1.Expression of human papillomavirus L1 capsid protein and tumor suppressor gene p16INK4A and their relevance in cervical lesions

Objective To observe the expression and relevance of human papillomavirus L1 (HPVL1)capsid protein and tumor suppressor gene p16~4A in cervical lesions.Methods The expression of HPVL1 capsid protein and p16INK4A in liquid-based cytology specimens and organization from 210 cases infected with HPV virus were detected by Max-vision immunohistochemistry method.Results In different grade of liquidbased cytology specimens,the positive rates of HPVL1 capsid protein had statistic difference (x 2 =70.50,P < 0.005).The rate in LSIL couples was 68 % (34/50),which was the highest in all couples.The positive rates of tumor suppressor gene p16INK4A were increased gradually.In SCC couples,the positive rates was 100 %(30/30),which was the hishest.In LISL couples,the rate of HPVL1+ p16INK4A-was 32 % (16/50),which was the hishest.In SCC couples,the rate of HPVL1-p16INK4A+ was 100 % (30/30),which was the highest.In orgnizational specimens,the positive rates of HPVL1 capsid protein in cervical intraepithelial neoplasia had statistic difference (x2 =54.37,P < 0.005).The rate in CIN Ⅰ group was 60.4 % (32/53),which was the highest.The positive rates of tumor suppressor gene p16 were increased gradually.In cervical cancer group,the positive rate was 100 % (28/28),which was the highest.In CIN Ⅰ group,the rate of HPVL1+ p16INK4A-was 45.3 % (24/53),which was the highest.In cervical cancer couples,the rate of HPVL1 (-) p16INK4A(+) was 100 % (28/28),which was the highest.Conclusion Detection on the expression of HPVL1 capsid protein and tumor suppressor gene p16INK4A can diverse different levels of cervical lesions and separate cases from aggravated or self-healing,to avoid over-treatment or misdiagnose.
2.Clinical analysis of deep fungal infections in ICU hospital

ObjectiveTo investigate the clincal status and countermeasures for intensive care unit (ICU) hospital deep fungal infections.Methods108 cases of ICU hospital deep fungal infections patients were divided into comprehensive treatment group and converntional treatment group,54 cases in each group.The morbidity and treatment effects were observed.ResultsThe cure rate of observation group was 96.3%,significantly higher than 79.6%in control group.The observation group complication rate was 9.2%,significantly lower than the control group 20.4%,the difference was significant (P < 0.05 ).ConclusionIn the ICU nosocomial fungal infection treatment,the scientific and effective means of comprehensive prevention and control best practices,could significantly improve patient outcomes,improve treatment,reduce hospital stay and reduce complications.
3.Effects of Exposure to Arsenic through Drinking Water on Simple Visual Reaction Time and IQ of Combined Raven's Test in Children

Objective To explore the intellectual effects of arsenic in drinking water on children. Methods Intellectual effects were evaluated by computer-based reaction time and Combined Raven's Test. Children aged 9 to 11 years living in three villages of Inner Mongolia were investigated in April, 2006. The arsenic concentrations in the drinking water of the three villages were 0.16 mg/L, 0.09 mg/L and less than 0.05 mg/L (control group). Results The mean, fastest and slowest visual reaction time of children exposed to the high levels of arsenic through drinking water were longer than those of children in control. The differences of mean, fastest and slowest visual reaction time between girls exposed to 0.16 mg/L arsenic and control were significant; the differences of the mean and slowest visual reaction time between 10-year old children exposed to 0.16 mg/L arsenic and control were significant. IQ of girls exposed to 0.16 mg/L arsenic was significantly lower than that in control. Conclusion Intellectual function of children can be affected by arsenic in drinking water, and the intellectual effects of arsenic on girls are more obvious.
4.Impact of health literacy on education retention of stroke patients

Objective To describe the health literacy level and to explore the effect of health literacy level on education retention of patients with stroke.Methods Totally 100 stroke survivors were selected as the subject by convenience sampling.They were evaluated by Short Test of Functional Health Literacy in Adults on admission,and they accepted 60-90 minutes education intervention during hospitalization.Evaluation retention were measured before discharge.Results The score of the overall health literacy was (15.60±1.36) points,among of them,low level was 37 cases,general level was 24 cases,high level was 39 cases.Age,education level and Mini-Mental Status Exam score had a significant effect on the health literacy of patients with stroke (x2/F=14.248,12.082,11.329,P< 0.05).After education intervention,the score of education retention was (6.69±2.00)points,and the score of education retention in low,general,high level health literacy were (5.08±2.06),(7.46±1.61),(7.51± 1.76) points,and there were significant differences(F=14.223,P < 0.05).Heahh literacy had a effect on education(P < 0.01).Conclusions The level of health literacy of stroke patients has the effect of improving the education retention.Improving and maintaining the health literacy of stroke patients should be the development direction of the prevention and treatment of stroke diseases.
5.Study of the level of ERM proteins in pulmonary microvascular endothelial cells induced by tumor necrosis factor-α

