1.Establishment and application of networked information management system for clinical laboratories in community of Suzhou City

Objective To establish a networked information management system for clinical laboratories in community of Suzhou and evaluate its clinical application.Methods Based on dedicated network of public health information in Suzhou City,laboratory information management system (LIS) and hospital information management system (HIS),the management for centralized detections of clinical samples from the health institutions of intra-regional community were performed as the key mission,and a regional,networked and informationized management system was designed for the clinical laboratories of community.Results The informationized management of whole process including test application,specimen collection,delivery and receiving,report releasing and payment,etc.was achieved on line.The information of clinical laboratories among the communities was transferred and shared in whole Suzhou City.Conclusion The established system should be safe,convenient and efficient online network-based information management system between the Suzhou clinical laboratory center and the laboratories in each local community,by which the centralized detections in the clinical laboratories of community could be performed readily.
2.The Expression of Hyaluronan Synthase mRNA in the Endolymphatic Sac Cells and Renal Tubules

Objective To study the difference between the mRNA expressions of hyaluronan synthase in the epithelial cells of endolymphatic sac and renal tubules with the oligonucleotide probe. Methods The oligonucleotide probe of hyaluronan synthase was designed and synthesized, the mRNA expressions of hyaluronan synthase in the epithelial cells of endolymphatic sac and renal tubules were detected with hybridization in situ. Results mRNA of hyaluronan synthases were strongly expressed in some epithelial cells of endolymphatic sac and renal tubules. Conclusion It is confirmed that a dual regulatory system for hyaluronan /hyaluronidase exists in the epithelial cells of endolymphatic sac and renal tubules at the level of molecular biology.

4.Study on CE fingerprint of Salvia miltiorrhiza

Objective: To investigate the CE fingerprint of Salvia miltiorrhiza . Methods : Separation was performed on a 50?m?50cm uncoated capillary with 20mmol?L -1 borate solution as CE buffer. The run voltage was 15kV and the UV detection was set at 210nm. Results : Fingerprint consisted of 11 common peaks. The validation of methods meet the requirements for SDA's technical regulations. Conclusion : The method was accurate and simple for quality control of Salvia miltiorrhiza.
5.Beta-lactamase Gene and intⅠ1 Gene in ESBLs-producing Klebsiella pneumoniae

OBJECTIVE To investigate the beta-lactamase gene and intⅠ1 gene in ESBLs-producing Klebsiella pneumoniae.METHODS Beta-lactamase gene and intⅠ1 gene were analyzed by PCR.RESULTS The positive rates of TEM,SHV,OKP,CTX-M-1group,CTX-M-2group,CTX-M-9group,GES,PER,VEB,OXA-10,ACT-1,LEN,DHA,and intⅠ1 were 51.4%,0%,0%,17.1%,0%,11.4%,0%,0%,0%,0%,0%,5.7%,17.1%,and 60%,respectively.Beta-lactamase gene was found in 62.9%,intⅠ1 gene was found in 60%.CONCLUSIONS There are very high positive percentages of beta-lactamase gene and intⅠ1 gene in ESBLs-producing K.pneumoniae.
6.The effects of the neotype hand-arm sling on shoulder subluxation and transferring movement in hemiplegic stroke patients

Objective To explore the effects of the neotype hand-arm sling on shoulder subluxation and transferring movement in hemiplegic stroke patients.Methods Thirty hemiplegic stroke survivors with shoulder subluxation and poor elbow flexor muscle tone (<2 on the Modified Ashworth Scale).were selected and randomly divided into three groups.Group 1 was trained in static upright standing,plus 10 min of sit-to-stand training and 20 min of walking training with no support for the hemiplegic,subluxed shoulder.Group 2 did the same training using a bagtype shoulder sling,and group 3 used the neotype hand-arm sling.Index finger palpation was used to evaluate the degree of shoulder subluxation,and the Five Times Sit-to-Stand (FTSST) and Timed Up-and-Go (TUG) times for the three groups were compared.Results In the static standing training,both the bag-type sling and the neotype hand-arm sling could effectively reduce shoulder subluxation.However,after 10 min of sit-to-stand training the neotype hand-arm sling was found to be significantly superior to the bag-type shoulder sling in improving shoulder subluxation,with the latter having no effect on shoulder subluxation.In the 20 rin of walking training,the neotype hand-arm sling provided significantly more effective support than the bag-type shoulder sling,although both could support the shoulders.Those using the neotype sling had shorter FTSST times,though there were no significant differences in the average TUG times.Conclusions The neotype slings can more effectively counter shoulder subluxation in upright stance,especially during sit-to-stand maneuvers and walking.It might have positive effects on transferring ability.

