1.Effects of gel gatifloxacin versus levofloxacin in treatment of bacterial conjunctivitis

Objective To assess the clinical effectiveness and safety of gatifloxacin gel to bacterial conjunctivitis. Methods Double-blind and random selection were designed for the study,with levofloxacin gel as the control medicine.Thirty-six eyes of the experimental group and 36 of the control group were eligible for evaluation.Each eye received the gel one drop a time and three times per day.The gels were applied to the conjunctiva sac for 7 d. Results The clinic effectiveness of experimental group and control group were 88.89% and 91.67%,which indicated no significant difference.Microbial eradication rates were 90.48% and 93.75%,no significant difference either.Safety composite scores were similar between groups. Conclusion Gatifloxacin ophthalmic gel is safe and effective for treatment of bacterial conjunctivitis,and contains certain clinical value.
2.The detection of coagulation in diabetic mellitus in Xinjang Uygur and Han

Objective To explore the changes of coagulation factors in Xinjiang Uygur and Han patients with diabetic mellitus in order to provide reference for the prevention and treatment of diabetic mellitus patients.Methods 2 100 patients with diabetic mel-litus were taken as the patient group which were diagnosed in our hospital between February 2014 and April 2015,another 446 healthy people were collected as the control group.The difference of coagulation between diabetic mellitus group and control group was compared.Coagulation between Uygur and Han was compared for 2 100 diabetic mellitus patients.Results The APTT of dia-betic mellitus group were significantly less than that in control group(P <0.05).FIB of diabetic mellitus group were significantly higher than that in control group(P <0.05).The difference of PT between diabetic mellitus group and control group were not sig-nificant(P >0.05).The APTT of Uygur group were significantly less than Han group(P <0.05).FIB of Uygur group were signifi-cantly higher than Han group(P <0.05).PT between Uygur group and Han group was not significant(P >0.05).Conclusion Fac-tors were difference of diabetic mellitus patients and healthy people,coagulation factors in diabetic mellitus patients of Xinjang Uygur and Han.
3.Current research situation and prospect of auricular acupuncture for epilepsy.

Epilepsy is a group of chronic diseases characterized by recurrent and transient brain dysfunction induced by abrupt and intermittent abnormal discharge of neurons in the brain, which is difficult to be cured. Thee auricular concha are is he zone of visera in TCM auricular points, but also the innervating area of the auricular brach of the vagus nerve. Auricular acupuncture, as a special treatment, has superior therapeutic effect for epilepsy. In recent years, remarkable progress has been made in the theoretical basis and clinic application of auricular acupuncture for epilepsy, and the ear-vagus percutaneous electrical stimulation apparatus (auricular acupuncture apparatus) is developed for clinical trial researches. From the basic study to clinical research of auricular acupuncture for epilepsy, the epilepsy pathogenesis as well as the intervention pathway and clinical efficacy of auricular: experimental support and rich clinical experience for auricular acupuncture apparatus.

4.Analysis of normal eye anterior segments in different ages

Objective To measure and conduct statistic analysis of normal eyes by Pentacam. Methods Two hundred and twenty-one healthy people (442 eyes) underwent examinations of Pentacam, and they were divided into 5 groups by age: <40 years old group (50 people, 100 eyes), 40-49 years old group (50 people, 100 eyes), 50-59 years old group (50 people, 100 eyes), 60-69 years old group (50 people, 100 eyes) and >70 years old group (21 people, 42 eyes). Results With the increase of age, there were significant differences in chamber volume, chamber angle and anterior chamber depth among age groups (P<0.05). In elder age groups, there were significant differences in these parameters between males and females (P<0.01). Conclusion Anterior segment parameters are significantly correlated with age. After 60 years old, the anterior segment parameters become significantly different with gender.
5.Application of heteroduplex mobility assay in quick identification of adenovirus subtypes in epidemic keratoconjunctivitis

Background The commonly used method of typing the adenovirus (AdV) for epidemic keratoconjunctivitis is direct DNA sequence.However,heteroduplex mobility assay (HMA) is found to be a faster method to identify the subtypes of adenovirus and is more conforming to the principle of cost and economic benefit.There are few studies to illustrate the application of HMA in epidemic keratoconjunctivitis.Objective This study analyzed the conserved region of hexon coding sequence and compared the outcomes between direct DNA sequence and HMA for classification of adenovirus in epidemic keratoconjunctivitis patients.The validity of HMA is evaluated by comparing the result of both studies Methods Two hundreds and fourteen patients with suspicious epidemic keratoconjunctivitis were included in Shanghai Eye Disease Prevention and Treatment Center or the clinical sites supervised by the Shanghai Prevention and Monitoring Office of Acute Hemerragic Conjunctivitis from January 2010 to December 2012.Sacconjunctival swab samples were collected from each patient under the informed consent.DNA of pathogens was extracted from the samples using QIA-amp minikit,and the conserved sequence with 366 bp at hexon protein coding region was amplified by PCR and sequenced subsequently to determine the infected adenovirus and their subtypes.These samples were simultaneously assayed by HMA,and the outcomes between DNA sequence and HMA were compared.Results Extracted DNA presented a yellow fluorescence band with the fragment size 35 kb and absorbance ratio at the wavelength 260 nm and 280 nm (A260/A280) was I.7.In the 214 samples,AdV type 1 (AdV1) was found in 4 samples,AdV2 in 33 samples,AdV3 in 15 samples,AdV4 in 12 samples,AdV8 in 19 samples,AdV19 in 15 samples and AdV37 in 8 samples.HMA showed the same outcome for the identification of AdV1,AdV2,AdV3,AdV8,AdV19 and AdV37 with direct DNA sequence.AdV4 was not feasible to HMA owing to 59.6% (over 10%) mutation sites.Conclusions Direct DNA sequencing for conserved regions in coding sequence of hexon is an important way to identify causing-disease adenovirus subtypes in the patients with epidemic keratoconjunctivitis.HMA can offer a consistent result with the DNA sequencing,and it might be used as a suitable tool for large-scale molecular epidemiology researches.
6.Application of hexon gene sequencing of adenovirus in pathogen detection of epidemic keratoconjunctivitis

