1.Functional effect of the long non-coding RNAs on HIV-1 infection in monocytes

Objective To identify the expression of long non-coding RNAs(lncRNAs) and their putatively modulated cytokines in human immunodeficiency virus(HIV)-1 infected monocytes and to explore the role of lncRNAs in immunomodulation of HIV-affected host cells.Methods RNAs arrays were used to screen the lncRNAs differentially expressed in HIV-infected monocytes and subsequently confirmed by quantitative real time PCR (qRT-PCR).A siRNA was designed and transfected to the human monocytes, followed by interleukin(IL)-1β, IL-10 and tumor necrosis factor(TNF)-α detection with ELISA.Results The results of the screening assays and qRT-PCR showed that the expression of lncRNA-n266623 was up-regulated in monocytes after HIV-1 infection;the secretions of IL-1β,IL-10 and TNF-α in human monocytes were significantly enhanced following transfection of siRNA targeting lncRNA-n266623 in the monocytes.Conclusion The HIV-1 infection can promote the expression of lncRNA-n266623 which may inhibit the expressions of IL-1β, IL-10 and TNF-α in monocytes and negatively regulate the immune response to HIV-1 infection.
2.Expressions of vascular en dothelial growth factor and tranforming growth factor-α in condyloma acuminatum tissues

Objective To investigate the role of vascular endothelial growth factor (VEGF) and transforming growth factor-α (TGF-α) in the angiogenesis of condyloma acuminatum (CA). Methods Tissue specimens were obtained from the lesions of 30 patients with CA and foreskin of 15 normal human controls. Reverse transcription (RT)-PCR and immunohistochemical method were utilized to measure the protein and mRNA expressions of VEGF and TGF-α in the specimens. Microvessel density (MVD) was determined by staining with anti-CD34 polyclonal antibodies. Results A statistical elevation was observed in the expression of VEGF and TGF-α mRNA as well as MVD in CA specimens compared with the normal control specimens (P < 0.001, 0.001, 0.01). The expressions of VEGF and TGF-α protein were observed in 90% and 86.7% of the CA specimens, and in 40% and 26.7% of the control specimens (both P < 0.01). MVD was positively correlated with the expression of VEGF and TGF-α protein in CA tissues (both P < 0.01). Conclusion The overexpression of VEGF and TGF-α in CA tissue may accelerate angiogenesis in, and participate in the development and progression of, CA.
3.Problems and practice in clinical theory teaching of foreign students

4.Preliminary exploration on curriculum reform for foreign clinical medical students

5.Analysis of Distortion Product Otoacoustic Emission in Pilots Under Physical Examination for Modification to Equipment of High Performance Aircraft

Objective To evaluate the hearing function of pilots under physical examination for modification to equipment of high performance aircraft,using pure tone audiometry(PTA) and distortion product otoacoustic emission(DPOAE).Methods Analyzed were all the results of PTA and DPOAE of 75 pilots (150 ears) which accepted physical examination for modification to equipment of high performance aircraft in our department from March 2008 to February 2009.75 cases were divided into two groups according to their flight time.The difference between these two groups was studied.Results According to the Equipments of Physical Constitution for Flying Persons of Air force,PLA,all 75 pilots passed the ENT assessments as being qualified for flying.Of all 150 ears,abnormal DPOAEs were obviously higher than that of abnormal PTA tests.128 ears had normal PTA results.92 ears had normal DPOAE results.36 ears (27.6&) showed abnormal DPOAEs at one or more frequencies,especially at 1,3 and 4 kHz.22 ears had abnormal PTA results.20 ears had noticeable V or U shaped thresholds at 3 kHz and/or 4 kHz.The DPOAE amplitudes showed similar curves.The other two ears showed depressions at all frequencies,in both PTA and DPOAE tests.No statistically significant difference was found between the two groups in DPOAE.But statistical difference was found in DPOAE at 4 kHz compared with other frequencies.No statistically significant differences were found between the two groups in DPOAE amplitudes at 1,2,3 and 4 kHz.but at 6 and 8 kHz.Conclusion The application of DPOAE test helped find pilots who were on the early stage of noise-induced hearing loss.These pilots were the most important objects for us to follow up.
6.Effects of an anti-CD86 chimeric antibody (ch1D1) on autoreactive B lymphocytes isolated from pa-tients with SLE