AIM: To optimize the technical parameters of total arasaponin'S purification with macroporous resin. METHODS: Conditions for the elution were studies under the conditio n of the content of total arasaponin. RESULTS: The purity of total arasaponin was up to 83.3% after d riing the part of 70% ethanolic elution. CONCLUSION: The method was a good purification for total arasaponi n.
8.Comparison on Intestinal Absorption of Pectolinarin and Pectolinarigenin by Everted Rat Intestinal Sac Method

This article was aimed to study the intestinal absorption about main active ingredients of pectolinarin and pectolinarigenin in Carboned Cirsium japonicum DC. The absorption rate and absorption rate constant were taken as indicators. The intestinal absorption of pectolinarin and pectolinarigenin were compared by everted rat intestinal sac method among different parts of the small intestine. The results showed that the absorption rate constant of pectoli-narin among duodenum, jejunum, ileum and colon parts were 0.505 1 ± 0.192 7, 0.936 0 ± 0.187 2, 0.732 0 ±0.133 5, 0.251 3 ± 0.027 6 (μg·h-1·cm-2). The absorption rate constant of pectolinarigenin among the duodenum, je-junum, ileum and colon were 0.059 1 ±0.008 3, 0.093 3 ±0.029 2, 0.112 3 ± 0.035 6, 0.029 4 ± 0.009 1 (μg·h-1·cm-2). It was concluded that the absorption of both ingredients increased over time. The absorption of both ingredi-ents in the jejunum and ileum was higher than other parts of the small intestine. The absorption rate of pectolinarin in the entire small intestine was much higher than the absorption rate of pectolinarigenin.
9.Protective effect and mechanism of sodium tanshinone II A sulfonate on microcirculatory disturbance of small intestine in rats with sepsis.

To explore the protective effect of sodium tanshinone IIA sulfonate (STS) on microcirculatory disturbance of small intestine in rats with sepsis, and the possible mechanism, a rat model of sepsis was induced by cecal ligation and puncture (CLP). Rats were randomly divided into 3 groups: sham operated group (S), sepsis group (CLP) and STS treatment group (STS). STS (1 mg/kg) was slowly injected through the right external jugular vein after CLP. The histopathologic changes in the intestinal tissue and changes of mesenteric microcirculation were observed. The levels of tumor necrosis factor-α (TNF-α) in the intestinal tissue were determined by using enzyme-linked immunoabsorbent assay (ELISA). The expression of intercellular adhesion molecule-1 (ICAM-1) in the intestinal tissue was detected by using immunohistochemisty and Western blot, that of nuclear factor κB (NF-κB) and tissue factor (TF) by using Western blot, and the levels of NF-κB mRNA expression by using RT-PCR respectively. The microcirculatory disturbance of the intestine was aggravated after CLP. The injury of the intestinal tissues was obviously aggravated in CLP group as compared with S group. The expression levels of NF-κB p65, ICAM-1, TF and TNF-α were upregulaed after CLP (P<0.01). STS post-treatment could ameliorate the microcirculatory disturbance, attenuate the injury of the intestinal tissues induced by CLP, and decrease the levels of NF-κB, ICAM-1, TF and TNF-α (P<0.01). It is suggested that STS can ameliorate the microcirculatory disturbance of the small intestine in rats with sepsis, and the mechanism may be associated with the inhibition of inflammatory responses and amelioration of coagulation abnormality.
10.Effects of aspirin on cell cycle in pancreatic carcinoma cells

AIM: To observe the effects of aspirin and prostaglandin E_2 (PGE_2) on the cell viability and cell cycle in SW1990 human pancreatic carcinoma cell lines, and to investigate the mechanisms of aspirin-induced growth inhibition and cell cycle arrest. METHODS: After incubated with aspirin or PGE_2 and their combination, the viability of SW1990 cells was measured by MTT assay. The levels of intracellular PGE_2 were determined by ELISA. The effects of aspirin or PGE_2 on cell cycle were investigated by flow cytometry (FCM). The expression of p21~ Wafl/cipl and p27~ Kipl/pic2 (the cyclin-dependent kinase inhibitors) were analyzed by Western blotting. RESULTS: Aspirin could inhibit the growth of cells and level of intracellular PGE_2 in a dose-dependent manner. Aspirin enhanced the expression of p21~ Wafl/cipl and p27~ Kipl/pic2 and induced cell cycle arrest at G_0/G_1 phase. PGE_2 increased the cell viability of SW1990 cells. However, it couldn't antagonize the changes of cell viability and cell cycle that induced by aspirin. CONCLUSIONS: The inhibitory effects of aspirin on growth and cell cycle of pancreatic carcinoma cells might not be mediated by a COX-dependent pathway completely. Cell cycle arrest induced by aspirin might be associated with up-regulation of p21~ Wafl/cipl and p27~ Kipl/pic2 .
