1.The inhibitory effects of dexamethasone on cisplatin induced apoptosis of human lung adenocarcinoma cell SPC-A1 and its molecular mechanism.

The aim of this study is to investigate the apoptotic inhibition and its molecular mechanism of dexamethasone (DEX) acting on cisplatin (CDDP)-induced apoptosis of human lung adenocarcinoma cell SPC-A1; SPC-A1 cells were pre-cultured in vitro for 24 hours with DEX in different concentrations and then CDDP was added in different concentrations for culturing for further 48 hours. The survival rates of the cells were determined by MTT. The expression of serum/glucocorticoid-induced kinase (SGK-1) and mitogen-activated protein kinase phosphatase-1 (MKP-1) in SPC-A1 cells after being cultured by 1 micromol/l DEX at different time was detected by semi-quantitative RT-PCR technology. The expression of glucocorticoid receptor (GR) in SPC-A1 cells was measured by immunohistochemistry (IHC) with biotin-labeled anti-GR. The results of MTT showed that SPC-A1 cells had resistance to CDDP-induced apoptosis with pre-cultured DEX and the resistance intensity presented DEX concentration-dependent. The expressing quantity of SGK-1 in SPC-A1 cells stimulated by DEX could be elevated and increased with intention of time, but the express of MKP-1 was not detected. Up-regulated expression of GR in SPC-A1 cells stimulated by DEX was detected by IHC. The number of cells expressing GR in SPC-A1 cells was significantly higher than that in the control group. The results showed that DEX inhibited apoptosis of SPC-A1 cells induced by CDDP. The possible molecular mechanism is that elevated expression of GR induced by DEX up-regulates the expression of SGK-1 which locates at the downstream of anti-apoptosis pathway. The apoptosis resistance of SPC-A1 cells may account for all above the factors.

2.Effects of surface sealing on the shear bond strength of the brackets

Objective: To investigate the effects of surface sealing on the shear bond strength(SBS) of the brackets. Methods: Twenty extracted premolars for orthodontics were cut into the buccofacial teeth pieces and etched by Transbond~TM Plus self-etching primers for 30 seconds firstly, then divided into two groups with 10 teeth pieces in each group randomly. ① experiment group, the buccal surface of the enamel was sealed with Filtek Flow 1400A3 flowable composite resin before bracket bonding;②control group, the bracket was bonded to the buccal surface of the enamel directly. All specimens were stored in deionized water at 37 ℃ for 48 h, thermocycled for 1 000 times at 30-second intervals between 5 ℃ and 55 ℃ water baths, and debonded with a universal testing machine. The amounts of resin remaining on the teeth pieces after debonding by using the resin remnant index (RRI), which were examined under 10×magnification stereo-microscope. Results: The SBS(MPa) between experiment group and control group was 8.08±1.38, 8.34±1.37 respectively, whereas the RRI was 2.80±1.03, 3.20±1.03 respectively. The SBS, RRI between experimental group and control group revealed no statistically significant differences. Conclusion: Surface sealing shows adequate bond strength, which is considered a clinically acceptable bond strength. The debonding of bracket is not affected by the surface sealing.

4.Analysis of the Utilization of Fentanyl Transdermal Patch for Cancer Pain in Our Hospital

OBJECTIVE:To provide reference for rational use of Fentanyl transdermal patch in the clinic. METHODS:By ex-tracting data from hospital information system and digital medical record management system,medical records of 150 cancer pain inpatients receiving Fentanyl transdermal patch were collected from our hospital during Jun.-Dec. 2014,and then investigated and analyzed retrospectively. RESULTS:Numeric rating scale (NRS) score of 150 cancer pain patients receiving Fentanyl transdermal patch were as follows:pain scores of 17 patients (11.3%) were 1-3 points,those of 63 patients (42.0%) were 4-6 points and those of 5 patients(3.3%)were 7-10 points;the rest of the patients were not rated. Among those 150 cases,88 cases were well tol-erated with opioid and 62 patients were considered to be opioid intolerant. Those intolerant patients were who could not be treated orally or could not bore side effects of opioid hepatic or renal dysfunction. 25 cases used Fentanyl transdermal patch without defi-nite evidence. As to drug combination,22 cases additionally received Morphine hydrochloride injection;12 cases additionally re-ceived Bucinnazine hydrochloride injection;8 cases additionally received Morphine tablet;10 cases additionally received Oxycodo-ne hydrochloride sustained-release tablets and Morphine hydrochloride sustained-release tablets;8 cases additionally received Oxyco-done and acetaminophen tablets;3 cases additionally received Carbamazepine tablets. CONCLUSIONS:Fentanyl transdermal patch are widely used for cancer pain treatment in our hospital. There still is irrational use of it,such as absence of pain score before use,no medication indication,irrational drug combination. Physicians should obey medication indication strictly,and clinical phar-macists should strengthen communication with physicians and pay attention to medication education for patients so as to guarantee rational drug use.