Objective To investigate the effect of tumor necrosis factor-α (TNF-α) on the levels of ezrin-radixin-moesin (ERM) proteins and the phosphorylated ERM proteins (p-ERM) in rat pulmonary microvascular endothelial cells (PMVEC),and to explore Rho kinase (ROCK) influencing on modulation of the ERM proteins phosphorylation.Methods Cultured rat pulmonary microvascular endothelial cells were randomly divided into dose-dependent and time-dependent groups.In dose-dependent group,cells were cultured with different doses of TNF-α (0,0.1,1,10 μg/LTNF-α) for 60 min.In time-dependent group,cells were cultured with TNF-α (10 μg/L) for 0,15,30,60,90,120,180 min.In ROCK inhibitor (Y27632) intervention group,cells were cultured with TNF-α (10 μg/L) or Y27632 (30 μmol/L) + TNF-α (10 μg/L) for 60 min respectively.The levels of ERM proteins and p-ERM were determined by western blot.One way analysis of variance (ANOVA) was employed for statistical analysis by using SPSS version 16.0 software to compare values among all groups.A significant difference was presumed as a P value < 0.05.Results Western blot revealed that ERM and p -ERM proteins were present in rat PMVEC.Stimulation withTNF-α gradually up-regulated the level of pERM proteins in a dose-dependent manner [0 μg/LTNF-α group:(0.648 ± 0.102),0.1 μg/LTNF-αgroup:(0.728-±0.082),1 μg/LTNF-α group:(0.926±0.121),10 μg/LTNF-α group:(1.245 ±0.134),all P =0.000].In time-dependent group,the level of p-ERM proteins rose at 15 min (0.777 ±0.151),peaked at 90 min (1.295 ±0.176),then decreased gradually at 120 min (0.802 ±0.139),but stayed higher level at 180 min (0.669 ±0.128) than that in un-stimulated 0 min group (0.631 ±0.123,P=0.004,0.000,0.001,0.016,respectively).When PMVEC pre-incubated with ROCK inhibitor and TNF-t,the level of p-ERM proteins caused a marked attenuation of TNF-αstimulation [(0.634 ± 0.112) vs.(0.875 ± 0.164),P =0.002],however,there are no significant differences compared to ROCK inhibitor alone group (0.661 ± 0.108) and no intervention group (0.654 ± 0.125),P =0.973,P =0.900,respectively).Conclusions TNF-α could induce up-regulation of the level of the phosphorylated ERM proteins in rat PMVEC,and ROCK signal molecules might involve in modulation of the ERM proteins phosphorylation.

7.Effects of 6-week voluntary wheel exercise on sex-hormones and hypothalamic-pituitary-testicular axis in high-fat fed SD rats

Objective To observe the effects of high fat diet and 6?week voluntary wheel running on the structure of hypothalamus, pituitary and testis and sex hormone levels in pubertal male rats. Methods Forty 3?week?old male Spra?gue?Dawley rats were randomly divided into following groups: the control group fed with normal diet (C), training group fed with normal diet ( CE) , control group fed with high fat diet ( D) , and training group fed with high fat diet ( DE) , 10 rats in each group. The groups C and D were bred for 8 weeks freely, and the groups CE and DE were assigned to have vol?untary wheel running twice/d, 1 h/time, 5 d/w, for 6 weeks after a 2?week adaptive feeding. Eight weeks later, blood sample was collected to detect the serum T, E2 , FSH and LH, and the E2 and T of testis were also detected. The histology of hypothalamic, pituitary, and testis tissues was observed by light microscopy. Results ( l) Compared with the group C, the group D had significantly decreased levels of serum T and E2 and testicular T (P<0?05), and significantly increased serum E2, FSH and LH levels (P<0?05). In the group D, vacuolar lipid droplets were increased in the hypothalamus, e?osinophils and basophils were reduced in the pituitary, and the area of seminiferous tubules, percentage of sperm cells, and quantity of Leygid cells were significantly decreased in the testis. (2) Compared with the group D, the serum T and testic?ular T concentrations were increased, but the serum E2, FSH and LH were decreased significantly (P<0?05) in the group DE, and vacuolar lipid drops were increased in the hypothalamus, eosinophil cells were increased in the pituitary gland, and the area of seminiferous tubules was increased but not significantly in the testis. Conclusions 6?week voluntary wheel exercise can improve the high fat diet?induced abnormal secretion of sex hormones, but not effectively improve the histologi?cal changes in hypothalamus, pituitary gland and testis.

Objective To investigate the effects of low temperature adjustable sodium dialysis combined Astragalus injection in patients with chronic cardiac insufficiency uremia combined the clinical curative effect of dialysis related hypotension .Methods Fourty‐nine patients with uremia combined chronic cardiac insufficiency and suffered repeated dialysis hypotension were involved in this study ,and they were divided into treatment group(n=25) and control group(n=24) .Patients in the control group were treated with hemodialysis for low temperature (35 .5 ℃) ,and sodium curve dialysis ;patients in the treatment group were treated with the same method in that of control group ,and they were treated with astragalus injection 40 mL continuous intravenous drip .Then ob‐serve the blood pressure changes and treatment effect of the two groups of before and after treatment .Results Systolic pressure of the treatment group and the control group was (92 .6 ± 8 .4) and (89 .5 ± 9 .0) mm Hg ,and that of after treatment was (99 .9 ± 8 .0) and (94 .9 ± 9 .3) mm Hg respectively ,the difference before and after treatment in each group was significant (t=3 .147 , 2 .044 ,P<0 .05) ,and the difference of systolic pressure before and after treatment in each group was significant (t=2 .020 ,P<0 .05) .Before the treatment ,the left ventricular ejection fraction score of treatment group and control group was (45 .80 ± 3 .62)%and (46 .01 ± 3 .59)% ,and that of after treatment was (59 .05 ± 3 .15)% and (53 .85 ± 1 .60)% ,respectively ,and the difference be‐fore and after treatment was significant (t=13 .906 ,9 .847 ,P<0 .05) ,and the difference of the left ventricular ejection fraction score after treatment in the two groups was significant (t=7 .328 ,P<0 .05) .Conclusion For patients with uremia combined chronic car‐diac insufficiency and suffered repeated dialysis hypotension ,low temperature adjustable sodium dialysis can significantly reduce the incidence of hypotension in the process of dialysis .But combined huangqi injection in the treatment group could significantly in‐creased the left ventricular ejection fraction ,thus improve the left ventricular function in patients with dialysis .
