Objective To observe the effects of estrogen on the behavior and the norepinephrine level in the habenular nucleus of depressive rats,and explore the mechanism of estrogen involved in the process of antidepression.Methods Thirty rats were chosen from forty male Wistar rats,which had the similar scores in open field test.The depression models of rats were established by chronic mild stress.Open field test and forced swimming test were performed to detect the behavioral changes between rats with ovary excision(OVX) and with estradiol-treated ovary excision(OV/ER).High performance liquid chromatography(HPLC) was used to observe the changes of norepinephrine level in habenular nucleus.Results After 21-day chronic mild stress,the open field test showed that the vertical motion and cross motion in OV/ER group were increased(P

Objective:To study the feasibility of surgical techniques and clinical experience of the airbag-free separation of nail and the whole extraperitoneal laparoscopic hernia repair(TEP).Methods:Ninty-three cases of inguinal hernia.Patients were treatecl in our hospital from March 2005 to January 2009,with 39 cases of regular.Free air and non-stapling of mesh TEP and 54 cases of routine open tension-free hernioplasty.Results:The operation times were as follows:TEP Group,unilateral hernia(101.3?15.6 min),bilateral hernia(138.4?17.3 min);Open Group,unilateral hernia(81.6?7.6 min),bilateral hernia 175.8?22.0 min.The couplications in TEP Goup inclucled 3 cases of retroperitoheal rupture,3 cases of intra-operative vascular injury,and 2 cases of postoperative scrotal hematoma.Nine cases of sacrotal hematoma,4 cases of infective and 6 cases of long-term pain in the groin area happened in open Group.Conclusion:TEP of free balloon separator with no stapling patch is safe,reliable,less post-operative complications and especially suitable for bilateral hernia.

Objective To investigate the effects of ubiquitin isopeptidase inhibitor Ⅰ on proliferation and apoptosis of human leukemic K562 cells.Methods Human leukemic K562 cells were treated with different concentration of ubiquitin isopeptidase inhibitor Ⅰ.Cell proliferation was monitored by MTT assay.Cell apoptosis was analyzed by flow cytometry after Annexin V/PI staining,and mRNA expression levels of bcl-2,bax and Caspase-3 were quantified by reverse transcription-polymerase chain reaction (RT-PCR).Results MTT assay showed that the proliferation of K562 cells was markedly inhibited by ubiquitin isopeptidase inhibitor Ⅰ in a time-and-dose dependent manner.The Annexin V positive rate of K562 cells after treatment with ubiquitin isopeptidase inhibitor Ⅰ was significantly increased by FCM analysis (P ＜ 0.05).The early apoptosis rate of K562 cells treated with 100 nmol/L ubiquitin isopeptidas inhibitor Ⅰ by 72 h,was obviously increased compared to control cells [(15.4±1.3) ％ vs (4.1±0.9) ％,P ＜ 0.01].The mRNA levels of Caspase-3 and bax were up-regulated and bcl-2 was down-regulated after treated with ubiquitin isopeptidase inhibitor Ⅰ,and the differences were statistically significant from control cells (P ＜ 0.05).Conclusion Ubiquitin isopeptidase inhibitor Ⅰ has inhibitory effect on proliferation and inductive effect on apoptosis of K562 leukemia cells,implying its possible application in treating leukemia.

Pancreatic cancer is one of the most aggressive malignancies in the world.Little progress has been made on the treatment of advanced pancreatic cancer in the past 20 years.miRNAs perform a regulatory role in the determination of the final phenotype of cancer cells,including carcinogenesis,metastatic potential,and chemosensitivity.These features of miRNAs have turned them into one of the most popular and promising fields of scientific and clinical research.Issues such as miRNA toxicity to non-target tissues or cells and lack of proper delivery systems for human pancreatic cancer will be the challenges for the field to overcome.

