1.Immunogenicity and safety of the acellular pertussis, diphtheria, tetanus,inactivated poliomyelitis, Haemophilus influenza type b conjugate vaccine (DTaP-IPV/Hib combined vaccine):a meta-analysis

Objective To evaluate the effectiveness and safety of DTaP-IPV/Hib combined vaccine in comparison with commercially available DTaP (diphtheria, tetanus and pertussis), Haemophilus influenzae type b (Hib), tetanus conjugate and IPV monovalent vaccine. Methods Randomized controlled trials (RCTs) on DTaP-IPV/Hib were retrieved by searching interna-tional and national databases. The pooled mean difference and relative risk and 95% CI were assessed by meta analysis with RevMan 5.0 software. Results Totally 6 studies were included for the final analysis. The seroprotection/seroconversion level of the Anti-PT (RR=0.26, 95%CI: 0.14, 0.48) in combination vaccine was higher. The antibody titer levels of Anti-PT (WMD=21.11, 95%CI:9.36, 32.86), Anti-polio type1 (WMD=59.15, 95%CI:2.81, 115.48), Anti-polio type 3 (WMD=169.82, 95%CI:75.33, 264.30) were higher respectively. But the antibody titer level of Anti-PRP (WMD=-3.58, 95%CI:-5.52,-1.64) in the com-bination vaccine group was lower. Redness (RR=0.82, 95%CI:0.72, 0.93) and Tenderness (RR=0.45, 95%CI:0.30, 0.65) were lower in the combination vaccine. Swelling (RR=2.03, 95%CI:1.02, 4.01) was more common in the patients given the combina-tion vaccine. Conclusions This study supports the conclusion that the DTaP-IPV/Hib combination vaccine is equivalent to the separate injections based on similar antibody responses to the vaccine antigens, effectiveness and safety after primary doses.
2.The effect of health education intervention on iodine deficiency disorders in Chinese pupils and housewives:a Meta-analysis

Objective To comprehensively evaluate the effect of health education on intervening iodine deficiency disorders(IDD) in Chinese pupils and housewives,and to provide a basis for prevention and intervention of the disease.Methods A computerized literature search was carried out in Chinese Biomedical Database (CBM),Chinese National Knowledge Infrastructure(CNKI),VIP,Wangfang and PubMed to collect articles published from 1997-2012 concerning the effect of education intervention on iodine deficiency disorders (IDD).The study was a self-controlled intervention study.Meta-analyses were performed to assess the knowledge on IDD prevention and behavior changes.Fixed and random effect models were employed to combine results after a heterogeneity test,and rate different(RD) was used as an indicator of the intervention effect.Results A total of 20 controlled before and after studies were included in our meta-analysis.The quality of individual studies was assessed by EPOC,and all studies were grade B.Because the heterogeneity was higher,each knowledge point was stratified analyzed according to the baseline rate.The results showed that the lower the baseline rate,the better the effect of the intervention.The analysis indicated that the RD for the main harm of IDD,the RD for the key measure to prevent IDD,the RD for the meaning of the pattern on the bags,and the RD for knowledge dissemination of IDD were increased by 35%,29%,27%,42% and 47%,respectively.Conclusion Health education is helpful to Chinese pupils and housewives in improving their IDD related knowledge and their attitudes toward IDD.

Objective To explore the effects of repetitive transcranial magnetic stimulation on behaviors and hippocampus neuronal apoptosis in rats with chronic stress-induced depression.Methods 40 male SD rats of SPF grade were randomly divided into normol control group (n =8) and model preparation group (n =30) after screening.Rats in model preparation group were singly housed and given chronic unpredictable mild stress(CUMS) to build depression model.Excluding unsuccessful modeling rats,the model preparation group was divided into three groups:model group(n=8,without any treatment),rTMS group (n=8,with the intervention of 10 Hz rTMS) and shame group (n=8,simulation of rTMS environment without rTMS stimulus).The changes of behaviors in each group were detected by weight measurement,sucrose consumption test and open-field test.The changes of morphology of hippocampal neurons were detected by Nissl's staining.The changes of Bax in hippocampal neuron were detected by Immunohistochemical staining.Results (1) Behavioral results showed stress for 21 d could make rat behavior scores decrease significantly(all P<0.05),and rTMS intervention could significantly improve their behavior scores (all P<0.01).Compared with model group,the weight reduction rate (0.32±0.05)%,the score of sucrose consumption test(7.03 ± 1.02) and the score of open field test(8212.41 ± 1416.15,8.75 ± 1.58) in rTMS group was higher(P <0.01).(2) Nissl staining showed stress for 21 d could make the number of hippocampal CA3 neurons was reduced,cell morphology was poor,and the number of Nissl bodies was reduced.rTMS intervention could increase the number of hippocampal CA3 neurons,cell morphology was integral and the number of Nissl bodies was increased.(3) Immunohistochemistry results showed stress for 21 d could cause the number of Bax cell were significantly increased(P<0.01),and rTMS intervention can make the number of Bax cell were significantly lower(P<0.01).Conclusion rTMS intervention improves the depressive behavior in chronic stress depression model rats and inhibits the apoptosis,which might work through inhibition of neuron apoptosis and decline of Bax expression in hippoeampal neurons.
5.Marrow mesenchymal stem cell transplantation in treatment of ischemic heart disease in rabbits

