1.Analysis of protoscoleces-specific antigens from Echinococcus granulosus with proteomics combined with Western blot.

OBJECTIVETo establish and optimize the proteomic analysis of protoscoleces-specific antigens from Echinococcus granulosus. To provide a foundation for identifying specific antigens in the soluble proteins of E. granulosus protoscoleces for further research.
METHODSBrood capsules were collected aseptically from fertile E. granulosus cysts from the livers of an infected patient. The fertile E. granulosus cysts were fractured, and protoscoleces were collected by centrifugation. The soluble proteins of protoscoleces were acquired using the 2D Quant kit according to the manufacturer's instructions. We employed two-dimensional electrophoresis (2-DE) combined with immunoblot assay (Western blot) to analyze the soluble components of E. granulosus protoscoleces antigens. The 2-DE and immunoblot maps obtained were analyzed with PDQuest 8.0 image analysis software.
RESULTSAbout 233 soluble protein spots were identified with Coomassie-stained gels. Most of the proteins had a molecular weight of 16,000 Da to 117,000 Da, and an isoelectric point value of 3.0 to 10.0. 2-DE immunoblot was conducted and 57 specific antigen spots were observed, among which 23 spots were identified.
CONCLUSION2-DE combined with Western blot is the key to successful proteomic analysis and presents a new possibility for searching the specific E. granulosus protoscoleces antigens.

3.Longitudinal stress and strain of normal human vascular system of the extremities, and the blood vessels following venous repair

Objective To investigate the longitudinal residue strain and stress-strain relation of normal human vascular system of the extremities and the relation between the different defect length and the option of repair, as well as a comparison of the different selection of the methods of repair judged from the longitudinal biomechanical effect. Methods A mechanical analysis including stretch experiment and longitudinal stretch ratio measurement were undertaken to evaluated the longitudinal residue strain and stress-strain relation of arteries and veins of the extremities in human. The results of different methods for repair of the defect of the same kind of vessels were compared with Student-t test. The length precinct to repair arterial injuries between end-to-end anastomoses and vein graft through 95% distribution region was evaluated. Results The longitudinal stretch ratio of arterial system of the extremities decreased from the proximal to the distal ends of the vascular branches and the stress-strain relation curve of artery turned gradually to the right side, while that of veinous system exhibited a contrary picture. 4.An Evaluation of the Mutations of Common Susceptive Genes Related Hearing Loss for 161 Patients in Liuzhou

Objective To investigate the epidemiological characteristics of three common susceptive gene retared hearing loss in the patients with the congenital deafness in Liuzhou.Methods 161 patients with congenital hearing loss were diagnosed with audiologic evolutions,including newborns and outpatients.The blood samples of all patients were taken for the extraction of DNA which was amplified by PCR.The common mutationsl hot spots of the mitochondrial DNA 12SrRNA,GJB2 and SLC26A4 were examined by restricted enzyme and directed sequencing.Results 1 case(0.62%)was found to carry mitoehondrial DNA 12SrRNA A1555G and 4 patients(2.48%)carried heterozygotes or homozygotes pathologic mutations of GJB2.10 patients(6.21%)were heterozygous carriers with pathologic mutations,IVS7-2 A>G,in the SLC26A4 gene.The detection rate of GJB2,mitochondrial DNA A1555G and SLC26A4 mutations in 161 patients were 9.31%.Conclusion The patients with congenital hearing loss distributed different minority groups in liuzhou zone.The mutational frequencies of the three common gene related hearing loss in the patients of Liuzhou were noticeably lower than the data reported in other regions in China.The gene screening for deafness was very important for early diagnosis and treatment.
5.Behavior problems and family factors in children of off-farm workers of Shanghai