AIM: To analyze the direction and degree of static cyclotorsion component (SCC) and dynamic cyclotorsion component (DCC) in corneal refractive surgery.
METHODS:Retrospective analysis. Totally 130 patients (260 eyes) with corneal refractive surgery in our hospital, according to the operation method were divided into femtosecond laser - assisted laser in situ keratomileusis (FS-LASIK) group and T-photorefractive keratectomy (T-PRK) group, the differences of the parameters of the two groups were compared; the differences of SCC success rate, SCC, DCC, and the eyeball rotation direction were compared between the two groups; correlation analysis on SCC, DCC and the parameters of postoperative patients were performed.
RESULTS: High order aberrations and spherical aberration in the T-PRK group after operation was higher than those of FS - LASIK group, and the difference was statistically significant (P<0. 05); in T-PRK group SCC in the operation was successful in 98 eyes, the success rate was 81. 7%; in FS-LASIK group SCC in the operation was successful in 82 eyes, the success rate is 58. 6%, the difference of SCC success rate between the two groups was statistically significant ( P < 0. 05 ); SCC in T - PRK group was 3. 52o ±2. 17o and FS-LASIK group was 3. 49o ±2. 26o, there was no significant difference (P>0. 05); DCC in T-PRK group (2. 86o±1. 14o) was higher than that of FS-LASIK group ( 2. 17o ± 1. 09o), and the difference was statistically significant (P<0. 05). There was no statistical difference in the direction of rotation of the eyeball in operation between the two groups (P>0. 05). The SCC of subjects in operation was positively correlated with UCVA, BCVA, spherical equivalent refraction and high order aberrations ( P < 0. 05 ); the DCC of subjects in operation was positively correlated with UCVA and high order aberrations (P<0. 05).
CONCLUSION: The success rate of SCC in T - PRK surgery is higher than that in LASIK, DCC in T - PRK surgery is higher than that in LASIK, and accurate measurement of SCC and DCC can be effective to compensate for it.

Objective To evaluate the efficacy and adverse effects of docetaxel combined with 5-fluorouracil in treating patients with advanced gastrice cancer.Methods 130 patients with advanced gastric cancer were randomly divided into observation group and control group,observation group treated with docetaxel combined with 5-fluorouracil,the control group treated with 5-fluorouracil and cisplatin,the clinical efficacy after the treatment was observed.Results The clinical efficiency rate of the observation group was significantly higher than that of the control group,the rate of leukopenia,thrombocytopenia and gastrointestinal reactions of the observation group was significantly lower than the control group,the quality of life scores of the two groups after treatment were all improved,but that of the observation group was significantly higher than the control group,the difference was statistically significant(P ＜ 0.05).Conclusion Docetaxel combined with 5-fluorouracil was effective in treating advanced gastric cancer.It worth further clinical research and application.

Objective To investigate the effects of an electromagnetic field on the extra-cellularly regulated kinase(ERK)signalling pathway and to determine the impact of electromagnetic activation on osteogenic proliferation and differentiation in rat bone marrow mesenchymal stem cells.Methods Rat bone marrow mesenchymal stem cells were isolated and cultured in vitro.The third-passage cells were divided into 4 groups(Control,PD98059,EMF and EMF+PD98059).Western blotting Was used to detect the activation of the ERK signal pathway after exposure to an electromagnetic field.MTT assay Was used to determine the activation of proliferation in the celb in the different groups.The cells' alkaline phosphatase activities were also detected. Results (1)The ERK signal pathway in these rat bone marrow mesenchymal stem cells was activated after exposure to a 15 Hz.1 mT,sine wave form electromagnetic field for 5 min.Activation remained high for at least 1 h.PD98059 can effectively block the activation of the ERK signal pathway.(2)Cell proliferation was promoted after exposure to the electromagnetic field,and this effect could be significantly inhibited by PD98059.(3)Alkaline phosphatase was significantly elevated in these bone marrow mesenchymal stem cells after exposure to the electromagnetic field.The activation in the EMF+PD98059 group Was slightly greater than in the EMF group.Conclusion Electromagnetic fields of 15 Hz and 1 mT can activate the ERK signal pathway and alter proliferation and osteogenic differentiation in the bone marrow mesenchymal stem cells of rats.

OBJECTIVE To find out the status about detection and drug resistance of Ureaplasma urealyticum(Uu) and Mycoplasma hominis(Mh) of urogenital tract infection in Shaoxing area of Zhejiang Province.METHODS The detections and drug sensitive tests of Uu and Mh adopting the reagent boxes produced by bioMerieux Company in France were made.RESULTS In 2678 samples of suspicious urogenital tract infection patients,980 samples cultured of mycoplasma were positive,and the positive rate was 36.6%.Among these cases,763 samples(28.5%) were Uu positive;116 samples(4.3%) were Mh positive.In the drug sensitive test,Uu and Mh were most sensitive to PRI,JOS,DOX and TET.To CFLX,OFL,ERY,CM and AM,they had the highest level of drug resistance.CONCLUSIONS The clinical therapy of mycoplasma infection ought to choose the appropriate antibiotics based on the results of the drug sensitive test of the laboratory.
