1.Expression of TNF\|α and CD68 in nasal polyps tissue and their significance

Objective: To study the expression of TNF\|α and CD68 in nasal polyps tissue and their significance. Methods. Paraffin sections of 19 nasal polyps and 8 chronic rhinitis samples were studied with immunohistochemical technique. Results. The expression of TNF\|α were stronger in nasal polyps tissues than that in controls. Numerous CD68 positive cells were found in epithelium laryer and in the lamina propria of nasal polyps. There were positive relationship between the expression of TNF\|α and that of CD68(r=0.615). Conclusion. It was suggested that activized macrophage as antigen\|presenting cell in the development of nasal polyps, may play a important role through upregulated TNF\|α′s synthesis and secretion.
2.Influence of anisodamin on guinea pig experimental nasal hypersensitivity

Objective:To observe the influence of anisodamin on guinea pig experimental nasal hypersensitivity.Method:Experimental animals were divided into normal control group, TDI group,reserpine group and anisodamine group.The scores of symptom of nose and the cytologic changes of nasal mucosa were parameters of observing.Result:The nasal symptom of reserpine group was significantly stronger than the TDI group,and the eosinophilia evidently increased (both P<0.01).The nasal symptom of the anisodamine group reduced and the eosinophilia decreased.It is significantly difference between the TDI group and the reserpine group (both P<0.01).Conclusion:Anisodamine may play a role in therapy of the experimental nasal hypersensitivity with hyperfuction of parasympathetic nerve.

4.The expression and effect of IL-4 in human palatine tonsil

AIM: To study the allergic disease in recurrent tonsillitis. METHODS: Immunohistochemical technique was used to investigate interleukin-4(IL-4) production in tonsil. RESULTS:The result showed that: a higher incidence of IL-4 producing cells were found in the tonsils of all ages, and a high density of IL-4 was found in the reticular crypt epithelium and extrafollicular, but low density in germinalcenter. IL-4 has been shown play a crucial role in the pathogensis of allergic disease. CONCLUSION:Allergic disease is an important reason of recurrent tonsillitis. The conclusion will give a new way to cure recurrent tonsillitis.
5.Nasal cavity volume and nasopharyngeal cavity volume in adults measured by acoustic rhinometry

Objective:To investigate the range of normal value of total nasal cavity volume(NV)and total na-sopharyngeal cavity volume (NPV)of healthy adults. Method: Six hundred and fifty-nine healthy adults andeighty-two adults of chronic rhinitis were measured with acoustic rhinometry. Result:The range of NV was29. 922~37. 481 cm3 and NPV was 29. 369~44. 159 cm3. Comparing healthy adults with adults of chronic rhini-tis,there was a significant difference in NV. Conclusion:Acoustic rhinometry suited for objective assessment ofthe nasal airways in adults. It was demonstrated that these data could provide available information for the studyof nasal physiology and pathophysiology,as well as for the diagnosis and judgement of therapeutic effectiveness ofnasal diseases.
6.The impact of lateral rhinotomy on nasal airway

Objective:In order to ascertain the lateral operation and medial maxillectomy affect on nasal airway.Method:Report 16 cases of nasal cavity or nasal sinus neoplasm who were therapied by lateral rhinotomy and medial maxillectomy.They were observed by acoustic rhinometer (AR) and CT examination in 1~2.5 years after the surgery. Result:The feeling of nasal obstruct were disappeared in 3 months after the surgery. The nasal airway resistance(NAR) of the ill nasal cavity was markedly decreased after the surgery, and the total NAR were decreasing unmarkably. The mucous proliferation was notable only in the first year after surgery, and especially on remains of the inferior turbinate or extra wall of nasal cavity.Conclusion:Lateral rhinotomy operation and medial maxillectomy have less impact on nasal airway and the functions.
7.Sodium channels in the apical membrane of human nasal epithelial cells

Objective To study the electrophysiological properties of sodium channels in the apical membrane of human nasal epithelial cells. Method Nasal epithelial cells of human inferior turbinate from patients with obstructive sleep apnea syndrome were cultured in serum free medium on collagen gel-coated membranes at an air-liquid interface and studied by a patch clamp technique. Results In cell-attached patches, a typical single channel current with a conductance of 21.09pS and reversal potential of -50.96 were recorded. The permeability ratio PNa/PK was more than 5.80. In the presence of 10-4 mmol/L amiloride in the pipette, the incidence of sodium channels decreased from 26.67% to 5.13%. This revealed that a population of channels were inhibited by amiloride at a dose of 10-4 mmol/L. Ca2+ at dose of 10-3 mmol/L did not influence the incidence of sodium channels. There was no obvious association between voltage and the open probability of the channels. Conclusions Our results indicate that most Na+ channels in cell-attached patches of human nasal epithelial cells are amiloride-sensitive and Na+ selective. Only a few channels are amiloride-insensitive. The channels were not activated by extracellular Ca2+ and the open probability followed a voltage-independent manner.

9.Protein kinase C in proliferation and infiltration of eosinophils in nasal polyp.

OBJECTIVETo explore the significance of protein kinase C (PKC) in proliferation and infiltration of eosinophils in nasal polyps.
METHODSWith in situ hybridization and immunohistochemistry staining methods, PKC, pro-apoptotic, and anti-apoptotic gene (Bax, bcl-2) expressions were measured in nasal polyp tissues from 26 patients and inferior turbinate mucosa tissues (ITMTs) from 20 healthy persons. The May-Grünwald-Giemsa (MGG) staining method was used to identify eosinophils.
RESULTSIn eosinophils, the positive cell expressive rates of Bcl-2 mRNA and its protein were significantly higher in the group with nasal polyps than in the ITMT group (P < 0.01). Although the positive cell expressive rate of Bax mRNA and associated protein were a little higher in the group with nasal polyp tissues than in the ITMT group, the difference was not significant (P > 0.05). There was PKC expression in the eosinophils of 26 cases of nasal polyps, but occasional PKC expression in 7 of 20 ITMT cases. In the two groups, PKC positive cell expression was significantly different, and the expression of PKC and bcl-2 mRNA as well as associated protein in eosinophils of nasal polyps showed a remarkably positive relationship (r1 = 0.0875, r2 = 0.0823, P < 0.01).
CONCLUSIONSIncreased PKC expression in eosinophils of nasal polyp tissues is closely associated with apoptosis inhibition, and it is presumed that eosinophil apoptosis inhibition in nasal polyp tissues is obtained by activation of the PKC signal transduction pathway.

10.Expression and distribution of aquaporin 1 in laryngeal carcinoma.

OBJECTIVE:
To detect expression level and distribution of aquaporin 1 (AQP1) in laryngeal carcinoma tissues and try to find out the significance about the mechanism of AQP1 in the invasion of laryngeal carcinoma.
METHOD:
Twenty cases of laryngeal carcinoma tissues and 15 cases of normal tissues beside tumors were acquired immediately after surgery. The expression and distribution of aquaporin 1 in laryngeal carcinoma were examined by RT-PCR, western blot analysis and immunohistochemical technique.
RESULT:
AQP1 expresses on the capillary endothelia of laryngeal mucous lamina propria and laryngeal mucous glands of normal tissues and mainly on the capillary endothelia cells, tumor cells and cancer nests of laryngeal carcinoma tissues. AQP1 mRNA and protein expression level in laryngeal tumor tissues is remarkably stronger than that in normal tissues beside tumors.
CONCLUSION
The expression of AQP1 was increased in laryngeal carcinoma tissues over normal laryngeal tissues. It suggests that the mechanism of AQP1 in laryngeal carcinoma etiopathogenesis needs to be studied.
