1.Influence of long cold ischemia preservation time on ultrastructural morphology of donor heart

Objective To investigate the impacts of long cold ischemic preservation time on the ultrastructural changes of donor heart and to provide the data for expanding the heart donor pool. Methods Heart was obtained from brain dead donor and preserved in the the University of Wisconsin (UW) solution at 4℃. Cardiac tissues were harvested at different time points of cold ischemic preservation (6, 8, 10, 12, 14 h) and observed under electron miscroscope. Results Donor heart did not have significant pathologied and ultrastructural changes when cold ischemic preservation time was 6 h. After that, time related impairment of myocardia and endothelium of coronary artery was seen.When ischemic time was longer than 12 h, focal myocardial necrosis and complete loss of the endothelium were detected. Conclusions Myocardial ultrastructure is an important index to evaluate the donor heart quality. Heart, which underwent 10 h of cold ischemia preservation time, causes no significant irreversible and pathological ultrastructural changes, and could be used for heart transplantation. When ischemia time was over 10 h, the donor heart presented with irreversible change and was nolonger unsuitable for transplantation.
2.Diagnosis value of 128-slice spiral CT in patients with adult intussusception

Objective To investigate the diagnosis value of 128-slice spiral CT in patients with adult intussusception. Methods Direct features of 128-slice spiral CT of 63 adult intussusception patients confirmed by operation and pathology were analyzed retrospectively. Results In 63 cases of intussusceptions, 1 case had idiopathic intussusception, and the other 62 cases had secondary intussusception (1 case with multiple intussusceptions). Direct signs included:target signin 60 cases,double intestines signin 59 cases,blood vessel curling signin 56 cases, andcomet-tail signandkidney signin 51 cases. Lipoma was the most common benign lesions of intussusception, and colon cancer was the most common malignant lesions of intussusception. Conclusions The 128-slice spiral CT combined with multiple plane restructuring has the important value in diagnosing adult intussusception.
3.Effects of paclitaxel on the intimal proliferation in rat aortic allografts

Objective To investigate the effect of paclitaxel on the intimal proliferation in rat aortic allografts and the possible mechanism on preventing graft arteriosclerosis.Methods Thirty-two inbred(Wista)r rats and 16 SD rats were divided into three groups:the isografts control group(Wistar/Wistar),the(allografts) control group ((Wistar)/SD) and test group(Wistar/SD) randomly(16 rats each groups).The rat abdominal aortic allograft model was used.The rats in test group were treated with paclitaxel after operation and those in control group with(0.9 %) normal saline.The grafts were removed and measured by means of pathology and immunohistochemistry 30 days later.Results The results showed that the thickness of the(aortic) intima,the degree of inflammatory cells infiltration in adventitia,stenosis ratio and the expression of(PCNA) were decreased in test group as compared with the allografts control group.Conclusions Paclitaxel can inhibit intimal proliferation in aortic allografts and prevent the graft arteriosclerosis.The mechanism is related to inhibition of vascular smooth(muscle) cell proliferation and alleviation of aortic allografts rejection.
4.The Effects of Ulinastatin on Proinflammatory Cytokine and Oxygen Free Radicals During Acute Lung Injury in Rabbits

Objective To study the effects of ulinastatin on proinflammatory cytokine and oxygen free radicals during acute lung injury in rabbits. Methods Eighteen health New Zealand rabbits were randomly divided into 2 groups: oleic acid group(n=9) and ulinastatin group(n=9). The rabbits of oleic acid group were intravenously injected analytical pure oleic acid(0.12ml/kg) to set up a model of acute lung injury. The rabbits of ulinastatin group were intravenously injected ulinastatin 50000u/kg before injecting oleic acid and received intravenous infusion of ulinastatin 5000u?kg -1 ?h -1 after injecting oleic acid. Before injecting oleic acid and 1h, 2h, 3h and 4h after injecting oleic acid,venous blood samples were taken to determine the concentration of plasma IL-8, MDA and active oxygen,and arterial blood samples were also taken for gas analysis. Results The concentration of IL-8, MDA and active oxygen and PaCO 2 gradually increased,and PaO 2 gradually decreased after injecting oleic acid in the both groups of rabbits. The changes of the above indices in the oleic acid group were larger than those in the ulinastatin group, and the difference between the two groups was significant. Conclusion Ulinastatin inhibited the release of proinflammatory cytokine and reduced the production of oxygen free radicals during acute lung injury.
5.Superparamagnetic iron oxide labeled mesenchymal stem cells and their magnetic resonance imaging in vitro

