1.The studies on phosphorylated-p38 MAPK in hypothalamus from restrain stress rat and the regulation of p38 MAPK by electroacupuncture

Objective To observe the effects of restrain stress on the phosphorylation of p38 mitogen-activated protein kinase ~MAPK) in rat hypothalamus and the regulation effects of electroacupuncture on the phosphorylated-p38 MAPK. Methods Twenty-eight rats were randomly divided into a control group, a restrain stress group and a restrain stress plus electroacupuncture group. The Western blot was employed to observe the changes of phosphorylated-p38MAPK in the three groups. Results Western blot analysis demonstrated a higher level of phosphorylation of p38 MAPK in the hypothalamus from restrain stress group than that from control. The higher level of phosphorylated-p38 MAPK could last for 3 hours after stress was relieved. The level of phosphorylated-p38MAPK could be decreased to some extent when the Zusanli acupoint of the restrained rat was acupunctured. The effects of electroacupuncture of Zusanli on phosphorylated-p38MAPK in hypothalamus could still be retained although the electroacupuncture was ended for 3 hours. Conclusion The results suggested that restrain stress can lead to a higher level of phosphorylation of p38 MAPK in hypothalamus, which might be an important event in the response of hypothalamic-pitutary-adrenal ~HPA) axis. The mechanism underlying the regulation of HPA by electroacupuncture might be (related) to the regulation of phosphorylated-p38 MAPK in hypothalamus.

To investigate the effect of calmodulin (CaM) signal system on the glycogen synthesis in skeletal muscle cells from glimepiride (GM)-treated normal rats. The results showed that GM could activate CaM and CaM played a key role in GM inducing insulin-mediated glycogen synthesis.

4.Expression of FOXO1 and FOXO3a on peripheral blood mononuclear cells and their clinical roles in patients with systemic lupus erythematosus

Objective To study the expression of transcription factor FOXO1 and FOXO3a on peripheral blood mononuclear cells (PBMC) in patients with active or inactive systemic lupus erythematosus (SLE) and investigate the effect of FOXO1 and FOXO3a on the clinical features of SLE. Methods Thirty SLE patients and 10 healthy controls were enrolled. PBMC were separated from the peripheral blood. Reverse transcription-polymerase chain reaction (RT-PCR) and western blot were applied to analyze the expression of FOXOI and FOXO3a. Results The level of FOXO1 expression was significantly decreased in active SLE patients compared with controls and patients with inactive SLE (P<0.05). The level of FOXO1 expression in inactive SLE patients was lower than that of the controls (P<0.05). The expression of FOXO1 mRNA was negatively correlated to SLEDAI. However, the level of FOXO3a was similar among the three groups. Conclusion The result suggests that FOXO1 may be involved in the pathogenesis of SLE and the expression level of FOXOI may be a good indicator for the disease activity of SLE.
5.Expression of VEGF-C and VEGFR-3 in hepatocellular carcinoma and their significance

Objective To investigate vascular endothelial growth factor-c (VEGF-C) and vascular endothelial growth factor receptor-3(VEGFR-3) mRNA expression, microvessels density (MVD) and lymphatic microvessels density (LVD) in human hepatocellular carcinoma and normal liver tissue. Try to illuminate the relationship among VEGF-C,VEGFR-3,MVD,LVD and the clinical pathological features of hepatocellular carcinoma. Methods Liver tissue of 60 cases definitely diagnosed as hepatocellular carcinoma and 20 normal cases were collected. VEGF-C and VEGFR-3 mRNA expression were examined by RT-PCR, MVD and LVD were examined by immunohistochemistry staining. Relationship between these indexes and clinical pathological features of hepatocellular carcinoma was also analysed. Results VEGF-C and VEGFR-3 mRNA expression, MVD and LVD in hepatocellular carcinoma were higher than those in normal liver tissue (P<0.01); In hepatocellular carcinoma tissue, expression of VEGF-C mRNA positively related with VEGFR-3 mRNA, MVD and LVD(P<0.01). VEGF-C and VEGFR-3 expression positively related with portal vein tumor thrombus, intrahepatal metastasis and lymph node metastasis (P<0.01). MVD positively related with portal vein tumor thrombus and intrahepatal metastasis (P<0.01). LVD positively related with lymph node metastasis (P<0.01). Conclusion VEGF-C and VEGFR-3 expression increase in hepatocellular carcinoma tissue. They might play roles in tumor invasion and metastasis by inducing angiogenesis and lymphangiogenesis.
6.Imprinting effects of prenatal overexposure to glucocorticoids on 11?-hydroxysteroid dehydrogenase type 1 expression in rat offspring liver

