1.Effects of tetramethylpyrazine on learning and memory in aluminium trichloride induced Alzheimer's disease mice

Objective To discuss the effects and mechanisms of tetramethylpyrazine(TMP)on learning and memory abilities of Alzheimer's disease(AD)mice.Methods The animal models of AD were induced by aluminium trichloride(AlCl3)intragasticly for forty days.Then,TMP was intragastricly administered for twenty days.The escape latency of the mice in water maze test was recorded;and AChE and SOD activities and MDA levels in the brain were detected by chemical eoiorimetrie method.The expressions of At3 and NF-κB in the cerebral cortex were measured by immunohistochemistry method.Results 1)TMP has markedly decreased escape latency in AlCl3 induced AD mice(P<0.05).2)In the brain of AlCl3 induced AD mice,TMP reduced AChE activity 19%[(1.37±0.13)U·mgpro-1vs(1.69±0.27)U·mgprot-1,P=0.016],decreased MDA levels 34% [(43.63 4-13.27)nmol·mgprot-1 vs(28.59±8.52)nmol·mgprot-1,P=0.023]and increased SODD activity 39%[(55.81±10.25)U·mgprot-1 vs(40.04±13.06)U·mgprot-1,P=0.026],respectively.These resuits showed significant difference with model mice group(P<0.05).3)TMP also decreased AB and NF-κB expressions in the cerebral cortex of AD mice models(P<0.05).Conclusion In AlCl3 induced AD mice,TMP can remarkably enhance the learning and memory abilities,presumably in relation to raise the activity of SOD,reduce the activity of AChE,the levels of MDA and the expressions of AB and NF-κB in the brain.
2.Study of the panel of 6 microsatellites in diagnosis of bladder cancer

Objective To evaluate the significance of the panel of 6 microsatellites in detection of bladder cancer. Methods In the tumor tissue and urine sediment of 32 cases of bladder cancer 10 microsatellites were chosen and PCR-SSLP silver staining assay was conducted according to the methods described in the literature and our previous study.15 cases of non-bladder cancer served as controls. Results Microsatellite alternate (MA) was found in 30 out of 32 cases of bladder cancers,the sensitivity being 93.8%.The MA of urine sediment of 15 cases of non-bladder cancer was negative,the specifity being 100.0%.Among the 10 microsatellites, 6 ones were chosen;the MA positivity of the panel of the 6 ones was 90.6% (29/32).This result was not significantly different from that of the panel of 10 microsatellites. Conclusions MA assay is a sensitive,effective method for detection of bladder cancer.Compared with the panel of 10 microsatellites,the panel of 6 microsatellites may be a better tool for detection of bladder cancer.
3.THE STUDY OF MICROSATELLITES ALTERATION IN DIAGNOSES OF BLADDER CANCER

Objective To investigate the possibility of microsatellite alteration (MA) in diagnosis of bladder cancer of Chinese people, and find the better panel which will be used in clinic. Methods A total of 6 and 10microsatellite markers were chosen, PCR-SSLP silver staining assay was performed in 31 and 32 bladder cancers tissue,exfoliate cells in urine and 10, 15 non-bladder cancers exfoliate cells in urine, respectively. Results MA (+) was found in 28 out of 31, 30 out of 32 bladder cancers, and the sensitivity was 90.3%, 93.7% respectively. The MA of urine sediment of 25 non-bladder cancers was negative, and the specificity was 100%. The cytology was carried out among 19 out of 31, 20 out of 32 bladder cancers at the same time, 2 cases ( 10.3 %) and 3 cases ( 15 % ) were found cancer positive, and the sensitivity is significantly lower than that by the analysis of MA in exfoliated cells. Conclusion MA was not associated with grade and stage of the bladder cancer. MA assay is a sensitive and effective method for the early detection of bladder cancer and post-operation surveillance.
4.Introspective reports of attention deficit hyperactivity disorder and its pathogenetic characteristics

AIM:To reveal the characteristics and patterns of mental activity in the cerebral cortex of patients with attention deficit hyperactivity disorder (ADHD) and explore the pathogenetic characteristics of ADHD.METHODS: The ADHD patients were asked to record their mental activity, general health condition and obsessions before the onset of pathological symptoms by means of psychological introspective reports.RESULTS: The direct cause of attention distraction and failure of attention focus is the patients' inability to perform cognitive tasks (including perception, memory and recollection, imagination, association and thought)in a continuously ongoing manner in relaxation. The frequent stagnancy and pause during the course of recognition precipitated the patients in a stressful state, which contributed to the onset of various hyperactivities and also resulted in the acquisition of "silent verbal thinking". This is an important psychopathological phenomenon long been neglected. All theses abnormalities caused the patients much discomfort, which was naturally relieved by means of impulsive and irritable behaviors.CONCLUSION: A series of clinical symptoms of ADHD arises from the same pathogenic factor of difficulty in mental activity and inability to carry out the activities in the cerebral cortex in a relaxed manner. The difficulty in mental activity is the key problem of ADHD and the source of various synptoms, which should be ;reated for the first place. The cause of this key problem is possibly lowered metabolism of the cerebral cortex, and the effect of "instinct" can not be neglected in the attempt to explain the pathogenesis of ADHD.
5.Chiropractic and acupuncture therapy in the treatment of spleen deficiency anorexia in children: clinical observation of 50 cases

