1.Relationship between the expression of Ki-67 and molecular classification and clinical patho-logical features in breast cancer

Purpose To study the expression of Ki-67 in breast cancer with different molecular classification, and to discuss the rela-tionship between the expression of Ki-67 and clinical pathological features in breast cancer. Methods All 245 patients with breast cancer were divided into different molecular classification through detecting the expression of ER, PR, HER-2 by immunohistochemical method. The expression of Ki-67 was also detected, and to study relationship between the expression and clinical pathological features. Results The difference of Ki-67 index was statistically significant in different molecular classification of breast cancer. Ki-67 index in patients with lymph node metastasis and larger tumor size were higher than that with no metastasis and smaller tumor size. Ki-67 index in patients with ER and PR positive were lower than that negative, the difference was statistically significant. Ki-67 expression differ-ence was not statistically significant between the two groups according to the median age (50 years old). But when patients were divid-ed into young and elderly groups by≤40 and≥60 years old, Ki-67 index in young group was more higher than that in elderly group. Conclusion Ki-67 index in patients with triple-negative breast cancer, young age, lymph node metastasis, larger tumor size and ER and PR negative were higher. Ki-67 can be used as an important indicator to determine breast cancer prognosis.
2.Comparison of clinical-pathological features and Ki-67 expression profile between triple-negative breast cancer (TNBC) and non-triple-negative cancer

Objective To compare clinical-pathological features and Ki-67 expression profile between TNBC and Non-triple-negative cancer (NTNBC). Methods The expressions of ER, PR and HER2 were examined by immunohisto?chemical Envision double staining in 226 cases of breast cancer. All cases were divided into triple-negative and non-triple-negative group according to expressions of these three episodes. And Ki-67 expressions in two groups were assessed by im?munohistochemical method. The difference of Ki-67 positive index between these two groups and the difference of Ki-67 positive index between different clinical-pathological features were both analyzed by comparative analysis. Results In pa?tients with TNBC, tumor size, lymph node metastasis rate and Ki-67 index were all significantly higher than that in NTNBC. No significant difference in age and histological grade were detected between these two groups. The expression of Ki-67 was not significant correlated with age, tumor size,lymph node status and histological grade in these two groups. Conclusion Patients with triple-negative breast cancer were with larger tumor size, higher proliferation, more lymph node metastasis and poorer prognosis.
3.Treatment of 41 Cases of Prostate Hyperplasia by Elongated Needle

To observe the therapeutic effect of acupuncture plus medicament in the treatment of prostatic hyperplasia, Zhibian (BL 54) and Zhongji (CV 3) were deeply punctured with elongated needles and enema was applied with self-prepared medicinal herbs. Then Qugu (CV2),Zhongliao(BL 33) and Xialiao(BL 34) were selected and connected with a herbal Ion Introduction Apparatus. The total effective rate was 92.4% in 41 cases. Elongated needle and herbal ion introduction can relieve the clinical symptoms of prostatic hyperplasia and improve the patients' life quality.
4.Effects of chemotherapy on free radicals in the rat glioma

AIM: To observe the role of free radicals in the inhibitory effect of chemotherapy on glioma cells. METHODS: C6 glioma cells were cultured in vitro , and treated with carmustine (B), teniposide (V), or/and nimodipine (N). Furthermore, the glioma-bearing rats were treated with B plus N, B+V+lisplatin (D)+N, or B+V+D+N+angiotensin Ⅱ. The MDA content and superoxide dismutase (SOD) activity in the culture supernatant and cortical brain tissue were assayed. RESULTS: B, V and N significantly decreased MDA content and SOD activity in the supernatant of glioma cell culture and C6 glioma cells. Chemotherapy reduced MDA content and increased SOD activity in the cortical brain tissue of tumor-bearing rats, with highest efficiency in B+V+D+N+angiotensin Ⅱ group. The survival time of tumor-bearing rats in B+V+D+N+angiotensin Ⅱ group was longer than that in other chemotherapy group. CONCLUSION: The antitumor effects of combined chemotherapy may be involved in the free radical metabolism. [
5.Expression of glucose regulated protein 78 and the pathogenesis of gestational diabetes mellitus

