1.Therapeutic effects of sitai tablet on blood pressure, blood lipid, blood sugar and insulin of rats with insulin resistance syndrome

AIM: To explore the effects of sitai tablet on blood pressure, blood lipid,blood sugar and insulin of rats with insulin resistance syndrome(IRS).METHODS: A total of 40 male clean SD rats were divided into normal control group, model group, sitai group and captopril group by randomized block design. The rats were fed with high-fat high sugar diet for 8 weeks to induce IRS. Then, sitai tablets were given to rats with IRS to observe the influence of sitai tablet on blood pressure, blood sugar, blood lipid, insulin levels and insulin sensitivity index(ISI) of them.RESULTS: The systolic pressure reduced from ( 158 ± 8), ( 157 ± 7) mm Hg before treatment to (128 ± 4), (110 ± 2)mm Hg after treatment in sitai group and captoril group respectively. There was also significant difference on systolic pressure of between sitai group, captopril group and model group [ ( 165 ± 6) mm Hg] ( t = 11.86 - 28.99, P < 0.01 ). In sitai group, the total cholesterol (TC), triacylycerol (TG)decreased clearly than those in model group( t = 44.27, 9. 14, P < 0.01 ), but high density lipoprotein cholesterol (HDL-C) increased( t = 703, P < 0. 01 ) . There were increased fasting insulin(FINS) and decreased ISI in model group. There was significant difference on these indes between model group and normal control group( t= 7.14, 9.35, P < 0.01 ) . The insulin resistance was evident so that the model was successfully established. The fasting blood sugar and FINS all reduced when comparing it between sitai group, captopsil group with model group, there was significant difference( t = 11.95, 6.71, P < 0.01). When comparing the fasting blood sugar in captoril group and sitai group, there was also difference( t = 8.33, P < 0.01 ) . The ISI all increased in sitai group and captoril group, there was difference between them and model group (t=9.82,9.35, P <0.01).CONCLUSION: Sitai tablet can remarkably reduce the blood pressure,blood sugar and blood fat, and increase the sensibility of insulin so that it can improve the general effects of IRS.
2.The Effect of Yinchenwulin Powder on the c-myc mRNA Expression of VSMC in Atherosclerosis Rats

Objective To observe the effect of Yinchenwulin(YCWL) powder on the c-myc rnRNA expression in vascular smooth muscle cell (VSMC) of atherosclerosis(AS) rats. Methods 70 Wistar rats were randomly divided into four groups: normal, model, Gypenosides treatment and YCWL powder treatment groups. AS models of rats were set up by feeding with high fat diet for one month. One month after treatment the level of serum lipid and blood rheology were measured, the phathological change of aorta was observed by HE staining, the ultrastructure change of aorta was observed by transmission electronic microscope, and c-myc mRNA expression in VSMC was detected by RT-PCR. Results Compared with Gypenosides treatment, YCWL powder could obviously decreased the levels of serum TC,TG and LDL, increase serum HDL level, reduce plasma viscosity,whole blood mid-cut,hematocrit and platelet adhesion rate, maintain aorta tissue structure, and down-regulate c-myc mRNA expression. Conclusion YCWL powder has therapeutic effect for atherosclerosis by regulating c-myc mRNA expression.
3.Effects of different serum microenvironments on culture of rat bone marrow mesenchymal stem cells in vitro

BACKGROUND:During culture of bone marrow mesenchymal stem cells(BMSCs),a certain serum is commonly added in the basic medium,such as calf serum and fetal bovine serum,but there are potential biological safety risks.OBJECTIVE:To study the effects of different serum microenvironments on in vitro culture of rat BMSCs.METHODS:BMSCs were harvested from adult rat bone marrow,and cultured in vitro by whole bone marrow adherence method.The cells were cultured under the following serum microenvironment.The primary cells of autoserum group were cultured with autoserum,changing the medium with fetal bovine serum after passage.The primary calls of homogeneity foreign serum group were cultured with homogeneity foreign serum,changing the medium with fetal bovine serum after passage.The primary cells of fetal bovine serum group were cultured with fetal bovine serum,and cultured with fetal bovine serum after passage.The primary cells of Dulbecco's modified Eagle's medium(DMEM)group were cultured with serum-free DMEM,changing the medium with fetal bovine serum after passage.The morphologic changes in BMSCs were detected under an inverted phase contrast microscope.Attachment rate and growth curve were measured.Surface marker CD11b,CD45 and CD90 expression was analyzed by flow cytometry.RESULTS AND CONCLUSION:In autoserum and homogeneity foreign serum groups,the homogenicity and degrees of fusion of call morphology were improved in comparison with other two groups and the day of first passage was less than other groups.The attachment rate was greater in the autoserum,homogeneity foreign serum and fetal bovine serum groups than the DMEM group at 24,48,72 hours(P<0.01).Doubling rate was fastest in the growth curve of autoserum group,followed by homogeneity foreign serum group and fetal bovine serum group.However,no doubling phenomenon was found in the DMEM group.Flow cytometry results demonstrated that the rates of CD11b-positive and CD45-positive cells at passage 3 were above 98% under medium containing serum,and CD90-positive rate was less than 2%.We could obtain BMSCs of higher purity.However,CD11b-positive rate was 95.83%,CD90-positive rate was 2.07%,but CD45 positive rate was only 64.79% under serum-free microenvironment.BMSC purity was significantly lower under serum-free microenvironment than under serum microenvironment.Results indicated that the microenvironment of rat autoserum can improve the attachment rate,growth and purification of BMSCs.
4.Impact of sulfentanyl on ropivacaine epidural block during abdominal panhysterectomy