Objective To explore the Chinese massage therapy for the treatment of infantile anorexia of spleen and stomach type clinical curative effect.Methods 2 to 6 years old anorexia children treated from September 2010 to September 2012 for general treatment and health care in our hospital were randomly recruited into a Qi deficiency of both spleen and stomach group and a Yin deficiency of both spleen and stomach group with 25 cases in each group.Both groups were treated with chiropractic therapy.On this basis,Qi deficiency of both spleen and stomach group was applied point-massage on Weishu,Pishu,and Zusanli; Yin deficiency of both spleen and stomach group was applied point-massage on Weishu,Ganshu,and Shenshu.Results ① The comparison of disease and syndromes:the total effective rate was 92% (23/25)and 88% (22/25) in Qi deficiency of both spleen and stomach group and Yin deficiency of both spleen and stomach group separately,showing no significant difference (x2=0.886,P>0.05); the total symptoms improving rate was 92%(23/25) and 76%(19/25)in Qi deficiency of both spleen and stomach group and Yin deficiency of both spleen and stomach group separately,the difference showing statistically significant (x2=0.625,P<0.05).② Symptom score comparison before and after treatment:The main symptoms score and accompanied symptoms score before and after the treatment were [(18.32±2.42) %,(4.12±0.17) points] and [(16.80±3.77) %,(5.30 ±0.41) points] in the Qi deficiency group and [(18.18±2.10),(4.45±0.30) points] and [(19.66 ± 3.62)%,(6.01 ± 1.02) points] in the Yin deficiency group.Both groups showed obvious improvements in the main and accompanied symptom scores after the treatment (P<0.01),but no significant difference between the two groups (P>0.05).Conclusion The treatment of anorexia due to deficiency of both spleen and stomch with chiropractic and point-massage was effective.
6.Evaluation of left ventricular twist function by speckle tracking echocardiography in aortic stenosis patients with preserved ejection fraction before and after aortic valve replacement

Objective To assess the effect of aortic valve replacement (AVR) on left ventricular (LV) twist function in severe aortic stenosis (AS) patients with preserved LV ejection fraction (LVEF) by speckle tracking imaging (STI).Methods Twenty-eight severe aortic stenosis (AS) patients (17 male,age 64.1 ± 10.4 years) with normal LVEF (≥50%) were examined by STI before and six months after AVR.The standard mitral valve and apical short-axis views were obtained to analyze LV basal and apical peak systolic rotation.LV peak systolic twist was calculated as the different between apical rotation and basal rotation.LV fractional shortening (LVFS) was calculated as the percentage fall of LV systolic dimension with respect to diastolic dimension.The data were compared with 28 age and sex-matched normal controls.Results In patients,LVEF remained unchanged after AVR.LV twist increased before (19.7° ± 5.7° vs 12.9° ± 3.2°,P <0.001) because of increased apical rotation (13.0° ± 5.8° vs 7.6° ± 2.6°,P <0.001),and normalized after AVR (14.4°± 5.2°,P <0.001).In controls,LV twist correlated with LVFS (r =0.81,P < 0.001),this relationship was reversed in patients before (r =0.52,P <0.01) and after AVR (r =0.34,P >0.05).Conclusions In patients with severe AS and normal LVEF,LV twist is exaggerated suggesting potential compensation for reduced long axis function.These disturbances normalize within six months of AVR but lose their relationship with basal LV function.
7.Effects of carbon monoxide releasing molecule-2 on post-resuscitation myocardial dysfunction