Objective To investigate the relationship between the expression of glucose regulated protein 78 (GRP78) in the placental trophoblast cells and the pathogenesis of gestational diabetes mellitus (GDM).Methods All the patients were recruited from Qingdao Municipal Hospital from May 2013 to May 2014.Among them, fifty women with GDM were assigned to the GDM group, and fifty healthy women were defined as the control group.All of them received cesarean section because of breech presentation, contracted pelvis, scarred uterus or on mother's demand.Real-time PCR was conducted to analyze the expression of GRP78 mRNA in the trophoblasts.Immunohistochemistry was performed to detect the localization of GRP78 protein in the placentasl trophoblast cells.Results (1) GRP78 mRNA expressed in the cytoplasm of trophoblasts of both the GDM group and the control group.The GRP78 mRNA levels in the GDM group and the control group were 15.6±0.4 and 6.0±0.7, respectively.The relative expression level of GRP78 mRNA in the GDM group was 2.6 times of that in the control group, with statistically significant difference (P<0.01).(2) The expression of GRP78 protein was found in the cytoplasm of the trophoblasts of the GDM group.It showed in deep, light brown or yellow after staining, according to the expression degree.The expression of GRP78 protein was also found in the cytoplasm of the trophoblasts of the control group, but it mainly showed yellow color (38/50).The strong positive rate of GRP78 protein in the GDM group (96%, 48/50) was higher than that in the control group (22%, 11/50;P<0.01).Conclusion The expression of GRP78 increased in the placental trophoblast cells of GDM patients.It might suggest that GRP78 had some effect on the pathogenesis of GDM.
6.Effects of transient receptor potential V6 silence on proliferation and apoptosis of trophoblasts

Objective To investigate the effects of the transient receptor potential V6 (TRPV6) gene silencing on the proliferation and apoptosis of trophoblasts HTR-8/SVneo cells.Methods siRNA sequences targeting the TRPV6 gene were constructed and then transfected into HTR-8/SVneo cells mediated by liposome.The cells were divided three groups,including blank control (add the reagent of transfenction),negative control groups (transfecting nonspecific siRNA) and experimental groups (transfecting TRPV6-siRNA).Those cells in every group were collected at 24,48,72 hours after transfecting.The expression levels of TRPV6 mRNA were detected by reverse transcription (RT) PCR at different times after transfecting.The effects of siRNA on the proliferation and apoptosis of the cells were assayed by methyl thiagolyl tetragolium (MTT) and flow cytometry at different times after transfecting.Results siRNA TRPV6 transfection could inhibit the expression of TRPV6 mRNA in the HTR-8/SVneo cells.The expression was decreased with the extension of time,by 0.72 ± 0.02,0.54 ± 0.02 and 0.29 ± 0.01 after 12,48 and 72 hours of siRNA transfection as compared with the blank control and the negative control groups (P <0.01).The rates of proliferation inhibition were (19.29 ± 1.23) %,(32.12 ± 1.35) % and (46.51 ±1.42) % at 24,48 and 72 hours respectively when compared with the blank control (2.12 ± 0.03)%,(2.42 ± 0.02) %,(3.13 ± 0.04) % and the negative control groups (2.37 ± 0.01) %,(2.61 ± 0.05) %,(2.93 ± 0.03) % (P < 0.01).The apoptosis rates of HTR-8/SVneo cells was 16.21% at 48 hours after transfected with siRNA TRPV6,which were significantly higher than 3.27% in the blank control and 5.34% in the negative control groups (P < 0.05).Conclusion Silenceing of TRPV6 genen could inhibit the proliferation and increase the apoptosis of extravillous trophoblas of human placenta.
7.Establishment of three-dimensional finite element digital foot model

BACKGROUND:To overcome the disadvantages of traditional mechanical analysis of specimens, and establish the finite element model of realistic foot, are the important basements for the finite element mechanical analysis on foot.
OBJECTIVE:To establish three-dimensional finite element model of foot and lay the foundation for the finite element analysis of normal foot and foot injury.
METHODS:A healthy female volunteer was involved in this study and was detected with spiral CT scanning on the feet. The resulting image was used to reconstruct the three-dimensional model by using Mimics software. Then entity model was generated in Geomagic software. Final y three-dimensional finite element model was established based on the digital main structure in Ansys. RESULTS AND CONCLUSION:The established finite element digital model of human foot included al bone, cartilage and ligament, skin and soft tissue. The three-dimensional finite element model of human foot was established based on CT data and using Mimics, Geomagic, Ansys softwares. The established model had similar size and shape with skeletal mode, and can rotate freely in any angle for a variety of measurement, the foot bones can be arbitrarily split or merge, which is suitable for biomechanical analysis.
8.Analysis of the factors affecting the community of Alzheimer's disease