Objective To explore the optimal situation of labeling bone mesenchymal stem cells (BMSCs) with superparamagnetic iron oxides (SPIO) mediated by poly-L-lysine (PLL), and determine the most optimal protocol of magnetic resonance imaging according to the patterns of MR in vitro. Methods BMSCs were isolated from white rat and purified, incubated with SPIO-PLL complexes at the range of concentrations (0, 4.2, 8.4, 21, 42, 84 ?g Fe per ml medium). The labeling ratio and distribution of SPIO particles in BMSCs, and the morphological evidence of abnormal visualization were evaluated by Prussian blue staining, fluorescent microscope and electron microscopy. MTT growth curves and magnetic resonance imagings were obtained at the range of concentrations. Trypan blue exclusion test was performed to elevate the viability of BMSCs labeled with PLL at the range of concentrations (0, 0.05, 0.25, 0.5, 1.0, 5.0 ?g PLL per ml medium). Objective To investigate changes in the number of endothelial progenitor cells (EPC) from peripheral blood and pathological feature in the development of transplant arteriosclerosis in mouse abdominal aortic allografts, and discuss their correlations. Methods A segment of abdominal aorta was transplanted orthotopically from C57BL/6 to Balb/c mice. The grafts were harvested at 3rd day, 2nd week, 4th week and 6th week after the operation and studied by light and electronic microscopy. Regional changes in the lumen and intima were measured with computer imaging analysis system. EPC from peripheral blood were quantified by flow cytometry. Results Endothelium injury and inflammatory cells infiltration were seen in the aortic allografts at 3rd day after transplantation.Neointimal lesions and acute rejection were observed as early as 2nd week after surgery. The lumen of allografts was significantly narrowed due to neointima hyperplasia and had progressed at 4th and 6th week postoperatively. The number of circulation EPC was increased from 1 st day after operation and reached the peak at 3rd day. Thereafter the number of EPC was decreased rapidly and significantly less at 14th and 28th day postoperation than that pre-operation. Conclusion Abdominal aortic transplantation from C57BL/6 to Balb/c mice presents typical pathological feature of transplant arteriosclerosis. The number of EPC from peripheral blood is related to the process of injured endothelial repair and neointima formation of aortic grafts. EPC count may be considered a novel biological marker and therapeutic intervention for transplant arteriosclerosis.
7.Clinicopathology, prognosis and EG-1 mRNA in breast cancer

Objective To investigate the correlation between the expression of EG-1 in breast cancer and the clinicopathological factors and prognosis of breast cancer.Methods EG-1 mRNA expression in 72 malignant and 18 benign breast tissues were evaluated by RT-PCR method,and its correlation with clinical characteristics and prognosis were retrospectively analyzed.Results EG-1 expression level was higher in malignant tissue than in the corresponding benign breast tissue(71%vs.24%,P<0.05).EG-1 expression was significantly correlated with lymph node metastasis and protein VEGF (P<0.05)and was not significantly with age,menopause,tnmor size,grade,hormone status,protein Her2 and pS3.The metastastic rate and recurrence rate in EG-1 positive patients was higher than that in EG-1 negative patients.Multivariate Cox model showed that EG-1 positive was an independent factor affecting overall survival and disease free survival.Conclusions EG-1 expression was up-regulated in breast cancer and significantly correlated with lymph node metastasis and protein VEGF.
8.Complex of growth differentiation factor-5 and fibrin gel for repair of lumbar disc injury in rabbits

Objective To investigate the result of local injection of growth differentiation factor-5 (GDF-5) and fibrin gel for treatment of lumbar disc injury in rabbits.Methods Lumbar puncture with a 20-gauge needle was performed at L3/4,L4/5,and L5/6 discs of 40 New Zealand white rabbits.After needle puncture,L3/4 discs were injected with GDF-5 and fibrin gel (compound group) ; L4/5 discs fibrin gel (fibrin gel group) ; L5/6 discs nothing (blank control group).Two weeks later,intervertebral disc degeneration in each group was observed via radiography,MRI and nucleus proteoglycan content detection and histological examination.Results At postoperative 2-,4-,8-,and 12-weeks,X-ray films revealed a gradual decrease in disc height index (DHI) among the three groups,but the decreasing velocity was lower in compound group than in other two groups (P < 0.05).On MRI,the signal of intervebral discs among the three groups diminished progressively with time,but a relatively lower decreasing was observed in compound group (P < 0.05).At postoperative 4-week,proteoglycan content of the nucleus pulposus was (6.3-± 0.4) in compound group,higher than (5.9-0.4) in blank control group and (5.8-± 0.3)in fibrin gel group (P <0.05).At postoperative 2-week,histological evaluation showed (5.28 ±0.41)points in compound group,lower than (7.54 ± 0.53) points in blank control group and (7.21 ± 0.44)points in fibrin gel group (P < 0.05).Conclusion Local injection of GDF-5 and fibrin gel facilitates the restoration of the injured discs and delays further disc degeneration.
9.Effects of bevacizumab on antioxidative function in human retinal pigment epithelial cells

