Journal of Peking University(Health Sciences) 2003;0(06):-
Objective: To dicuss the regulation of expression of cGMP-dependent protein kinase I_? (PKG I_?) by calcineurin (CaN) in vascular smooth musle cells (VSMCs) proliferation. Methods: Cultured wistar rat aortic VSMCs were used as an experimental model. CaN was inhibited by its special inhibitor cyclosporin A (CsA). Phenylephrine (PE) was given to stimulate VSMCs to proliferate. All of cultured cells were divided into four groups:control group, 0.5 mg/L CsA group, 5 mg/L CsA group and 5 mg/L CsA+10 ?mol/L PE group. The mRNA and protein expressions were assayed by RT-PCR, immunocytochemistry and Western blot analysis. Results: The OD ratio of PKG I_? mRNA expression in 0.5 mg/L CsA group resembled that in the control group while that in 5 mg/L CsA group was significantly higher than that in the control group (P

Objective To study how to use the machine of B.BRAUN's Diapact continuous renal replacement therapy (CRRT) to achieve multiple models of plasma treatment by analyzing the characteristics of the machine. Methods Based on the machine of B.BRAUN's Diapact CRRT,assisted by extra single pump, the pipeline connection of the machine was reformed and the appropriate parameter was reset. Results After the reconstruction, the new equipment could succeed in achieving special plasma treatment such as double filtration plasmpheresis (DFPP)and plasma adsorption (PA).Conclusions The reconstructed equipment can attain special plasma treatment.Compared with plasma exchange (PE),the equipment has some characteristics such as few syndromes and without external plasma.According to its safety and stability, this new method is suitable for clinical application.

Objective To investigate the clinical outcomes of arteriovenous anastomosis by microvascular anastomotic coupler device. Methods From January 2013 to November 2013,23 cases of microvascular anastomotic coupler device were applied in 18 patients with vascular injury of upper limb, and anastomosis time and patency rate was observed. Results In 18 patients, the brachial artery injury in 2 cases, the radial artery injury in 11 cases, the ulnar artery injury in 8 cases, the cephalic vein injury in 1 case, and basilic vein injury in 1 case.23 microvascular anastomotic coupler devices were applied in vascular injury. The post procedure vascular patency rate was 100% and the average time of anastomosis was 3min 59s.Conclusion Microvascular anastomotic coupler device is charactered by higher patency rate and faster operation. It can not only be used in venous anastomosis of tissue graft, and also has a certain value in the repair of arteriovenous injury.

AIM: To investigate the effect of ginsenoside-Rg3 on the expression of vascular endothelial growth factor (VEGF) and tumor necrosis factor-α (TNF-α) in retina with diabetic rats and its roles in preventing neovascularization in diabetes. METHODS: Sixty male Wistar rats were divided into 3 groups randomly: negative control group, diabetic control group and ginsenoside-Rg3 treatment group (5mg/kg, 0.2mg/mL) followed by establishing diabetic model. The expression of VEGF and TNF-α were measured after 8 weeks. RESULTS: There were significant differences among negative control group, diabetic control group and ginsenoside-Rg3 treatment group in the expression of VEGF and TNF-α (F=129.363, 211.992; all the P<0.01). VEGF and TNF-α expression were significantly higher in diabetic control group and ginsenoside-Rg3 treatment group than that in negative control group (P<0.01), with a significant reduction in ginsenoside-Rg3 treatment group than that in diabetic control group (P<0.01). CONCLUSION: Ginsenoside-Rg3 can down-regulate the expression of VEGF and TNF-α in retina, which may interfere in the development of diabetic retinopathy.

Xanthine-Xanthine oxidase ( X-XOD) added to the culture medium of cultured cardiac cells in rats may damage cell membrane. Electrophysiological findings indicated that APA, MDP, OS,Vmax, were decreased and the SDF was increased. Astragalus polysaccharide (APS) could protect the cells from being damaged by X-XOD. APS recoverted all the de-creased cardiac functional parameters in free-radical-damaged rats by X-XOD. APS had anti-free-radical damage action on the cultured my-ocardiocytes and the myocardial contractility of the isolated rat working heart.

Objective To explore the clinical effect of the improved operation of endoscopic thoracic sympathectomy(ETS)for treatment of palmar hyperhidrosis. Methods The improved operation of ETS was used to 105 patients with palmar hyperhidrosis.T3 sympathetic was blocked in 105 cases with palmar hyperhidrosis. Results Hand sweating disappeared and axillary sweating discreased significantly in all patients. No one had any serious complications such as bleeding and Horner Syndrome except for five patients with pneumothoraces.Compensatory sweating was found in 15 patients following up from 1~12 months after operation. Conclusion The improved operation of ETS is a simple, safe, effective and less complications procedures.

Objective To establish a sensitive, reliable and simple method for determination of selenium(Se) in salt. Methods The salt samples were dissolved in water and added hydrochloric acid, thiourea, placed for 30 minutes under the room temperature, then used HG-AFS to determine Se. Results The linear range was 0.5-20 ?g/L, the detection limit was 0.06 ?g/L, RSD