Objective To understand the risk factors of the elderly in community of Haining city in patients with Alzheimer's disease.Methods Take the elderly Chang street Haining city selected cluster sampling method 7 437 ≥60 years as the research subjects,by using a self-designed questionnaire Alzheimer's disease causes and clinical dementia rating scale,understanding the factors that may affect Alzheimer's disease and aggravating factors,and the data of multi factor regression analysis.Results Multivariate Logistic regression analysis showed that women (Wald x2 =19.96,P < 0.01),aged (Wald x2 =96.01,P < 0.01),widowed (Wald x2 =5.09,P < 0.05),smoking/drinking (Wald x2 =47.77,P < 0.01),living alone (Wald x2 =16.14,P < 0.01),depression (Wald x2 =12.05,P < 0.01) were the risk factors of Alzheimer's disease; Rural (Wald x2 =458.11,P < 0.01) ; Lowculture (Wald x2 =121.39,P <0.01) were protective factors for Alzheimer's disease.According to the clinical dementia rating scale of disease severity assessment and pathogenesis of polytomous Logistic regression,the results showed that the elderly (Wald x2 =50.37,P < 0.01),lowculture (Wald x2 =30.39,P < 0.01),widowed (Wald x2 =5.20,P < 0.05),living alone (Wald x2 =16.50,P < 0.01),somatic diseases (Wald x2 =16.58,P < 0.01) and large maternal age (Wald x2 =10.53,P < 0.01) were risk factors for disease.Conclusion Age,widowed,solitude are common risk factors of Alzheimer's disease and aggravating factors,should be combined with risk factors for chronic disease management actively take measures to prevent and delay the occurrence,the development of Alzheimer's disease.
9.Usefulness of different-b-value diffusion-weighted imaging in early cerebral infarction

Objective To investigate the usefulness of different-b-value diffusion weighted imaging (DWI) in patients with early cerebral infarction and obtain the optimal b value of early cerebral infarction.Methods DWI at b-value of 1,000,2,000,and 3,000 s/mm2 was performed for 40 patients within 72 h after the onset of stroke using a GE Signa HDx 3.0T MRI scanner.Post-processing was done by the DWI specific software Function Tool to gain signal intensity and mean apparent diffusion coefficient in the lesions center and the contralateral normal brain tissue,respectively.The sensitivity of conventional MRI and different-b-value DWI was calculated in the diagnosis of early cerebral infarction.Results In 40 patients with early cerebral infarction,the sensitivity of b-values of 1,000,2,000,and 3,000 s/mm2 (DWIb=1 000,DWIb=2 000,DWIb=3 000) and conventional MRI in the diagnosis of early cerebral infarction were 100%,97.5%,97.5%,72.5%,respectively.With b value increased,signal intensity and ADC value declined.Under the condition of different b values,signal intensity and ADC value were statistically significant in region of interest (P<0.05).Conclusion DWI is superior to conventional MRI scan in monitoring early cerebral infarction.With the increase of b value,the sensitivity is the higher to hyperacute cerebral infarction,the signal contrast is obvious,while signal to noise ratio of the image reduces.It is suggested that brain tissue contrast and the sensitivity to the new cerebral infarction is more predictable on b value =1,000 DWI than on the other DWIs.
10.Analysis on the pathogenic bacteria and their drug susceptibility in 552 cases of pregnancy combined with urinary tract infection in Qingdao

Objective To investigate the distribution and drug resistance of pathogenic bacteria in patients with preg-nancy combined with urinary tract infection. Methods Mid-stream urine culture and drug susceptibility analysis were conducted on 552 patients with pregnancy combined with urinary tract infection. Results The top 3 pathogenic bacteria for pregnancy combined with urinary tract infection were escherichia coli, enterococcus faecium and acinetobacter bau-mannii. Escherichia coli remained low resistance rate to piperacillin-tazobactam (1.99%), imipenem (1.49%), amikacin (4.47%), cefperazone-sulbactam (2.98%) and macrodantin (3.98%), but high resistance rate to ciprofloxacin (73.13%) and levofloxacin (67.16%). Conclusion The spectrum and drug resistance rate of pathogenic bacteria in pregnancy combined with urinary tract infection are changing. The pathogenic bacteria in urine and their drug resistance should be closely monitored to provide reference for clinical drug use.