Objective To investigate the effects of bevacizunab on the antioxidative function of human retinal pigment epithelium (RPE),in order to explore the possible mechanism of macular atrophy induced by the application of anti-vascular epithelial growth factor (VEGF) agents in age-related macular degeneration.Methods Human RPE cells were incubated in DMEM/F12 medium containing 0.25 g · L-1 bevacizumab and divided into 5 groups according to incubation period:0 hour(control),12 hours,24 hours,48 hours and 72 hours,and then the oxidative stress was induced by adding H2O2.Cell viability was measured by the CCK8 assay.MitoSox Red was used to determine mitochondrial reactive oxygen species (mtROS) production.Mitochondrial membrane potential was measured using the JC-1 assay.The expression levels of NOX4 and HO-1 were detected by RT-PCR and Western blot,respectively.Results CCK8 assay determination showed that the above treatment had no significant effect on cell viability,the cell viability of 0 hours,12 hours,24 hours,48 hours and 72 hours were (100.2 ±3.3)%,(99.2 ±2.7)%,(102.5 ±6.4)%,(103.9 ±3.7)%,(103.6 ±3.3)%,the difference was not statistically significant (P > 0.05).Compared with the control group,the levels of mtROS increased at 12 hours,24 hours,48 hours and 72 hours,the difference was statistically significant (P < 0.05).Mitochondrial membrane potential at 12 hours,24 hours,48 hours,72 hours were lower than the control group,the difference was significant,48 hours reached the lowest,72 hours significantly increased,but still lower than the control group.RT-PCR and western blot results demonstrated that the expression of NOX4 mRNA and protein increased at 12 hours,24 hours,48 hours and 72 hours,and reached the highest at 24 hours,then decreased significantly,but still higher than the control group,the difference was statistically significant (P <0.01).Compared with the control group,the expression of HO-1 mRNA decreased at 24 hours,48 hours and 72 hours,while the expression of HO-1 protein decreased at 48 hours and 72 hours,the difference was statistically significant (P < 0.05).Conclusion The clinical concentration of bevacizumab can reduce the anti-oxidative function of RPE cells,which may be one of the causes of progressive macular atrophy after long-term anti-VEGF therapy.
10.Effects of Sodium Tanshinone ⅡA Sulfonate on Cardiac Hypertrophy and MAPK Signaling Pathway

Objective To determinate the effects of sodium tanshinone ⅡA sulfonate(STS)on cardiomyocyte hypertrophy and explore the relative effects of STS on mitogen-activated protein kinase signal transduction system in rats with cardiomyocyte hypertrophy through constricting the thoracic aorta.Methods The models of cardiomyocyte hypertrophy were established in vivo,and the thoracic aorta was partially tied between the right innominate and the left common carotid arteries.The rats were randomly divided into 6 groups(n=8/group)as follows:①sham,②transverse aortic constriction(TAC),③TAC+low-dose Tan(TAC+LT)(5 mg/kg),④TAC+middle-dose Tan(TAC+MT)(10 mg/kg),⑤TAC+high-dose Tan(TAC+HT)(20 mg/kg),and ⑥ TAC+Val(10 mg/kg).After treatment for 8 weeks,echocardiography was performed to observe the changes in hypertrophy and heart function,and heart samples were cut into transverse sections and stained with hematoxylin and eosin(H&E).The MAPKs protein expression in the cardiomyocytes was detected by Western blot.Results The heart weight index(HWI),left ventricular mass index(LVMI)and cross-sectional diameter of cardiomyocytes(CD),left ventricular posterior wall thickness(LVWT),and interventricular septal thickness(IVS)were significantly increased in TAC group as compared with sham group.The relative parameters in STS groups and Val group were reduced as compared with those in TAC group.Western blot analysis revealed the p-ERK and p-p38 expression was significantly decreased in TAC group as compared with sham group(P<0.01).The p-ERK expression was significantly decreased in STS groups and Val group as compared with TAC group(P<0.05).The TAC+HT group,TAC+MT group and Val group had significantly higher p-p38 expression than TAC group(P<0.05).Conclusion Tanshinone ⅡA could regulate the expression of protein in MAPK pathway to exert its inhibitory effects on hypertrophy of cardiomyocytes.
