1.Regulation of expression of cGMP-dependent protein kinase I_? by calcineurin in vascular smooth mucle cells proliferation

Objective: To dicuss the regulation of expression of cGMP-dependent protein kinase I_? (PKG I_?) by calcineurin (CaN) in vascular smooth musle cells (VSMCs) proliferation. Methods: Cultured wistar rat aortic VSMCs were used as an experimental model. CaN was inhibited by its special inhibitor cyclosporin A (CsA). Phenylephrine (PE) was given to stimulate VSMCs to proliferate. All of cultured cells were divided into four groups:control group, 0.5 mg/L CsA group, 5 mg/L CsA group and 5 mg/L CsA+10 ?mol/L PE group. The mRNA and protein expressions were assayed by RT-PCR, immunocytochemistry and Western blot analysis. Results: The OD ratio of PKG I_? mRNA expression in 0.5 mg/L CsA group resembled that in the control group while that in 5 mg/L CsA group was significantly higher than that in the control group (P

Objective To study how to use the machine of B.BRAUN's Diapact continuous renal replacement therapy (CRRT) to achieve multiple models of plasma treatment by analyzing the characteristics of the machine.Methods Based on the machine of B.BRAUN's Diapact CRRT,assisted by extra single pump,the pipeline connection of the machine was reformed and the appropriate parameter was reset.Results After the reconstruction,the new equipment could succeed in achieving special plasma treatment such as double filtration plasmpheresis (DFPP)and plasma adsorption (PA).Conclusions The reconstructed equipment can attain special plasma treatment.Compared with plasma exchange (PE),the equipment has some characteristics such as few syndromes and without external plasma.According to its safety and stability,this new method is suitable for clinical application.
4.Cultivation and drug sensitive test of bile bacteria in patients with bile duct diseases during endoscopic retrograde cholangiopancreatography

Objective To observe the bacterioflora of bile and it' s drugs sensitivity in patients with bile duct diseases to serve as a guidance in medication. Methods Bile of 156 cases of patients with biliary disease was collected and cultured respectively during endoscopic retrograde cholangiopancreatography (ER-CP) by deep cannulation and sucking out bile through the catheter. Forty eight blood samples among them were cultured simultaneously. Ten kinds of drug papers were used to investigate the bacterial sensitivity. The data were analysed statistically- Results Bacteria positive rate of these 156 bile samples was 62. 8% , including Gram - negative bacteria (81.6% ) and Gram - positive bacteria (18. 4% ). These bacteria consist of Pseudomonas aeruginosa (35. 1% ) , Escherichia coli(22. 8% ) , Streptococcus faecalis (16.7%) ,Entero bacilliaerogenes(7. 9% ) ,Klebsiella(7. 0% ) , Citrobacter(6. 1% ) ,Acinetobacler lwqffi(2. 6% ) and Staphylo-coccus aureus( 1. 7% ). The bacteria positive rate was only 4. 2% in the 48 blood samples. Drug sensitive rates of bacteria against 10 kinds of drugs were changed in recent years. The result indicated that the sensitive rates of bacteria were significantly higher in ciprofloxacin, fortum, cefoperazone, sulbactam + cefoperazone and cilastatin than those in ampicillin,azlocillin, cefazolin sodium, eefuroxime(P
5.Preparation of biomimetic multilayered scaffolds using collagen/chitosan for cartilage tissue engineering

Objective Prepare biomimetic muitilayered scaffold which has similar structure of natural cartilage.Method By lyophilizing the scaffolds which were prefrozen at-20℃ and in liquid nitrogen successively,we prepared double-layered spongy scaffolds.By partially thawing the prefrozen samples and refreezing them in liquid nitrogen before the final liyophilization,we prepared biomimetic multilayered scaffolds with about 2mm thickness.XRD and FT-IR were used to confirm the interaction between collagen and chitosan.SEM was used to observe the morphologies of the scaffolds.The mechanical properties of pure chitosan scaffolds,pure collagen scaffolds,composite single-layered scaffolds and biomimetic multilayered scaffolds were compared both in dry and wet conditions.Results There was chemical interaction between collagen and chitosan.Composite materials will form better pore structure.The biomimetic multilayered scaffolds have upright pores,round pores and a dense layer from bottom to top of the scaffolds.The scaffolds have quite different mechanical properties between dry and wet state.Under wet state,the different layers of the biomimetic muitilayered scaffold have different mechanical properties.Results The biomimetic structure of the multilayered scaffold is very close to that of the natural articular cartilage,and the different layers of the biomimetic muitilayered scaffold had different mechanical properties under wet state.These are hopefully beneficial to help maintain the phenotypes of chondrocytes and promote the repairing effect of cartilage defects.
6.Effect of aminophylline and pentoxyverine on serum CRP, MCP-1 and immune factors in elderly patients with sleep apnea syndrome

