1.Effects of hemorrhagic shock and sodium cyanide intoxication on rabbit serum oxidative stress

Objective To investigate the effects of sodium cyanide (NaCN) and hemorrhagic shock on the level of serum oxidative stress in rabbits. Methods A total of 15 Japanese white rabbits were randomly divided into 5 groups: sham operation, hemorrhagic shock (HS), NaCN intoxation, HS+NaCN and HS+NaCN+4-dimethy laminophenol (4-DMAP). The rabbits were anesthetized for left arteria carotis communis cannulation, and connected with three-way tube and RM6240 electronic physiological signal recording system. The models of hemorrhagic shock and/or sodium cyanide intoxation (blood pressure was controlled at 5.3 kPa) were established. At 0.5, 1 and 2 h after the intraperitoneal injection of 2.5 mg/kg NaCN and/or 3.2 mg/kg 4-DMAP, the blood were drawn. Objective To investigate the expression of STAT3 signaling pathway genes including Survivin and COX-2 in cholangiocarcinoma,as well as the relationship between expression of these genes and prognosis of patients with cholangiocarcinoma.Methods The tumor and normal tissue samples were respectively collected from 43 patients with cholangiocarcinoma and 12 patients with intra-and extrahepatic bile duct stones or hepatic duct injury in the Affiliated Provincial Hospital of Anhui Medical University from September 2007 to July 2012.The expression of STAT3,phosphorylated-STAT3 (p-STAT3),Survivin and COX-2 were examined using immunohistochemistry,and the relationship between the expression of these genes and the clinical pathological features and prognosis of patients with cholangiocarcinoma was analyzed.Patients were followed-up through outpatient examination and telephone interview until March 2014.Categorical data were analyzed using the chi-square test.Correlation analysis was done by Spearman's method.The survival curve was generated using the Kaplan-Meier method,and the survival analysis was conducted using the log-rank test.Results The positive expression rates of STAT3,p-STAT3,Survivin and COX-2 in the tumor samples were 69.8% (30/43),65.1% (28/43),72.1% (31/43),79.1% (34/43),respectively,which were compared with 41.7% (5/12),8.3% (1/12),16.7% (2/12) and 41.7% (5/12) in the normal tissue samples,showing a significant difference for the last 3 indexes (x2=12.136,9.811,4.679,P < 0.05).Overexpression of p-STAT3,Survivin and COX-2 protein was correlated with lymph node metastasis (x2 =14.700,5.959,4.075,P < 0.05).Overexpression of p-STAT3 was also related to neural invasion (x2=10.384,P < 0.05).Expression of Survival and COX-2 protein was not associated with lymph invasion (x2=2.718,3.024,P > 0.05).Expression of p-STAT3,Survivin and COX-2 was however not associated with gender,age and tumor location,differentiation and diameter (x2=0.148,0.720,1.835,1.040,0.236 ; 0.001,0.009,0.029,1.863,0.197 ; 0.433,0.686,0.002,2.974,0.029,P > 0.05).Expression of Survivin and COX-2 protein was positively correlated to p-STAT3 protein (r =0.524,0.583,P < 0.05).All the 43 patients were followed up for 6-60 months.Among the 17 patients with hilar cholangiocarcinoma,the median survival time was 7,9,9 months for patients with positive expression of p-STAT3,Survivin and COX-2 protein,compared with 18,11 and 11 months for patients with negative expression of these proteins.The survival rates of the patients with positive and negative expression of p-STAT3 protein were 33.3% and 68.6%,respectively,with a statistical significance for p-STAT3 protein (x2=12.916,P < 0.05).Of the remaining 26 patients with common bile duct carcinoma,the median survival time was 9,10 and 9 months for patients with positive expression of p-STAT3,Survivin and COX-2 protein,compared with 20,20 and 20 months for patients with negative expression of these proteins.The survival rates of the patients with positive expression of p-STAT3,Survivin and COX-2 protein were 20.8%,9.4% and 8.5%,which were lower than 37.5%,37.5% and 50.0% of patients with the negative expression of these proteins,with a statically significance for all the 3 proteins (x2=12.787,6.245,11.161,P < 0.05).Conclusions The p-STAT3,Survivin,COX-2 proteins are highly expressed in the cholangiocarcinoma and the expression levels of these proteins are positively correlated.The survivin and COX-2 may be the downstream genes of STAT signaling pathway,which are involved in the progression and prognosis of cholangiocarcinoma.
3.Clinical characteristics of rheumatoid arthritis caused by pauciarticular arthritis