Objective To investigate the protective role of carbon monoxide releasing molecule-2 (CORM-2) in post-resuscitation myocardial dysfunction (PRMD) in rat models of cardiopulmonary resuscitation (CPR).Methods Cardiopulmonary resuscitation model was established after cardiac arrest induced by ventricular fibrillation.Male healthy Sprague-Dawley (SD) rats were randomly (random number) divided into 4 groups according to random number table:control group,CORM-2 group,inactive CORM-2 (iCORM-2) group and Sham group,in which the equal volume (1 mL) of 0.2% DMSO,50 μmol/kg CORM-2,50 μmol/kg iCORM-2 and 0.2% DMSO were respectively administered into the rats of these groups after resuscitation.The ejection fraction (EF) of left ventricle and myocardial performance index (MPI) were measured to detect the myocardial function by echocardiography at 12 hours after resuscitation.Mitochondrial respiration was assessed with Clark oxygen electrode at the same time.Western blot was used to determine the ratio of mitochondrial cytochrome c (cyt c) to cytoplasmic cyt c as well as caspase-3 level.Multiple comparisons were made by analysis of variance.Results Compared with the control group,higher EF and MPI,higher state Ⅲ respiration rate and respiratory control rate (RCR) of mitochondria,and decreased ratio of mitochondrial cytc/cytoplasmic cyt c and lower caspase-3 level were observed in the CORM-2 group (P < 0.05).However,there were no significant differences in above biomarkers found between iCORM-2 group and control group (P > 0.05).Conclusions The CO released from CORM-2 might improve mitochondrial respiration and PRMD by inhibition of myocardial apoptosis via a mitochondrial pathway.
8.The Experimental Investigation on Teaching Reform of Orthopedics of TCM Based on Network Resources

Objective:To have an experimental investigation on teaching reform of Orthopedics of TCM based on network resources.Method:Two comparison groups were set and observed when we were teaching the orthopedics students of 2003 grade.One was taught by traditional problem-based learning(PBL) method,the other was taught by PBL method based on network resources.Results:The creative thought and research ability of the students who had been taught by PBL method based on network resources were greatly improved.Conclusion:The teaching method based on network resources has so many advantages such as students being able to self-learn and research without time and space limit that it can be used more in medical students teaching.
9.Correlation and Clinical Significance of Ezrin and β-catenin Protein Expression in Sporadic Breast Cancer

Objective To explore the clinical significance of Ezrin and β?catenin in breast cancer. Methods Immunohistochemical staining method was adopted to detect Ezrin and β?catenin protein expression level in 145 cases of breast cancer tissues,and their correlation with clinical data and prognosis of breast cancer was analyzed. Results Ezrin was expressed in 70 cases(48.3%),β?catenin was expressed in 82 cases (56.6%),and there was significantly negative correlation(r=0.267,P=0.001). The higher histologic grade of breast cancer,the higher expres?sion level of Ezrin(P=0.007),and the lower expression level of β?catenin(P<0.001). Ezrin expression level was increased significantly(P=0.027),but β?catenin expression level was reduced significantly(P=0.011)in lymph node positive breast cancer tissue. Ezrin expression was sig?nificantly correlated with shorter overall survival(P=0.004)and disease free survival(P=0.017)of breast cancer patients,but β?catenin expres?sion was significantly correlated with longer overall survival(P<0.001)and disease free survival(P=0.001)of breast cancer patients. However , Ezrin and β?catenin were not the independent risk factors of breast cancer patients as determined by multivariate Cox regression. Conclusion Ez?rin was significantly negative correlated with β?catenin in breast cancer. They play a role in the progression and poor prognosis of breast cancer , which can be used as breast cancer treatment targets.
10.The changes of CD27+ B cells in peripheral blood of the patients with HBV related disease and its clinical significance

Objective To observe the changes of peripheral blood CD27+B cells percentage in patients with HBV-related disease of different severity and the clinical significance.Methods 20 cases of chronic viral hepatitis B,23 cases of HBV-related liver cirrhosis,21 cases of liver cancer were selected,25 cases of healthy controls in the corresponding period who had the physical examination were set as the healthy control group.The peripheral blood in the healthy controls and the patients with HBV-related disease was collected.The cellular immune level changes and CD27+ B cells percentage was detected by flow cytometry,the humoral immunity was detected by immune transmission turbidty method:IgM,IgD.Results (1)Compared with the healthy control group,CD27+B cells percentage significantly decreased in the three groups of patients with HBV-related disease [(5.16 ± 0.36) % vs.(4.52 ± 0.22) %,(2.24 ± 0.15) %,(0.58 ± 0.02) %,F =4.32,P < 0.05],and the downward trend gradually obvious as the disease degree exacerbated.nnnnn(2)IgM,IgD in the three groups of HBV related disease patients rised obviously,and the increase range became more obvious as the disease degree exacerbation(F =3.29,5.23,P =0.02,0.03).Conclusion CD27+B cells has a close relationship with HBV-related disease,and rebuilding the body's immune defense system is great importance for evaluating prognosis and the clinical guidance in HBV-related disease.
