2.Experimental Study of Anti-inflammation of Durian Peel Extract

Objective To investigate the anti-inflammatory effects and mechanism of durian peel extracts (DPE). Methods The in vivo anti-inflammation effects of DPE were examined by carrageenin-induced mice paw edema test and allergic contact dermatitis test induced by 2, 4-DNFB. And the in vitro anti-inflammation effects of DPE were evaluated with methyl thiazolyl tetrazolium ( MTT) assay in RAW 264.7 cell model of inflammation induced by lipopolysaccharide (LPS). Results The results of animal experiments showed that DPE groups could markedly relieve mice paw edema induced by carrageenin ( P<0.01 or P<0.001 compared with blank group). DPE could effectively inhibit the allergic contact dermatitis induced by 2, 4-DNFB in mice, showing good dose-effect relationship. The results of in-vitro test showed that DPE at the given concentrations had no influence on RAW 264.7 cell proliferation. Tumor necrosis factor alpha ( TNF-α) , interleukin 6 ( IL-6) , interleukin 1 beta (IL-1β), nitric oxide (NO) and nuclear factor kappaB (NF-кВ ) were observably inhibited, and anti-inflammatory cytokine IL-10 was enhanced by 25 and 50 mg/L of DPE. Conclusion DPE exert potential anti-inflammation effect, and the mechanism might be related to its inhibition of NF-кВsignal pathway.
3.MRI characteristics of enthesitis in peripheral ankylosing spondylitis

Objective To explore the MRI findings of enthesitis in peripheral ankylosing spondylitis (AS). Methods We retrospectively reviewed the MRI obtained from 13 patients with clinically-diagnosed peripheral AS. MRI finding attributable to enthesitis could be visualized as tendon or ligament enthesitis, bone marrow edema (BME), synovitis, bursitis, tenosynovitis. Results MRI findings of knee showed enthesitis in 8 knees, mainly involved in the insertions of the quadriceps tendon at the upper patellar pole , the patellar ligament at the lower patellar pole and the tibial tubercle, BME in 8 knees, synovitis in 5 knees, bursitis in 3 knees, and tenosynovitis in 4 knees. MRI findings of ankle showed enthesitis in 3 ankles, involved in plantar insertion into calcaneus and Achilles tendon insertion into the calcaneus, BME in 1 ankle, and tenosynovitis in 3 ankles. Conclusion The lower extremities are more often involved in peripheral AS than the upper extremities. MRI characteristic of enthesitis may contribute to the diagnosis of peripheral AS early combined with HLA-B27.

5.Effect of Chinese Daoyingong method on knee osteoarthritis

ObjectiveTo explore the effect of Chinese Daoyingong method on knee osteoarthritis.Methods Eighty patients with knee osteoarthritis were randomly divided into the experiment group and control group in equal number.The control group was given the routine medicine combined with Chinese fumigation and the experiment group was treated with Chinese Daoyin method beside the treatment in the control group.The two groups were compared in terms of the score on visual analog scale of pain and the score on knee function before and after treatment.Results The visual analog scale of pain in the experiment group was significantly lower than that in the control group after intervention and the score on knee function was significantly higher than that of control group (P<0.001). Conclusion The intervention with Chinese Daoyingong method plus routine medication and Chinese herbal fumigation is effective in the treatment of knee osteoarthritis.
6.Effect of ribosomal protein S3 Ser209 on NF-κB transcriptional activity and its DNA binding capacity

Objective To construct expression vectors of human ribosomal protein S 3(RPS3) and RPS3Ser209 mutant in orcler to investigate the effect of RPS3Ser209 mutant on NF-κB signaling pathway and DNA binding capacity .Methods The vector RPS3-myc was amplified by polymerase chain reaction ( PCR) from the human liver cDNA and subcloned into pcDNA-3.1myc-HisB.RPS3S209A represented mutant RPS3 expression vectors, in which the designated amino acid was mutated to an alanine residue .Dual luciferase reporter gene assay was used to detect the NF-κB transcription activity in HEK293 cells,immunofluorescence to detect RPS3 location, and EMSA to examine NF-κB DNA-binding activity.Results The expression vectors of RPS3-myc and RPS3S209A-myc were constructed.Compared with wild-type RPS3,the nucleus translocation, transactivation activity of NF-κB and DNA binding ability of RPS3S290A were reduced significantly .Conclu-sion The impact of RPS3 on NF-κB signaling pathway depend on its serine 209.
7.Comparison of MRI and arthroscopy in classification of mediopatellar plica and grading of cartilage injury in knee