Objective To improve the comprehensive understanding and treatment level of the disease by analyzing the clinical characteristics of rheumatoid arthritis (RA) caused by pauciarticular arthritis.Methods The method was retrospective analysis and summary of clinical and laboratory data of 198 cases of patients with RA,in which 98 cases of pauciarticular arthritis belonged to the observation group and 100 cases of polyarticular arthritis belonged to the control group.Results Male patients in observation group were obviously more than those in the control group (t =2.456,P =0.015).Courses of disease was obviously shorter than those in the control group (t =-2.450,P =0.018).The number of involved joints was obviously less than those in the control group (t =-6.316,P <0.001).The incidence of morning.stiffness was significantly less than those in the control group (t =-3.884,P < 0.001).Rating scores of disease activity score 28 were significantly lower than those in the control group (t =-8.694,P < 0.001).And positive rate of anti-cyslic citrullinated peptide antibody is significantly higher than those in the control group (t =-2.299,P =0.022).Thyrotrophin levels were significantly higher than those in the control group (t =3.809,P < 0.001).There was no significant differences in age,erythrocyte sedimentation rate and C reactive protein,immunoglobulin and complement level,free triiodothyronine,free thyroxine,blood lipids,blood system and kidney involvement between the two groups (all P > 0.05).Conclusions The conclusion turns out to be that there are no typical clinical manifestations showing that rheumatoid arthrifts is caused by pauciarticular arthritis.Maybe it is the early performance of disease.It is worth attention that concurrent subclinical hypothyroidism turns out to be more.
4.Cytotoxic action of sulfur mustard on cultured lymphocytes and their caspase-3 expression

Objective To investigate the manner and molecular mechanism of lymphocyte cytotoxic action induced by sulfur mustard. Methods Lymphocytes were separated from the spleen of Wistar rats and were cultured. DNA strand breaks were detected by fluorospectrophotometry. RT PCR and Western blotting were employed to detect the gene and protein expressions of caspase 3 and its activation. Results Sulfur mustard induced DNA strand breaks in the lymphocytes. Gene and protein expressions of caspase 3 were increased in a time dependent manner. Conclusion Sulfur mustard induces lymphocyte cytotoxic action by means of depending on the expression and activation of caspase 3.
5.Interventional effect of tetrapeptide on sulfur mustard-induced lymphocyte apoptosis

Objective To investigate the changes of caspase 3 activity and the interventional effect of tetrapeptide AC DEVD CHO on sulfur mustard induced apoptosis of lymphocyte. Methods The activity of caspase 3 was detected by fluorospectrophotometry. DNA ladder and apoptosis were detected by DNA agarose gel electrophoresis and flow cytometry. Results Sulfur mustard enhanced the activity of caspase 3 and its polypeptide inhibitor (AC DEVD CHO) partially blocked apoptosis, which was mainly presented as an obscure phenomenon of DNA ladder and a decrease in percentage of apoptosis. Conclusion Sulfur mustard can induce the apoptosis of lymphocyte by means of activating caspase 3. The inhibitor may have an interventional effect on lymphocyte apoptosis induced by sulfur mustard.

Objective To detect the mutagenicity of sulfur mustard in the tk gene of L5178Y3.7.2c tk +/- mouse lymphoma cells. Methods The plating efficiency and the mutant frequency were detected by the microtiter procedure. Results Sulfur mustard induced tk locus mutation with mutation frequency 2-15 times higher than that of spontaneous mutation frequency of L5178Y3.7.2c tk +/- cells. There were two different phenotypes of mutation colonies induced, which were large and small colonies, but most of them were small colonies. Conclusion Sulfur mustard is characterized by marked mutagenic activity and cytotoxicity. The compound may result in large range of DNA damage.