Objective To investigate the effects of ch1D1, an anti-CD86 chimeric antibody, on autoreactive B lymphocytes isolated from patients with systemic lupus erythematosus ( SLE) . Methods Flow cytometry analysis was performed to measure the expression of CD86 on the surface of B cells isolated from patients with SLE and to analyze the effects of ch1D1 on the activation of CD4+T cells. The method of magnetic bead sorting was used to separate B cells, NK cells and CD4+T cells from PBMC collected from healthy subjects and patients with SLE for subsequent experiments. Antibody-dependent cell-mediated cyto-toxicity (ADCC) and complement-dependent cytotoxicity (CDC) that were mediated by ch1D1 were meas-ured with LDH release assay. Effects of ch1D1 on the secretion of auto-antibodies and the proliferation of CD 4+ T were detected by ELISA and 3 H -thymidine ( 3 H-TdR) incorporation assay, respectively. Results The levels of CD80 (68. 08±14. 28 vs 46. 10±12. 14, n=24, P<0. 000 1) and CD86 (44. 72±14. 90 vs 13. 99±10. 74, n=24, P<0. 000 1) expressed on the surface of B cells isolated from patients with SLE were significantly higher than those from the healthy subjects, suggesting the abnormal activation of B cells. Com-pared with the negative control group and the murine monoclonal antibody 1D1, ch1D1 was more effective in mediating the ADCC and CDC responses (P=0. 017 2, P=0. 038 8). Activated T cells significantly en-hanced the secretion of auto-antibodies by B cells isolated from patients with SLE. Compared with the nega-tive control group, the enhanced secretion of auto-antibodies was significantly inhibited by treatment with ch1D1 (P=0. 001 9). Moreover, ch1D1 significantly inhibited the proliferation and activation of CD4+T cells induced in patients with SLE (P=0. 002 4, P=0. 049 5). Conclusion ch1D1, the anti-CD86 chim-eric antibody, could effectively mediate the ADCC and CDC responses against autoreactive B cells isolated from patients with SLE, inhibit the secretion of auto-antibodies and suppress the proliferation and activation of auto-reactive CD4+T cells. It might be a potential immunotherapy agent for the treatment of SLE.
7.Study on the immunosuppressive activities of myeloid derived suppressor cells in patients suffering from recurrent spontaneous abortion

normal pregnancy and RSA (P=0.0250).Compared with women with normal pregnancy, those with RSA showed significantly decreased levels of Arg-1 ( P=0.0474) and suppressed activities of MDSCs against CD8+T cell proliferation (P=0.0230).Conclusion The MDSCs might play an important role in the main-tenance of maternal immune tolerance.Decreased number and inhibited function of MDSCs in the peripheral blood of women with RSA might be responsible for the development of RSA.
8.Association between body mass index and mortality among older Chinese: evidence from CHARLS

Objective:
To investigate the association between body mass index ( BMI ) and mortality risk among older Chinese based on the China Health and Retirement Longitudinal Study ( CHARLS ).
Methods:
The demographic features, BMI, prevalence of chronic diseases and mortality among the elderly at ages of 60 years and greater were captured from the CHARLS database from 2011 to 2018. A multivariable Cox proportional hazards regression model was used to examine the association between BMI and the risk of death.
Results:
Totally 6 023 subjects were enrolled, including 3 006 men ( 50.09% ) and 3 017 women ( 49.91% ), and 68.69% of the participants ( 4 137 subjects ) were at ages of 60 to 69 years. There were 637 subjects ( 10.58% ) with underweight, 1 544 ( 25.63% ) with overweight, and 557 ( 9.25% ) with obesity. During the follow-up period ( 35 091 person-years ), 1 035 subjects died. Multivariable Cox proportional hazards regression analysis revealed an increased risk of mortality among the underweight elderly ( HR=1.496, 95%CI: 1.261-1.775 ) and a reduced risk of mortality among the obese elderly ( HR=0.671, 95%CI: 0.511-0.881 ) relative to the elderly with normal weight, after adjustment for age, gender, smoking, household registration, administration of anti-diabetic drugs, administration of anti-dyslipidemia drugs, and administration of anti-hypertensive drugs.
Conclusion
It is found that the risk of mortality among the Chinese elderly correlatives with BMI through the analysis of CHARLS data.
9.Feasibility of school health management model based on Internet technology

Objective:
In order to understand and supervise the current situation of school health in Shanghai, the feasibility of internet-technology (IT) based school health management model is exarnined.
Methods:
Questionnaire survey, The feasibility analysis of IT based school health management model is discussed.
Results:
Principal directors from educational departments and health supervision centers more optimistic about the school health supervision model than school teachers(80.0%, 95.5%, 52.0%;73.3%, 90.9%, 55.1%). However, the three departments all in the publishing the information of school health(0, 13.9%, 6.3%).
Conclusion
IT based school health management model will become one of the most important supervision methods in the future. School Health management model based on IT is feasible.
10.Bridge internal fixation system for periprosthetic femoral fracture of Vancouver type B1

Objective:To investigate the therapeutic effects of bridge internal fixation system on the treatment of periprosthetic femoral fracture of Vancouver type B1.Methods:From June 2013 to October 2018, 10 patients with periprosthetic femoral fracture of Vancouver type B1 were treated by bridge internal fixation system at Department of Orthopedics, Zhucheng People's Hospital Affiliated to Weifang Medical College. They were 3 males and 7 females, aged from 65 to 84 years (average, 73.6 years). All patients had received hip replacement due to femoral neck fracture, including 6 hemi-hip replacements and 4 total hip replacements. Fracture had occurred in 9 cases after the primary hip replacement and in one case after revision. The time from primary hip replacement to the present surgery ranged from 1.5 to 4.0 years (average, 2.5 years). Recorded were operation time, intraoperative blood loss, fracture healing time, hip joint function and complications at the last follow-up.Results:In this group, operation time ranged from 65 to 114 min (average, 82 min), intraoperative blood loss from 110 to 320 mL (average, 145 mL). The 10 patients were followed up for 12 to 18 months (average, 15 months). Their X-ray films showed that bony union was achieved in all after 3 to 6 months (average, 4.3 months). According to their hip Harris scores at the last follow-up, 7 cases were rated as excellent, 2 as good and one as fair. Follow-ups revealed no loosening or breakage of implants, infection, femoral prosthesis loosening, fracture or femoral prosthesis displacement.Conclusion:Bridge internal fixation system is a good way to treat periprosthetic femoral fracture of Vancouver type B1, leading to satisfactory short-term outcomes and fine functional recovery.