Background Epidemic keratoconjunctivitis is a common eye disease,and adenovirus is one of the common pathogens.The hexon protein,one main capsid protein of the virus,is an important target of antibody binding.Thus,sequencing the coding region of the hexon protein is an important way for adenovirus fast typing.Objective This study was to complete a molecular epidemiology survey of epidemic keratoconjunctivitis and investigate its association with adenovirus in Shanghai area by sequencing the coding region of hexon protein.Methods Two hundred and fourteen sacconjunctival swab specimens were collected from 214 patients with suspicious epidemic keratoconjunctivitis who visited Shanghai Eye Disease Prevention and Treatment Center and the clinical sites supervised by the Shanghai Prevention and Monitoring Office of Acute Hemerragic Conjunctivitis under the informed consent from January 2010 to December 2012.DNA was extracted from the specimens and then the 140 bp conserved sequence in hexon protein coding region was amplified by PCR initially to determine an adenovirus pathogen.Furtherly,956 bp conserved sequence of the hexon codind district was sequencied to clarify the serotype of adenovirus in the adenovirus-positive specimens.Results 50.93% patients (109/214) were detected to be adenovirus-positive by generic PCR,in which AdV1 + was in 4 patiens,AdV2+ was in 33 patients,AdV3+ was in 15 patients,AdV4+ was in 12 patients,AdV8+ was in 19 patiens,AdV19+ was in 15 patients,AdV37+ was in 8 patients.The subgenus D adenoviruses,including AdV8+,AdV19+ and AdV37+ often resulted in corneal inflammation,pseudomembranous conjunctivitis and preauricular lymph nodes;while subgenus B adenovirus induced much frequent tract infection and less corneal response.Conclusions PCR-sequence of conserved region of hexon protein coding district is applicable for the detection and serotyping of adenovirus in epidemic keratoconjunctivitis.
7.Investigation on Effects and Mechanism of Acupoints

Acupoints are surface reaction and treatment spots under the pathological conditions which have functions of diagnosis and treatment. For the study of acupoints, we can carry it out from the aspects of structure and function. The two aspects have a close relationship. Structure is the material basis of function, while function is the manifesta-tion of structure. Therefore, the study of acupoints is not only significant to reveal the essence of acupoint, but also has enormous value in the guidance of acu-moxibustion for clinical practice. Staring from the normality of structure and variability of function, we explored the essence of acupoints, and further revealed ordered regulation rule and its biological basis, so that the essence of acupuncture points will be illustrated with scientific evidences.

Wnt signaling transduction pathway regulates cell growth,motility and differentiation,and plays a crucial role in the regulation of embryonic development and tumor genesis. This article reviewed the advances in study on Wnt signaling transduction pathway and colorectal cancer.
9.5-LOX inhibitor MK886 suppresses the stemness of human colon cancer HT-29 cells

Objective It remains a controversy whether 5-lipoxygenase (5-LOX) is associated with colon cancer stem cells.This study was to investigate the effect of the 5-LOX inhibitor MK886 in maintaining the stemness of the human colon cancer cell line HT-29.Methods Using CCK-8 assay, we examined the inhibitory effects of different concentrations of MK886 (12.5, 25, 50, 75, 100, and 200 μmol/L) on the colon cancer HT-29 cells cultured in vitro and calculated its half-inhibitory concentration (IC50).Then, we detected the effects of MK886 IC50 on the clone-and sphere-forming abilities of the cells, determined the mRNA expressions of the stemness markers CD133, Lgr5, Oct4 and Ascl2 by real-time PCR after 24 and 48 hours of MK886 IC50 intervention, and measured their protein expressions by Western blotting after 24, 48 and 72 hours of MK886 IC50 intervention.Results The inhibition rates of MK886 on the HT-29 cells at 24 and 48 hours were significantly increased in a time-and dose-dependent manner ([14.99±3.06] and [19.98±0.57]% at 12.5 μmol/L, [20.46±1.14] and [34.97±6.02]% at 25 μmol/L, [50.76±5.94] and [66.90±5.74]% at 50 μmol/L, [66.84±1.77] and [73.11±2.48]% at 75 μmol/L, [72.67±2.36] and [77.78±3.30]% at 100 μmol/L, [83.67±0.24] and [84.69±2.24] % at 200 μmol/L) as compared with the blank control (0% and 0%) (P<0.05).The clone-forming rate and number of spheres formed were remarkably lower in the MK886 intervention than in the control group ([10.60±1.71] vs [44.67±3.21]%, P<0.05;6.00±1.60 vs 19.07±2.89, P<0.05).After 24 and 48 hours of MK886 intervention, the mRNA expression of CD133 in the HT-29 cells was markedly up-regulated in comparison with that at 0 hour (0.72±0.10 and 0.39±0.07 vs 1.66±0.33, P<0.05), and so were those of Lgr5, Oct4 and Ascl2 (P<0.05).Conclusion The 5-LOX inhibitor MK886 can inhibit the proliferation and clone-and sphere-forming abilities of human colon cancer HT-29 cells by down-regulating the expressions of the stemness markers and thus suppressing the stemness of the colon cancer stem cells.