Purpose:To Explore the requirements and accuracy for applying Microwave-ELISA Two-Steps-Test to the factory physical examination.Methods:Do the Microwave-ELISA Two-Steps-Test for 1,128 factory physical examination specimens that need the HBsAg and HBeAg inspection;comparing the result to the conventional ELISA test;and retest the specimens with the HBsAg-positive results.Results:There are 119 HBsAg-positive and 32 HBeAg-positive results for the ELISA test;there are 122 HBsAg-positive and 34 HBeAg-positive results for the microwave test;all the HBsAg-positive results are proved to be positive by using the colloid gold HbsAg rapid testing paper.Conclusion:Microwave-ELISA Two-Steps-Test is timesaving,simple and high sensitive for testing Hepatitis-B signs,which can reduce the hook phenomenon and can be used to do the large number of HBsAg and HBeAg checks in factory physical examinations.It can hasten the result report and greatly improve the efficiency.
8.Construction of a path for optimizing the health management of HBV infections among pregnant and lying-in women based on a Delphi method

Objective:
To establish an optimized path for health management of HBV infections among pregnant and lying-in women based on a Delphi method, so as to provide the evidence for intensifying the interruption of the mother-to-child transmission of HBV.
Methods:
Based on literature review and previous studies, the preliminary framework and contents of the optimized path for health management of HBV infections were constructed. Experts from epidemiology, clinical medicine and maternal and children healthcare were invited to participate in two-round Delphi consultations, and the preliminarily designed indicators were screened and revised. The score for feasibility of each indicator was calculated, and the weight of each indicator was estimated using a proportional distribution method.
Results:
Sixteen experts participated in the consultation, including 13 women. The participants had a mean age of (45.69±5.71) years, and a mean employment duration of (23.06±7.05) years. All participants had a degree of bachelor and above, and there were 14 experts with vice senior professional titles. The mean positive coefficient was 96.88% and the mean authority coefficient was 0.790 during the two-round expert consultations. There were significant differences in the coordination coefficient of importance, necessity and feasibility of indicators at all levels (P<0.05), and the coefficient of variation of the feasibility was all less than 0.250. The final optimized path for health management of HBV infections among pregnant and lying-in women included 6 primary indicators, 17 secondary indictors and 73 tertiary indicators. Among the primary indicators, delivery management (0.173 4), screening and evaluation (0.172 8) and pregnancy management (0.172 7) had a high weight.
Conclusion
A scientific and reliable optimized path is created for health management of HBV infections among pregnant and lying-in women, which has a potential value for improving the interruption of mother-to-child transmission of HBV.
9.Research progress of cell senescence in diabetic foot ulcers

Diabetic foot ulcers are common in elderly people with diabetes.Ongoing research is aimed at unraveling the complicated molecular mechanisms underlying diabetic foot ulcers.The role of cellular aging in delayed healing of diabetic foot ulcers has been recognized.Analyzing the relationship between cellular aging and diabetic foot ulcers, examining how cellular aging delays the healing of diabetic foot wounds, and identifying treatment methods for the removal of senescent cells and for slowing down cellular senescence will provide new therapeutic targets and strategies for the development of drugs for diabetic foot ulcers.
10.Preliminary studies on effects of methylprednisolone and dexamethasone on Th cytokines in asthmatic children

Objective This study was carried out to compare chnical efficiency of methylprednisolone (MP) and dexamethasone (Dex) and their effects on Th cytokines in asthmatic children.Methods A total of 39 children with moderate and severe asthma at acute exacerbation were randomly divided into two groups, one (A) with MP 1 -2 mg/kg (n=21) and the other (B) with Dex 0.25 -0.75 mg/kg (n= 18),every 12 -24 h by intravenous drip.Scores of respiratory effort and peak expiratory flow rate prior to and three days after treatment in the asthmatic children aged over five years were evaluated,respectively,as well as the time of wheezing vanishing after treatment.Serum levels of interleukin-2 (IL-2) and IL-4 were determined by radioimmunoassay (RIA) prior to and three days after treatment,respectively.Results There was statistically significant difference in the time of wheezing vanishing between groups A and B (P = 0.042).Three days after treatment,PEER was significantly higher in group A than that in group B (P = 0.025).No statistically significant difference in serum IL-2 and IL-4,as well as ratio of IL-2/IL-4,was found in group B prior to and three days after treatment (P>0.05).Statistically significant difference in serum IL-2 was not observed in group B prior to and three day after treatment (P>0.05),and serum IL-4 decreased significantly (P=0.001) and ratio of serum IL-2/IL-4 (P=0.027) increased significantly three days after treatment than those prior to treatment.No significant correlation between respiratory-effort scores and ratio of serum IL-2/IL-4 prior to treatment was found (P=0.613).Conclusions Up-regulation of IL-2,or inhibition of release of IL-4,probably is not the main anti-inflammatory mechanism of dexamethasone.Methylprednisolone has little effect on serum IL-2,but can effectively reduce serum IL-4, thus increasing the ratio of serum IL-2/IL-4 and counterbalancing function of the Thl/Th2 cells.