Objective:To investigate the interpersonal attribution style of the baccalaureate nursing female students(BNFS)and to explore the difference of attribution style between the female students majoring in clinic medicine and nursing.Methods:A cross-sectional study was carried out by using the Interpersonal Relationship Subscale of Multidimensional-Multiattributional Causality Scale by Lefcourt et al. 500 college female students,from clinic medicine and nursing science of Central South University Xiangya School of Medicine, were recruited by using stratified random sampling.Results:The higher Grade, the higher Interpersonal Relationship Attribution Score (IRAS) of BNFS(44.7?7.8~50.8?9.5,45.9?8.0~51.2?7.9). BACKGROUND: Na+/dicarboxylate transporter protein (NaDC3) participates in the metabolism of energy, which may has effect on cellular senescence; however, the mechanism is not clear. OBJECTIVE: To investigate the role of human NaDC3 in cellular senescence by means of cell cycle and P16 measurements. DESIGN, TIME AND SETTING: The single sample observation was performed in the Nanjing Drum Tower Hospital from December 2007 to May 2008. MATERIALS: The WI-38 cell was purchased from ATCC Company, the retrovirus vector pLNCX2, sense vector PLNCX2-hNaDC3 and antisense vector PLNCX2-AshNaDC3 was provided by Clontech Company. METHODS: hNaDC3 was introduced into WI-38 cells with retroviral virus-mediated transfection. The integration and expression of exotic gene were confirmed. Whereafter, to examine the effects of hNaDC3 on cellular senescence of WI-38 cells by cell cycle and expression of P16. MAIN OUTCOME MEASURES: The integration and expression of hNaDC3, the cell morphology and generation of WI-38, the cell cycle and expression of P16 were observed. RESULTS: The results of Northern blot and Western blot revealed that hNaDC3 were expressed in WI-38/ hNaDC3 cells at gene and protein level, while, in WI-38/As hNaDC3 cells, there were integration of hNaDC3 mRNA with not protein expression. Compared with the control cells, WI-38 cells transfect with hNaDC3 cDNA showed cell generation decreased by eight to twelve passages, which in a low cell growth rate with cell cycle arrested at G1 phase, and the expression of P16 was elevated. CONCLUSION: Human NaDC3 gene may contribute to the process of cellular senescence of human diploid fibroblasts by increasing expression of P16.
7.Effects of Mindfulness Behavior Training on Mood States and Quality of Life in Old Patients post Fixation for Hip Fracture

Objective To investigate the effects of mindfulness behavior training on mood states and quality of life in old patients post fixation for hip fracture. Methods Patients were randomly divided into experiment group (n=30) and control group (n=30). The control group received routine rehabilitation, while the experiment group received mindfulness practice therapy in addition. They were evaluated with Profile of Mood States (POMS), Short Form of Health Survey (SF-36), and Mindful Attention Awareness Scale (MAAS) before and 5 weeks after treatment. Results There was no difference in the scores of POMS, MAAS and SF-36 before treatment (P>0.05). The scores of MAAS and SF-36, the subscores of tension-anxiety, depression-dejection, fatigue-inertia and vigor-activity of POMS improved more in the experiment group than in the control group (P<0.05). Conclusion Mindfulness behavior training can improve the mood state and quality of life of old patients post fixation for hip fracture.
8.Influence of salvianolic acid B on expressions of TGF-β1/Smad signaling pathway related proteins in human lung fibroblasts and its mechanism

