1.Determination of Hesperidin in Henggu Gushangyu Tablets by HPLC

Objective To determine the content of Hesperidin in Henggu Gushangyu Tablets by high performance liquid chromatography. Methods The determination was conducted on Shim-pack-C18 column (150 mm?4.6 mm, 5 ?m) with mobile phase of acetonitrile-0.4% phosphoric acid water (20∶80). The detection wavelength was 283 nm. Results Good linear relationship of Hesperidin between area and amount was noted for 0.194~0.970 ?g, with a correlation coefficient of 0.999 9. The average recovery was 98.50%, and RSD was 0.63%. Conclusion The established method is simple, accurate and sensitive, and can be applied to the quality control of the preparation.
2.Influence of tanshinone on rabbit malignant arrhythmias and calmodulin signal transduction pathway after myocardial infarction

Objective: To explore influence of tanshinone on rabbit malignant arrhythmias and calmodulin signal transduction pathway after myocardial infarction. Methods: A total of 30 rabbits were randomly and equally divided into sham operation group (sham group), model group and tanshinone group. Acute myocardial infarction model was established through ligating left anterior descending branch of coronary. After modeling, the rabbits were randomly and equally divided into model group and tanshinone group. Incidence rate of malignant arrhythmias, action potential duration (APD) of three myocardial layers (namely endocardium layer, myocardium layer and epicardium layer), transmural dispersion of repolarization, Ca2+ concentration, compared among three groups. Results: Incidence rate of malignant arrhythmias in tanshinone group was significantly lower than that of model group (20.0% vs. 70.0%, P<0.01); compared with sham group, there were significant rise in transmural dispersion of repolarization, 90% APD, Ca2+ concentration, expression levels of calmodulin and calmodulin kinase Ⅱ of three layers in model group and tanshinone group (P<0.01 all) ; compared with model group, there were significant reductions in transmural dispersion of repolarization[(46.2±10.9) ms vs.(35.5±8.8) ms], 90% APD [epicardium layer, (231.5±17.4) ms vs.(211.0±16.3) ms], Ca2+ concentration [epicardium layer, (132.0±12.3) mmol/L vs.(102.3±10.3) mmol/L], expression levels of calmodulin [epicardium layer, (0.724±0.014) vs. (0.563±0.014)] and calmodulin kinase Ⅱ[epicardium layer, (0.759±0.019) vs. (0.589±0.017)] in tanshinone group, (P<0.05~0.01). Conclusion: Tanshinone possesses anti-malignant arrhythmias effect after myocardial infarction. Its mechanism may be regulating calmodulin and calmodulin kinase Ⅱ signal transduction pathway.
3.Effect evaluation of high quality nursing service in patients with artificial joint replacement

Objective To explore the effects of high quality nursing service mode on nursing of patients with artificial joint replacement.Methods 82 patients from January to December 2010 were set as the control group,another 82 patients from January to November 2011 were named as the observation group.The observation group was given high quality nursing service mode,while the control group received conventional nursing.The satisfaction degree of patients to nursing,average hospitalization time and cost and incidence of complications were compared between two groups.Results The satisfaction degree of the observation group was 98.78%,significantly higher than 93.90% in the control group.The average hospitalization time was(14.68±5.88)days in the observation group,shorter than(19.22±6.78)days of the control group.The average hospitalization expense in the observation group was (25 665±528)yuan,less than (30 866± 732)yuan of the control group.The complication rate in the observation group was 21.95%,significantly lower than 46.34% of the control group.Conclusions Targeted high quality nursing service mode can effectively improve the quality of nursing,decrease the incidence of complications,reduce the patient's hospitalization time and cost,as well as improve the patients' satisfaction degree.
4.The obtaining of an anti-human CD40 mono-clonal antibody with special functions and the analysis of it's biological effects

objective : To prepare mouse anti-human CD40 antigen functional monoclonal antibody and to further study it's biological effects by triggering the Cd40 molecules on B cells and dendritic cells(DCs) Methods: Using cell fusion, McAb screening immunofluorescence analysis immunoblotting and competition test obtain mouse anti-human CD40 McAb;by the proliferation assay of B cells and DCsand the analysis of expression of differentiation antigens on DCs. Results: On the basis of phenotype analysis, Western blotting and competition test, it is verified that 5Cll recognizes human CD40 antigens specially; 5Cll can augement the proliferation of tonsil B cells in the LCD32cells and IL-4 culturing system; 5Cll can medicate DCs to get proliferation and maturatio. Conclusion: 5Cll+LCD32 +IL4 can make tonsil resting B cells survive and long-term proliferate in vitro, the setup of CD40 System furnish necessary tool for the study of B cells; McAb5Cll also triggered the generation, proliferation and maturation of dendritic cells from peripheral blood monocytes. Thus 5Cll is a McAb withspecial function and important application value.
5.Effects of paclitaxel on the intimal proliferation in rat aortic allografts