Objective To explore the MRI features in mediopatellar plica and cartilage injury.Methods The MRI appearances of 30 patients with mediopatellar plica were divided into four types and chondral injury were divided into five grades,then the characteristics of MRI imaging were analyzed,and compared with arthroscopy classification.Results The MRI manifestations of A type (n =7) of mediopatellar plica showed a cordlike elevation on the synovial wall;B type (n =13) showed shelf-like appearance but did not cover the anterior surface of the medial femoral condyle;C type (n =8) showed shelf-like appearance and covered the anterior surface of the medial femoral condyle and D type (n =2) showed a central defect in the mediopatellar plica.The MRI manifestations of chondral injury grades included zero grade (n =10),Ⅰ grade (n =2),Ⅱ grade (n =7),Ⅲ grade (n =7) and Ⅳ grade (n=4).There was no statistical significance in terms of classification of mediopatellar plica and grading of cartilage injury via MRI and arthroscopy.Conclusion MRI provides solid evidence for preoperative assessment and choice of operation scheme for classification of mediopatellar plica and grading of cartilage injury in knee.
8.Effect of Consciousness-restoring Needling Combined with Comprehensive Rehabilitation Training on Motor Function and Activities of Daily Living of Poststroke Patients with Hemiplegia

Objective To observe the therapeutic effect of consciousness-restoring needling combined with comprehensive rehabilitation training on motor function and the activities of daily living of poststroke patients with hemiplegia. Methods Sixty qualified patients were evenly randomized into observation group and control group. All of the patients were given conventional medicine treatment and conventional rehabilitation training, and the treatment group was given consciousness-restoring needling additionally. The therapeutic effects were compared at the end of first session of treatment for 4 weeks and 12 weeks after the first session of treatment. The Fugl-Meyer motor assessment scale (FMA), modified Barthel Index (MBI) and Stroke-Specific Quality of Life Scale ( SS-QQL) were taken as the main evaluation indexes. Results The differences of FMA, MBI and SS-QOL scores were insignificant between the two groups before treatment ( P>0.05). At the end of treatment for 4 weeks, FMA, MBI and SS-QOL scores were increased in the two groups (P<0.05), and the increase was more obvious in the observation group (P<0.05). The results of follow-up till the 12th week showed that FMA, MBI and SS-QOL scores were still higher than the baseline level ( P<0.05). Conclusion Consciousness-restoring needling combined with comprehensive rehabilitation training has better effect on improving motor function and the activities of daily living of poststroke hemiplegia patients than comprehensive rehabilitation training alone.
9.Preparation of Tilmicosin Imprinted Polymers and Its Application to Solid Phase Extraction

Molecularly imprinted polymers (MIPs) with high selectivity to tilmicosin (TIM) were prepared using tylosin(TYL) as dummy template, methacrylic acid(MAA) as monomer, ethylene glycol dimethacrylate (EGDMA) as cross-linker.The effects of 4 porogens including dimethyl formamide, methanol, acetone, and chloroform on the recognition capability of MIPs were investigated.Orthogonal test was used to optimize the preparation of MIPs, and the optimal composition was as follows; 1.0 mmol TYL, 8.0 mmol MAA, 20.0 mmol EGDMA, 6.0 mL chloroform, 20.0 mg azobisisobutyronitrile.The solid phase extraction condi tions and characteristics of MIPs as adsorptive material for the selective extraction and enrichment of TIM were also studied.The recovery of TIM was above 90% when the following procedure was applied to MIPs cartridge: conditioning with methanol and water(pH 9.0), loading with acetonitrile, cleaning with methanol and chloroform respectively, and eluting with 3 mL methanol-ammonia(95:5, V/V).The recovery of TIM on non-imprinted polymers cartridge was only 32%.
10.Expression of connective tissue growth factor induced by parathyroid hormone via MAPK signaling pathway in human renal proximal tubular cells

Objective To evaluate the effect of parathyroid hormone (PTH) on the expression of connective tissue growth factor (CTGF) in human renal tubular epithelial cells, and to explore the role of MAPK signaling pathway. Methods Real time RT-PCR, Western blot, and reporter gene assay were employed to detect PTH-induced CTGF expression in HK-2 cells. Inhibitors (PD98059 and U0126) of MAPK signaling pathway were used to confirm involved signal pathway. Results HK-2 cells had basic expression level of CTGF mRNA and protein, which were increased significantly after treatment with PTH. The luciferase activity was up-regulated to a higher level as compared with control group after treatment with 10-10 mol/L PTH for 12 h [(1.8884±0.0780) vs (0.9891±0.0300) A, P<0.01]. Moreover, a small amount of p-ERK1/2 was detected in normal HK-2 cells, but it was increased significantly in response to PTH activation, most remarkably when treated with 10-10 mol/L PTH for 30 min. Inhibitors of MAPK signaling pathway, PD98059 and U0126, noticeably inhibited the expression of CTGF mRNA and protein as well as gene promoters in HK-2 cells. Conclusion PTH can induce higher expression of CTGF in HK-2 cells probably via MAPK signaling pathway.
