1.Research progress in lncRNAs and the diagnosis, prognosis, and drug resistance of gastric cancer
Chinese Journal of Clinical Oncology 2015;(1):71-76
Long non-coding RNAs (lncRNAs) are noncoding RNAs with lengths exceeding 200 bp. These lncRNAs cannot code complete proteins but control gene expression. Recent studies have shown that lncRNAs are closely related to the diagnosis, prognosis, and drug resistance of gastric cancer. This article summarizes the recent progress of studies in China and abroad on lncRNAs relative to the diagnosis, prognosis, and drug resistance of gastric cancer.

Objective To investigate the effects of taurine treatment on retina ultrastructure and glutamate receptor GLAST mRNA expression in diabetic rat. Methods Diabetic rats were fed with 1.2% taurine for 3 months.Reverse transcription and polymerase chain reaction(RT-PCR) were employed to detect the expression of GLAST mRNA.Changes of retinal ultrastructure of rats were observed with transmission electronic microscope. Results From the first to the third month, the(mRNA) expression of GLAST gradually decreased in diabetic rat retina.Taurine could up-regulate the expression in this period.Membrane disc arrange was disordered and mitochondria swelled in STZ diabetic rat retina, which were ameliorated after taurine treatment. Conclusion Taurine can up-regulate the mRNA expression of GLAST and restrain excitotoxicity of glutamate. These observations suggest that taurine may play a role of nerve protection.

Objective: The purpose of this study was to determine whether taurine had retinal neuroprotective effects against glutamate-induced excitototxicity, especially in retinal ganglion cells (RGCs) damage in adult rat. Methods: We used a rat model of retinal glutamate excitotoxicity induced by intravitreal injection of 40 nmol glutamate in one eye. The retinal damage was evaluated by counting the number of cells in the ganglion cell layer (GCL) in flat mount preparations of whole retinas, examining the a- and b-waves in the electroretinogram (ERG), hematoxylin-eosin (HE) staining, and observing ultrastructure of retinas. Results: After intravitreal injection of glutamate, thinning of inner retinal layers, smaller a- and b-waves in the ERG were observed, and degenerative changes were observed in inner nuclear layer and RGCs by electron microscopy. These damages could be prevented by 25mg/kg bw taurine (intraperitoneal injection). 5mg/kg taurine had unremarkable effect. In animals that received intravitreal injection of glutamate, there was significant reduction (53%) in total cell numbers in GCL, with 75% reduction in RGCs. 25mg/kg bw taurine-treated animals showed a significant reduction in glutamate- induced RGCs loss. Conclusion:Repeated application of taurine can prevent glutamate-induced retinal neuronal cell damage, especially in RGCs and retinal functional damage in rats.

Objective To observe the effect ofmycophenolate mofetil combined with prednisone in treatment of old patients with refractory nephrotic syndrome. Methods From June 2009 to June 2010,67cases of old patients with refractory nephrotic syndrome were divided into two groups by random digits table,32 cases in control group were treated with cyclophosphamide combined with prednisone and 35 cases in observation group were treated with mycophenolate mofetil combined with predrisone. The clinical effect and the levels of urinary protein, serum creatinine and serum uric acid between two groups were compared. Results There was no significant difference in total effective rate between two groups [68.8%(22/32) vs.74.3% (26/35)] (P > 0.05 ).After treatment, the levels of urinary protein, serum creatinine and serum uric acid in two groups were decreased (P< 0.05 ),but there were no significant differences between two groups (P > 0.05 ).The adverse reaction rate in observation group was significantly lower than that in control group (P < 0.05).Conclusion Mycophenolate mofetil combined with prednisone in treatment of old patients with refractory nephrotic syndrome has good effect and low adverse reaction, and it can be applied in clinic.

International Journal of Traditional Chinese Medicine 2009;31(5):393-394
Objective To investigate the effect of Xuebijing injection on release of high mobility group box 1 (HMGB1) in endotoxin-induced cells. Methods HMGB1 concentration and mRNA expression were analyzed by enzyme-linked immunosorbent assay (ELISA) and RT-PCR, respectively. The effects of Xuebijing injection with 2, 10 and 50 mg/ml on HMGB 1 concentration in the culture medium of 200 ng/ml lipopolysaccharide-induced macrophage-like RAW 264.7 cells and BRL-3A hepatocytes, and HMGB1 mRNA expression in lipopolysaccharide-induced macrophages were observed. Results 50 mg/ml Xuebijing injection significantly decreased HMGB1 concentration in the culture medium of lipopolysaccharide-induced macrophages and hepatocytes(P< 0.01), and inhibited HMGB1 mRNA expression in macrophages. Conclusion Xuebijing injection inhibits endotoxin-induced release of HMGB1.

Objective It has been known that NSAIDs can inhibit the growth of tumors through cyclooxygenase, but it is unknown that whether there are other mechanisms involved. It is essential to detect the effect of indomethacin for proliferation and apoptosis in the HCT116 and explore its anti tumor mechanism. Methods Human colon adenocarcinoma cell line HCT116 was treated with different concentration of indomethacin for 24 h and CDK 2,CDK 4, Bcl 2, Bax and p21 WAF1/CIP1 protein were detected by Western blot. Results Indomethacin down regulated the expression of CDK 2, CDK 4, Bcl 2 and up regulated the expression of p21 WAF1/CIP1 , however, the expression of Bax remained unchanged. Conclusions Inhibiting proliferation and inducing apoptosis contribute to the anti tumor activity of indomethacin by down regulating the expression of CDK 2, CDK 4, Bcl 2 and up regulating the expression of p21 WAF1/CIP1 .

Objective To analyse the detective rate and significance in diagnosis of cerebral and meningeal metastases by MR.Methods 115 cases with cerebral and meningeal metastasis were proved by operation and pathology. There were male 86 and females 29 ranged in age from 17-77 years old, plain MR scan and enhanced MR scan were performed in all cases. Results In this group included 46 cerebral metastases(40.0%),33 meningeal metastasis(28.7%),36 both cerebral and meningeal metastases(31.3%). Different degree of enhancement was showed on enhanced MRI in all cases, "mouse tail signs"or"target signs" were appeared in which cerebral metastasis with the adjoining meninges involved. Conclusion MRI is a good method for diagnosing cerebral and meningeal metastasis.Enhanced MRI has an important value.

Objective To analyse the MRI appearances of giant cell tumors of the bone and the relationship with their pathologic bases. Methods Histologic examinations were conducted in 21 cases, and their correlations with the MRI appearances were further evaluated. Results There were 3 cases of the Ⅰ grade,3 cases of the Ⅰ~Ⅱ grade,13 cases of the Ⅱ grade,2 cases of the Ⅲ grade of the giant cell tumors. Most tumors were composed of solid content that presented sligthly low to intermediate signal intensity on T 1WI,sligthly high to high signal intensity on T 2WI.The minority of the tumors were composed of the half of the solid and cystic contents expectively.Specific tumors was mainly composed of cystic content or combined with fluid-fluid level on T 2WI.Soft tissue masses were found in 8 cases. Conclusion The location and their extension into the soft tissue of the tumors can be clearly demonstrated on MR imaging, but there is no relationship between the grades of the tumors and their MRI appearances.