Objective To investigate the effect of paclitaxel on the intimal proliferation in rat aortic allografts and the possible mechanism on preventing graft arteriosclerosis.Methods Thirty-two inbred(Wista)r rats and 16 SD rats were divided into three groups:the isografts control group(Wistar/Wistar),the(allografts) control group ((Wistar)/SD) and test group(Wistar/SD) randomly(16 rats each groups).The rat abdominal aortic allograft model was used.The rats in test group were treated with paclitaxel after operation and those in control group with(0.9 %) normal saline.The grafts were removed and measured by means of pathology and immunohistochemistry 30 days later.Results The results showed that the thickness of the(aortic) intima,the degree of inflammatory cells infiltration in adventitia,stenosis ratio and the expression of(PCNA) were decreased in test group as compared with the allografts control group.Conclusions Paclitaxel can inhibit intimal proliferation in aortic allografts and prevent the graft arteriosclerosis.The mechanism is related to inhibition of vascular smooth(muscle) cell proliferation and alleviation of aortic allografts rejection.
6.Scheme of hospital information network based on power cat technology

Objective To develop a high stability and wide coverage network with power cat technology to promote hospital informatization.Methods The common network layout methods were compared,and the advantages of power cat technology were introduced when used for network layout.A series or parallel connection method was used to lay the power cat in the existing power line,and to achieve the full coverage of the hospital network.Results Power cat technology gained advantages in high efficiency,low cost and high stability when used for network layout.Conclusion A stable,economic and convenient network is constructed in the hospital with power cat technology.
7.Effect of somatostain intra-articular injection on metabolism of free radicals in rabbits with knee osteoarthritis

Objective To discuss the effect of somatostain SS on metabolism of free radicals in animal models of osteoarthritis,so to discuss the treatment mechanism of intra- articular injection of somatostatin on osteoarthritis.MethodsForty experimental Japanese big-ear rabbits were randomly divided into four groups:normal control group,blank model group,normal saline group and SS group,n =10 for each group.After modeling,the rats in SS group and normal saline group received intra- articular injection of somatostain and saline respectively on the right knee.Ten weeks after the modeling,we measured the movement of right knee joint in rabbits,the levels of nitric oxide ( NO ) and hyaluronic acid (HA) in serum from the heart blood,and the activity of SOD and the content of MDA in synovial tissue.Results Compared with the blank model group,the movement of the knee joint was significantly increased in SS group,(96.01 ± 1.06)vs (50.21 ± 1.80) (P < 0.01 ).In the blank model group,the levels of NO and HA (μmoL/mL) in serum were ( 111.60 ± 2.76) and ( 309.11 ± 1.89 ),which was significantly decreased in SS group,which was( 80.14 ± 1.67 ) and ( 133.73 ± 2.75 ) ( P1 =0.0049,P2 =0.0052,P < 0.01 ).Compared with the blank model group,the SOD activity was (15.77 ± 2.76) and the MDA in serum nmol/mg prot was( 1.33 ±1.03),while was significantly increased in SS group,(24.74 ± 1.67) ( P < 0.01 ),and the levels of MDA in serum was significantly decreased in the SS group,(0.89 ±1.46) (P<0.01).Conclusion Intra-articular injection of somatostatin in knee osteoarthritis can improve the oxidative stress,enhance the activity of the knee joint.
8.Influence of long cold ischemia preservation time on ultrastructural morphology of donor heart

Objective To investigate the impacts of long cold ischemic preservation time on the ultrastructural changes of donor heart and to provide the data for expanding the heart donor pool. Methods Heart was obtained from brain dead donor and preserved in the the University of Wisconsin (UW) solution at 4℃. Cardiac tissues were harvested at different time points of cold ischemic preservation (6, 8, 10, 12, 14 h) and observed under electron miscroscope. Results Donor heart did not have significant pathologied and ultrastructural changes when cold ischemic preservation time was 6 h. After that, time related impairment of myocardia and endothelium of coronary artery was seen.When ischemic time was longer than 12 h, focal myocardial necrosis and complete loss of the endothelium were detected. Conclusions Myocardial ultrastructure is an important index to evaluate the donor heart quality. Heart, which underwent 10 h of cold ischemia preservation time, causes no significant irreversible and pathological ultrastructural changes, and could be used for heart transplantation. When ischemia time was over 10 h, the donor heart presented with irreversible change and was nolonger unsuitable for transplantation.
9.Preclinical study of recombinant adenovirus carrying p53, B7-1, and GM-CSF in the treatment of human laryngeal squamous carcinoma

Objective To study the effect of a recombinant adenovirus carrying p53 gene, B7 1 gene and GM CSF (BB 102) on the nude mice transferred with laryngeal carcinoma. Methods Nude mice model bearing laryngocarcinoma was established using human laryngeal squamous carcinoma cell line (Hep 2). Large amounts of recombinant adenovirus (BB 102) were injected into the tumor. Changes in carcinoma treated with recombinant BB 102 adenovirus were observed under light and electron microscopes. Results The difference between experimental and control groups was statistically significant. The morphology of cells infected with BB 102 was analyzed for evidence of apoptosis by transmission electron microscopy. It has been shown that wild type P53 protein can inhibit cell growth and induce apoptosis, while B7 1 and GM CSF have no such effects. Conclusions The results showed that recombinant adenovirus BB 102 has significant efficacy in suppressing tumor cell growth and in inducing their apoptosis, suggesting that BB 102 might be developed into a therapeutic agent in clinical therapy of laryngeal carcinoma