Objective To survey prevalence of behavior problems in children of off-farm workers and effects of family factors on children' s behavior problems.Methods The study was done in the whole 7 schools for off-farm workers' children in Shanghai Baoshan district,including all 3 ~ 6 grades students of primary school and 1 ~3 grades students of junior high school.Totally 4350 students finished survey (95.4%).Achenbach' s child behavior checklist ( CBCL,parent' s version) and general family questionnaire were used.Results ( 1 ) Prevalence of behavior problems in children was 11.2% ; 12.8% 12 ~ 16 years children had behavior problems( 14.7%boys and 10.4% girls) ; 11.4% of one-child children had behavior problems compared with that of 11.2% child with siblings children ( x2 =0.038,P > 0.05 ).(2) Total score ( 20.64 ± 17.67 ) and factor scores of thought problems,depressed,hyperactivity,delinquent rule-breaking behavior were higher than those of Chinese norm (P <0.05 or P < 0.01 ) among 6 ~ 11 years boy group.Total score ( 17.23 ± 14.73 ) and scores of thought problems,depressed,social problems,hyperactivity,delinquent rule-breaking behavior were higher than those of Chinese norm (P < 0.01 ) among 6 ~ 11 years girl group.Among 12 ~ 16 years boy group,total score (20.44 ± 19.32) and all factor scores of CBCL were higher than those of Chinese norm(P<0.01 ).Total score ( 17.02 ± 16.72) and scores of immature,delinquent rule-breaking behavior,brutality were higher than those of Chinese norm(P < 0.05 or P <0.01 ) among 6 ~ 11 years girl group.(3)Multi-regression analysis showed that poor parent-child relationship was the most important predict factor for higher score of CBCL among all family risk factors (P < 0.05 ).Conclusion Children of the off-farm workers and their families need special mental health care program.
6.Study of effect of mifepristone on apoptosis of human ovarian cancer cell line 3AO

Objective To investigate the effect of mifepristone on the activity of proliferation and the apoptosis, the expression of estrogen receptor (ER),progesterone receptor (PR) protein and morphology changes of human ovarian carcinoma cell line 3AO and SKOV3 in vitro. Methods The proliferative activity of 3AO and SKOV3, which were cultured in vitro, was measured by tetrazolium-based colorimetric assay (MTT assay). Flow cytormetry (FCM) was used to measure the expressive rate of ER, PR , p53 protein, bcl-2 protein, cell apoptotic rate and cell proliferative cycle of 3AO cells, which were cultured with different concentration and duration of mifepristone. The morphologic and ultrastructure changes of apoptotic 3AO cells was observed by the light and electron microscopy. Results Mifepristone inhibited significantly the proliferation of 3AO cells in dose-time dependent manner in vitro. The inhibitory rate of 3AO cells growth, which were cultured with different concentration of mifepristone( 5,10,20,40,80 ?mol/L) and duration (24,48,72 h) was from 1.7% to 75.0%(P0.05). 3AO cells apoptosis activity appeared the positive correlation with the dose of mifepristone and cultured duration (P
7.Biofilm inhabitation and representation on the surface of polyethylene