Objective To investigate the feasibility of treating ischemic heart disease with transplantation of marrow mesenchymal stem cells.Methods Marrow mesenchymal stem cells were collected from New Zealand rabbit's sternum and cultured.A myocardial infarction model was created by ligation of the distal left anterior descending artery in New Zealand rabbit.MSCs were injected into the region of myocardial infarction.The size of the myocardial infarction area was measured by PET and the cardiac function was assessed by measuring the pressure change of left ventricle(dp/dt).Results The size of the myocardial infarction area diminished and the cardiac function was improved after the transplantation of marrow mesenchymal stem cells.Conclusion Transplantation marrow mesenchymal stem cells may improve cardiac function of ischemic heart disease in animal model of New Zealand rabbit.
6.The effect of repetitive transcranial magnetic stimulation on behaviors and hippocampal GR protein ex-pression in depression model rats

Objective To explore the effect of repetitive transcranial magnetic stimulation (rTMS) on behaviors and hippocampal glucocorticoid receptor (GR) protein expression in chronic stress depression model rats and the possible antidepressant mechanism of rTMS. Method Seventy-five male Sprague-Dawley rats were randomly divided into the blank control group (n=15) and the stress-induced group (n=60). Singly housing and chronic unpredictable mild stress (CUMS) were used to induce the depression model in stress-induced group. Forty-five CUMS rats were selected and ran?domly divided into rTMS group (receiving 10 Hz rTMS intervention for 3 weeks), sham group (receiving pseudo rTMS treatments for 3 weeks) and depression group (with no further treatment). Body weight measurements and performance in the sucrose consumption and forced swimming test (FST) were evaluated before modeling, after modeling and after inter?vention. The GR protein and GR mRNA expression level in the hippocampus were examined after intervention. Results Compared with control group, the body weight growth rate and the sugar water preference were significantly lower in stress-induced group (P<0.01), and the immobility time of FST was significantly longer (P<0.01). After the 3-week rTMS intervention, the body weight growth rate and the sugar water preference in rTMS group, which were insignificantly differ?ent from control group (P>0.05), were higher than those in sham group and depression group (P<0.01). The immobility times of FST in rTMS group and control group were shorter than sham group and depression group (P<0.01). Compared with rTMS group and control group, GR and GR mRNA expression levels in the hippocampus were significantly reduced in sham group and depression group (P<0.01). Conclusion rTMS can improve depression behavior of CUMS rats, which may be associated with upregulation of GR expression in the hippocampus.

To further investigate the possible molecular mechanism of rTMS treatment for depres?sion. Methods Forty male Sprague-Dawley rats of SPF grade were randomly divided into the blank control group (n=8) and the stress-induced group (n=30). Singly housing and chronic unpredictable mild stress (CUMS) were used to induce the depression model in stress-induced group. Twenty-four model rats were divided into three groups:model group (with no further treatment), rTMS group (receiving 10 Hz rTMS intervention for 3 weeks) and shame group (receiving pseudo TMS treatments for 3 weeks). Weight measurement, sucrose consumption test and open-field test were used to assess the behavior changes. The rat hippocampal CA3 area of BDNF positive staining cell number and expression levels of BDNF mRNA in hippocampus were examined after intervention. Results The weight reduction rate, score of sucrose consump?tion test and the score of open field test were significantly higher in rTMS group than in model group (P<0.05). The num? ber of BDNF staining positive cells in the hippocampal CA3 area was lower in model group and shame group than in the blank control group whereas was higher in the rTMS group than in the model group (P<0.01). Compared with the model group, the BDNF mRNA relative expression was significantly increased in the hippocampus of rTMS group (P<0.01). Conclusion rTMS can improve depressive behaviors of CUMS rats probably through the increase in expression of BDNF in the hippocampal neurons and neuronal regeneration.
8.Evaluation of long-term-siRNA treatment with HBV transgene mice on inhibit replication of hepatitis B virus