Objective To explore the impact of sulfentanyl on sufentanil epidural block during abdominal panhysterectomy. Methods 90 patients scheduled for panhysterectomy were randomly divided into three groups. Tthe control group received epidural administration of 1% ropivacaine of 0.2 mL/kg after 2% idocaine of 3 mL , while the study group 1 received 10μg sufentanil and the study group 2 received 20μg sufentanil in addition to the medications used in the control group. The anesthetic effect, changes in vital signs, and incidence of adverse reactions were compared among the three groups. Results In group S1 and group S2, the onset of epidural anesthesia was faster , time to the highest plane of sensory blockade and time to degree 3 in the Bromag scores were faster , duration of sensory blockade was longer , and OAA/S score was better , as compared with group D , with significant statistical significances (P<0.01);and the effect was better in group S2 than in group S1. There was no difference among the three groups in adverse reactions. MAP , HR and SPO2 were lower in groups S1 and S2 than in group D during the procedure, with a statistical difference (P<0.05). Conclusions Proper dose of sufentanil plays a positive role in ropivacaine epidural block during panhysterectomy , not only increases the onset of anesthesia, but also makes the anesthestic effect better, and has higher safety It is worth popularizing clinically.
5.Effect of combined use of different dose of exmedetomidine and ultra-low dose naloxoneon postoperative hyperalgesia induced by remifentanil

Obejective To explore the effect of combined use of dexmedetomidine and ultra-low dose naloxone on postoperative hyperalgesia induced by remifentanil. Method 80 ASA gradeⅠ-Ⅱ female patients who were scheduled to perform endoscopic sinus surgery (ESS) were randomly divided into four groups: purely remifentanil group (group R) and remifentanil plus different dosage dexmedetomidine and ultra-low dose naloxone group (group RDN1 ~ RDN3). Then the postoperative VAS on 30 min, 1 h, 2 h, 4 h, 8 h, 12 h and 24 h together with the firstpostoperative pain time , PCA press numbers , fentanyl usage and adverse reaction rate were recorded and evaluated. Results Average VAS of group R are higher than other groups in all 7 time points at the adjusted significant level of 0.0083, and group R > group RDN1 > group RDN2 > group RDN3, while differences between group RDN2 and RDN3 in all time points are not statistical significant.There is no difference between group RDN1 and group RDN2 inthe first postoperative pain time and the PCA press time at 1h after operation. Difference between group RDN2 and group RDN3 in the PCA press time at 24 h after operation was not significant, and the fentanyl usage of RDN groups are significantly less than group R. Conclusions Combined use of dexmedetomidine and ultra-low dose naloxone induced by remifentanil can improving patients′postoperative hyperalgesia , effect increase with the dose of dexmedetomidine increas , and the increasement is more sensitive in acute pain.
6.Effect of Three-stage Traditional Chinese Medicine Rehabilitation on Neurological and Motor Function in Patients with Cerebral Ischemic Stroke

Objective To explore the effect of three-stage Traditional Chinese Medicine (TCM) rehabilitation on neurological function and motor function in patients with cerebral ischemic stroke. Methods 100 patients with acute cerebral ischemic stroke were randomly divided into experimental group (n=50) and control group (n=50). The three-stage TCM synthesis rehabilitation was used in the experimental group; the 2-week western medicine rehabilitation was used in the control group. The scores of National Institutes of Health Stroke Scale (NIHSS) and Fugl-Meyer assessment (FMA) were evaluated. Results There was no significance in the scores of NIHSS and FMA between both groups before and 4 weeks after treatment (P>0.05). The score of NIHSS was lower (P<0.01), and the score of FMA was higher (P<0.05) in the experimental group than in the control group in the 12th week. Conclusion Three-stage TCM synthesis rehabilitation can facilitate the recovery of neurological and motor function of patients with acute cerebral ischemic stroke.
7.Bioinformatics Analysis on Proteomics of Rat Bone Marrow Mesenchymal Stem Cells Intervened by Active Principle Region of Yang-Xin Tong-Mai Formula