Objective To investigate the effect of aminophylline and pentoxyverine on serum CRP, monocyte chemotaxis protein 1(MCP-1) and serum immune factors in elderly patients with sleep apnea syndrome .Methods 112 elderly patients with apnea syndrome were divided into the control group and the experiment group by order of medical treatment and 56 cases in each group.The control group were treated by rosiglitazone hydrochloride tablets and bay three ramie snore capsule and the experiment group were treated on the control group with Aminophylline Tablets and Pentoxyverine Citrate Tablets.Follow up 3 months, the CRP, MCP-1,IFN-γ,IL-4,IL-8 and IL-10 were tested and compared.Results Compared with the control group after treatment, the CRP,MCP-1,IFN-γand IL-8 level were lower(P <0.05),the IL-4 and IL-10 level were higher(P <0.05).Conclusion Aminophylline and pentoxyverine can be effective in treatment of elderly patients with apnea syndrome and its complications , suggesting that the mechanism may be related to reduce the patients serum CRP, MCP-1, IL-8 and IFNγ-levels, is related to the increasing of the levels of IL-4, IL-10.
7.Cloning of nm23-H1 cDNA gene and construction of recombinant adenovirus vector

Objective:To clone human nm23-H1 c DNA gene and to recombine it into adenovirus vector.Methods:Nm23 -H1cDNA fragment was amplified from human liver by reverse transcription-polym erase chain reaction(RT-PCR) and cloned into pMD18-T plasmid. The gene was exa mined by sequencing. Then pAd-Shuttle-CMV vector containing nm23-H1 was const ructed. Results:The results showed that the gene fragment cloned in pMD18-T was coincident with the sequence of nm23-H1.A pAd-Shuttle -CMV v ector containing nm23-H1 gene with correct order was con firmed by Kpnl digestion. Conclusion:nm23-H1 may be cloned into recombined adenovirus vector pAd-Shuttle-CMV.

9.Environmental Mercury Contamination Monitoring by Using Freshwater Fishes

Objective The present paper makes attempt to study the enrichment features of mercury in freshwater fishes and to evaluate the mercury contamination in environment with the fishes.Methods The fish samples of silver carp,common carp,johnny carp,shorthead catfish and catfish were collected from Songhua River and Heilongjiang River.The content of mercury in freshwater fishes,water and sediment was determined,the biological characteristic of mercury contamination in aquatic animal and the applicability of mercury contamination in aquatic amimal and its estimation were analyzed in Sep.,2007.Results The levels of mercury in river water and sediment were 0.053 ?g/L and 0.27 mg/kg for samples collected from Jiamusi section of Songhua River,and 0.034 ?g/L and 0.16 mg/kg for samples collected from Fuyuan section of Heilongjiang River.The concentrations of total mercury in the muscles of Songhua River’s silver carp,common carp,johnny carp,shorthead catfish and catfish was(0.070 ? 0.036),(0.120?0.071),(0.113?0.067),(0.152?0.107) and(0.196?0.095) mg/kg respectively.The concentrations of total mercury in the muscles of Heilongjiang River ’s silver carp,common carp,johnny carp,shorthead catfish and catfish was(0.042 ? 0.034),(0.103?0.077),(0.095?0.072),(0.157?0.118),(0.174?0.101) mg/kg respectively.The mercury content in fish in the same water environment was significantly positive correlated with its weight(P
10.Investigation of Lepisma Breeding in Libraries of Universities

Objective To investigate the species and ecosystem of lepisma breeding in libraries of universities.Methods The lepisma samples were collected from the libraries of universities in Anhui by trapping bottle and chemical stimulating and the life cycle and the biocycle were observed meanwhile,from June 2005 to June 2007.Results 38 libraries were examined and 3 748 lepismas were gathered,belonging to 1 family 1 genus and 4 species.The first activity peak appeared in May,and the second in October.Lepismas were active from 20:00 to 5:00.The activity peak appeared at 24:00.This was identical with the living habits of their hiding by day and coming out at night.Conclusion The lepisma pollution in libraries of universities is serious in Anhui province and the Ctenolepisma villosa is the dominant species.