BACKGROUND: Studies have demonstrated that intervention material retention for a period of time in vivo causes pathogenic bacteria surface attachment, proliferation even forming biofilm to become potential source of high polymer catheter-associated infection. Inhibition of biofilm formation significantly reduces infection.OBJECTIVE: To evaluate the antimicrobial effect of anti-infective polyethylene material and its inhibiting effect to biofilm on the surface.DESIGN, TIME AND SETTING: Single sample observation was performed at the National Engineering Research Center for Engineering Plastics, Technical Institute of Physics and Chemistry, Chinese Academy of Sciences from June 2008 to May 2009, and the antimicrobial test was performed at the General Hospital of General Staff Headquarters of PLA in March 2009.MATERIALS: Organic antibacterial agent2,4,4'-trichloro-2'-hydroxydiphenyl ether; IONPURE~(R) Silver-Inorganic anti-microbialreagent; low-density polyethylene (LDPE).METHODS: LDPE was respectively blended with organic antibacterial agent and inorganic antibacterial agent, and the LDPE slice was prepared by injection molding process.MAIN OUTCOME MEASURES: The antimicrobial efficacy of anti-infective polyethylene was determined by the plate counting method. The ultrasonic-plate method and scanning electron microscopy were introduced to observe the biofilm formation.biofilm maturation. The propagation of microbe was found to be depressed remarkably on the surfaces of modified samples with organic antibacterial agent, and the formation of biofilm was prevented. The observation of SEM confirmed that no biofilm was formed on the surfaces of PE added organic antibacterial agent. The results showed that a large amount of bacteria and extracellularpolysaccharide matrix adhered on the surfaces of PE added inorganic antibacterial agent and untreated PE.CONCLUSION: Anti-infective polyethylene added organic antibacterial agent possesses excellent antimicrobial performance against S. aureus and E coli, and it can inhibit the formation of biofilm on the surfaces, which provides a promising approach to prevent catheter-related infections.
8.Vertebroplasty with artificial bone repairs vertebral compression fractures in the elderly:3-month follow-up results

BACKGROUND:Bone cement used in the vertebroplasty treatment of osteoporotic vertebral compression fractures is nonabsorbable and has stronger intensity than normal bone tissue, which can cause further fractures of adjacent segments. OBJECTIVE:To investigate the short-term effect of vertebroplasty with artificial bone in the treatment of osteoporotic thoracolumbar fractures. METHODS:A total of 21 patients with osteoporotic vertebral compression fractures were enrol ed, including 7 males and 14 females, aged 61-89 years old. Al the patients were subjected to vertebroplasty with calcium sulfate injection. Visual analog scale and Oswestry Disability Index assessment were used before and at 0, 24, 48 weeks after treatment;meanwhile, X-ray and CT were used to observe the fil ing and leakage of artificial bone. RESULTS AND CONCLUSION:After treatment, al the patients showed improvements in the scores of visual analog scale and Oswestry Disability Index (P<0.01). X-rays and CT scans showed that there were three cases of leakage but without spinal cord and nerve compressions. After 3 months, most of the artificial bone was absorbed. These findings indicate that vertebroplasty with artificial bone can ease pain and improve living conditions of patients with osteoporotic vertebral compression fractures.
9.Laparoscopic versus open D2 gastrectomy for advanced gastric cancer: A meta-analysis on long-term outcomes

Objective To evaluate the long-term clinical outcomes between laparoscopic gastrectomy and open gastrectomy with D2 lymph dissection for advanced gastric cancer.Methods Clinical studies that compared clinical outcomes of laparoscopic gastrectomy and open gastrectomy for advanced gastric cancer were searched from PubMed,EMBASE,Medline,Cochrane Library,WanFang,CNKI,CMCC and VIP database with the Gastric neoplasms Laparoscopy Gastrectomy Long-term outcomes Meta-analysis between Jan.2002 and Oct.2016.Data of long-term survival and recurrence were analyzed by using of RevMan 5.2 software.Survival data were present by the odds ratio(OR) and 95% CI.The heterogeneity of the data was analyzed using the I2 test.Results Fifteen studies including 4,053 cases were enrolled.There were 2,091 patients in LG group and 1,962 patients in the open gastrectomy group.There was no significant difference in the 3-year overall survival rate(OR =1.00,95% CI:0.83-1.20,P =0.98),5-year overall survival rate (OR =1.14,95% CI:0.95-1.36,P =0.15),5-year disease-free survival rate(OR =1.13,95% CI:0.93 ~ 1.39,P =0.22)and cancer recurrence rate (OR =0.96,95% CI:0.79 ~ 1.18,P =0.71)between the patients treated with laparoscopic gastrectomy,or open gastrectomy (P > 0.05).Conclusion Laparoscopic gastrectomy with D2 lymph dissection for advanced gastric cancer has similar long-term outcomes as compared to open gastrectomy.