This study was aimed to analyze the bioinformatics of proteomics of rat bone marrow mesenchymal stem cells (MSCs) intervened by active principle region of Yang-Xin Tong-Mai Formula (apr-YTF). The latest versions of bioinformatics tools including DAVID (http://david.abcc.ncifcrf.gov/) and GO (http://www.geneontology.org/) were combined to assign a precise function to rat bone marrow MSCs intervened by apr-YTF. KEGG and VISANT were assigned with a precise function to rat bone marrow MSCs intervened by apr-YTF. The results showed that a total of 102 biological processes were mainly involved, with 35 cellular components and 6 molecular functions. These proteins interacted in 3 signal transduction pathways. It was concluded that the following proteins and signal transduction pathways played an important role in the process of apr-YTF inducing BMSCs differentiation into cardiomyocytes. Presenilin-1 and Presenilin-2 were in the Notch signaling pathway. And syntaxin-4 protein was in soluble N-ethylmaleimide sensitive fusion protein attachment protein (SNARE). The apr-YTF played a role on MSCs from multiple sites, with multiple links through different biological processes. The bioinformatics of proteomics can predict action mechanism of traditional Chinese medicine (TCM) from the holism concept. The validation in combination with molecularbiology was a good way for TCM modernization.
8.Application of ibutilide in clincal atrial arrhythmia

Objective To study the efficacy and safety of ibutilide for AF and atrial flutter.Methods Thirty-two AF and atrial flutter patients with arrhythmia ≤3 months were randomly divided into ibutilide treatment group (n=17) and amiodarone treatment group (n=15).The patients in ibutilide treatment group were treated with 10 ml 5% glucose injection containing 1 mg ibutilide,which was repeated after 10 min if it was ineffective and those in amiodarone treatment group were treated with 10 ml 5% glucose injection containing 150 mg amiodarone,which was repeated after 10 min if it was ineffective.Results The total recovery rate of AF and atrial flutter was significantly higher in ibutilide treatment group than in amiodarone treatment group (64.7% vs 40.0%,P<0.05).The mean recovery time of AF and atrial flutter was significantly shorter in ibutilide treatment group than in amiodarone treatment group (29.28±12.57 min vs 70.59±16.83 min,P<0.01).Conclusion Ibutilide can rapidly recover AF and atrial flutter with a high success rate and a reliable safety.The therapeutic effect of ibutilide is better than that of amiodarone for AF and atrial flutter.

10.Effect of methane on P2X 7R/NLRP3 signaling pathway during inflammatory responses in a rat model of spinal cord ischemia-reperfusion

Objective:To evaluate the effect of methane on purinergic ion channel type 7 receptor (P2X 7R)/nod-like receptor protein 3 (NLRP3) signaling pathway during inflammatory responses in a rat model of spinal cord ischemia-reperfusion (I/R). Methods:Fifty-four clean-grade healthy male Sprague-Dawley rats, aged 3 months, weighing 350-380 g, were divided into 3 groups ( n=18 each) using a random number table method: sham operation group (group S), spinal cord I/R group (group I/R) and methane group (group M). Rats underwent sham operation in group S. Spinal cord ischemia was induced by occlusion of the thoracic aorta combined with controlled hypotension for 9 min, followed by reperfusion in anesthetized animals in group I/R.Methane-rich saline 10 mg/kg was intraperitoneally administered immediately after onset of reperfusion in group M. Motor sensory deficit index (MSDI) in hind limbs was measured at 12, 24 and 48 h of reperfusion.The L 3-5 segment of spinal cord was removed at 48 h of reperfusion for determination of the number of normal neurons (by Nissl staining), the number of activated microglia and expression of P2X 7R, NLRP3, apoptosis-associated speck-like protein containing CARD (ASC), cysteine-requiring aspartate protease (caspase-1), interleukin-1beta (IL-1β) and IL-18 (by immunofluorescence staining) in anterior and posterior horns of spinal cord. Results:Compared with group S, the MSDI was significantly increased at 12, 24 and 48 h of reperfusion, and the number of normal neurons in anterior and posterior horns of spinal cord was decreased, the number of activated microglia was increased, and the expression of P2X 7R, NLRP3, ASC, caspase-1, IL-1β and IL-18 was up-regulated at 48 h of reperfusion in group I/R ( P<0.01). Compared with group I/R, the MSDI was significantly decreased at each time point of reperfusion, and the number of normal neurons in anterior and posterior horns of spinal cord was increased, the number of activated microglia was decreased, and the expression of P2X 7R, NLRP3, ASC, caspase-1, IL-1β and IL-18 was down-regulated at 48 h of reperfusion in group M ( P<0.05). Conclusion:The mechanism by which methane reduces inflammatory responses is related to inhibiting P2X7R/NLRP3 signaling pathway in a rat model of spinal cord I/R.