Objective To investigate the influence of salvianolic acid B on the expressions of TGF-β1/Smad signaling pathway related proteins in human lung fibroblasts,and to explore the mechanism of the inhibitory effect of Sal B on TGF-β1-induced lung fibroblast activation.Methods The human embryonic lung fibroblasts (MRC-5) were cultured in vitro and randomly divided into control group ( the cells were cultured with DMEM without TGF-β1 or Sal B),Sal B group (the cells were cultured with 10μmol·L-1 Sal B),TGF-β1 group (the cells were cultured with 10μg·L-1 TGF-β1),and TGF-β1 (10μg·L-1 )+ Sal B (10μmol·L-1 )group.The protein levels of p-Smad2,p-Smad3,TβRⅠ,and Smad7 in the fibroblasts in various groups were detected by Western blotting method.Results Compared with control group,the expression levels of p-Smad2,p-Smad3,and TβRⅠproteins in TGF-β1 group were significantly increased (P<0.05),and the expression level of Smad7 protein was decreased (P<0.05).Compared with control group,the expression levels of p-Smad2,p-Smad3,TβRⅠ,and Smad7 proteins in lung fibroblasts in Sal B group had no significant change (P>0.05).Compared with TGF-β1 group,the expression levels of p-Smad2,p-Smad3,and TβRⅠ in TGF-β1+ Sal B group were descended (P<0.05),and the expression level of Smad7 was increased (P<0.05).Conclusion Sal B could suppress the TGF-β/Smad signaling pathway in lung fibroblasts and to inhibit the TGF-β1-induced lung fibroblast activation.
9.Effect of Ethyl Pyruvate on Expression of Caspase-3 in Hippocampus and Learning and Memory in Rats with Hypoxic Ischemic Brain Damage

Objective To investigate the effect of ethyl pyruvate (EP) on the expression of caspase-3 in the hippocampus and the learn-ing and memory ability in neonatal rats with hypoxic ischemic brain damage (HIBD). Methods 90 Wistar rats of 7 days old were randomly divided into sham operation group (n=15), HIBD group (n=15), EP1 group (n=15, 10 ml/kg), EP2 group (n=15, 30 mg/kg), EP3 group (n=15, 50 mg/kg) and EP4 group (n=15, 100 mg/kg). The model was established with Rice's method. 30 minutes before operation, and every 24 hours after operation, EP groups were injected with 10 ml/kg, 30 mg/kg, 50 mg/kg, 100 mg/kg EP in abdomen respectively, for 2 weeks. Af-ter treatment, the caspase-3 positive cells were observed by immunohistochemical staining, and the latency and the times crossing the target quadrant were tested by Morris water maze test. Results The caspase-3 positive cells were less in EP groups than in HIBD group (P<0.05), expect EP1 group (P>0.05), especially in EP3 group (P<0.01). The latency and the times crossing the target quadrant were better in EP groups than in HIBD group (P<0.05), expect EP1 group (P>0.05), especially in EP3 group (P<0.01). Conclusion Ethyl pyruvate can de-crease the expression of caspase-3 in hippocampus, and improve the ability of memory and learning in neonatal rats with HIBD.
10.Effect of autologous cytokine induced killer cell therapy in patients with lung cancer

Objective To evaluate the clinical effect of cytokine induced killer cellular immunotherapies ( CIK) on the patients with advanced lung cancer.Methods Peripheral blood mononuclear cell from 32 cases of patients with advanced lung cancer were isolated.Cells were cultured in DMEM supplemented with IFN-γ,IL-2,anti-CD3 monoclonal antibody and other cytokines,and transfused back into the patients in several times after 15 days of culture.Subsequently,we observed the size of tumor,the change of immunologic function by CIK treat-ment,the status of clinical symptoms,and the toxic and side effect.The clinical curative effect was also evaluated. Results The symptoms in 32 patients who received CIK therapy were reduced,and the side effects were light. Immunophenotype analysis showed that CD3 /CD4 coexpression were found in 34.4%patients,CD3 /CD8 co-expression in 68.2%,and CD3 /CD56 co-expression in 13.7% patients.CT images about 15 days after CIK cell transfusion showed that pleural effusion were decreased.Clinical evaluation effective rate was 56.0%,and disease control rate( CR +PR +SD) was 84.3%.Conclusion CIK cell immunotherapies of cancer patients can improve immunologic function of cancer patients and inhibit tumor growth and relieve the clinical symptom of patients,and improve the quality of life.