Objective:To investigation of the long-term-siRNA treatment with HBV transgene mice on inhibit replication of hepatitis B virus .Methods:The constructed siRNA expressed vectors was transfected HBV transgene mice by hydrodynamics -based in-jection via vena caudalis .Different groups were set including:specificity siRNA groups ( pSilencer5.1/C2,pSilencer4.1/C2,pSilenc-er3.1/C2),PBS group and negative vector group (n=10).The effect was observed in different periods (6 d,21 d,1 months,3 months, 6 months and 9 months after injection ) .HBsAg was analyzed by Chemiluminescence method , HBV-DNA was analyzed by real time quantitative PCR ( RQ-PCR) .Results:Compared with the PBS group , specificity siRNA groups showed decreased levels of HBsAg and HBV-DNA (P<0.05).Negative vector group did not show such changes ,there were no significant differences (P>0.05).Conclu-sion:The siRNA based on the expression vector can suppress the expression and replication of HBV in HBV transgene mice .The inhi-bition effects of long-term-siRNA treatment was specific .
9.Selective nerve excitability induced by symmetric biphasic pulses

BACKGROUND:Studies have shown that the application of electrical stimulation of peripheral nerve fibers can recover partial muscle functions due to the loss of central nervous control.
OBJECTIVE:To verify the feasibility of selective nerve excitatory effectively used the symmetric biphasic pulses under the bipolar electrodes stimulation of 1 mm.
METHODS:Eight adult Wistar rats were selected to expose the sciatic nerves after anesthesia and then the electrodes were placed on the sciatic nerves careful y to establish the model of selective nerve stimulation. Experimental electrode was homemade Cuff bipolar electrode, and the electrode stimulators were Grass S88
stimulator and AWG2005 arbitrary waveform signal generator. The two-way dual-electrode stimulation was used. The distance between two electrodes was 1 mm, and the stimulation waveform was symmetric biphasic pulse
with the width of 0.2 ms. The output pulse amplitude, pulse width and delay could be adjusted. The stimulation
intensity was adjusted, and the law of nerve excitability was detected under two-way dual-electrode stimulation, in order to achieve selective nerve excitability, and the feasibility of two-way dual-electrode stimulation to achieve
selective nerve excitatory was verified with“col ision”method.
RESULTS AND CONCLUSION:The change of nerve action potential was amplified by P511 amplifier and then linked into oscil oscope for displaying, and dual-electrode stimulation waveform was the symmetric biphasic pulse with the width of 0.2 ms. With the increasing of stimulus amplitude, we achieved the selective nerve excitatory. The results indicate the selective nerve excitatory can be achieved with the closed (1 mm) dual-electrode symmetric pulse, and the feasibility and effectiveness of this method can be verified with“col ision”principle.
10.Establishment and application of HCV genotype liquichip detection method

Objective To establish a liquichip method for detecting 6 sub-genotypes of hepatitis C virus(HCV),including 1a, 1b,2a,3a,3b and 6a.Methods The coupling method of PCR amplification and nucleic acid probe was established.The PCR product and the microspheres mixture of the coupled nucleic acid probe were hybridized for establishing the liquichip detection method.The sensitivity and specificity of the established liquichip detection method were evaluated.Nucleic acid in 93 serum samples was detec-ted by this method..Results The established HCV nuclei acid liquichip genotype detection method had the higher specificity and sensitivity,which could detect and classfy 6 HCV sub-genotypes.The sensitivity for HCV 1a,3a and 6a sub-genotypes was 1× 105 copies/PCR;the sensitivity for HCV 1b,2a and 3b sub-genotypes was 1×104 copies/PCR.The detection results in 93 serum samples showed that the this genotyping method had the characteristics of high throughput,rapidness,sentsitivity and specificity. Conclusion This method can be used for the simultaneous and quick detection of 6 HCV sub-genotypes and provides a new meth-od for the genotyping detection of HCV.
