1.The inhibitory effects of dexamethasone on cisplatin induced apoptosis of human lung adenocarcinoma cell SPC-A1 and its molecular mechanism.

The aim of this study is to investigate the apoptotic inhibition and its molecular mechanism of dexamethasone (DEX) acting on cisplatin (CDDP)-induced apoptosis of human lung adenocarcinoma cell SPC-A1; SPC-A1 cells were pre-cultured in vitro for 24 hours with DEX in different concentrations and then CDDP was added in different concentrations for culturing for further 48 hours. The survival rates of the cells were determined by MTT. The expression of serum/glucocorticoid-induced kinase (SGK-1) and mitogen-activated protein kinase phosphatase-1 (MKP-1) in SPC-A1 cells after being cultured by 1 micromol/l DEX at different time was detected by semi-quantitative RT-PCR technology. The expression of glucocorticoid receptor (GR) in SPC-A1 cells was measured by immunohistochemistry (IHC) with biotin-labeled anti-GR. The results of MTT showed that SPC-A1 cells had resistance to CDDP-induced apoptosis with pre-cultured DEX and the resistance intensity presented DEX concentration-dependent. The expressing quantity of SGK-1 in SPC-A1 cells stimulated by DEX could be elevated and increased with intention of time, but the express of MKP-1 was not detected. Up-regulated expression of GR in SPC-A1 cells stimulated by DEX was detected by IHC. The number of cells expressing GR in SPC-A1 cells was significantly higher than that in the control group. The results showed that DEX inhibited apoptosis of SPC-A1 cells induced by CDDP. The possible molecular mechanism is that elevated expression of GR induced by DEX up-regulates the expression of SGK-1 which locates at the downstream of anti-apoptosis pathway. The apoptosis resistance of SPC-A1 cells may account for all above the factors.

2.Periopertive nursing of five children with floating thumb treated with composite tissue flap grafting

Objective To summarize experience of periopertively nursing children with floating thumb treated with composite tissue flap transplantation. Method Health education was done to 5 children with floating thumb treated with composite tissue flap grafting as well as their families including preparing preoperational checks, preventing crisis of blood circulation, medication and pain nursing and paying attention to rehabilitation instruction. Result All thumbs survived successfully and one patient developed with crisis of blood circulation, which disappeared in 15 minutes. Conclusion Good preoperative nursing and effective health instruction are critical for nursing of the children with floating thumb treated with composite tissue flap transplantation.
3.Effect of Na~+/dicarboxylate transporter protein on cell cycle and cyclin P16 in human fibroblasts

BACKGROUND: Na+/dicarboxylate transporter protein (NaDC3) participates in the metabolism of energy, which may has effect on cellular senescence; however, the mechanism is not clear. OBJECTIVE: To investigate the role of human NaDC3 in cellular senescence by means of cell cycle and P16 measurements. DESIGN, TIME AND SETTING: The single sample observation was performed in the Nanjing Drum Tower Hospital from December 2007 to May 2008. MATERIALS: The WI-38 cell was purchased from ATCC Company, the retrovirus vector pLNCX2, sense vector PLNCX2-hNaDC3 and antisense vector PLNCX2-AshNaDC3 was provided by Clontech Company. METHODS: hNaDC3 was introduced into WI-38 cells with retroviral virus-mediated transfection. The integration and expression of exotic gene were confirmed. Whereafter, to examine the effects of hNaDC3 on cellular senescence of WI-38 cells by cell cycle and expression of P16. MAIN OUTCOME MEASURES: The integration and expression of hNaDC3, the cell morphology and generation of WI-38, the cell cycle and expression of P16 were observed. RESULTS: The results of Northern blot and Western blot revealed that hNaDC3 were expressed in WI-38/ hNaDC3 cells at gene and protein level, while, in WI-38/As hNaDC3 cells, there were integration of hNaDC3 mRNA with not protein expression. Compared with the control cells, WI-38 cells transfect with hNaDC3 cDNA showed cell generation decreased by eight to twelve passages, which in a low cell growth rate with cell cycle arrested at G1 phase, and the expression of P16 was elevated. CONCLUSION: Human NaDC3 gene may contribute to the process of cellular senescence of human diploid fibroblasts by increasing expression of P16.

6.Interpersonal Attribution Style of Female Students of Nursing School and Medical School

8.Biological activity of components, protein concentration of althernaria alternate lysates by different methods

Objective To find the best methods to make the extraction of alternaria alternate. Methods Alternaria alternate was harvested from air and identified, cultured at 26 ℃ for four weeks. Protein was extracted by different methods and its concentration was determined using the Bradford assay, its bioactivity was tested by RAST inhibition, and residual proteins were analyzed by SDS-PAGE.Results The extraction methods like grinder(a), grinder plus ultrasonic(b), ground with liquid nitrogen(c) and liquid nitrogen plus ultrasonic(d) were applied in the extraction procedure. The concentrations of protein extractions were as follows (g/mL): a was 0.44?0.04;b was 0.75?0.03;c was 0.72?0.03 and d; 1.29?0.05. From SDS-PAGE, we found that the most component in the extraction was that obtained by liquid nitrogen plus ultrasonic. From RAST, 50% inhibition was as follows(g/mL): a was 8.5, b was 9.4, c was 7.0,d was 3.7. Conclusion Ground with liquid nitrogen plus disintegration by ultrasonic was the most effective method of extraction.
9.Using the first page data of medical record to analyze medical service quality of different disease entities

Objective:To search for a method for analyzing medical service quality of different disease entities using the first page data of medical records.Methods: Based on documents review and real data analysis,the basic principles,methods and steps of disease entity dividing and selecting,evaluation index selecting,and calculating were introduced.Special attention was paid to certain problems in the discussion.Results: When the inpatients of selected disease entities covered 60% of total hospitalized patients,the medical quality information included exceeded 80% of the total information.For 97.66% disease entities,their inpatient cost and length of hospital stay were logarithmic-normally distributed.Conclusion: Dividing disease entities by fixed digits of ICD-9 code is easily performable.This method should be used when the disease entities include about 60% of total inpatients,and geometric mean should be used in calculating average length of hospital stay and average cost.
10.Clinical application of transdermal fentanyl for 30 patients with moderate or severe cancer pain

Purpose:To study analgesia effect ,adverse reactions and life quality changes of transdermal fentanyl in patients with cancer pain.Methods:30 patients with moderate or severe cancer pain were treated by transdermal fentanyl .The pain intensity, quality of life and adverse reactions were observed before and after the treatment.Results:After using transdermal fentanyl ,all the patients obtained moderate to good remission.The excellent,marked and moderate rates were 43.33%,53.33% and 3.33% respectively. The adverse reactions included nausea, vomiting,constipation,dizziness and lethargy.The rate of the adverse reactions was less.The quality of life improved markedly. Conclusions:Transdermal fentanyl is a safe,effective and simple way to relieve cancer pain in patients with moderate or severe pain.It can improve the quality of life markedly.Its adverse reactions are mild.
