cohorts and 2 case-control studies on the incidence and prognosis of metformin and ovarian cancer. The results suggest that the use of metformin may improve the prognosis of ovarian cancer, however, due to the limited epidemiological studies of the disease, its relationship with ovarian cancer still needs further exploration.
