Objective To investigate the effects of hyperuricemia on vascular endothelial function. Methods With high-resolution ultrasound,flow-and nitroglycerin induced dilation, the vasodilation of brachial artery was measured in 30 hyperuricemia male patients and 30 healthy male subjects(control group). Both serum NO and plasma endothelin-1(ET-1) levels were determined at the same time. Results In patients with hyperuricemia,flow-induced vasodilation was much reduced compared with that in the control subjects(P0.05).Plasma NO level in the hyperuricemia group was lower than that in the control group (P

Objective To clone the human Islet neogenesis associated protein(rhINGAP)gene, express the gene extraeellulary in Pichia yeast for.further study on biological function and animal test on INGAP.Methods INGAP gene Was amplified with PCR and inserted into the recombinant plasmidα/pUC18.Then,the fusion gene of α and INGAP was digested and inserted into the expression plasmid pPIC9K.The positive recombinant plasmid which integrated INGAP Was confirmed by restriction enzyme digestion and sequencing, and it Was linearized with Sal Ⅰ digestion and transfered into the yeast host strain GS115 through electroporation. The yeast transformants that harbor the desired gene INGAP with high copy were selected by the auxotroph mediam G418,and verified by PCR.The condition of hake-flask culture was optimized, and the recombinant human INGAP Was induced expression with methanol as the only Carbone source. The antigen activity of the desired protein Was detected by Western blotting and ELISA method. Results Recombinant plasmid αINGAP/pPIC9K were successfully constructed and three positive Pichia yeast transformants were obtained. The expressed protein had satisfactory antigen activity, which Was confirmed by the Western blotting and ELISA method. Conclusions Pichia yeast expressing human Islet neogenesis associated protein (rhINGAP)gene was successfully constructed.

This study was aimed to observe the clinical effect of Pingai Mixture intervention on HIV/AIDS patients in order to clarify the characteristics of Pingai Mixture for AIDS treatment. A total of 41 HIV/AIDS patients who met the inclusion criteria were selected. Pingai Mixture was applied in the treatment. Three months was one treatment course. The treatment was given for two courses. Clinical symptoms, physical signs and changes in CD4+ lymphocytes of HIV/AIDS patients before and after treatment were observed. The SPSS software was used for statistical analysis. The results showed that the posttreatment total score (primary, secondary symptoms and the main physical sign cred-its) was significantly reduced compared with pretreatment (P < 0.01). The posttreatment spontaneous perspiration, night sweats, hair loss and other symptoms of a separate score was significantly reduced compared with pretreatment (P< 0.05). There were no significant changes on other symptoms or physical sign indicators. There was no significant difference in the Karnovsky score after 6-month treatment compared with pretreatment. It was concluded that Pingai Mixture is effective in AIDS treatment. It can improve the clinical symptoms and the administration is safe.

Objective To study the association between white matter hyperintensities(WMH)and cognitive function in silent lacunar cerebral infarction patients and its risk factors. Methods One hundred and seventy-four silent lacunar cerebral infarction patients were included in this study. They underwent MRI to show the site and severity of their WMH,including degrees 0-3of periventricular WMH in 21,102,39,12patients respectively, and deep WMH in 19,53,86,16patients respectively. Then, their cognitive function scores were compared. Results The degree of WMH was significantly higher and the age was significantly older in silent lacunar cerebral infarction patients(P<0.05).The situation memory, working memory and Simplified Intelligent State Scale score were significantly different in patients with different degrees of WMH in peripheral brain(P<0.05,P<0.01).The working memory was significantly different in patients with different degrees of WMH in deep brain(P<0.05).Degrees 1and 2of periventricular WMH were the independent risk factors for the impaired working memory(β=-0.264,P=0.038;β=-0.325,P=0.011)while degree 3of periventricular WMH was the independent risk factor for the impaired cognitive function(β=-0.273,P=0.014).Conclusion The memory is easy to be impaired in silent lacunar cerebral infarction patients with WMH of aging brain.Periventricular WMH is the major risk factor for cognitive impairment.

OBJECTIVE:
To investigate the anti-inflammatory effect and mechanisms of interleukin-35 (IL-35) in inflammatory bowel disease.
METHODS:
BALB/c mice were divided into three groups with 10 mice in each group:control group, model group (oral administration of 4% glucan sodium sulfate for 7 d) and IL-35-treated group (oral administration of 4% glucan sodium sulfate for 7 d, intraperitoneal injection of 2 μg IL-35 at d2-5). Disease activity index (DAI) was scored every day. After 7 d, the mice were sacrificed, and the serum and intestinal tissue samples were collected. The gross morphology of the colon was observed; HE staining was used to observe the pathological changes of colon tissue; flow cytometry was employed to detect the change of macrophage polarization ratio in colon tissue; the mRNA expression levels of cytokines IL-6, TNF-α, IFN-γ, IL-10 and SHIP1 in colon tissue were determined by real-time quantitative RT-PCR; the expression and distribution of SHIP1 in colon tissue was measured by immunohistochemistry; Western blotting was adopted to detect the expression level of SHIP1 protein in colonic intestinal tissues of each group.
RESULTS:
The DAI scores of the mice in the model group were higher than those in the control group, while the DAI scores in the IL-35-treated group were lower than those in the model group (all <0.01). Compared with the control group, the colon length was significantly shortened in the model group (<0.05), while the colon length of the IL-35-treated group had an increasing trend compared with the model group, but the difference was not statistically significant (>0.05). Compared with the model group, microscopic inflammatory infiltration score was decreased and microscopic crypt destruction and score was significantly lower in IL-35-treated group (all <0.05). The relative expression of proinflammatory cytokines IL-6, TNF-α and IFN-γ in the colon tissue of IL-35-treated group was decreased compared with the model group, while the relative expression of IL-10 mRNA was higher than that of the model group (all <0.05). Compared with the control group, the proportion of M1 macrophages in the model group increased (<0.05), while the proportion of M1 macrophages in the IL-35-treated group was lower than that in the model group (<0.05). The relative expression of SHIP1 mRNA and protein in the colon tissue of IL-35-treated group was higher than that in the model group (all <0.05).
CONCLUSIONS
IL-35 can inhibit the polarization of M1 macrophages and regulate inflammatory cytokines to promote anti-inflammatory effect on mice with colitis.