Objective:To observe the imprinting effects of prenatal overexposure to glucocorticoids on 11?-hydroxysteroid dehydrogenase type 1(11?-HSD1) expression in the rat offspring liver. Methods: Pregnancy was determined by examining vaginal smear. Eight pregnant rats were divided into dexamethasone(DEX) group and normal saline(NS) group at random with each group containing 4 rats.DEX group was given subcutaneous injection of DEX [0.05 mg/(kg?d)] and NS group was given injection of NS.Western blot was adopted to determine 11?-HSD1 protein in the liver of the offspring at the age of 4 months;RT-PCR was used to determine 11?-HSD1 mRNA and hepatic phosphoenolpyruvate carboxykinase(PEPCK) mRNA;and glucose oxidase method was used to measure the blood glucose concentration. Results: Western blot and RT-PCR showed that prenatal overexposure to DEX increased the expression of 11?-HSD1 protein and mRNA in the liver of the female offsprings at 4 months old, but not in male offsprings.Prenatal overexposure to DEX also increased the expression of PEPCK mRNA and blood glucose level in the female offsprings at 4 months old.Conclusion:Our study suggests that prenatal overexposure to glucocorticoids throughout gestation can imprint the expression of 11?-HSD1 in rat offsprings liver, which may lead to the PEPCK overexpression and blood glucose elevation.Development of diabetes may be induced in individuals overexposed to glucocorticoids during fetal period.

Objectives To explore the feasibility ofdiagnosis of Perimembranous Ventricular Septal Defect with Right Aortic Valve Prolapse by ultrasonic echocardiography. Methods 131 VSD patients aged 1 to 22 years, weight 8.0 to 54.0 kg, underwent transthoracic echocardiography (TTE) and left ventricular angiography to show the diameter of VSD and the degree of AVP. Results Among 131 patients, 87 cases were diagnosed as VSD,14 as VSD with slight AVP, 12 as, VSD with middle AVP, and 18 as VSD with severe AVP by ultrasonic echocardiography respectively. After the left ventricular angiography and aortic root angiography , it proved that positive rate of VSD was 44.8%, positive rate of VSD with slight AVP was 42.86%, positive rate of VSD with middle AVP was 58.3%,and positive rate of VSD with severe AVP was 83.3%. Conclusion The transthoracic echocardiography (TTE) contributes to diagnosis of severe degree of AVP,while TTE evaluates the slight degree of AVP incorrectly enough. Left ventricular angiography and aortic root angiography are needed to realize the di-agnosis of AVP.
10.Ultrasound microbubble contrast agent enhances TRAIL gene transfection into hepatocellular carcino-ma cells

Objective To evaluate the impact of the recombined plasmid vector with enhanced green fluorescent protein (EGFP) encoding soluble tumor necrosis factor related apoptesis inducing ligand (pIRES-EGFP-sTRAIL) on proliferation and apoptosis of human hepatocellular carcinoma cell line HepG2, and investi-gate the feasibility and efficiency of the transfection of pIRES- EGFP- sTRAIL into HepG2 by ultrasound micro-bubble contrast agent. Methods pIRES-EGFP-sTRAIL was constructed and transfected into HepG2 cells by using different types of mediated methods: microbubble echocontrast agent combining appropriate dose of ultra-sound irradiation, liposome method, microbubble echocontrast agent only or blank medium treatment. Transfec-tion efficiency was evaluated by EGFP-expressed cell count; proliferation-lnhibiting rate and the apoptosis rate of HepG2 cells were determined by MTT method and flow cytometry analysis; changes of cell morphology were examined by microscopy with Hoechst33258 dyeing; expression of caspase-8 and caspase-3 was detected by Western blot. Results Ultrasound microbubbh enhanced pIKES-EGFP-sTRAIL uptake by HepG2 cells, and the transfection efficiency was significantly higher in ultrasound microlmbble group than that in other groups( P<0.05 ) ; pIRES- EGFP- sTBAIL effectively inhibited HepG2 cell proliferation and induced cell apoptosis by triggering caspase cascade. Both the inhibiting rate and apoptosis rate were significantly higher in ultrasound microbubble group than those in other groups(P<0.05). Conclusion pIRES-EGFP-sTRAIL expresses ef-fectively in HepG2 cells, sTRAIL has a potential role on the inhibiting proliferation and inducing apoptosis of HepG2 cells by triggering caspase cascade, and this role can be enhanced by the administration of low-intensity ultrasound and microbubble echecontrast agent.
